Cloning, expression and characterization of a cDNA (6A8) encoding a novel human alpha-mannosidase.

A 3300-bp cDNA (6A8) has been isolated from a human tonsil cell lambdagt11 cDNA library (GenBank accession number: AF044414). The 6A8 gene is localized on human chromosome 13q31-32. Its cDNA has an open reading frame from position 57 bp to 3243 bp, encoding a 1062 amino-acid polypeptide. The sequence of the polypeptide has 89% identity to rat liver ER alpha-mannosidase. Homogenates of COS-7 cells transfected with 6A8 cDNA showed an enhanced enzymatic activity with p-nitro-phenyl-alpha-D-mannopyranoside, which was not inhibited by swainsonine. These data suggest that 6A8 alpha-mannosidase belongs to the class II alpha-mannosidase. Western blot analysis showed a band for 6A8 cDNA encoded protein of approximately 120 kDa. Northern blot analysis revealed two 6A8 mRNA transcripts with different tissue distribution. Enhanced concanavalin A (ConA) binding to CNE-2L2 cells transfected with a reverse 6A8 DNA was observed, indicating that the 6A8 protein is an important cellular alpha-mannosidase.

Effects of a mandibular repositioner on obstructive sleep apnea.

The purpose of this study was to investigate the effects of a mandibular repositioner on airway, sleep, and respiratory variables in patients with obstructive sleep apnea. Twenty-two patients selected for this study were confirmed with a diagnosis of obstructive sleep apnea based on initial nocturnal polysomnography. The patients were fitted with a mandibular repositioner designed to hold the mandible anteroinferiorly. Six months later, an outcome polysomnographic study was undertaken for each patient with the appliance in place. Lateral cephalometric radiographs in the upright position were also obtained before and after 6 months of treatment. The respiratory disturbance index decreased in 21 of the 22 patients with the appliance in place. The mean respiratory disturbance index of the 22 patients decreased significantly from 40.3 to 11.7 events per hour (P <.01). Some 59.1% of subjects were considered a treatment success with follow-up respiratory disturbance index < 10 events per hour. The mean minimum blood oxygen saturation level during sleep also improved significantly from 73.4% to 81.3% (P <. 01). The mandibular repositioner was constructed to position the mandible at 75% of the maximal mandibular advancement and with a 7 mm opening between the upper and lower incisors, and no aberrant effect on temporomandibular joint was noted. The retropalatal airway space increased and the cross-sectional area of the soft palate and the vertical distance of the hyoid bone to the mandibular plane decreased significantly. The tongue posture became significantly flatter. A significant linear correlation was found between the reduction in apnea index and specific craniofacial skeletal structures (length of anterior cranial base, mandibular plane angle, and upper to lower facial height ratios, P <.05). Subjects with a smaller reduction in apnea index tended to have shorter anterior cranial bases, steeper mandibular planes, and smaller upper to lower facial height ratios. We conclude that a mandibular repositioner may be an effective treatment alternative for obstructive sleep apnea and that a reduction in the frequency of apneic episodes is mainly attributed to the effects of the appliance on oropharyngeal structures.

Evidence for the existence of insect defensin-like peptide in scorpion venom.

Insect defensin refers to a group of antibacterial peptides derived from a variety of insect species as well as from scorpion and possessing a three-dimensional structure highly similar to that of scorpion toxins. A full-length cDNA encoding an insect defensin-like peptide was isolated from the venom gland cDNA library of the Chinese scorpion Buthus martensii Karsch. The precursor, the overall organization of which is similar to that of insect defensins, consists of 61 amino acid residues with a putative signal peptide of 15 residues, a propeptide of 7 residues, and a mature peptide of 39 residues (named BmTXKS2). The positions of six cysteines and a conserved glycine in mature BmTXKS2 are the same as those in LqDef, the first defensin found in scorpions, which suggests these peptides should present a similar cysteine-stabilized alphabetamotif. Phylogenetic analysis further shows that the structure of BmTXKS2 is closer to that of ancient defensins (e.g., LqDef and AaDef, two insect defensins present in the scorpion hemolymph) than to scorpion toxins.

