Protective effect of recombinant human augmenter of liver regeneration on CCl4-induced hepatitis in mice.

OBJECTIVE: To observe the anti-injury role of recombinant human augmenter of liver regeneration. METHODS: To establish liver injury models induced by CCl4 in vivo and in vitro, cell survival rates and LDH release rates served as observing index in experiments in vitro. Survival rates of liver failure animals, serum concentration of ALT, LDH, DNA and pathological examination were selected as parameters in experiment in vivo. RESULTS: rhALR could increase the survival rates and decrease the LDH release rates of injured hepatocytes in vitro. rhALR could also increase the survival rates of liver failure animal, promote hepatocyte proliferation and decrease the serum level of ALT and LDH in vivo. CONCLUSION: ALR is an important stimulator for liver regeneration and may play important role in liver damage repair.

[Study on endothelin-1 positive expression and quantitative analysis in lung cancer].

OBJECTIVE: In order to study endothelin-1 (ET-1) positive expression in lung cancer and the relationship between the ET-1 quantitative analysis and the types, grades of lung cancer. METHOD: ET-1 positive expression and quantitative analysis were detected using avidin-biotin-peroxidase complex method and image analysis technology. RESULT: The ET-1 positive expression were mainly located in the cytoplasm in 104 cases with various types of lung cancer. The positive rate of ET-1 in adenocarcinoma, squamous-cell carcinoma and large-cell-lung cancer were 71.4%, 57.1% and 40.0% respectively. The positive rate of ET-1 in small-cell-lung cancer was 21.4%, significantly lower than others. The results of image analysis on adenocarcinoma and squamous-cell carcinoma showed that the lower lung cancer differentiation, the higher ET-1 quantitative. CONCLUSION: There is ET-1 positive expression in all of types of lung cancer, but there is a high ET-1 positive expression in adenocarcinoma and squamous-cell carcinoma. The results of image analysis indicated that ET-1 quantitive expression was somehow related to the differentiation degree of neoplasm, so ET-1 quantitative analysis may be as a monitoring index of adenocarcinoma and squamous-cell carcinoma growing.

[Effect of Shenmai injection on morphological structure of kidney and peritoneum in rats with 5/6 nephrectomy].

OBJECTIVE: To study the protective effect of Shenmai injection (SMI) on kidney and peritoneum in rats with 5/6 nephrectomy. METHODS: By using cell morphological quantification, light microscopy and scanning electron microscopy, the morphological structure changes of peritoneum and kidney in rats with 5/6 nephrectomy caused chronic renal failure model after the intramuscular or intraperitoneal injection of SMI were observed. RESULTS: The SMI injection could relieve glomerular sclerosis, promote the recovery and proliferation of injured peritoneal mesothelial cell. The effect of the SMI intraperitoneal injection is better than that intramuscular of injection. CONCLUSION: The SMI has the protective effect on kidney and peritoneum in rats with 5/6 nephrectomy.

[Clinical study of puerarin in treatment of patients with unstable angina].

OBJECTIVE: To observe the clinical efficacy of puerarin in treating patients with unstable angina (UA). METHODS: Frequency of angina events, consumed doses of nitrates, heart rate, blood pressure, rest electrocardiogram (ECG) and myocardial oxygen consumption were observed before and after drug therapy in puerarin group (21 cases) and control group (18 cases) respectively. RESULTS: The puerarin group not only showed more effective than that in control group in reducing frequency of angina events and consumed doses of nitrates and improving abnormal rest ECG, but also appeared to be preferable to the control group in reducing myocardial oxygen consumption and increasing exercise duration (P < 0.05). CONCLUSION: Puerarin is a safe and effective drug in treating patients with UA.

[Interventional thrombolysis for treatment of intravascular thrombus].

OBJECTIVE: To evaluate the interventional thrombolysis for treatment of untravascular thrombus and disscuss the problems of its clinical use. METHODS: 18 patients with intravascular occlusive disease were treated with pulsed-spray thrombolysis(PST). The concentration of urokinase was 10,000 U/ml. The dose of urokinase was 10,000 U/min, the total dose was from 300,000 U to 800,000 U, and the average dose was 380,000 U. RESULTS: In 18 cases, 14 were obtained thrombolysis completely(77.8%), the cases of thrombolysis partly and unaffected were 2(11.1%) and 2(11.1%) in number respectively. The mean time of thrombolysis was 2.8 hours. CONCLUSION: The treatment of thrombolysis is highly beneficial to the patients with intravascular occlusive disease.

