[The expression of uridine diphosphate glucuronosyltransferase mRNA regulated by alcohol].

OBJECTIVE: To investigate whether ethanol and isopentanol could modulate the drug metabolism by regulating uridine diphosphate glucuronosyltransferase (UGT) expression in rat livers and cultured rat hepatocytes. METHODS: Rat livers and primary rat hepatocyte cultures have been used to determine the mRNA levels of five UGT isoenzymes following treatment with ethanol and isopentanol. RESULTS: Following treatment with alcohol in rats, UGT1A1 mRNA and UGT1A5 mRNA were increased to a mean of 177% and 166% of control, respectively. The mRNA levels of UGT2B1 and UGT2B3 were also increased in alcohol-treated rats to 178% and 132% of control, respectively. Incubation of hepatocytes with ethanol and isopentanol significantly increased the mRNA expression of UGT1A1, UGT1A5, UGT2B1 and UGT2B3 mRNA. CONCLUSIONS: The expression of UGT can be regulated by ethanol and isopentanol. As a result, chronic alcohol consumption may modify the metabolism of numerous endogenous and exogenous compounds, in particular some drugs, which are substrates of these UGT isoenzymes.

Post-stroke dementia. Nootropic drug modulation of neuronal nicotinic acetylcholine receptors.

Nefiracetam is a new pyrrolidone nootropic drug that is being developed for clinical use in the treatment of post-stroke vascular-type and Alzheimer's-type dementia. Among a few neuroreceptors that have been identified as potential targets of nootropics, neuronal nicotinic acetylcholine receptors (nnAChRs) are deemed the most important since they are related to learning, memory, and Alzheimer's disease dementia. We have recently found potent stimulating action of nefiracetam on nnAChRs. Rat cortical neurons in long-term primary culture expressed nnAChRs. Whole-cell patch clamp experiments revealed two types of currents induced by ACh, alpha-bungarotoxin (alpha-BuTX)-sensitive, rapidly desensitizing, alpha 7-type currents and alpha-BuTX-insensitive, slowly desensitizing, alpha 4 beta 2-type currents. Although alpha 7-type currents were only weakly inhibited by nefiracetam, alpha 4 beta 2-type currents were potently and efficaciously potentiated by nefiracetam. Nefiracetam at 0.1 nM reversibly potentiated ACh-induced currents to 200-300% of control. Very high concentrations (about 10 microM) also potentiated these currents, but to a lesser extent, indicative of the bell-shaped dose-response relationship known to occur for nefiracetam, even in animal behavior experiments. Three specific inhibitors of each of PKA and PKC did not prevent nefiracetam from potentiating ACh-induced currents, indicating that these protein kinases are not involved in nefiracetam action. Pretreatment with pertussis toxin did not alter nefiracetam potentiation, indicating Gi/Go proteins are not involved. Pretreatment with cholera toxin did abolish nefiracetam potentiation. Thus, nefiracetam potentiation is mediated via Gs proteins. In conclusion, nefiracetam stimulates alpha 4 beta 2-type nnAChRs via Gs proteins at nanomolar concentrations. The potentiation of alpha 4 beta 2-type nnAChRs is thought to be at least partially responsible for cognitive enhancing action.

Nationality differences in distributions of serum lipids, lipoproteins and apolipoproteins levels in Xinjiang China.

OBJECTIVE: To reveal the distribution characteristics of serum lipids, lipoproteins, and apolipoproteins levels in different nationalities. METHODS: Quantitative levels of those traits mentioned above were determined and body height (H), weight (W) and body mass index (BMI = W/H2) were assessed in 773 Kazaks (men 360 and women 413) and 911 Han nationality (men 466 and women 445) from the Xinjiang Autonomous Region of China. RESULTS: Kazaks men and women, respectively, had significantly higher serum levels of HDL-c and ApoA1 (all P < 0.001) and significantly lower serum levels of TG (men P < 0.01, women P < 0.001), Lp(a) (all P < 0.005), ApoB (men P < 0.005, women P < 0.001) and ApoB/A1 (men P < 0.01, women P < 0.001) than their Han nationality controls. The threshold points of ApoA1 (< 1.2 g/L) and ApoB (> 1.2 g/L) are higher in Hans than in Kazaks for ApoA1 (20.7 vs 9.8%) and ApoB (18.6 vs 14.3%); the serum levels of TG, HDL-c, Lp(a), ApoA1 and ApoB/A1 are all closely correlated with nationality (P < 0.001, P = 0.001, and P < 0.05, respectively). CONCLUSION: The results suggest that the nationality differences exist in serum levels of lipids, lipoproteins, and apolipoproteins, and Kazaks have a superior serum lipid pedigree to Hans. This differences may come from genetic differences, which affect the serum levels of lipids, lipoproteins, and apolipoproteins by controlling lipid metabolism patterns. Future study will be needed to dissect to the role of genetic factors on serum lipids.