The Saccharomyces cerevisiae centromere protein Slk19p is required for two successive divisions during meiosis.

Meiotic cell division includes two separate and distinct types of chromosome segregation. In the first segregational event the sister chromatids remain attached at the centromere; in the second the chromatids are separated. The factors that control the order of chromosome segregation during meiosis have not yet been identified but are thought to be confined to the centromere region. We showed that the centromere protein Slk19p is required for the proper execution of meiosis in Saccharomyces cerevisiae. In its absence diploid cells skip meiosis I and execute meiosis II division. Inhibiting recombination does not correct this phenotype. Surprisingly, the initiation of recombination is apparently required for meiosis II division. Thus Slk19p appears to be part of the mechanism by which the centromere controls the order of meiotic divisions.

CXC chemokine receptor CXCR2 is essential for protective innate host response in murine Pseudomonas aeruginosa pneumonia.

Pulmonary infection due to Pseudomonas aeruginosa has emerged as a leading cause of mortality. A vigorous host response is required to effectively clear the organisms from the lungs. This host defense is dependent on the recruitment and activation of neutrophils and macrophages. A family of chemotactic cytokines (chemokines) has been shown to participate in this protective response. In this study, we assessed the role of the ELR(+) (glutamic acid-leucine-arginine motif positive) CXC chemokines and their CXC chemokine receptor (CXCR2) in lung antibacterial host defense. The intratracheal administration of Pseudomonas to mice resulted in the time-dependent influx of neutrophils to the lung, peaking at 12 to 24 h after inoculation. The influx of neutrophils was associated with a similar time-dependent expression of the ELR(+) CXC chemokines, KC, macrophage inflammatory protein 2 (MIP-2), and lipopolysaccharide-induced CXC chemokine (LIX). Selective neutralization of MIP-2 or KC resulted in modest changes in neutrophil influx but no change in bacterial clearance or survival. However, neutralization of CXCR2 resulted in a striking increase in mortality, which was associated with a marked decrease in neutrophil recruitment and bacterial clearance. Conversely, the site-specific transgenic expression of KC resulted in enhanced clearance of bacteria after Pseudomonas challenge. This study indicates that ELR(+) CXC chemokines are critical mediators of neutrophil-mediated host defense in Pseudomonas pneumonia.

Purification and characterization of the DeoR repressor of Bacillus subtilis.

Transcription of the Bacillus subtilis dra-nupC-pdp operon is repressed by the DeoR repressor protein. The DeoR repressor with an N-terminal His tag was overproduced with a plasmid under control of a phage T5 promoter in Escherichia coli and was purified to near homogeneity by one affinity chromatography step. Gel filtration experimental results showed that native DeoR has a mass of 280 kDa and appears to exist as an octamer. Binding of DeoR to the operator DNA of the dra-nupC-pdp operon was characterized by using an electrophoretic gel mobility shift assay. An apparent dissociation constant of 22 nM was determined for binding of DeoR to operator DNA, and the binding curve indicated that the binding of DeoR to the operator DNA was cooperative. In the presence of low-molecular-weight effector deoxyribose-5-phosphate, the dissociation constant was higher than 1,280 nM. The dissociation constant remained unchanged in the presence of deoxyribose-1-phosphate. DNase I footprinting exhibited a protected region that extends over more than 43 bp, covering a palindrome together with a direct repeat to one half of the palindrome and the nucleotides between them.

The N-terminal domain of p73 interacts with the CH1 domain of p300/CREB binding protein and mediates transcriptional activation and apoptosis.

