Percutaneous optical imaging system to track reporter gene expression from vasculatures in vivo.

This study develops a percutaneous optical imaging system for tracking fluorescent reporter gene expression in vasculatures. We build a percutaneous optical imaging system that primarily comprised a 1.5-mm, semi-rigid, two-port optical probe. The performance of the optical probe is first tested in vitro with cell phantoms, and then the feasibility of the percutaneous optical imaging system is validated in vivo in eight femoral artery segments of two pigs. The green fluorescent protein (GFP) gene is locally delivered into four arterial segments, while saline is delivered to the four contralateral arterial segments as controls. The targeted arteries are localized using color Doppler, and thereafter the optical probe is positioned to the target arterial segments under ultrasound guidance. Optical imaging captures are obtained using different exposure times from 10 to 60 s. Subsequently, the GFP- and saline-targeted arteries are harvested for fluorescent microscopy confirmation. The percutaneous optical probe is successfully positioned at a distance approximately 2 mm from the targets in all eight arteries. The in-vivo imaging shows higher average signal intensity in GFP-treated arteries than in saline-treated arteries. This study demonstrates the potential using the percutaneous optical imaging system to monitor, in vivo, reporter gene expression from vasculatures.

Bronchial tuberculosis. Cytologic diagnosis of fiberoptic bronchoscopic brushings.

OBJECTIVE: To determine the cytomorphologic features of bronchial tuberculosis on fiberoptic bronchoscopic brushings and its diagnostic value. STUDY DESIGN: Seven hundred forty-six bronchoscopic examinations were retrospectively studied. The patients' macroscopic findings made by fiberoptic bronchoscopic examination, as well as brush cytologic and biopsy findings, were reviewed. RESULTS: Of 746 bronchoscopic brushing specimens, 23 were diagnosed as endobronchial tuberculosis by cytomorphology; that accounted for about 3.1% of cases. The same or a similar diagnosis was made for both specimen types in 16 of the specimen pairs. Acid-fast stain showed that only one brushing was positive (4.3%); all the biopsy specimens were negative. Epithelioid cell granulomas and/or caseous necrosis were found in 23 cases of brush smears, while tuberculous granulomas accounted for 16 bronchial biopsies. Seven were nonspecific chronic bronchitis or normal bronchial mucosa. CONCLUSION: The cytomorphologic alterations of tuberculosis are suitable for diagnosing bronchial tuberculosis on fiberoptic bronchoscopic brushings. Its sensitivity for defining bronchial tuberculosis is not lower than that of bronchial histologic biopsies or bacteriologic examination.

Effectiveness of a worksite nutrition education activity in a factory in China.

To evaluate the effectiveness of a worksite nutrition education activity in China, a two-group pretest-posttest nutrition education lesson was designed and carried out in two divisions (N = 240 in each division) of a steel tube factory in Chengdu, Sichuan. Special features of the program were nutrition education materials developed from both (a) the results of a pretest survey of the employees, and (b) focus group discussions conducted with factory hospital and workteam staff; a colloquial style; a slogan; and an illustrated handout depicting the new Chinese Dietary Guidelines and a proposed Food Guide. Analysis of variance, chi-square, and t-tests showed both significant increases (p < 0.05) in nutrition knowledge and attitude scores and significant improvements in dietary practices in the group receiving the education. It is concluded that the method is a useful and practical model for designing and developing worksite nutrition education in China.

Diphosphoinositol polyphosphates: the final frontier for inositide research?

The diphosphoinositol polyphosphates comprise a group of highly phosphorylated compounds which have a rapid rate of metabolic turnover through tightly-regulated kinase/phosphohydrolase substrate cycles. The phosphohydrolases occur as multiple isoforms, the expression of which is apparently carefully controlled. Cellular levels of the diphosphoinositol polyphosphates are regulated by cAMP and cGMP in a protein kinase-independent manner. These inositides can also sense a specific mode of intracellular Ca2+ pool depletion. In this review, we will argue that these are characteristics of highly significant cellular molecules.

Determination of LD-Ig complex with counter immunoelectrophoresis using terylene cellulose acetate membrane as supporting media.