[Treatment of acute aplastic anemia with allogeneic peripheral blood stem cell transplantation: a case report and literature review].

OBJECTIVE: To report a patients with acute aplastic anemia(AAA) successfully treated with allogeneic peripheral blood stem cell transplantation(allo-PBSCT). METHODS: A 30 years old patient with AA received allo-PBSCT from a HLA-compatible sibling donor. PBSCs mobilization regimen was G-GSF 250 micrograms/d x 6. The conditioning regimen included (CTX 50 mg.kg-1.d-1) x 4 and (ATG 10 kg-1.d-01) x 2. 8.97 x 10(8) nucleated cells/kg, 9.20 x 10(6) CD34+ cells/kg, 11.20 x 10(5) CFU-GM/kg were transplanted. RESULTS: Hematopoietic reconstitution obtained on day 18 after allo-PBSCT. Monitoring of engraftment with three microsatellite markers showed mixed chimerism on day 18 and complete chimerism after day 25. No acute or chronic graft versus host disease (GVHD) occurred. In the follow-up duration of 240 days, the blood and bone marrow pictures were normal and the living status was good. CONCLUSION: Allo-PBSCT was an effective therapy in the treatment of acute aplastic anemia.

Autologous peripheral blood stem cell transplantation in the patients with hematologic malignancies and solid tumors.

OBJECTIVE: To evaluate the long-term therapeutic effects of autologous peripheral blood stem cell transplantation (auto-PBSCT) on the treatment of hematological and solid tumors. METHODS: Fifty-one patients were recruited in this auto-PBSCT study, in which several potentially important parameters were studied including the optimal time for stem cell collection, the dose of stem cell reinfusion, the time of hematopoietic reconstitution, the disease free survival (DFS) and overall survival (OS), complications related to transplantation, and maintenance chemotherapy after auto-PBSCT. RESULTS: After APBSCT, 3-year and 5-year survival rates of NHL were 83.3%; those of AML were 74.7%; those of MM were 37.9% and 19%; those of ALL were 40% and 0% respectively. Hematopoietic reconstitution was greatly promoted by granulocyte colony stimulating factor (G-CSF). The mean time for patients' neutrophil to recover up to > 0.5 x 10(9)/L after APBSCT was 11.14 days in the group of the patients receiving G-CSF in contrast to 17.6 days in the group receiving no G-CSF. The most common complications of transplantation were fever, liver dysfunction and hypokalaemia, which were curable. No death was due to transplantation related complications. CONCLUSION: Comparing with conventional chemotherapy, our study suggests that auto-PBSCT is a very important therapeutic option that can significantly improve the prognosis in the patients with hematological and solid tumors, especially in the patients with AML and NHL.

[Different serum lipid adjustment drugs for the treatment of hyperlipidemic fatty liver].

OBJECTIVE: To research the effect of different serum lipid adjustment drugs, Simvastatin and Fenofibrate, on hyperlipidemic fatty liver. METHODS: The rat models of hyperlipidemic fatty liver induced by high fat diet were treated with Fenofibrate and Simvastatin respectively to investigate the change of the liver index, liver function, the serum and liver lipid, MDA, and pathologic histology. RESULTS: The liver index and the content of triglyceride (TC1.80 +/-0.20/2.10+/-0.33), total cholesterol (TG 0.76+/-0.18/1.09+/-0.31), MDA both in the serum and the liver in the model group were significantly higher than those in control group. Slight or moderate hepatic steatosis was observed in model group. Fenofibrate decreased serum triglyceride, total cholesterol, and MDA both in serum and liver significantly, but increased the ALT, ALP, liver index, liver lipids, and finally leading to severe hepatic steatosis. Simvastatin lowered the level of lipid and MDA both in serum and liver, and liver histology was improved nearly to normal. Moreover, little side effect on the liver was observed. CONCLUSIONS: Simvastatin can be applied for prevention and treatment of hyperlipidemic fatty liver safely and effectively, while Fenofibrate can not.