A comparative study of transvaginal ultrasonography and pelvic arteriogram in assessment of patients with gestational trophoblastic tumour.

OBJECTIVE: To compare the reliability of transvaginal ultrasonography with pelvic arteriography in the assessment of patients with gestational trophoblastic disease. METHODS: Transvaginal ultrasonography was performed in 24 patients with gestational trophoblastic tumour. Within one week after ultrasound investigation, pelvic arteriography was carried out in each patient. Of 24 cases, 16 patients hadn't been treated by chemical reagent, 5 had accepted 2 to 5 courses of chemotherapy, and 3 had achieved complete remission before both investigations performed. RESULTS: In 3 patients with complete remission, 2 had no evidence of abnormal findings either on transvaginal ultrasonography or on pelvic arteriography, 1 showed intramyometrial lesions by both methods. In the remaining 21 patients, all demonstrated a abnormal uterine image, and 5 of them accompanied with the finding of parametrium metastatic signs by transvaginal ultrasonography; these abnormal results were confirmed by pelvic arteriographic imaging. However, in two cases without clinical and ultrasonic signs of parametrium metastasis, pelvic arteriography indicated the early metastasis of parametrium vessels. CONCLUSIONS: Even though it is difficult to predict the early parametrium metastasis in patients with gestational trophoblastic disease by B-ultrasonic investigation, our data would support the introduction of transvaginal ultrasonography in the diagnosis and evaluation of gestational trophoblastic tumour.

[Quantitative studies of taste and fungiform papillae on the anterior human tongue].

OBJECTIVE: To study qantitatively in the taste and fungiform papillae on the anterior human tongue. METHODS: Sixty normal adults were evaluated the spatial taste and the fungiform papilla density at spatially-matched sides on the right and left sides of the anterior tongue. The taste stimulative solutions were delivered to a spatially-matched flow chamber attached to the surface of the right or left anterior tongue. RESULTS: (1) There is no statistical difference in number of the fungiform papillae, taste threshold and magnitude matching between right/left, male/female and races; (2) There is a significant difference in fungiform papilla density and taste functions in different individuals; (3) The fungiform papilla density is the key to taste function on 2/3 anterior of tongue, and is positively correlated to it; (4) The use of a spatially-matched flow chamber is a valid test system for overall, objective and accurate evaluating taste on the 2/3 anterior of tongue, and can be used to study quantitatively the fungiform papilla density. CONCLUSION: It is of clinical significance for the diagnosis of taste diseases and the dynamic studies of the taste and the fungiform papillae after lingual nerve injury and repair.

[The effect of chitosan in prevention of fibrous scar tissue formation after laminectomy].

OBJECTIVE: To study the effect of chitosan in the prevention of fibrous scar formation in the epidural space after laminectomy. METHOD: Fourty-eight SD rats were used as the animal model. Laminectomy was performed in lumber 1, and the exposed dura was covered with hyaluronate (group A), free fat (group B), chitosan (group C), and saline solution (group D) respectively. At 4, 8 and 12 weeks postoperation, scar formation was observed. RESULT: The results revealed that scar formed in the group D at the fourth week reached a peak at the eighth week and began to soften and constrict at the twelfth week. Sodium hyaluronate and chitosan had a significant efficacy in the prevention of scar formation, but the effect of chitosan was superior to that of sodium hyaluronate. CONCLUSION: The findings of this study indicate that chitosan has a preventive effect on scar tissue formation.

[Long-term outcome of selective posterior rhizotomy for spastic cerebral palsy].

OBJECTIVE: To investigate the long term outcome and complication of selective posterior rhizotomy for spastic cerebral palsy. METHOD: 26 patients with cerebral palsy who had received SPR were followed up for four years. RESULT: In the 9 patients who could walk and stand up from squatting position independently, 7 could stand steadily on one foot after operation. In the 12 who patients could walk and stand up from squatting position dependently, 7 could walk independently in the room after operation. In 5 patients couldn't walk and stand up from squatting position, 3 patients could walk dependently in the room after operation. Complications occurred in 3 patients. CONCLUSION: Selective posterior rhizotomy is effective in the treatment of spastic cerebral palsy. Strictly selecting candidates for the operation and good rehabilitation training before and after operation should be emphasized for good results.

