Lysostaphin is effective in treating methicillin-resistant Staphylococcus aureus endophthalmitis in the rabbit.

PURPOSE: To determine the effectiveness of lysostaphin treatment of experimental endophthalmitis caused by methicillin-resistant Staphylococcus aureus (MRSA). METHODS: In one experiment, rabbits were injected in the mid-vitreous with 50 or 200 CFU of S. aureus; untreated groups and groups injected intra-vitreally at 8 or 24 hours postinfection with vehicle or lysostaphin (0.1 mg/ml) were compared in terms of CFU/ml vitreous at 24 or 48 hours postinfection. Histopathology of untreated and treated eyes was also compared. To quantify the potency of lysostaphin, additional rabbits were injected with 50 CFU of S. aureus and untreated eyes and eyes treated at 8 hours with 0.001, 0.01 or 0.05 mg/ml were compared in terms of CFU/ml vitreous at 24 hours postinfection. RESULTS: Vitreous of untreated eyes or vehicle-treated eyes injected with 50 or 200 CFU of S. aureus contained 5-10 million CFU/ml at 24 or 48 hours postinfection. All eyes treated with lysostaphin at 8 hours postinfection had less than 1 log CFU/ml in the vitreous (P >or= 0.0001). Similarly, eyes treated with lysostaphin at 24 hours postinfection had approximately 1 log of CFU/ml at 48 hours postinfection. None of the untreated eyes were sterile and 88% or 50% of the eyes treated at 8 or 24 hours postinfection, respectively, were sterile. Eyes treated with lysostaphin at 8, but not 24, hours postinfection had less pronounced pathologic changes than the untreated eyes (P = 0.002). A significant reduction in the CFU/ml vitreous at 24 hours postinfection was obtained by treating infected eyes at 8 hours postinfection with lysostaphin at concentrations of >or=0.001 mg/ml (P <or= 0.0034). CONCLUSIONS: Lysostaphin is effective in treating experimental endophthalmitis mediated by MRSA.

Rhenium-188 HEDP to treat painful bone metastases.

PURPOSE: Rhenium-188 hydroxyethylidine diphosphonate (HEDP) is a new and attractive radiopharmaceutical that localizes in skeletal metastases and emits beta particles that may be therapeutically beneficial. In this study, the therapeutic efficacy of Re-188 HEDP was investigated in an uncontrolled initial trial of 61 patients with different types of advanced cancer for the palliation of painful bone metastases. MATERIALS AND METHODS: Sixty-one patients with painful bone metastases of lung, prostate, breast, renal, rhinopharyngeal, and bladder cancers were treated with 1.1 GBq (31 mCi) to 6.9 GBq (188 mCi) Re-188 HEDP. After treatment, the patients were followed at weekly intervals for the first 2 months and monthly thereafter for as long as 1 year. Hematologic function tests were also performed before and after treatment for 6 weeks. Pain responses were scored according to a three-point pain-rating scale as complete, significant, and minimal. RESULTS: Prompt and significant relief of bone pain occurred in 80% of patients overall. Of the specific tumor types, pain relief was achieved in 77% of patients with lung cancer, in 80% with prostate cancer, in 83% with breast cancer, in 100% with bladder cancer, in 50% with renal cancer, in 50% with rhinopharyngeal cancer, and in 87% of patients with other tumor types, with no severe side effects or hematopoietic toxicity. CONCLUSION: This large clinical trial verified that Re-188 HEDP is a useful radiopharmaceutical agent to treat painful bone metastases from various tumor types.

Gender- and age-related differences in corneal topography.

PURPOSE: To investigate gender- and age-related differences in the corneal topography of a normal population. METHODS: One hundred thirty-two topographic examinations were collected from 100 patients ranging in age from 23 to 83 years (average, 57.35+/-17.38 years). Data were segregated by gender and further divided into younger (less than 50 years) and older (50 years or more) age groups. The topographic indices of Surface Regularity Index, Surface Asymmetry Index, Irregular Astigmatism Index, Standard Deviation of Corneal Power, Corneal Eccentricity Index, Coefficient of Variation of Corneal Power, Simulated Keratometry 1 and 2, and Average Corneal Power were examined. The astigmatism pattern and corneal irregularity were determined and compared with respect to gender and age. RESULTS: The corneas of older men were flatter than those of older women (p < 0.001). The vertical corneal meridian, but not the horizontal meridian, showed statistically significant gender-related changes with aging (p < 0.001). Older men had a significantly higher potential for against-the-rule astigmatism than women (p < 0.001). Corneal irregularity (measured in terms of the Surface Regularity Index and Irregular Astigmatism Index) increased with age (p < 0.001 and p < 0.001, respectively), although there was no gender-related difference. In the younger group, no gender-related differences in corneal curvature or astigmatism pattern were found. CONCLUSION: Aging influences changes in patterns of astigmatism differently in men and women. Decreases in levels of sex hormones may play a role in gender-related changes in corneal structure with age.