[Study of the route of intrahepatic metastasis of primary hepatic carcinoma].

OBJECTIVE: To study the relationship between pylic countercurrent blood-flow and intrahepatic metastasis of hepatocarcinoma. METHODS: Portal vein-hepatic artery fistula and pylic countercurrent blood flow in the liver were examined by color Doppler ultrasound. Patients examined were divided into four groups: group 1, hepatocarcinoma with background of hepatitis, group 2, hepatocarcinoma without background of hepatitis, group 3, hepatitis and group 4, normal liver. RESULTS: Pylic countercurrent flood flow was most frequently observed in group 1 patients. Intrahepatic metastasis was more frequent in group 1 than in group 2 patients. There was no statistically significant difference in the frequency of portal vein-hepatic artery fistula formation around and inside of tumor between group 1 and group 2 patients. CONCLUSION: Intrahepatic metastasis of hepatocarcinoma is due to countercurrent blood flow in the portal system.

[Study on the constituents from freeze-dried power of blackberries (Rubus ursinus)].

Three Compounds were isolated from freeze-dried powder of Blackberries (Rubus ursinus L.) which showed an activity on inhibition of chemocarcinogen. The structures of them were identified as stigmasta-5,22-dien-3-ol],beta-sitosterol and beta-sitosterol-3 beta-D-glucose. All these compounds were isolated at the first time from the plant.

[Further study on cyclin D1 expression and function in human nasopharyngeal carcinoma cell line].

OBJECTIVE: In the present study, we tried to further elucidate the expression and function of cyclin D1 in human nasopharyngeal carcinoma (NPC) cell line. METHODS: Western blot was used in the analysis of D-type cyclin expression profile. The distribution of this protein was determined in the double parameter flow cytometry (FCM). The function of cyclin D1 in nasopharyngeal carcinoma cell line was determined in the antisense-oligonucleotied-cyclin D1 and antibody mediated knock-out experiments. RESULTS: The immunoblot analysis of two NPC cell lines showed that the spectra of D-type in NPC cell lines founded their expressions in D1, D2, and D3. It also suggested that the overexpression of cyclin D1 could lead to deregulation of G1/S control. An additional double parameter FCM showed a characteristic variation in HNE1 which was consistent with the cell-cycle oscillation and the peak level expressed in G0/G1 phase and the lowest level in the S-phase. Two experimental approaches aimed at specific functional knock-out of cyclin D1 were employed; the cyclin D1-ASPODN experiment showed an inhibition of cyclin D1 expression at mRNA level and a down-regulated cyclin D1 partial inhibition of progression into S phase, resulting in cell growth arrest while the electroporation of neutralizing antibodies demonstrated a specific delay of S-phase entry in HNE1 and the requirement for the cyclin D1 in cell cycle progression of HNE1 cell line in regulation of G1/S. CONCLUSION: These data reveal that the cell-cycle regulatory function of cyclin D1 is essential for cell cycle progression in the G1 of nasopharyngeal carcinoma cell line and suggest that cyclin D1 play an important role in NPC carcinogenesis.

[An exponential universal formula and evaluation model of sustainable development for cities].

In order to study an evaluation model of sustainable development for cities, analysis was performed to study the features of indicator system of community, economy and environment. Based on the genetic algorithms optimum, an exponential universal formula of single indicator was proposed. Evaluation model of sustainable development about the coordinative development of community, economy and environment was developed using weighting method combining the general contrast algorithm with analytic hierarchy process for different structure. The evaluation results of sustainable development for examples using this model showed that the exponential formula of single indicator based on parameters optimum and the evaluation model are not only simple and practical, but also comparisonable, realizable and general.

[Values of carbon dioxide emission from different land-use patterns of alpine meadow].

