Novel cationic selenium-cluster nitride species [SenN]+(n = 1-11) formed by laser ablation of a Se target in the presence of N2.

Nitride cations of selenium clusters [SenN]+ (n = 1-11) were readily produced by laser ablation of a selenium disk that was surrounded by a trace amount of nitrogen seeded in helium and followed by supersonic expansion into a high vacuum. Even at high nitrogen partial pressures, the cluster mononitride cations were found to be essentially the only nitride products in the whole size range we studied. The exception was [Se3N2]+, which is known to be a stable five-membered ring with seven pi electrons. We propose that, in the laser-ablation plasma, the selenium clusters with n > 2 take on a chain conformation, and that the N species links the two ends of the selenium chains, thus forming stable mononitride cations of the cyclic selenium clusters. Their stability is supported by the results of ab initio calculations (at both B3LYP/ 6-31 + G* and MP2/6-31 + G* levels) and of mass-selected cluster-ion photodissociation experiments.

The role of Alzheimer's disease-related presenilin 1 in intercellular adhesion.

Most cases of familial early-onset Alzheimer's disease are caused by mutations in the presenilin 1 (PS1) gene. However, the cellular functions of PS1 are unknown. We showed predominant localization of PS1 to cell-cell contacts of the plasma membrane in human prostate epithelial tissue and in a human epithelial cell line HEp2 stably transfected with an inducible PS1 construct. PS1 co-immunoprecipitated with beta-catenin from cell lysates of stable transfectants. Conversely, PS1 lacking the PS1-beta-catenin interaction site did not co-immunoprecipitate with beta-catenin and was not recruited to the cell-cell contacts. L cells, which do not form tight intercellular contacts, formed clusters of adhered cells after stable transfection with GFP-PS1 cDNA and demonstrated a clear preference for independent aggregation in the mixed cultures. However, L cells transfected with mutant GFP-PS1 constructs, which had a truncated N-terminus of PS1 or deleted PS1-beta-catenin interaction site, failed to form intercellular contacts. In addition, in primary cultures of mouse cortical neurons PS1 was highly concentrated on the surface of extended growth cones. Taken together, our results suggest an important role of PS1 in intercellular adhesion in epithelial cells and neurons.

Laboratory investigation of the migration of hydrocarbons in organobentonite.

To investigate the migration behaviors of hydrocarbons through a compacted organobentonite and to evaluate the effectiveness of this material as a liner for gasoline storage tanks, a demonstration-scale permeability test with detailed core sampling and mass balance of gasoline were developed for laboratory investigation. Results from this demonstration-scale test show that organobentonite not only effectively prevents the advective migration of hydrocarbons but also retards the transport of hydrocarbons. Due to the effect of surface tension between gasoline and water, the movement of gasoline through organobentonite is impeded by the repellence of molding water. It is also found that benzene has the highest mobility and is the least attenuated among the four hydrocarbons of interest (benzene, toluene, ethylbenzene, and xylene isomers; i.e., BTEX). The effluent concentrations of BTEX in the water phase are generally not very high. Moreover, substantial amounts of hydrocarbons, up to 150% of the dry weight of the organobentonite, can be retained in the compacted liner. This indicates that compacted organobentonite can also act as an adsorbent to attenuate organic contaminants.

A crossed molecular beam study of the O(1D) + C(3)H(8) reaction: multiple reaction pathways.

The O((1)D) + C(3)H(8) reaction has been reinvestigated using the universal crossed molecular beam method. Three reaction channels, CH(3) + C(2)H(4)OH, C(2)H(5) + CH(2)OH, and OH + C(3)H(7), have been observed. All three channels are significant in the title reaction with the C(2)H(5) formation process to be the most important, while the CH(3) formation and the OH formation channels are about equal. Product kinetic energy distributions and angular distributions have been determined for the three reaction channels observed. The oxygen-containing radicals in the CH(3) and C(2)H(5) formation pathways show forward-backward symmetric angular distribution relative to the O atom beam, while the OH product shows a clearly forward angular distribution. These results indicate that the OH formation channel seems to exhibit different dynamics from the CH(3) and C(2)H(5) channels.