Bystander effects of cancer cell lines transduced with the multisubstrate deoxyribonucleoside kinase of Drosophila melanogaster and synergistic enhancement by hydroxyurea.

The multisubstrate deoxyribonucleoside kinase of Drosophila melanogaster (Dm-dNK) can be expressed in human cells with retained enzymatic activity. The cells expressing Dm-dNK exhibit increased sensitivity to several cytotoxic nucleoside analogs. In this study, we further evaluated Dm-dNK as a potential novel suicide gene in combination with (E)-5-(2-bromovinyl)-2'-deoxyuridine (BVDU) as the prodrug. We used two human cancer cell lines transduced with a retrovirus encoding the Dm-dNK cDNA and investigated whether the cells expressing the enzyme can induce cell death of untransduced cells, a phenomenon known as the "bystander effect". A bystander effect was observed in a thymidine kinase-deficient human osteosarcoma cell line but not in the MIA PaCa-2 human pancreatic adenocarcinoma cell line. The cytotoxicity of BVDU increased in both cell lines when the compound was used in combination with subtoxic concentrations of hydroxyurea. Hydroxyurea also enhanced the bystander effect in the osteosarcoma cells, but not in the MIA PaCa-2 cells, treated with BVDU. These findings indicate that BVDU phosphorylated by Dm-dNK in transduced cancer cells may also induce bystander cell death in certain cell lines.

Effect of famciclovir on herpes simplex virus type 1 corneal disease and establishment of latency in rabbits.

Famciclovir (FCV) is efficacious in the treatment of acute herpes zoster and recurrent genital infections but has not been used to treat ocular herpes simplex virus (HSV) infections. We evaluated the efficacy of orally administered FCV in treating HSV-1 epithelial keratitis and determined its effects on the establishment of latency and subsequent reactivation. Rabbits were inoculated with HSV-1 strain 17 syn+ and treated twice daily with increasing concentrations of FCV (60 to 500 mg/kg of body weight). This resulted in a significant, dose-dependent improvement in keratitis scores, as well as prolonged survival. Regardless of the dose of drug used, all groups exhibited the high rates of spontaneous and induced reactivation characteristic of 17syn+. The efficacy of 250 mg of FCV per kg was also compared to topical treatment with 1% trifluorothymidine (TFT). Although TFT treatment was more effective at reducing eye disease, FCV-treated rabbits had a better survival rate. Real-time quantitative PCR analysis of rabbit trigeminal ganglia (TG) demonstrated that FCV significantly reduced the HSV-1 copy number compared to that after treatment with TFT or the placebo but not in a dose-dependent manner. In summary, oral FCV treatment significantly reduces the severity of corneal lesions, reduces the number of HSV-1 genomes in the TG, improves survival, and therefore may be beneficial in reducing the morbidity of HSV keratitis in the clinic.

Rapid detection of Mycobacterium tuberculosis in contaminated BACTEC 12B broth cultures by testing with Amplified Mycobacterium Tuberculosis Direct Test.

Contamination of broth cultures of acid-fast bacilli (AFB) by bacterial species other than Mycobacterium species frequently occurs. Many of these contaminated cultures require redecontamination and reincubation before the appropriate tests can be performed for identification, significantly affecting the turnaround time for reporting culture results. In this study, the Amplified Mycobacterium Tuberculosis Direct Test (MTD; Gen-Probe) was performed to detect the Mycobacterium tuberculosis complex (MTBC) in 125 BACTEC 12B broth cultures with positive growth indices. Among these, 41 grew non-AFB bacteria only, and all 41 were negative by the MTD. The remaining 84 bottles contained contaminated cultures that grew both AFB and other bacteria or yeasts. Repeat decontamination and reincubation of these specimens required a mean time of 13 days (range, 3 to 40 days). The MTD results were positive for 10 samples, 9 of which were MTBC culture positive and 1 of which grew Myobacterium celatum, a species known to cross-react in the MTD. All cultures growing other mycobacterial species were negative by the MTD. The results of this study demonstrate that the MTD is both sensitive and specific in detecting MTBC in contaminated broth cultures and that, when used selectively, the MTD can potentially rule in or out a diagnosis of MTBC as much as 12 days earlier than using nonamplified DNA probe testing alone can.