The newly identified p53 homolog p73 mimics the transcriptional function of p53. We have investigated the regulation of p73's transcriptional activity by p300/CREB binding protein (CBP). p73-p300 complexes were identified in HeLa cell extracts by cofractionation and coimmunoprecipitation assays. The p73-p300 interaction was confirmed in vitro by glutathione S-transferase-protein association assays and in vivo by coimmunoprecipitating the overexpressed p300 and p73 in human p53-free small-cell lung carcinoma H1299 or osteosarcoma Saos-2 cells. The N terminus but not the N-terminal truncation of p73 bound to the CH1 domain (amino acids [aa] 350 to 450) of p300/CBP. Accordingly, this p73 N-terminal deletion was unable to activate transcription or to induce apoptosis. Overexpression of either p300 or CBP stimulated transcription mediated by p73 but not its N-terminally deleted mutant in vivo. The N-terminal fragment from aa 19 to 597, but not the truncated fragment from aa 242 to 1700 of p300, reduced p73-mediated transcription markedly. p73-dependent transcription or apoptosis was partially impaired in either p300- or CBP-deficient human breast carcinoma MCF-7 or H1299 cells, suggesting that both coactivators mediate transcription by p73 in cells. These results demonstrate that the N terminus of p73 directly interacts with the N-terminal CH1 domain of p300/CBP to activate transcription.

The disability assessment database model.

Collecting disability-related information in order to prevent and treat specific disabilities is important and necessary, but does it meet the major needs of individuals with disabilities? This article presents preliminary research performed to assess those needs as well as a Disability-service Assessment Database (DAD) model. This model provides local disability service data and surveys users so that their input is used to modify existing data. The content of the database and Web site are discussed and diagramed. A major goal of the DAD is to serve as a model to other communities.

Changes in genioglossus muscle activity in obstructive sleep apnea patients with and without snore guard.

OBJECTIVE: To study the effect of Snore Guard on the genioglossus (GG) muscle activity of obstructive sleep apnea (OSA) patients. MATERIALS AND METHODS: Fifteen male patients with mild to severe OSA were diagnosed by overnight polysomnographic studies, and were reexamined after using Snore Guard for 2-6 months. The difference in GG muscle activity was then compared. RESULTS: The overnight GG muscle activity decreased significantly with Snore Guard. The levels of GG muscle activity during awake quiet breath and obstructive sleep apnea were higher than in sleepstate quiet breath without Snore Guard, and significantly decreased with Snore Guard. The increase in GG muscle activity during sleepstate quiet breath with Snore Guard was not statistically significant. The fluctuating GG muscle activity without Snore Guard was effectively improved by treatment with the appliance. CONCLUSION: The effect of Snore Guard on OSA patients is a mechanical enlargement of upper airway volume. The genioglossus muscle is passive during treatment.

[Effect of cutting on vegetation composition and soil properties].

Different cutting experiments were executed on well-grown natural Imperata cylindrica vegetation in red soil region. The biomass of every 6 years cutting was the highest, and the annual cut was the lowest. Annual cutting resulted in the highest total accumulative biomass in 6 years, with a total production of 399.1 kg per plot. The species composition was also influenced by the length of cutting period. In the 6th year of experiment, the xylophyta biomass in the treatment of every 6 years cutting accounted for 41.5% of the total biomass, while the herb biomass in the treatment of annual cutting was 99.0% of the total. The length of cutting period also influenced the layer's development of soil profile and the physicochemical properties of soil. In comparing with the treatment of annual cutting, the soil bulk density in the treatment of every 6 years cutting was lower, and the contents of soil organic matter, total nitrogen, available phosphorus and available potassium were higher.

[A comparative study on chlorophyll content, chlorophyll fluorescence and diurnal course of leaf gas exchange of two ecotypes of banyan].

The morphological differences, chlorophyll contents, fluorescence and diurnal course of leaf gas exchange between terrestrial banyan and amphibious banyan were compared with pot culture. The amphibious banyan possesses well developed aerial and hydro-adventitious roots, and wider leaf with inclination of evolution toward mesophytic traits. The chlorophyll content of terrestrial banyan was higher than that of amphibious banyan. The diurnal course of leaf gas exchange indicated that net photosynthetic rate of terrestrial banyan was slightly higher than that of amphibious banyan grown in water, but much higher than that grown in soil. The amphibious banyan grown in water had the highest transpiration rate, the terrestrial banyan had a lower one, and the amphibious banyan grown in soil had the lowest. Linear regression analysis showed a positive correlation between net photosynthetic rate and stomatal conductance, implying that the stomatal conductance was dominant factor controlling the gas exchange. In this study, the term of intrinsic water use efficiency (net photosynthetic rate/stomatal conductance ratio, Intrinsic WUE) was applied to describe the photosynthesis and water properties, and the result showed that it was a more suitable measure compared to the usual WUE(net photosynthetic rate/transpiration rate). Among the three banyan plants examined, the amphibious banyan had the highest intrinsic WUE.