Drosophila stem loop binding protein coordinates accumulation of mature histone mRNA with cell cycle progression.

Replication-associated histone genes encode the only metazoan mRNAs that lack polyA tails, ending instead in a conserved 26-nt sequence that forms a stem-loop. Most of the regulation of mammalian histone mRNA is posttranscriptional and mediated by this unique 3' end. Stem-loop-binding protein (SLBP) binds to the histone mRNA 3' end and is thought to participate in all aspects of histone mRNA metabolism, including cell cycle regulation. To examine SLBP function genetically, we have cloned the gene encoding Drosophila SLBP (dSLBP) by a yeast three-hybrid method and have isolated mutations in dSLBP. dSLBP function is required both zygotically and maternally. Strong dSLBP alleles cause zygotic lethality late in development and result in production of stable histone mRNA that accumulates in nonreplicating cells. These histone mRNAs are cytoplasmic and have polyadenylated 3' ends like other polymerase II transcripts. Hypomorphic dSLBP alleles support zygotic development but cause female sterility. Eggs from these females contain dramatically reduced levels of histone mRNA, and mutant embryos are not able to complete the syncytial embryonic cycles. This is in part because of a failure of chromosome condensation at mitosis that blocks normal anaphase. These data demonstrate that dSLBP is required in vivo for 3' end processing of histone pre-mRNA, and that this is an essential function for development. Moreover, dSLBP-dependent processing plays an important role in coupling histone mRNA production with the cell cycle.

State-to-state dynamics for O(1D)+D2-->OD+D: evidence for a collinear abstraction mechanism.

The O(1D)+D2-->OD+D reaction at two different collisional energies, 2.0 and 3.2 kcal/mol, is investigated by using the H(D) Rydberg "tagging" time-of-flight technique. Experimental results in this study indicate that the OD product is clearly more backward scattered relative to the O(1D) beam direction at the collisional energy of 3.2 kcal/mol. The extra backward scattered OD products are mostly in the highly vibrationally excited states (upsilon = 4,5,6), which is typical of a collinear abstraction mechanism. The experimental results in this work also provide a good testing bed for quantitative theoretical investigations of this benchmark system.

The cell cycle machinery and asymmetric cell division of neural progenitors in the Drosophila embryonic central nervous system.

Asymmetric cell divisions can be mediated by the preferential segregation of intrinsic cell fate determinants into one of two sibling daughters. In dividing Drosophila neural progenitors the apical-basal orientation of the mitotic spindle, the basal cortical localization of the cell fate determinants Numb and/or Prospero as well as the coordination of these events are mediated by several proteins which include Bazooka (Baz), Inscuteable (Insc) and Partner of Inscuteable (Pins) which localize as an apical cortical complex starting at interphase. Here I will summarize data which suggest that the formation of this apical complex involves two distinct steps: (1) during the initiation of apical complex formation in interphase neuroblasts, there appears to be a hierarchical relation amongst these components where Baz recruits Insc and Baz/Insc in turn recruit Pins to the apical cortex/stalk; (2) while in delaminated mitotic neuroblasts the maintenance of the apical cortical localization of these proteins is dependent on the presence of all three components. Moreover, we show that the maintenance of this apical protein complex is essential for the correct execution of asymmetric division. Finally, the localization of the various asymmetrically localized proteins shows cell cycle dependence; however, the involvement of the cell cycle regulator in asymmetric cell divisions has not been previously shown. Here we present evidence from ongoing experiments which suggest a requirement for the key cell cycle regulator cdc2 in asymmetric cell divisions.

Characterization and mutant analysis of the Drosophila sema 5c gene.

