Characterization of the rat GRIK5 kainate receptor subunit gene promoter and its intragenic regions involved in neural cell specificity.

The GRIK5 (glutamate receptor ionotropic kainate-5) gene encodes the kainate-preferring glutamate receptor subunit KA2. The GRIK5 promoter is TATA-less and GC-rich, with multiple consensus initiator sequences. Transgenic mouse lines carrying 4 kilobases of the GRIK5 5'-flanking sequence showed lacZ reporter expression predominantly in the nervous system. Reporter assays in central glial (CG-4) and non-neural cells indicated that a 1200-base pair (bp) 5'-flanking region could sustain neural cell-specific promoter activity. Transcriptional activity was associated with the formation of a transcription factor IID-containing complex on an initiator sequence located 1100 bp upstream of the first intron. In transfection studies, deletion of exonic sequences downstream of the promoter resulted in reporter gene activity that was no longer neural cell-specific. When placed downstream of the GRIK5 promoter, a 77-bp sequence from the deleted fragment completely silenced reporter expression in NIH3T3 fibroblasts while attenuating activity in CG-4 cells. Analysis of the 77-bp sequence revealed a functional SP1-binding site and a sequence resembling a neuron-restrictive silencer element. The latter sequence, however, did not display cell-specific binding of REST-like proteins. Our studies thus provide evidence for intragenic control of GRIK5 promoter activity and suggest that elements contributing to tissue-specific expression are contained within the first exon.

Solution structure and interaction surface of the C-terminal domain from p47: a major p97-cofactor involved in SNARE disassembly.

p47 is the major protein identified in complex with the cytosolic AAA ATPase p97. It functions as an essential cofactor of p97-regulated membrane fusion, which has been suggested to disassemble t-t-SNARE complexes and prepare them for further rounds of membrane fusion. Here, we report the high-resolution NMR structure of the C-terminal domain from p47. It comprises a UBX domain and a 13 residue long structured N-terminal extension. The UBX domain adopts a characteristic ubiquitin fold with a betabetaalphabetabetaalphabeta secondary structure arrangement. Three hydrophobic residues from the N-terminal extension pack closely against a cleft in the UBX domain. We also identify, for the first time, the p97 interaction surface using NMR chemical shift perturbation studies.

Methylmercury and H(2)O(2) provoke lysosomal damage in human astrocytoma D384 cells followed by apoptosis.

Methylmercury (MeHg) is a neurotoxic agent acting via diverse mechanisms, including oxidative stress. MeHg also induces astrocytic dysfunction, which can contribute to neuronal damage. The cellular effects of MeHg were investigated in human astrocytoma D384 cells, with special reference to the induction of oxidative-stress-related events. Lysosomal rupture was detected after short MeHg-exposure (1 microM, 1 h) in cells maintaining plasma membrane integrity. Disruption of lysosomes was also observed after hydrogen peroxide (H(2)O(2)) exposure (100 microM, 1 h), supporting the hypothesis that lysosomal membranes represent a possible target of agents causing oxidative stress. The lysosomal alterations induced by MeHg and H(2)O(2) preceded a decrease of the mitochondrial potential. At later time points, both toxic agents caused the appearance of cells with apoptotic morphology, chromatin condensation, and regular DNA fragmentation. However, MeHg and H(2)O(2) stimulated divergent pathways, with caspases being activated only by H(2)O(2). The caspase inhibitor z-VAD-fmk did not prevent DNA fragmentation induced by H(2)O(2), suggesting that the formation of high-molecular-weight DNA fragments was caspase independent with both MeHg and H(2)O(2). The data point to the possibility that lysosomal hydrolytic enzymes act as executor factors in D384 cell death induced by oxidative stress.

IUdR polymers for combined continuous low-dose rate and high-dose rate sensitization of experimental human malignant gliomas.