Gap junction-mediated bidirectional signaling between human fetal hippocampal neurons and astrocytes.

Gap junctions are clusters of intercellular channels that connect the interiors of coupled cells. In the brain, gap junctions function as electrotonic synapses between neurons and as pathways for the exchange of metabolites and second-messenger molecules between glial cells. Astrocytes, the most abundant glial cell type coupled by gap junctions, are intimately involved in the active control of neuronal activity including synaptic transmission and plasticity. Previous studies have suggested that astrocytic-neuronal signaling may involve gap junction-mediated intercellular connections; this issue remains unresolved. In this study, we demonstrate that second-trimester human fetal hippocampal neurons and astrocytes in culture are coupled by gap junctions bidirectionally; we show that human fetal neurons and astrocytes express both the same and different connexin subtypes. The formation of functional homotypic and heterotypic gap junction channels between neurons and astrocytes may add versatility to the signaling between these cell types during human hippocampal ontogeny; disruption of such signaling may contribute to CNS dysfunction during pregnancy.

Flow-injection electrogenerated chemiluminescence determination of isoniazid using luminol.

Based on a new electrogenerated chemiluminescence (ECL) analytical idea, this paper explains a sensitive and selective flow-injection ECL method using luminol for the determination of isoniazid, based on the sensitizing effect of isoniazid for the weak ECL emission of electrochemically oxidized luminol. Under the optimum experimental conditions, the relative ECL intensity was linear with isoniazid concentration in the range of 4.0 x 10(-8) mol/L to 8.0 x 10(-6) mol/L and with a detecting limit of 2.8 x 10(-8) mol/L.

Increased severity of HSV-1 keratitis and mortality in mice lacking the 2-5A-dependent RNase L gene.

PURPOSE: The2',5'-oligoadenylate-dependent RNase L gene functions in the interferon-inducible RNA decay pathway known as the 2-5A system. The purpose of this study was to determine whether the absence of this gene affects the pathogenesis of herpes simplex virus type 1 (HSV-1) ocular infection in the mouse. METHODS: HSV-1 (strain McKrae) was applied bilaterally to unscarified corneas of RNase L-null mice and congenic controls. To evaluate the severity of herpetic keratitis, slit lamp examinations (SLE) were performed every other day for 14 days. To study corneal histology and apoptosis, HSV-1-inoculated RNase-L-null and congenic control mice, as well as mock-inoculated mice (apoptosis negative control), were killed at 6 and 18 hours postinoculation (PI). Uninoculated mice that underwent corneal scarification (apoptosis positive control) were killed 2 hours after scarification. Eyes were dissected and the corneas processed for light and transmission electron microscopy and the TUNEL assay. RESULTS: In comparison with the congenic control mice, RNase L-null mice showed significantly more severe herpetic keratitis (PI day 8, SLE score, mean +/- SEM: 3.27 +/- 0.10 vs. 2.34 +/- 0.06; P: < 0.001) and significantly higher mortality (PI day 14, 70% vs. 20%; P: < 0.001). Few apoptotic cells were seen in HSV-1-infected RNase L-null mice, although DNA fragmentation consistent with apoptosis was detected in the corneas of congenic control mice 6 and 18 hours after HSV-1 inoculation and in uninfected mice with scarified corneas. Signs of apoptosis were not present in the mock-infected corneas. Electron microscopic evidence of keratocytic apoptosis was detected only in the uninfected scarified corneas and the HSV-1-infected congenic control corneas. CONCLUSIONS: The increased severity of ocular disease and increased mortality in the RNase L-null mice provides evidence, for the first time, that the 2-5A system contributes to protection during ocular herpetic infection. The reduced frequency of apoptosis in these mice suggests that one possible mechanism for this protective effect could be the induction of apoptosis in corneal cells as a means of reducing the spread of infectious virus.

[A computer image processing system to analyze endothelial cell-platelet thrombus quantitatively].