Class V semaphorins are transmembrane glycoproteins characterised by the presence of thrombospondin type I (Tsp) repeats linked to their extracellular semaphorin domain. Sema 5C is the only class V semaphorin found in Drosophila. Dsema 5C RNA is maternally provided and its embryonic expression is prominent in the mesoderm and muscle attachment sites. Here, we show that DSema 5C exists in two protein isoforms as a result of alternative splicing and that both protein and RNA have similar expression patterns. Using a combination of various molecular markers, we show that the DSema 5C protein becomes enriched in mesodermal cells that would normally give rise to fat body and visceral structures. In late embryos, DSema 5C is expressed in segment boundary cells that would constitute subsets of muscle attachment sites. Both RNA and protein are excluded from the somatic precursors and the mature muscles. The expression data suggest DSema 5C localised to the epidermal component of muscle attachment sites. Mutations in Dsema 5C were isolated from a P-element excision screen and by blotting analysis. The Dsema 5C mutants are homozygous viable and show no obvious embryonic phenotypes, suggesting that the maternal and zygotic components of Dsema 5C are not essential for fly development.

Reexpression of a cluster of silenced transgenes is associated with their rearrangement.

Irreversible inactivation or silencing of tumor suppressor genes occurs frequently in the development of cancer. A similar process of silencing can occur after the integration of transfected or microinjected genes into the genomes of recipient cells. The inactivation of transfected genes seems particularly efficient in cells with stem cell characteristics. We have been studying the inactivation of genes transfected into cultured P19 embryonal carcinoma cells and found that the CpG-rich sequence comprising the coding region of the lacZ reporter gene becomes extensively methylated after integration into the genome. 5-Aza-2'-deoxycytidine (5AdC), an inhibitor of DNA methylation, induced the reexpression of silent transgenes in one clone of P19 cells studied in detail. However, the reexpressed genes remained heavily methylated over the lacZ coding sequence. We used pulsed-field gel electrophoresis to analyze the structure of the transgenic locus in the parental and in 5AdC-treated cells and found that, in each of the cells reexpressing the transgene, the cluster of transgenes had been rearranged. Each clone had undergone a different rearrangement that appeared to involve recombination within the tandemly repeated copies of the transgene. Our data seem consistent with the idea that 5AdC induces efficient DNA recombination between tandemly repeated genes and that the reexpression of silenced genes induced by 5AdC might be triggered by the chromatin reorganization at the site of DNA recombination.

Use of weighted p-values in regional inference procedures.

Our previous studies have demonstrated that the power to detect linkage was improved by calculating a moving average of consecutive p-values in a small region as compared with testing all single p-values. The goal of this study was to test whether the power can be improved further with an alternative method whereby the middle p-values in the sequence were given more weight than the others. We also wanted to compare the moving average tests with multipoint linkage tests. The simulated extended pedigree data from the general population was analyzed to identify two major genes (MG1 and MG5) underlying two quantitative traits (Q1 and Q5). We used the variance components method implemented in the GENEHUNTER program to test for linkage of 14-marker regions each on chromosome 19 and chromosome 1 to the adjusted quantitative traits Q1 and Q5, respectively, in all 50 replicates. As before, we found that the moving average test was more powerful than a test based on single p-values. In some cases, the weighting procedure increased the power further and was similar to that of multipoint analysis, but this was not consistently found. In addition, all methods had low power and it is not possible to make a general conclusion that some weighting schemes are better than others.

Influence of light and heat on the stability of rotundine sulfate injection.

The influence of both light and heat on the stability of rotundine sulfate injection was studied. Results show that in experiments with either isothermal heating or exposure to light at high temperatures, the drug coloration rate obeys zero-order kinetics. The total rate constant, k(total), caused by both light and heat can be divided into two parts: k(total)=k(dark)+k(light), where k(dark) and k(light) are the rate constants caused by heat and light, respectively. The k(light) can be expressed as k(light)=A(light)exp(-E(a,light)/RT)E, where E is the illuminance of light, A(light) is an experimental constant related to light sources, and E(a,light) is an experiment constant. Because the form of k(light) is similar to the Arrhenius equation, it is suggested that E(a,light) might be the observed activation energy of the rate-determining step of the subsequent processes of the photochemical reaction. This viewpoint is supported by the fact that E(a,light) is independent of light sources.