OBJECTIVE: In order to determine abnormal LD isoenzyme bands in serum, we established a method of counter immunoelectrophoresis using terylene cellulose acetate as supporting media. SETTING: Department of Clinical Laboratory, Southwestern Hospital, Chongqing 400038 China. PRACTICE DESCRIPTION: LD isoenzymes were electrophoretically fractionated with a Paragon electrophoretic system. LD anomaly was identified by counter immunoelectrophoresis using terylene cellulose acetate as supporting media. MAIN OUTCOME MEASUREMENTS: Identification of the LD-Ig complex using a simple and practical method. RESULTS: An abnormal LD-4 band and an extra band on the cathodic side of LD-5 were observed in the serum of a post-burn patient and proved to be an LD-IgG complex. CONCLUSION: We have established a useful and efficient protocol that uses both isoenzyme electrophoresis and counter immunoelectrophoresis to identify abnormal isoenzyme bands. The method of counter immunoelectrophoresis we describe, using terylene cellulose acetate as supporting media, is simple and suitable for routine clinical use.

Are temporary migrants escapees of the one-child-per-family population policy: a revisit to the detachment hypothesis.

Using Hubei province as a case study, this paper retests the detachment hypothesis against the three conventional hypotheses regarding migration-fertility linkage (i.e., selectivity, disruption, and adaptation hypotheses) in explaining migrant and non-migrant fertility differentials in China. The analysis of yearly order-specific birth probabilities suggests that temporary migrants exhibit a significantly higher probability of having a second or higher order birth than comparable permanent migrants and non-migrants. This higher fertility among temporary migrants occurs after migration; temporary migrants actually do not differ from non-migrants in fertility before migration. But permanent migrants experience no significant change in their fertility after migration. The results lend a strong support to the detachment hypothesis, which best explains the fertility differentials between migrant and non-migrant populations in contemporary China; the separation of temporary migrants' actual residence from their official one does lead to a greater likelihood among temporary migrants to have unplanned births.

A computer/GPS guidance system for emergency rescue vehicle location.

The system, as built, has been able to meet all of the original design criteria. In the original rudimentary tests we were sufficiently pleased with the performance to pursue the next step, scheduling trials in the real, small-agency, rural EMS environment. The Federal Communications Commission licensing changes necessary to add the digital data communications to the agency license were probably the most time-consuming portion of the test, with a major delay caused by the turnaround time in the wait for the new license to arrive. Construction of the device to allow "plug and play" installation is possible, with no change in the internal circuitry of the agency transceivers being necessary for the initial testing. To date, all testing has yielded positive results. Tracking the test EMS vehicle throughout the service area (approximately 700 square miles) should prove immensely satisfying.

Twenty-four hour intermittent perfusion storage of the isolated rat heart: the effect of perfusion intervals on functional preservation.

We investigated the effect of intermittent perfusion intervals on heart preservation. The isolated rat heart was flushed with a cardioplegic solution, CP-11EB, and stored at 0 degrees C for 24 hours. During storage, the heart was perfused periodically at 60 mm Hg for 3 minutes with 25 degrees C oxygenated CP-11EB. The perfusion schedules were as follows: group I, every 4 hours; group II, every 6 hours; group III, every 8 hours; group IV, every 10 hours; group V, 10 and 17 hours of storage; group VI, every 11 hours. Poststorage function was assessed after 30 minutes of working reperfusion. Function of the unstored hearts including aortic flow (51.5 +/- 2.5 ml/min), coronary flow (24.5 +/- 1.3 ml/min), cardiac output (75.6 +/- 3.0 ml/min) and work (85.0 +/- 5.4 g-ml/min) served as controls. Group V hearts were best preserved with aortic flow recovered to 65%; coronary flow 44%; cardiac output 58%; and work 53% of the control. Recovery in group IV was comparable with group V. Groups I, II, III, and VI recovered significantly less than group V. Myocardial adenosine triphosphate content (micromoles per gram dry) in group V was 23.8 +/- 1.7, 20.8 +/- 0.9, 13.5 +/- 2.4, and 15.0 +/- 4.0, at 0, 10, 17, and 24 hours of storage, respectively. Intermittent perfusion at 10 and 17 hours elevated adenosine triphosphate to 99% and 88% of prestorage level. Poststorage reperfusion did not improve improve adenosine triphosphate content (16.2 +/- 2.4) over the end-storage level.(ABSTRACT TRUNCATED AT 250 WORDS)

Impact of migration on fertility in sub-Saharan Africa.

Much lower levels of fertility in urban than rural areas throughout sub-Saharan Africa imply that fertility decline in the region may be facilitated by rapid urbanization and rural-to-urban migration. The present study uses data from Demographic and Health Surveys in six countries--Ghana, Kenya, Mali, Senegal, Togo and Uganda--to assess the impact of long-term rural-urban female migration on fertility. Results of logit analyses indicate that in most countries women who leave the countryside represent the higher fertility segment of the rural population in the years before migration. Migrants' risk of conception declines dramatically in all countries around the time of migration and remains lower in the long run among most migrant groups than among rural and urban nonmigrants. Descriptive analyses suggest that the decline in migrant fertility is related to the rapid and pronounced improvement in standard of living experienced by migrants after settling in the urban area and may be due in part to temporary spousal separation.