The value order of carbon dioxide emission from different land-use of alpine meadow was significantly with season changing. It was found that the Potentilla fruticosa shrub meadow(C, 1871.40 g/m2) > Kobresia humilis meadow(A, 1769.63 g/m2) > Degraded Potentilla fruticosa shrub meadow(D, 1495.60 g/m2) > Degraded Kobresia humilis meadow(B, 1191.26 g/m2) during growth season, A(661.46 g/m2) > C(550.90 g/m2) > B(502.50 g/m2) > D (384.50 g/m2) in rest period; and A(2431.09 g/m2) > C(2422.30 g/m2) > D(1880.10 g/m2) > B(1694.06 g/m2) for whole year, separately. This deference not only decided the soil microbial activity and soil characters, but also have the closely relationship to shortage of frozen period.

[Leaching and vertical transport of Zirconium-95 in soils].

The leaching and vertical transport of Zirconium-95 in three representational soils (paddy soil, red-yellow earth and sea mud) in Zhejiang province and the influence of salinity on the activities of Zirconium-95 in sea mud were studied with soil-pole installation. The results show that: 1. The content of Zirconium-95 in collected leachate was low after passing through soil column with 250 ml, 500 ml, 750 ml leaching amount. The amount of Zirconium-95 in collected leachate of paddy soil, sea mud and red-yellow earth was about 5.33%-7.68%, 0.77%-1.32% and almost 0 of labelled amount respectively. And with increasing the total amount of leaching water, the content of Zirconium-95 in collected leachate varied indistinctively, which indicated that once Zirconium-95 was absorbed by soils, it was very difficult to desorb. 2. In paddy soil, red-yellow earth and sea mud, 45.99%-50.02%, 96.02%-97.16% and 95.94%-98.01% radioactivity of Zirconium-95 was concentrated in the 0.4 cm upper layer soils respectively, which illustrated that most of Zirconium-95 remained in the surface soils. 3. With the increase of salinity in leaching water, the amount of Zirconium-95 in surface soils (0-0.2 cm) presented obviously increased, which demonstrated that increasing salinity could raise the adsorption rate of Zirconium-95 in soils.

[Analysis on the present situation of human rabies epidemic in China].

OBJECTIVE: To summarize and analyze human rabies epidemic situation in the last five years in China and to explore the possible factors causing the rising of epidemics to provide reference for the prevention and control of rabies in the future. METHODS: Collecting and analyzing data of rabies epidemics in the last five years in whole nation. RESULTS: Human rabies is continuing going up in the last five years in China with an increase of 44.65%, 1.74% and 45.93% in 1997, 1998, 1999. Human rabies incidence was also seen an increase from 0.0134/100,000 in 1996 to 0.02708/100,000 in 1999. The provinces of rabies increase were mainly located in south China. Among them, Jiangsu province reported 40 human cases in 1999, which increased 207.69% than the corresponding period in 1998. The cases in these high epidemic provinces took up 75% of the whole nation's cases. Number of reported rabies case was also rising. CONCLUSION: Facts as increasing number of dog raising, having not been injected with rabies vaccine and antirabies serum timely or full course after exposure, unstrictly controlling the quality of rabies vaccine as well as lacking good cooperation in different official department on rabies control might serve as factors responsible for the recurrent increase of human rabies.

[A pilot study on rapid glaucoma screening].

OBJECTIVE: To screen glaucoma effectively and rapidly and evaluate the specificity and sensitivity of the method. METHODS: 734 subjects were screened in 4 weekend days. All subjects underwent intraocular pressure (IOP) examination with Tonopen, anterior chamber depth measurement with Van Herick method, fundus photography without pupil dilatation and supra-threshold perimetry with Frequency Doubling Perimeter (FDP), Octopus and Humphery automatic perimeter. The cases with positive results in the above examinations should undertake threshold perimetry and fundus stereo photography for the evaluation of the retinal nerve fiber layer (RNFL). RESULTS: Of the 734 subjects with complete data, there were 55 cases with open angle glaucoma (7.5%), 59 angle-closure glaucoma (8.4%) and 98 glaucoma suspects (13.4%). The IOP of 34 patients was > 21 mm Hg in all the glaucomatous patients (29.8%). The abnormal rates of optic nerve and visual field were 94.5% and 67.3% respectively in the patients with open-angle glaucoma. The abnormal rates of anterior chamber, visual field and optic nerve were 89.8%, 72.9% and 45.8% respectively. CONCLUSIONS: The most efficient method for screening open-angle glaucoma is fundus stereo photography with evaluation by an expert. Anterior chamber evaluation is necessary in screening glaucoma. Supra-threshold perimetry has limited specificity, and FDP might be a promising method in screening glaucoma with relatively good specificity.