Cysteine, glutathione, and Percoll treatments improve porcine oocyte maturation and fertilization in vitro.

Several factors have been examined to improve in vitro fertilization and development of porcine oocytes. Cysteine is known to be beneficial for oocyte maturation and male pronuclear formation in pigs and glutathione is known to help prevent membrane disruption of sperm in other species, including human. It has also been reported that the presence of cumulus cells influences the outcome of in vitro fertilization in cattle. The objectives of the present study were to investigate the effects of several factors involved in porcine in vitro maturation (IVM) and fertilization (IVF) procedures on oocyte embryogenic competence. The following factors were examined: the effects of different concentrations (0, 0.285, 0.57, 1.14, 2.28 microM) and exposure duration (22 and 44 hr) of cysteine during IVM, glutathione inclusion and of cumulus presence during IVF, and the use of gradient Percoll (45%/90%) during sperm preparation. The presence of cysteine in maturation medium improved blastocyst development significantly regardless of the duration of exposure when compared to the control (11--16% vs. 4%, P < 0.01). However, no dose-responsive effect was observed at the concentrations tested. The use of gradient Percoll during sperm preparation significantly improved cleavage (85% vs. 57%, P < 0.01) and blastocyst development (24% vs. 6%, P < 0.01) over conventional sperm preparation. Significant improvement was also achieved by the addition of glutathione to Percoll gradient (30% vs. 20%, P < 0.05). In conclusion, cysteine and glutathione as well as Percoll and cumulus were beneficial to embryogenic competence of porcine oocytes in this study. Mol. Reprod. Dev. 59:330-335, 2001.

Isolation of an antimicrobial compound from Impatiens balsamina L. using bioassay-guided fractionation.

By using brine shrimp (Artemia salina) lethality test-guided fractionation, a single bioactive compound (LC(50)=26 ppm) was isolated from the 95% ethanol extract of the dried aerial parts of Impatiens balsamina L. and subsequently identified as 2-methoxy-1,4-naphthoquinone (MNQ). The structure of MNQ was confirmed by UV, FT-IR, MS, and 1-and 2-D NMR spectroscopy. The antimicrobial activity of MNQ was evaluated using 12 bacterial and eight fungal strains. Five gram-positive and two gram-negative bacteria as well as all eight fungi (including multi-drug resistant strains) tested were highly sensitive to MNQ. A tea prepared according to traditional methods was found to contain sufficient MNQ to account for its antimicrobial properties.

Pregnancy outcomes of patients who conceived within 1 year after chemotherapy for gestational trophoblastic tumor: a clinical report of 22 patients.

OBJECTIVE: The aim of this study was to explore the risk of pregnancy of patients who conceived within 1 year after successful chemotherapy for gestational trophoblastic tumor (GTT). METHODS: From 1966 to 1996, 22 patients who conceived within 1 year after chemotherapy were followed up and analyzed retrospectively. RESULTS: Among 22 patients, 9 had term deliveries and 1 had a premature birth, 6 had induced abortion at the patient's request, and 6 had therapeutic abortion because of various indications such as repeated hydatidiform mole (1 case), intrauterine death (1 case), inevitable abortion (1 case), and threatened abortion (3 cases). The fetal loss rate was 27.1% (6/22). The incidence rate of gestational trophoblastic disease (GTD) was 9.1% (2/22). The incidence rate of GTT was 4.5% (1/22). The average interval between completion of chemotherapy and pregnancy was 10.25 months in the group of term pregnancies and 5.86 months in that of fetal loss (P < 0.05), indicating that the longer the interval, the lesser the risk of GTD. CONCLUSION: The results suggest that contraception for 1 year is necessary in patients with GTT after successful chemotherapy. However, in the case of a patient who conceives within 1 year, it is not necessary to terminate pregnancy, but the pregnancy must be carefully watched.

