Current source density (CSD) analysis of retinal field potentials. I. Methodological considerations and depth profiles.

1. The technique of current source density (CSD) analysis was used to obtain depth profiles of light-evoked source/sink distributions in the retina of the frog. 2. The effects of a number of methodological considerations on the CSD profiles were explored. Adoption of best technique leads to minimal variability and noise in the CSD profiles. Several situations that give rise to artifactual source/sinks were identified. 3. At the time of the b-wave peak, there is a large sink near the outer plexiform layer (OPL), a source at the inner limiting membrane (ILM), and a complex response at the inner plexiform layer (IPL). 4. The IPL response at light onset consists of 1) an initial, sharp sink, followed by 2) a slower source with a time course similar to the b-wave, and last 3) an even slower sink with a time course similar to the M-wave. 5. The first component of the IPL response is the current sink for the proximal negative response (PNR) elicited by a diffuse light stimulus. The PNR has current sources both proximal and distal to this layer. 6. The OPL sink underlies the b-wave. The second component of the IPL response is a source for the b-wave. Part of the ILM source also might be a b-wave source. 7. A large part of the ILM source is the current source for the M-wave. The third component of the IPL response is the M-wave sink, but it is small because it subtractively interacts with the IPL b-wave source.

Current source density analysis of retinal field potentials. II. Pharmacological analysis of the b-wave and M-wave.

1. The actions of two pharmacological agents, barium ions (Ba2+) and picrotoxin (PTX), were examined on components of the electroretinogram (ERG) in frog retina. Depth profiles of light-evoked field potentials were recorded, and current source densities (CSDs) were computed from these. 2. Ba2+ abolished the M-wave, slow PIII, and the c-wave, but only decreased b-wave amplitude down to approximately 65% of control amplitude. 3. Ba2+ abolished a slow current sink in the inner plexiform layer (IPL) and the source at the inner limiting membrane (ILM). This IPL sink/ILM source appears to generate the M-wave. 4. Ba2+ decreased the current sink at the outer plexiform layer (OPL) to approximately 70% of control amplitude, and it increased an IPL source. This Ba(2+)-resistant OPL sink/IPL source appears to generate a significant portion of the b-wave. The Ba(2+)-sensitive portion of the b-wave might be generated by Müller cells. 5. PTX enhanced retinal field potentials, particularly the M-wave in the proximal retina. This enhanced M-wave was shown to originate from an enhanced IPL sink/ILM source. 6. Our results suggest that the M-wave originates from Müller cells, through the spatial buffering of the light-evoked increase in [K+]o of the proximal retina. A portion of the b-wave may also originate from Müller cells, but a stronger direct contribution from depolarizing bipolar cells is suggested.

Role of nitric oxide in the regulation of oxygen consumption in conscious dogs.

The role of nitric oxide (NO) in the regulation of O2 consumption was studied in chronically instrumented conscious dogs. A specific NO synthesis inhibitor, nitro-L-arginine (NLA, 30 mg/kg i.v.), significantly increased mean arterial pressure from 100 +/- 4 to 134 +/- 5 mm Hg (mean +/- SEM) and total peripheral resistance by 157 +/- 16% and reduced cardiac output by 47 +/- 3% and heart rate by 34 +/- 6% after 120 minutes. Changes in arterial blood gases were not observed. There were significant changes in PO2 (-14 +/- 2 mm Hg), O2 saturation (-21 +/- 2%), the percentage of hemoglobin as oxyhemoglobin (-21 +/- 2%), and O2 content (-3.0 +/- 0.9 vol%) and a significant increase in percent reduced hemoglobin (21 +/- 1%) in mixed venous blood, associated with an increase in O2 extraction (5.1 +/- 0.2 vol%) (all P < .01). O2 consumption was increased from 124 +/- 6 to 155 +/- 9 mL/min (P < .05). Methoxamine, titrated to have hemodynamic effects similar to those of NLA (eg, mean arterial pressure increased from 97 +/- 4 to 131 +/- 5 mm Hg), had much smaller effects on venous blood gases, hemoglobin, and O2 extraction (2.3 +/- 0.7 vol%) and no significant effect on O2 consumption. NLA also caused an increase in O2 consumption of 37 +/- 8% (P < .01) in quietly resting conscious dogs that had undergone pretreatment with hexamethonium and atropine, but no significant change in O2 consumption in dogs anesthetized with barbiturate.(ABSTRACT TRUNCATED AT 250 WORDS)

Age estimation by Chinese permanent teeth with image analysis.