[Detection of hypermethylation of p15 gene in patients with acute leukemia by polymerase chain reaction].

OBJECTIVE: To explore the role of hypermethylation of p15 gene in the pathogenesis of acute leukemias. METHODS: DNA from 36 cases of acute leukemia were investigated using restriction enzyme digestion combined with polymerase chain reaction (PCR) technique. RESULTS: Hypermethylations of p15 gene were found in 22 (61.1%) patients, including 17 cases of acute non-lymphoblastic leukemia (ANLL) and 5 of acute lymphoblastic leukemia (ALL). ANLL patients exhibited p15 gene hypermethylations had higher WBC in peripheral blood (P < 0.05) and higher percentages of leukemia cells in bone marrow (P < 0.01). CONCLUSION: Inactivation of p15 gene due to hypermethylation may be involved in the pathogenesis of some acute leukemias.

[Bax protein expression in the carcinogenesis of human oral mucosa].

OBJECTIVE: Bax gene is an important apoptosis-promoting gene. In order to investigate the expression of Bax in oral premalignant lesions and oral squamous cell carcinomas, a total of 38 samples are evaluated using a labelled streptavidin-biotin (LSAB) immunohistochemical assay. METHODS: A total of 38 specimens were studied, including normal oral mucosa, premalignant lesions and squamous cell carcinomas. The specimens were obtained and blocked, fixed with 10% buffered formalin and embedded in paraffin using conventional histopathological techniques. 3 microns-thick sections of paraffin-embedded tissues were cut, mounted onto slides coated with 5% APES (3-aminopropyltriethoxysilane), dried overnight at 56 degrees C, dewaxed in xylene and rehydrated through descending graded alchols to phosphate-buffered saline (PBS, pH 7.4). For antigen retrieval, slides were immersed in 10 mmol/L sodium citrate buffer (pH 6.0) and boiled twice for 5 min in a microwave oven (800 W). After treated for 10 min with 3% H2O2, 18% methanol in PBS, the slides were covered with 10% normal porcine serum for 10 min at 37 degrees C. Then slides were incubated with primary antibody (bax rabbit polyclonal antibody) for 60 min at 37 degrees C and were subsequently incubated with prediluted biotinylated antibody against rabbit immunogobulins, and streptavidinhorseardish peroxidase conjugate for 30 min at 37 degrees C. After washing, peroxidase activity was detected using 3,3'-diaminobenzidine as chromogen with H2O2 as substrate. The cells in the test specimens which demonstrated granular staining were considered as positive. A haemocytometer counter with 6. 6 framework were applied and only the positive cells on the cross were counted. The cell counting were processed in ten randomly chosen 400* microscopic fields and the mathematic mean was presented as the final counting of each sample. Statistical valuations were performed using the version 6.0 SPSS package. Positive controls were sections of bladder cancer tissues. Blank controls were fabricated for each specimen by the omission of the primary antibody, which was replaced with PBS. RESULTS: In the process of oral carcinogenesis, each stage had Bax expression. The positive staining appeared in cytoplasm. In the normal oral mucosal specimens Bax expression was evident in the prickle layer, but not in the basal cell layers. Various degrees of Bax expression were seen in the diseased tissues. The staining pattern of hyperkeratotic lesions was similar to the normal oral mucosa, but Bax expression were also seen in the basal cell layer. In the mild, moderate, severe dysplasia and squamous cell carcinomas, Bax expression were seen in all layers, however, the intensity of staining were greater in mild and moderate dysplasia. The number of positive cells tended to increase gradually with the development of cell malignancy in the tissues of hyperkeratosis, mild and moderate dysplasia (P < 0.05). In the tissue of squamous cell carcinomas the number of positive cells had no marked difference comparing with the normal oral mucosa. CONCLUSION: The expression of Bax is involved in oral carcinogenesis and the compensative increase of Bax protein expression may be an early response.