Local polymeric delivery enhances IUdR radiosensitization of human malignant gliomas (MG). The combined low-dose rate (LDR) (0.03 Gy/h) and fractionated high-dose rate (HDR) treatments result in cures of experimental MGs. To enhance efficacy, we combined polymeric IUdR delivery, LDR, and HDR for treatments of both subcutaneous and intracranial MGs. In vitro: Cells (U251 MG) were trypsinized and replated in triplicate 1 day prior to LDR irradiation in media either without (control) or with 10 microM IUdR. After 72 hr, LDR irradiation cells were acutely irradiated (1.1 Gy/min) with increasing (0, 1.25, 2.5, 5.0, or 10 Gy) single doses. Implantable IUdR polymers [(poly(bis(p-carboxyphenoxy)-propane) (PCPP): sebaic acid (PCPP:SA), 20:80] (50% loading; 10 mg) were synthesized. In vivo: For flank vs. intracranial tumors, mice had 6 x 10(6) subcutaneous vs. 2 x 10(5) intracranial cells. For intracranial or subcutaneous MGs, mice had intratumoral blank (empty) vs. IUdR polymer treatments. One day after implantation, mice had immediate external LDR (3 cGy/h x 3 days total body irradiation) or HDR (2 Gy BID x 4 days to tumor site) or concurrent treatments. For the in vitro IUdR treatments, LDR resulted in a striking increase in cell-killing when combined with HDR. For the in vivo LDR treatments of flank tumors, the growth delay was greater for the IUdR vs. blank polymer treatments. For the combined LDR and HDR, the IUdR treatments resulted in a dramatic decrease in tumor volumes. On day 60 the log V/V0 were -1.7 +/- 0.22 for combined LDR + HDR + IUdR polymer (P < 0.05 vs. combined LDR + HDR + blank polymer). Survival for the intracranial controls was 22.9 +/- 1.2 days. For the blank polymer + LDR vs. blank polymer + LDR + HDR treatments, survival was 25.3 +/- 1.7 (P = NS) vs. 48.1 +/- 3.5 days (P < 0.05). For IUdR polymer + LDR treatment survival was 27.3 +/- 2.3 days (P = NS). The most striking improvement in survival followed the IUdR polymer + LDR + HDR treatment: 66.0 + 6.4 days (P < 0.05 vs. blank polymer + LDR + HDR). The polymeric IUdR delivery plus combined continuous LDR and HDR treatments results in growth delay and improved survival in animals bearing the MG xenografts. This treatment may hold promise for the treatment of human MGs.

Complete genome sequence of an M1 strain of Streptococcus pyogenes.

The 1,852,442-bp sequence of an M1 strain of Streptococcus pyogenes, a Gram-positive pathogen, has been determined and contains 1,752 predicted protein-encoding genes. Approximately one-third of these genes have no identifiable function, with the remainder falling into previously characterized categories of known microbial function. Consistent with the observation that S. pyogenes is responsible for a wider variety of human disease than any other bacterial species, more than 40 putative virulence-associated genes have been identified. Additional genes have been identified that encode proteins likely associated with microbial "molecular mimicry" of host characteristics and involved in rheumatic fever or acute glomerulonephritis. The complete or partial sequence of four different bacteriophage genomes is also present, with each containing genes for one or more previously undiscovered superantigen-like proteins. These prophage-associated genes encode at least six potential virulence factors, emphasizing the importance of bacteriophages in horizontal gene transfer and a possible mechanism for generating new strains with increased pathogenic potential.

Photophysical processes of 1,3-dicarbazolypropane.

The photophysical processes of 1,3-dicarbazolypropane (DCZP), which possesses two fluorogen, carbazoly, and one carbonic chain composed of three carbon atoms, were studied. The formation of intermolecular excimer, intramolecular excimer and triple exciplexes have been investigated in the system of DCZP and 1,4-dinitrilebenzene (DNB) with steady state fluorescence spectroscopy. The experimental results show that with the addition of DNB into the lower concentration solution of DCZP in benzene, the fluorescence of intramolecular excimer of DCZP, (D-D)*, is quenched while the ((D-D)A)* type triple exciplex (one molecule of DCZP with one molecule of DNB) is formed and with the addition of DNB into higher concentration solution of DCZP, the formation of (DDA)* type triple exciplex (two molecules of DCZP with one molecule of DNB) is also confirmed.

Helper-dependent adenoviral vector-mediated gene transfer in aged rat brain.