Primary culture of porcine pancreatic acinar cells.

OBJECTIVE: To develop a method for the primary culture of porcine pancreatic acinar cells. INTERVENTIONS: Dispersed pancreatic acinar cells available utilizing RPMI-1640 medium containing collagenase III. After purification, the isolated acinar cells were cultured in RPMI-1640 medium with the addition of 2.5% fetal bovine serum. MAIN OUTCOME MEASURES: The morphological characteristics of acinar cells were described. (3)H-thymidine incorporation of acinar cells and the activity of amylase or lipase were determined during the culture process. RESULTS: There were no remarkable morphological changes in the pancreatic acinar cells during the 20 days culture. The acini showed a tendency to gather but did not attach to the walls of the culture disks. A good (3)H-thymidine incorporation of acinar cells in the primary culture was maintained. The secretion of amylase or lipase from the acini decreased with the length of time of the culture. DISCUSSION: The primary culture of acinar cells from a porcine pancreas which was carried out in this study maintained the normal morphology of the acinar cells and their ability to grow but not their secretion of amylase or lipase. The method would benefit by the further experiments on acini of porcine pancreas.

[Correlation of cell apoptosis induction with expression of human beta5 integrin on hematopoietic cells].

OBJECTIVE: To investigate the function of alpha(v)beta5 integrin in hematopoietic cells. METHODS: Beta5 integrin cDNA was expressed in K562 cells through a retroviral vector system. The changes of the alpha(v)beta5 and alpha(v)beta3 integrins expression in apoptosis and differentiation induced by serum depletion were observed. RESULT: The beta5 integrin cDNA failed to express in K562 cells after the introduction of the beta5 integrin retrovirus vector pG beta5CHT. Imbalanced expression of alpha(v)beta3 and alpha(v)beta5 integrins occurred during apoptosis and differentiation induced by serum depletion. Treatment of hematopoietic cells with anti-alpha(v)beta5 monoclonal antibody inhibited the cells apoptosis. CONCLUSION: Overexpression of the alpha(v)beta5 integrin cDNA in hematopoietic cells was associated with the inhibition of cell proliferation and apoptosis.

[Treatment of nuclein 32P colloid perfusion with chronic suppurative maxillary sinusitis].

OBJECTIVE: To determine the effect of nuclein 32P colloid in treatment of chronic maxillary sinusitis (CMS). METHOD: 22 patients (31 sides) with CMS after washing of sinucavity were perfused with nuclein 32P in sinucavity. 20 patients (31 sides) with routine conservative treatment were served as a control group. Two groups were followed up for one year. RESULT: In nuclein group, 27 sides were cured, 4 sides were recurred and the rate of cure was 87%. In routine group, 19 sides were cured, 12 sides were recurred and the rate of cure is 61%. There is significant difference between the two groups (P < 0.01). The treatment active of nuclein group is better than that of routine group. CONCLUSION: Nuclein 32P colloid with the function of ionizing radiation can increase the pathogenic tissue immunocompetence and metabolic course, obstruct the microvascular and promote the absorption of inflammatory exudation and edema subsidence. It can also make the proliferous tissue atrophy thence accelerate the rate of cure of CMS.

[Antioxidation of flavones of wheat germ on mammary tumor of rats].

The effect of flavonoids of wheat germ on mammary tumor of rats induced by 7,12-dimethylben(a) anthracene (DMBA) was investigated. Sprague-Dawley female rats (50 day-old, weighted around 176 g) were randomly divided into 4 groups. The negative and positive control group were fed on stoke diet. The high and low dose test groups were fed on diets with wheat germ flavonoids 10 and 2 g/kg respectively. Except rats in the negative control group, each rat was given DMBA 15 mg dissolved in 1.5 ml vegetable oil by tube feeding. After the administration of DMBA for 24 weeks, the incidence of tumor in the high dose test group was lower than that in the positive control group. The activity of blood and liver glutathione peroxidase (GSH-Px) and peroxidase dismutase(SOD) in the test groups was significantly higher than those in the positive control group, while the MDA level was significantly lower. The results suggested that the effect of flavonoids on inducing peroxidase might be one of the chemical prevention mechanisms on mammary tumors.