[The surgical treatment of tetralogy of fallot in 136 adults].

OBJECTIVE: To summarize the experience of surgical treatment of tetralogy of Fallot (TOF) in adults. METHOD: From April 1975 to December 1997, 136 adult patients underwent total corrective repair of tetralogy of Fallot. The patient age ranged from 14 to 52 years old. The ventricular septal defects were closed with Dacron patch in 112 patients, with pericardial patch in 24 patients. A right ventricular outflow patch was required to eliminate right ventricular outflow tract obstruction in 126 patients. RESULT: Five patients died during hospitalization, with a hospital mortality of 3.68%. Four early postoperative deaths were due to severe low cardiac output syndrome. CONCLUSION: In view of current improved and standardized techniques, we conclude that the total corrective repair of TOF may have an excellent result in adult patients.

[The influence of changes in vocal cords' cover-body on their vibratory pattern].

OBJECTIVE: To discuss the influence of changes in vocal cords' cover-body on their vibratory pattern. METHODS: Fifty adult patients with vocal polyps were observed by telescopic videolaryngostroboscopy and detected by electroglottography (EGG). We examined the vocal cords' vibratory pattern according to laryngostroboscopic image and discussed the change of vocal cords' vibratory parameters and EGG curves. RESULTS: The vocal cords' vibratory parameters and EGG curves changed with different vocal polyps. All vocal polyps affect the regularity(REG), amplitude (AMP), glottal closure(GLO) and mucosal wave(MUC) of laryngostroboscopic image; the symmetry(SYM), REG and GLO bore linear correlation to the notches in EGG curves, the REG and MUC bore linear correlation to the steep changes; micro-polyps with pedicles reduced the vibratory parameters CQ and F0, the main abnormalities were notches in EGG curves; the changes in macro-polyps with pedicles were the same as the micropolyps except increased CQ. All vocal polyps with wide bases either single or bilateral, had increased CQ and F0, the main abnormalities were steep in contact opening phase and flat in wave peak. CONCLUSIONS: Studies with laryngostroboscopy and EGG revealed how the vocal polyps affected vocal cords' cover-body, changed the vibratory pattern of vocal cords, and factors regulating vocal cords' vibratory pattern.

[Generation of anti-rhEPO ScFv by using phage display technology].

In this paper, the recombinant phage antibody techniques was used to construct, clone, screen, and express anti-rhEPO single chain antibody ScFv. The variable region genes of antibody were amplified by PCR from a hybridoma cell line D3 which secreted monoclonal antibody to rhEPO. The ScFv gene fragments were successfully cloned into phagemid vector pCANTAB5E. The recombinant phages were panned by rhEPO which was coated on a microtiter plate. After three rounds of panning, 8 clones were determined specifically binding to rhEPO antigen. The positive recombinant phagemids were extracted and transformed into non-suppressed E. coli HB2151. The soluble single chain antibody was expressed, and the specificity of the expressed ScFv was determined by ELISA. Western blot and Dot-blot. The result of SDS-PAGE indicated that the apparent molecular weight of the target peptide which is mainly in culture supernatant is 32 kD. The DNA sequence data showed that the ScFv gene included 783 bp, encoding 261 amino acids.

[Human herpesvirus-6 infection induces IL-10 production in monocytes].

In this report, interleukin 10(IL-10) and other cytokines were evaluated through reverse transcription-polymerase chain reaction (RT-PCR) and sandwich ELISA to explore possible immune pathogenesis of human herpesvirus 6 (HHV-6) infection. The results showed that IL-10 was expressed in HHV-6 infected monocytes. Kinetic studies found that transcription of IL-1 beta, IL-6 and TNF-alpha mRNA decreased with accumulation of IL-10 mRNA. When endogenous IL-10 induced by HHV-6 was blocked using anti-human IL-10 monoclonal antibody, the production and the expression of TNF-alpha, IL-1 beta and IL-6 markedly increased as compared with infection by HHV-6 alone, indicating that endogenous IL-10 inhibited cytokine production at the transcription level in HHV-6 infected monocytes. IL-10 has been shown to suppress Th1 response and down-regulate monocyte/macrophage function. The data from our studies suggest that disturbance of immune function and latent infection resulting from HHV-6 infection might partially contribute to endogenous IL-10 production.

[Detection of dengue virus genome RNA in some kinds of animals caught from dengue fever endemic areas in Hainan Island with reverse transcription-polymerase chain reaction].