A Computer Image Processing System for cone-plate rolling model which forms endothelial cell-platelet thrombus in vitro is presented in this paper. Some parameters such as endothelial cell shape parameters (area, perimeter, long axial length, short axial length, angle of orientation, and shape index), platelet adhesion coverage, platelet adhesion density, diffusivity and surface reactivity constant are measured during the reaction of endothelial cells and platelet under the fluid shear stress condition. The invitro thrombus model and the corresponding quantitative analysis system is useful in the studies on endothelial cells-platelets interaction and mechanism of thrombosis and atherosclerosis.

[Effect of Xue Zhi Kang on the formation of fatty liver in cholesterol-fed rabbits].

OBJECTIVE: To explore the effect of Xue Zhi Kang (XZhK) on the formation of fatty liver in cholesterol-fed rabbits. METHODS: XZhK was refined from Chinese traditional medicine Hong Qu and its main composition is HMG-CoA reductase inhibitor. Thirty New Zealand white rabbits were randomly assigned into 3 groups (control, hypercholesterol-fed, and XZhK groups) during the 12 week's experiment. The effects of XZhK (0.8g.kg(-1).d(-1)) were investigated in rabbits fed a l.5% cholesterol diet. The changes of serum TC, TG, HDL-C, and LDL-C concentrations and the histopathological changes of the liver were detected. RESULTS: The serum TC, LDL-C, and TG levels decreased in XZhK group compared with those in the cholesterol-fed control group (P<0.05 ). The serum HDL-C concentration slightly increased, but there was no statistical significance. The fatty degeneration of liver was ameliorated in XZhK group (P<0.05). CONCLUSIONS: XZhK can modulate the lipid metabolism and inhibit the formation of fatty liver in cholesterol-fed rabbits.

[Treatment of dying wastewater from a woolen mill with a pilot-scale anaerobic/oxic membrane bioreactor (A/O MBR)].

A pilot-scale (10 t/d) anaerobic/oxic membrane bioreactor (A/O MBR) was used for treatment of dying wastewater from a woolen mill. The results showed that when COD, BOD5 and color in the influent was 179-358 mg/L, 44.8-206 mg/L and 50-240 dilution times (DT), the average COD, BOD5 and color of A/O MBR effluent was 20.2 mg/L, 1.6 mg/L, 25 DT respectively. The removal of COD, BOD5, color, turbidity was 92.1%, 98.4%, 60.7% and 98.9% respectively. Each quota of the treated water met the gray water standards (CJ25.1-89). The A/O MBR process has many advantages, such as stable performance, simple operation, easy management etc. The result of this work could be reference for the designing of industrial scale A/O MBR process for treatment of the woolen mill wastewater.

[The development and identification of animal osteoblasts from bone marrow culture].

This study was aimed to evaluate a modified method for animal osteoblast marrow culture on the basis of previously published method. The marrow mononuclear cells(MMNC) in femoral bone-marrow of SD rat were gained with Ficoll-Hapaque centrifuge, which were then cultured in 5 x 10(5) cells/cm3 on the 96-well culture plates with RPMI 1640 medium containing 30% horse serum in a humidified atmosphere of 5% carbon dioxide at 37 degree Celsius. The medium was changed every three days. Cultured cells were identified with inverted microscopy for observing the shape of living cells, and with Wright-Geimsa mixture stain and Alkaline phosphatase (ALP) stain. In order to avoid false positive reaction, we caused damage to the ALP in cultured cells and stained the cells with ALP again for comparison. The results of inverted microscopy showed that the small spherical mononuclear cells became polygonal, spindle-shaped or squamous successively from the fifth day onward. Wright-Geimsa staining showed the specific property of osteoblast. ALP stain positive reaction was between "2+" and "3+", then ALP stain negative reaction was noticed after the damage to ALP. These data demonstrated that MMNC in SD rat could become osteoblast in RPMI 1640 medium containing 30% horse serum. The modified osteoblast marrow culture method has the advantages of short time, less risk of contamination and higher efficiency.

Retroviral vector containing human p16 gene and its inhibitory effect on Bcap-37 breast cancer cells.