Transfer of the neurotrophin gene into brain can attenuate age-related deficits such as neuronal atrophy and memory loss, but a suitable vector for this procedure has been lacking. The toxicity and immunogenicity of first-generation adenoviral vectors with E1 deletion (fgAdv) prohibit the application of gene transfer in the majority of central nervous system disorders. Here, we report less toxic and persistent gene expression mediated by helper-dependent adenovirus (hdAdv) in aged rat brain. After intrahippocampal or intraventricular inoculation of the vector, transgene expression was monitored by X-Gal staining and compared with fgAdv-mediated expression. Host inflammatory and immune responses against these vectors were evaluated by immunohistochemical detection of microglia, astrocytes, and infiltrating macrophages, as well as by enzyme-linked immunosorbent assay of cytokines TNF-alpha and IL-1beta. Transgene expression mediated by hdAdv persisted for more than 183 days regardless of inoculation site, as compared with 33 and 66 days for fgAdv-mediated expression after intraventricular and intrahippocampal inoculation, respectively. Inoculation with hdAdv was also associated with reduced numbers of activated microglial cells, astrocytes, and infiltrating macrophages in brain tissue. Secretion of the proinflammatory cytokines TNF-alpha and IL-1beta was minimal after hdAdv but not after fgAdv inoculation. These findings indicate that hdAdv would provide a safe and effective means to transfer therapeutic genes into aged brain.

Formation of bone-like apatite on poly(L-lactic acid) fibers by a biomimetic process.

Bone-like apatite coating on poly(L-lactic acid) (PLLA) fibers was formed by immersing the fibers in a modified simulated body fluid (SBF) at 37 degrees C and pH 7.3 after hydrolysis of the fibers in water. The ion concentrations in SBF were nearly 1.5 times of those in the human blood plasma. The apatite was characterized by scanning electron microscopy (SEM) with energy dispersive X-ray spectroscopy (EDX), X-ray photoelectron spectroscopy (XPS), thin-film X-ray diffraction, and Fourier transform infrared spectroscopy. After 15 days of incubation in SBF, an apatite layer with about 5-6 microm thickness was formed on the surface of the fibers. This apatite had a Ca/P ratio similar to that of natural bone. The mass of apatite coated PLLA fibers increased with extending the incubation time. After 20 days incubation, the fibers increased their mass by 25.8 +/- 2.1%. The apatite coating had no significant effect on the tensile properties of PLLA fibers. In this article, the bone-like apatite coating on three-dimensional PLLA braids was also studied. The motivation for this apatite coating was that it might demonstrate enhanced osteoconductivity in the future studies when they serve as biodegradable scaffolds in tissue engineering.

The cell wall and cell division gene cluster in the Mra operon of Pseudomonas aeruginosa: cloning, production, and purification of active enzymes.

We have cloned the Pseudomonas aeruginosa cell wall biosynthesis and cell division gene cluster that corresponds to the mra operon in the 2-min region of the Escherichia coli chromosome. The organization of the two chromosomal regions in P. aeruginosa and E. coli is remarkably similar with the following gene order: pbp3/pbpB, murE, murF, mraY, murD, ftsW, murG, murC, ddlB, ftsQ, ftsA, ftsZ, and envA/LpxC. All of the above P. aeruginosa genes are transcribed from the same strand of DNA with very small, if any, intragenic regions, indicating that these genes may constitute a single operon. All five amino acid ligases, MurC, MurD, MurE, MurF, and DdlB, in addition to MurG and MraY were cloned in expression vectors. The four recombinant P. aeruginosa Mur ligases, MurC, MurD, MurE, and MurF were overproduced in E. coli and purified as active enzymes.

QSAR study on the toxicity of substituted benzenes to the algae (Scenedesmus obliquus).

50% effective inhibition concentration 48h-EC50 of 40 substituted benzenes to the algae (Scenedesmus obliquus) was determined. The energy of the lowest unoccupied molecular orbital (E(LUMO)) was calculated by the quantum chemical method MOPAC6.0-AM1. By using E(LUMO) and the hydrophobicity parameter log K(OW) the quantitative structure-activity relationship model (QSAR) was developed: log1/EC50=0.272 logK(OW) - 0.659E(LUMO) + 2.54, R2 = 0.793, S.E. = 0.316, F = 71.07, n = 40. A series of equations were obtained about the measured EC50 values of different subclasses of compounds. For those compounds containing double -NO2, their toxicity may be related chiefly to the intracellular reduction of -NO2 obtaining electron, while for anilines and phenols, K(OW) contributes most to the QSAR and E(LUMO) very little.

Radiostereometric analysis of distal radial fracture displacement during treatment: a randomized study comparing Norian SRS and external fixation in 23 patients.