[Expression and insecticidal characteristics of cry3A in Lepidoptera-specific Bacillus thuringiensis].

The cry3A gene coding Coleoptera-specific protein was transformed into a wild strain YBT803-1 by electroporation, A transformant BMBY-001 was obtained. SDS-PAGE analysis demonstrated that cry3A could express 64 kD protein in BMBY-001. However, the expression of cry1Ab and cry1Ac were effected. Bioassay showed that BMBY-001 was not only highly toxic to Phyllodecta vulgatissima larva(LC50 0.413 microL/mL) but also toxic to Plutella xylostella(LC50 3.319 microL/mL).

[Determination of naringin and synephrine in fructus aurantii from different habitats by HPLC].

This paper reports the determination of Naringin and Synephrine in Fructus Aurantii from different habitats by HPLC. The result shows the highest content of Naringin in Citrus aurantium L. 'Xiangcheng' from genuine crude drug habitat Xingan city Jiangxi provice, the highest content of synephrine in Citrus aurantium L. 'Xiangcheng' from Yiyang city Jiangxi provice. Quality evaluation of genuine crude drug was studied preliminarily.

[Partial laryngectomy plus radiotherapy versus partial laryngectomy alone for laryngeal carcinoma].

OBJECTIVE: To investigate the effect of radiotherapy in the combined treatment of laryngeal cancer with partial laryngectomy. METHODS: One hundred and seventy-one cases of laryngeal cancer were randomly separated into three groups of partial laryngectomy, partial laryngectomy with preoperative radiotherapy (dose 40-50Gy) and partial laryngectomy with postoperative radiotherapy(dose 51-70 Gy). The surgical procedures included cordectomy, vertical partial laryngectomy, horizontal partial laryngectomy and near total laryngectomy. RESULTS: The overall 5-year survival rate was 82% (123/150). The 5-year survival rates for surgery alone group (56 cases), surgery with preoperative radiotherapy group(65 cases) and surgery with postoperative radiotherapy group(50 cases) were 85.7%, 80.7% and 79.5% respectively. There was no significant difference among the groups(chi 2 = 0.703, P = 0.704). Analysis of survival rates revealed a significant difference among the different stages(chi 2 = 12.248, P = 0.007). All 171 cases except 3 that had near total laryngectomy achieved satisfactory phonation. CONCLUSION: Neither preoperative radiotherapy nor postoperative radiotherapy can improve the 5-year survival rate of laryngeal cancer with partial laryngectomy. All patients with partial laryngectomy got satisfactory phonation.

[The clinical significance of poststyloid space extension in nasopharyngeal carcinoma].

OBJECTIVE: To explore the clinical significance of poststyloid space extension in nasopharyngeal carcinoma. METHODS: One hundred and ninety-eight patients with newly treated nasopharyngeal carcinoma (stage I-IV a) were analyzed. These patients were divided into two groups depending on whether the poststyloid space was extended or normal. The 5-year survival rate, relapse and distant metastasis after the treatment were estimated using the Kaplan-Meier, Log-Rank methods. RESULTS: The rates of patients with oropharyngeal extension for these two groups were 72.1% and 16.8% respectively (P < 0.001). The rates of the cases with involvement of the 9, 10, 11, 12th cranial nerves were 20.9% and 6.2% respectively (P < 0.01). The rates of metastasis to cervical lymph node were 69.8% and 35.4% respectively (P < 0.01). The 5-year survival rates were 46.76% and 68.88% respectively (P = 0.0027). The local control rates were 63.7%, 78.7% respectively (P = 0.0862). The distant metastasis free survival rates were 71.7% and 84.2% respectively (P = 0.0468). These results showed that the survival rate, the local control rate were reduced and the distant metastasis was increased for patients with poststyloid space extension. CONCLUSIONS: The poststyloid space extension was an important prognostic factor with poorer treatment outcome. It is reasonable to classify such condition as stage T3 of nasopharyngeal carcinoma.