Detection of dengue virus genome RNA in brain of bats caught from dengue fever endemic areas in Hainan Island were carried out with reverse transcription-polymerase chain reaction(RT-PCR). Positive result was demonstrated in 20 of 35 bats tested, with a positive rate of 57.14%; RT-PCR was applied for the detection of dengue virus genome RNA in sera of bats caught from the endemic areas in Hainan Island, 3(16.66%) of 18 sera were positive. Dengue virus genome RNA in female Aedes aegypti captured from dengue fever endemic areas in Hainan Island was detected by using RT-PCR, 1(33.33%) of 3 lots was positive. Similar examinations on bat brains and mosquitoes captured from non-endemic areas were all negative. In addition, monoclonal IgG antibodies against types 1-4 dengue viruses were added onto brain imprints of bats captured from endemic areas of dengue fever in Hainan Island, 16(80.00%) of 20 showed positive results for type 2 dengue virus antigen by direct immunofluorescent assay. The antibodies to various types of dengue virus were coincided to that of dengue virus genome RNA. The above results proved that bat is the reservoir of dengue virus, and this provides an important clue for the effective control of dengue fever epidemics in endemic areas.

Prejunctional 5-HT receptors enhance cholinergic neurosecretion in the rabbit and human iris-ciliary body.

PURPOSE: To characterize prejunctional 5-HT heteroreceptors which modulate 3Hacetylcholine (3H-ACh release) in isolated rabbit and human iris-ciliary bodies (ICB3). METHODS: ICB tissue segments were incubated with 3H-choline, superfused and electrically stimulated four times (S1, S2, S3, S4) at 3-10 Hz for 1 min to elicit 3H-ACh. secretion. Test agents (5-HT agonists and antagonists) were added before S2, S3 and S4 and their effects determined by the stimulation ratio (Sx/S1) of evoked 3H-ACh. release. 3H-ACh in superfusate fractions was fractionated and quantified by ion exchange chromatography. RESULTS: In rabbit ICBs, evoked 3H-ACh. release was enhanced in a concentration-dependent manner by 5-HT (10(-9)-10(-5) M, EC50 = 5.8 x 10(-8) M). The maximum effect of 5-HT (10(-6) M) corresponded to a 45.14 +/- 7.40%) (n = 6) increase in 3H-ACh release. Higher concentrations of 5-HT (> 10(-4) M) induced desensitization. The response to 5-HT (10(-6) M) in the presence of the 5-HT3/4 antagonist tropisetron (10(-9) M), 5-HT1 antagonist methiothepin ((10(-7) M), equated to 89.96% and 88.78% respectively of control value; in the presence of non-selective 5-HT antagonist mianserin, 5-HT4 antagonist SDZ-205557 (10(-7) M) was -15.20% and 32.84% of control. The 5-HT response was mimicked by the selective 5-HT4 agonist 5-methoxytryptamine (10(-9)-10(-4) M, EC50 = 7 x 10(-8) M), whereas the 5-HT3 agonist M-CPBG[(1-m-chlorophenyl)-biguanide] and phenylbiguanide, the 5-HT1 agonist 5-carboxamidotryptamine and the 5-HT2A agonist alpha-methyl-5-HT were ineffective. The selective 5-HT1A agonist (+)-8-OH-DPAT (10(-8)-10(-5) M) had no significant effect, but at concentration of 10(-4) M, inhibited evoked 3H-Ach. release. Similar results were obtained in the human ICB. The evoked 3H-Ach. release was enhanced in a concentration-dependent manner by 5-HT (10(-9)-10(-5) M, EC50 = 3.36 x 10(-8) M) or 5-MOT (10(-8)-10(-5) M, EC50 = 6.59 x 10(-7) M), The selective 5-HT4 antagonist, GR113808A (10(-8) M) inhibited the 5-HT-inducing increase of the cholinergic response, producing parallel right shits of the concentration-response curves to 5-HT. The selective 5-HT3 antagonist ondersetrone (5 x 10(-7) M) and tropisetron (10(-9) M) did not affect 5-HT inducing 3H-Ach release. CONCLUSIONS: Our results indicate that cholinergic terminals in the rabbit and human ICB contain facilitatory 5-HT heteroreceptors that belong to the 5-HT4 subtype. Their role in modulation of intraocular pressure remains to be elucidated.