Growth regulation of multidrug resistant ovarian cancer cells by 1D7 monoclonal antibody.

The small molecular weight protein (p7) was overexpressed in the human ovarian carcinoma cell lines, SKVLB600 (selected for vinblastine resistance from SKOV3 cells). OVCAR 4/ADR100 (selected for doxorubicin resistance from NIH:OVCAR4) and (OVCAR4/VBL200 (selected for vinblastine resistance from NIH:OVCAR4). Trace amounts of the protein were also found in the parent cell lines, SKOV3 and NIH: OVCAR4. An anti-p7 monoclonal antibody (1D7) specifically inhibited the proliferation of the drug resistant cancer cells. In order to assess the function of p7, we established a new cell line, SKVLB600R, which was maintained in drug-free medium for 16 months. This cell line stopped expressing Pgp that is responsible for its resistance to cytotoxic drugs, but continued to overexpress p7. The proliferation of SKVLB600R was also inhibited by 1D7. Our results indicate that p7 may be specifically involved in the proliferation of multidrug-resistant cells.

Physical activity in childhood and adolescence as predictor of physical activity in young adulthood.

BACKGROUND: Despite the general belief that physical activity in childhood and youth is an important prerequisite for the physical activity in adulthood, there is not much information based on reliable longitudinal studies about the continuity of physical activity from childhood and adolescence to adulthood. METHODS: As a part of a national-level research program called "Cardiovascular Risk in Young Finnus," we studied to what extent leisure-time physical activity at the age of 9, 12, 15, and 18 predicts physical activity nine and 12 years later. A total of 610 9-year-old, 624 12-year-old, 572 15-year-old, and 503 18-year-old boys and girls were studied in 1980. A follow-up measurement was carried out with the same subjects in 1983, 1986, 1989, and 1992. Accordingly, in 1992 they were 21, 24, 27, and 30 years of age. These data concern only the measurements taken in 1980, 1989, and 1992. Physical activity was measured by means of a short questionnaire. A sum index of physical activity (PAI) was computed with the help of five variables. RESULTS: The correlations between the indices derived in 1980 and 1989, and between those derived in 1980 and 1992, were, with the exception of one group, significant but low varying within a nine-year interval from .18 to .47, and within a 12-year interval from .00 to .27. Corresponding multiple correlations varied from .18 to .53 and from .18 to .30. Participation in competitive sport and the physical education grade were the best predictors of later physical activity. CONCLUSIONS: The results gave support to the conclusion that persistent participation in sport in particular increases the probability of a higher level of physical activity in later life.

Streptozotocin, an analog of N-acetylglucosamine, blocks the removal of O-GlcNAc from intracellular proteins.

Streptozotocin (STZ), an analog of N-acetylglucosamine (GlcNAc), is a specific toxin for the pancreatic beta cell. We found that treatment of rats with STZ results in an early beta-cell-specific increase in the level of intracellular protein modification by O-linked GlcNAc (O-GlcNAc). Using a model O-GlcNAc peptide based on the transcription factor Sp1, we show that treatment of cultured cells with STZ during peptide biosynthesis results in hyperglycosylation of the peptide as a result of the ability of STZ to specifically inhibit the activity of O-GlcNAc-selective N-acetyl-beta-D-glucosaminidase. Although this inhibitory activity of STZ probably can occur in all cells, we found, using in situ hybridization, that beta cells express very high levels of the mRNA encoding the enzyme responsible for cytoplasmic protein O-glycosylation, O-GlcNAc transferase (OGT). These findings suggest that the pancreatic beta cell is particularly sensitive to the toxicity of STZ because it expresses such high levels of OGT. When STZ blocks O-GlcNAc removal from intracellular proteins, the cell with the most rapid on-rate for O-GlcNAc, the beta cell, will experience the most rapid accumulation of this protein modification. Because we also show that the on-rate of O-GlcNAc is substrate driven in several cell types, we speculate that the beta cell, with its high level of OGT, may also respond to elevations of blood sugar with increased protein modification by O-GlcNAc. Thus, this proposed mechanism of STZ toxicity on the beta cell may result from an exaggeration of a heretofore unrecognized physiological response to glucose mediated through the high level of OGT in these cells.