A total of 567 human teeth except third molars were studied to estimate age, using a combined method of multiple regression analysis of data from a scoring system, direct measurements with image analyser, and logarithmic transformation. As a result, the standard errors of estimate were 4.8-5.4 years in the working sample, and 2.9-5.4 years in the control sample respectively, by using a single tooth. By testing the formulae on forensic autopsy cases in Guang Dong and Guang Xi, (People's Republic of China), it is demonstrated that the standard error of estimate is 3.7 and 2.6 years respectively for any single tooth (incisor, canine, premolar or molar) except the third molar. Age determination from multiple teeth of the same individual has also been studied. As a result, according to the different age grouping, the standard errors of estimate ranged from 0.8 to 2.6 years (2 teeth from the same individual) and 0.6 to 2.3 years (3 teeth from the same individual). It is concluded that the method presented here is helpful in improving the accuracy of age determination from teeth.

Control of communicable diseases in the People's Republic of China.

Communicable diseases were rampant in old China. Since the new China was founded, the Government has assigned high priority to the prevention of infectious diseases. A series of measures were adopted, such as instituting health policies in the vein of "Prevention First" as well as health laws and regulations, coordinating department participation and setting up the network of prevention and control of diseases. The incidence of leading infectious diseases has declined dramatically. Although great success has been achieved, infectious diseases still form one of the major public health problems in China. There are some existing problems which hinder the progress of infectious disease control. This situation is being addressed by the government and the slogan "Health for All in 2000" has become the focus of the nation.

Gender difference in smoking effects on adult pulmonary function.

Data on 1,618 male and 1,669 female adults aged 40-69 yrs, from China in the Beijing Respiratory Health Study, were analysed to investigate the gender differences in effects of smoking on pulmonary function. Smoking was characterized by total smoking-years, smoking status (former, transitional and constant), smoking type (cigarette, cigar and others). The effects of smoking on height-standardized forced vital capacity (FVC) and forced expiratory volume in one second (FEV1) were assessed by multiple regressions adjusting for age, education level, use of an indoor coal stove for heating, passive smoking, occupational dust and gas/fume exposure, and residence. Prediction equations were derived from nonsmoking asymptomatic subjects. As compared to women, men had a much higher smoking prevalence (78 vs 35%) but a lower quitting rate (14 vs 23%). Female lifetime nonsmokers had greater mean percentage predicted lung function values than male lifetime nonsmokers, whilst female cigarette smokers had lower values than their male counterparts. In both sexes, the highest mean percentage predicted lung function values were found in lifetime nonsmokers, whilst the lowest values were found among former smokers, the second lowest among transitional smokers, and constant smokers actually had greater values than both former and transitional smokers. These findings were confirmed by sex-specific regression analyses. A global test on the interactions between smoking and sex was highly significant. This study suggests that adverse smoking effects on pulmonary function were greater in women than in men.(ABSTRACT TRUNCATED AT 250 WORDS)

A maternal homeobox gene, Bombyx caudal, forms both mRNA and protein concentration gradients spanning anteroposterior axis during gastrulation.