Reduced skeletal muscle calpain-10 transcript level is due to a cumulative decrease in major isoforms.

The DNA polymorphism SNP-43 in the calpain-10 gene is associated with insulin resistance and reduced skeletal muscle transcript in Pima Indians. Alternative splicing generates transcript isoforms calpain-10a to -10h. We determined the contribution of calpain-10 mRNA isoforms to the decreased total skeletal muscle calpain-10 mRNA levels observed in the G/G homozygotes. The expression levels of the major isoforms, calpain-10a and -10f, were positively correlated with the total calpain-10 mRNA levels, indicating a cumulative effect.

Size effects of realgar particles on apoptosis in a human umbilical vein endothelial cell line: ECV-304.

Regulation of endothelial cell apoptosis is a critical modulator of normal and pathological angiogenesis. Inorganic arsenic, especially arsenic trioxide, has been demonstrated to induce clinical remission in patients with acute promyelocytic leukemia as well as those with several solid cancers. So, in this study, we examined the in vitro effect of realgar(As2S2), used to treat human diseases for centuries in traditional Chinese medicine, on cell survival and apoptosis in a human umbilical vein endothelial cell (HUVEC) line: ECV-304. Four different suspensions of realgar particles with diameters from 100-500 nm containing equivalent doses were investigated to determine their size effects. After 2, 4, 6 and 8 h, realgar particles of diameters 100 nm and 150 nm obviously reduced ECV-304 cell survival, which was evidenced by a reduction of cell viability in the MTT assay, whereas cells exhibited slight viability reduction in response to treatment with realgar particles of diameters 200 nm and 500 nm. Significant apoptosis was identified by morphological observation, flow cytometry assay and DNA gel electrophoresis in the ECV-304 cells by the presence of realgar particles of diameters 100 nm and 150 nm. It is demonstrated that particles of realgar with average diameters from 100 nm to 150 nm can induce ECV-304 cell apoptosis. These results suggest that particles of realgar smaller than 150 nm may provide a less toxic agent for anti-neoplasia by suppressing angiogenesis.

Hepatitis G viral RNA co-infection in plasma and peripheral blood mononuclear cells in patients with hepatitis C.

The incidence of the co-infection of hepatitis G virus (HGV) and hepatitis C virus (HCV) and its clinical implication was investigated and the difference in the positive rate of HGV RNA and HCV RNA between plasma and peripheral blood mononuclear cells (PBMCs) observed. By using reverse transcriptase polymerase chain reaction (RT-PCR) assay, HCV-RNA and HGV-RNA in plasma and PBMCs of 72 patients with hepatitis C was detected. It was showed that HGV RNA was positive in plasma of 11 patients, in PBMCs of 15 patients, and simultaneously in both of plasma and PBMCs of 10 patients with the co-infection rate being 22.2%. Nine patients were both HGV RNA and HCV RNA positive in plasma, 11 patients were both HGV RNA and HCV RNA positive in PBMC, and 6 patients were both HGV RNA and HCV RNA positive in both plasma and PBMC with the positive rate being 12.4%, 15.3% and 8.3% respectively. The positive rate of both HGV RNA and HCV RNA in PBMCs was higher than in plasma. It was concluded that the HGV co-infection rate in the patients with hepatitis C was 22.2%. Simultaneous examination of plasma and PBMC can improve clinically detectable rate.

Repair of atrial septal defect through a minimal right vertical infra-axillary thoracotomy in a beating heart.

We present a minimally invasive and cosmetic approach to repair atrial septal defect. Fifty-five atrial septal defects were successfully repaired through a minimal right vertical infra-axillary thoracotomy. Every repair was performed on the beating heart with continuous suction through aortic needle vents and with the head of the patient lowered. This approach can be used not only in adults but also in young girls.