OBJECTIVE: To study the effects of the p16 gene on tumor cell growth inhibition and cell cycle arrest. METHODS: The recombinant retroviral vector pLp16SN was constructed by cloning the human p16 cDNA into the retroviral vector pLXSN. Retroviruses with or without the p16 gene were obtained by transfecting pLp16SN and pLXSN vectors into PA317 cells. Bcap-37 human breast cancer cells were infected with these retroviruses followed by selection with G418. The expression of p16 was detected by Northern and Western blots. Cell biological characteristics, including cell growth rate, cell cycle and tumorigenesis in nude mice were assessed. RESULTS: Both mRNA and protein expression of p16 in Bcap-37 cells transfected with retroviral vector containing the p16 gene were much higher than that in Bcap-37 cells transfected with empty vector or parental Bcap-37 cells. Cell overexpressing the p16 gene exhibited a slower rate of growth, a higher percentage of cells in the G1 phase, and smaller tumors in nude mice, compared with parental Bcap-37 cells and cells transfected with empty vector. CONCLUSION: Overexpression of the p16 gene could suppress the growth of Bcap-37 breast cancer cells by arresting the cell cycle at the G1 to S-phase.

Optical coherence tomography in measuring retinal nerve fiber layer thickness in normal subjects and patients with open-angle glaucoma.

OBJECTIVES: To investigate image characteristics and thickness of the retinal nerve fiber layer (RNFL) in normal and glaucomatous eyes using optical coherence tomography (OCT), and analyze the relationship between RNFL thickness and visual field index. METHODS: Eighty-three normal persons (150 eyes) and 83 patients with primary open angle glaucoma (POAG, 149 eyes) underwent OCT examinations with 3.4 mm diameter circle scan to calculate the RNFL thickness. Statistical analysis was used to compare differences in RNFL thickness in quadrants and means between the normal and glaucomatous groups and the different stages of POAG. Linear correlation and regression analysis were used to show the correlation between RNFL thickness and visual field index of 115 eyes in glaucomatous patients. Reproducibility, sensitivity and specificity of RNFL measurements using OCT were evaluated. RESULTS: RNFL thickness measured by OCT in normal subjects was thicker in superior and inferior, less in temporal, and thinnest in nasal quadrants. The curve showed double peaks. RNFL of glaucomatous patients showed local thinning or defect, diffuse thinning, or both. The mean RNFL thicknesses of the normal group in the temporal, superior, nasal and infelor quadrants were 90.1 +/- 10.8 microns, 140.4 +/- 10.5 microns, 85.2 +/- 14.0 microns, and 140.4 +/- 9.7 microns, respectively with a mean of 114.2 +/- 6.0 microns. The numbers for the glaucomatous group were respectively 56.0 +/- 31.0 microns, 81.0 +/- 36.3 microns, 47.1 +/- 27.5 microns, and 73.4 +/- 38.4 microns for the four quadrants, with a mean of 64.6 +/- 28.8 microns. There was a significant difference in RNFL thickness between the normal and glaucomatous groups (P < 0.000), and the three stages (early, developing and late) of glaucomatous groups (P < 0.000). There was a close negative relationship between RNFL thickness and visual field index (r = -0.796, P < 0.0001). The sensitivity and specificity of RNFL thickness in POAG measured using OCT were 93.3% and 92.0%, respectively. CONCLUSIONS: OCT can quantitatively measure RNFL thickness differences between normal persons and glaucomatous patients. RNFL thickness gradually decreases while visual field defect increases with the development of POAG.

Regeneration of nNOS-containing nerve fibers in rat corpus cavernosum.

OBJECTIVE: To investigate the effect of cavernous nerve injury on the nNOS-containing nerve fibers in corpus cavernosum. METHODS: Thirty-three male Sprague Dawley (SD) rats were randomly divided into 3 groups: sham-operated controls (n = 5) underwent pelvic exploration without transection of the cavernous nerve; unilateral injury group (n = 14) had their cavernous nerve cut on one side; and bilateral injury group (n = 14) underwent neurotomy on both sides. Corpora cavernosa were harvested at the 3rd week and 6th month after surgery. nNOS-positive nerve fibers were examined with streptavidin-peroxidase immunohistochemistry techniques (SP method). RESULTS: After bilateral ablation, the nNOS-positive nerve fibers were significantly decreased at the 3rd week (17 +/- 4) and remained so at the 6th month (16 +/- 4). For the unilateral injury group, the nNOS-positive nerve fibers were similarly decreased on the side of the neurotomy at the 3rd week (18 +/- 6), but by the 6th month, the number increased significantly (61 +/- 9) and approximated the level on the contralateral side (81 +/- 13). CONCLUSION: Following unilateral cavernous nerve ablation in rats, nNOS-containing nerve fibers regenerate 6 months after surgery. This regeneration process does not occur in animals with bilateral cavernous nerve injury, suggesting that during radical pelvic surgery, the cavernous nerve has to be preserved at least on one side in order to maintain the capacity for penile erection.