In a randomized study, we included 23 osteoporotic patients with a distal radial fracture and loss of reduction after 1 week. The facture was re-reduced. In one group, a self-setting hydroxyapatite, Norian SRS, was injected into the fracture and the wrist was immobilized for 2 weeks with a dorsal splint (n 12). In the other group, the fracture was immobilized for 5 weeks with an external fixator (n 11). During the operation, the fracture fragments were marked with tantalum markers, so that loss of reduction during the immobilization and after mobilization could be studied with radiostereometric analysis (RSA). We found some recurrence of compression in the fracture in both groups during immobilization. After mobilization, the motion of the fracture, measured by displacement of the fragments along the longitudinal axis, was less than 2 mm, except in 3 cases treated with Norian SRS. A compression along the longitudinal axis of less than 2 mm is not likely to cause any problem in the long term. From the first to the last investigation, 7/12 patients with Norian SRS and 4/11 with external fixation lost more than 2 mm of the reduction along the longitudinal axis. We conclude that 5 weeks of immobilization is sufficient for healing with external fixation in this age group. This immobilization time might be reduced to 2 weeks for fractures treated with Norian SRS, but additional hardware should be used to ensure stability of the fracture system.

The accumulation effect of lead on DNA damage in mice blood cells of three generations and the protection of selenium.

The accumulation effect of lead on DNA damage and the protection of selenium against lead were studied in mice blood cells of three generations. The test animals were divided into four groups: controls, Pb group, Se group, Pb + Se group. Lead and Selenium were administered to mice in the drinking water. The concentrations of lead (in form of lead acetate) and selenium (in form of Kappa-selenocarrageenan) used in this paper were 1 microgram/ml and 0.25 microgram/ml respectively. The percentage of damaged cells (DNA comets) was examined and detected conveniently by the single-cell gel electrophoresis (comet) assay. Results showed that 1 microgram/ml lead did not damage the female mice blood cells of the first generation seriously. However, there was a significant damage effect in both sexes of the second and third generations, suggesting the accumulation effect of lead was very significant starting from the second generation. In addition, under normal physiological conditions, Kappa-Se did not appear to enhance the protective ability on DNA damage significantly, whereas under lead exposure conditions, administration of Kappa-Se displayed the occurrence of a good protection against lead intoxication which was started from the female mice of the second generation.

Regulation of rat carboxylesterase expression by 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD): a dose-dependent decrease in mRNA levels but a biphasic change in protein levels and activity.

Carboxylesterases play an important role in the metabolism of endogenous lipids and foreign compounds; therefore, xenobiotic regulation of carboxylesterase gene expression has both physiological and pharmacological significance. We previously reported that beta-naphthoflavone and 3-methylcholanthrene, two potent inducers for cytochrome P4501A enzymes, had opposing effects on the expression of hydrolase S, a secretory carboxylesterase. Beta-naphthoflavone caused suppression, whereas 3-methylcholanthrene caused induction of the expression of this enzyme. The aim of the present study was to determine the effects of 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD), another prototypical cytochrome P4501A inducer, on the expression of this and several other rat carboxylesterases (hydrolases A, B, and C) in liver and extrahepatic tissues. Rats received TCDD treatment at nonlethal (<or=10 microg/kg), sublethal (30 microg/kg), or lethal doses (90 microg/kg). The overall hydrolytic activity in the liver microsomes toward both 1-naphthyl- and para-nitrophenylacetate was markedly increased in the nonlethal dosage groups, but markedly decreased in the lethal dosage group. Consistent with such a biphasic dose-response relationship, the levels of enzyme proteins exhibited an initial increase, followed by a decrease in response to nonlethal and lethal doses, respectively. In contrast, treatment with TCDD caused a dose-dependent decrease on the levels of mRNA encoding these enzymes. All liver carboxylesterases showed a similar pattern of change on activity, protein, and mRNA levels, suggesting that TCDD coregulates the expression of these genes. In the extrahepatic tissues, a similar biphasic change was observed in activity and in protein and mRNA levels. In both liver and kidney, the expression of cytochrome P4501A1 (CYP1A1) was significantly induced in a dose-dependent manner. TCDD is known to upregulate the expression of CYP1A1 gene through the aryl hydrocarbon receptor (AhR). The differential effects on the expression of liver carboxylesterases and CYP1A1 suggest that TCDD regulates the expression of hydrolytic enzymes via a mechanism(s) other than the AhR-mediated transcription activation, as observed in the CYP1A1 regulation. The different patterns of change on protein and mRNA levels in the nonlethal dosage groups suggest that TCDD regulates the expression of hepatic carboxylesterases by acting on both transcription and translation.

Unraveling oligodendrocyte origin and function by cell-specific transgenesis.

Besides the role of mature oligodendrocytes in myelin synthesis during the development of the central nervous system (CNS), the oligodendrocyte lineage also encompasses the largest pool of postnatal proliferating progenitors whose behavior in vivo remains broadly elusive in health and disease. We describe here transgenic models that allow us to track the functions and origins of such cells by using proteolipid protein and 2',3'-cyclic nucleotide 3'-phosphodiesterase (CNP) gene promoters to direct oligodendroglial expression of different reporters, in particular the green fluorescent protein (GFP). We emphasize that the CNP-GFP mouse, which targets the entire oligodendroglial lineage from embryonic life to adulthood, provides an outstanding tool to study the in vivo properties of oligodendrocyte progenitor cells in normal and damaged CNS.

[Extraction technology of Fructus Psoraleae with orthogonal test].

The extraction conditions of Fructus Psoraleae were studied with orthogonal test, taking the contents of psoralen and isopsoralen as indexes and smesh degree, alcohol concentration, alcohol volume, immersion time and immersion times as factors. The results showed the optimal extraction process is pulverizing the herbs into 20 meshes powder, 6 times volume of 55% alcohol immersed three times and two days every time.

[Assessment of ecosystem health--concept framework and indicator selection].

Based on the concept framework of ecosystem health, this paper discussed the suitable indicators to synthetically assess the ecosystem health in governed landscape. Healthy ecosystem means not only ecological health itself, but also the long-term maintenance of healthy human population and the beneficial promotion of social and economic development. Resilience, biodiversity and productivity are the characteristics of healthy ecosystem. The first step in the structure of indicators to assess ecosystem health should be the determination of precise indicator principles. According to the purpose of assessment and the principles of selecting indicator, the indicator system for assessing ecosystem health can be divided into three categories, namely, biophysical indicators, ecological indicators, and social and economic indicators.

[Relationship between intrahepatic HCV-specific CTL activity and response to interferon therapy in chronic hepatitis C].

BACKGROUND: To study the relationship between intrahepatic hepatitis C virus-specific cytotoxic T lymphocyte (HCV-CTL) activity and response to interferon (IFN) therapy in chronic hepatitis C. METHODS: 45 patients with chronic hepatitis C who received IFN-alpha therapy were evaluated for HCV-specific CTL in the liver before treatment with standard chromium release assay. The relationship between treatment response and HCV-CTL activity was analyzed. RESULTS: HCV-CTL activity was detected in 20 (44.4%) of the 45 IFN-treated patients. Of the 42 patients who completed a 6-month course therapy, 19 had detectable HCV-CTL activity, 23 had no detectable HCV-CTL activity. After 6 months of IFN therapy, 18 of 42 (42.9%) patients received an end-of-treatment complete response (ETR). After 6-12 months of follow-up, 10 patients (23.8%) had a sustained response (SR). Of the 19 patients with detectable HCV-CTL activity, 15 (78.9%) developed an ETR. In contrast, only 3 of the 23 patients with no detectable HCV-CTL activity developed an ETR to IFN (P <0.01). All the 10 patients who got a sustained response were those with detectable HCV-CTL activity. Conclusion The host immune response, particularly that mediated by CD8+ HCV-CTL, may be very important in determining the outcome of IFN therapy for the patients with chronic hepatitis C.

[Effect of iron deficiency anemia on the reproduction of female rats and the development of their offspring].

The effects of iron deficiency anemia(IDA) on the reproduction of female rats and the development of their offspring were studied. Four-week-old female Wistar rats were randomly divided into two groups. The rats in experimental group and control group were fed on low iron or normal diet respectively for 14 weeks, and then they were mated with normal adult male rats. The development and growth of the newborn, weanling and four-day-old offspring was observed and the trace element Fe, Zn, Cu, Mn and Se in serum were measured. IDA made notable impacts on the reproduction of female rats with small sized litters, less living fetal rats. The survival rate of female rats in the IDA group was less than that in control group. Hemoglobin and packed cell volume in blood were lower in IDA group than those in control group(P < 0.0001). The content of serum Fe and Cu in IDA group was also lower than that in control group. Iron deficiency might cause metabolism disorders of other trace element.