We have isolated a caudal (cad) homologue from a cDNA library of Bombyx mori embryos. The Bombyx cad cDNA encodes a protein of 244 amino acids. The homology between Drosophila and Bombyx homeodomains is 80%. Similar to Drosophila cad, there is no YPWM peptide sequence along the upstream of homeodomain. Northern blot hybridization with a Bombyx cad probe revealed the presence of single maternal transcript of 2.3 kb. A stronger signal of the transcripts was detected in unfertilized eggs and in eggs up to 36 hours after deposition. The transcripts decreased rapidly by 2 days and a weak signal was maintained until hatching. To analyse its spatial expression pattern, we have established a novel frozen sectioning method for in situ hybridization and immunohistochemistry experiments. The results showed that Bombyx cad transcripts accumulated first in the nurse cells and transferred into the oocyte at a defined time during oogenesis. The maternal transcripts of Bombyx cad formed a concentration gradient spanning the anteroposterior axis during the gastrulation stage and were restricted to the anal pad, the most posterior domain, after 2 days of embryogenesis; the Drosophila cad mRNA revealed the corresponding expression profile during the syncytial blastoderm stage. The Bombyx cad protein was not detected in the ovary and the first 9 hours of eggs, but was first detected evenly during cellular blastoderm stage. During gastrulation, Bombyx cad protein concentration gradients shifted along the anteroposterior axis coinciding with the shifting of the mRNA concentration gradients.(ABSTRACT TRUNCATED AT 250 WORDS)

Estradiol regulation of mRNA expression of stimulatory G-protein alpha-subunit in white adipose tissue from female rats.

Adipose tissue has been recognized as a major peripheral metabolic target of estrogens. The present study was addressed to examine in female rats whether differences in the adipose tissue mRNA expression of alpha-subunit of stimulatory (Gs) and/or inhibitory (Gi) G-proteins exist between intact and ovariectomized rats, the latter with or without estradiol or testosterone treatment. The fat cell membrane protein amount of Gs and Gi alpha-subunit also was examined. All these parameters were evaluated in parametrial fat tissue samples obtained from 40 female Sprague-Dawley rats. A group of rats (N = 20) was investigated for evaluation of mRNA expression and another group (N = 20) for quantification of the protein amount of Gs and Gi alpha-subunit. Each group was represented by five control rats (sham-operated), five ovariectomized (OVX) rats, five ovariectomized rats treated with estradiol (OVXE) and five ovariectomized rats treated with testosterone (OVXT). Ribonucleic acid extracted from adipose tissue and analyzed by northern blot with G alpha s, G alpha i-3 cRNA probes revealed three major bands with estimated sizes of 1.9, 3.5 and 2.35 kb, respectively. Messenger RNA quantitative analysis, by a solution of hybridization RNAase protection assay on total nucleic acid samples, showed that the amount of G alpha i-1 and G alpha i-2 mRNA was similar within the different groups, whereas the G alpha s mRNA was significantly less abundant (p < 0.01) in OVX and OVXT rats than in control or OVXE rats. No difference in G alpha s mRNA content was found between control and OVXE rats.(ABSTRACT TRUNCATED AT 250 WORDS)

[Study on the apical sealability of warm lateral condensation techniques]

An apical leakage test and a visible light spectrophotmetric analysis were performed to compare the apical sealability of warm lateral condensation and conventional lateral condensation techniques. The results showed that warm lateral with sealer delivered best sealability. It was superior to warm lateral without sealer(P<0.05) and conventional lateral condensation technique(P<0.01).Because of its inconvenient operation causing by native-made putta-percha,it is difficult to use warm lateral condensation technique widely in our country.

Microprocessor-programmed infusion of theophylline rapidly attained expected steady-state level in rabbit plasma.

A self-made microprocessor-programmed (two-compartmental model) infusion controller was connected with an infusion pump, which achieved an expected steady-state plasma concentration (Cpss) rapidly (5 T1(2)alpha) and maintained the level. Theophylline was selected as an example, and its pharmacokinetic parameters of rabbits, expected Cpss, body weight (wt), and infusion time (t) were input. The programmed infusion rate (Kt) was determined by the following equation: (Kt) = Cpss.K10.Vc.wt (1 + [(K21-beta)/beta]EXP(-K21t)) and the predicted value was calculated by the formula: C(t) = Cpss X [1-EXP(-alpha t)]. The needed concentration and total volume of drug were automatically shown on the screen. The drug was automatically infused after pumping, and the plasma concentration of theophylline was measured by colorimetric method. The results showed that the median absolute value of the performance error (MAVPE) was 8.3%. Although T1(2)beta of theophylline was 6.08 h, the expected Cpss was attained in only 30 min (5 T1(2)alpha) after start of infusion and then well maintained.

[Mechanism of changes in coagulation after electric burns in rabbits].

The changes in local lesion after high-voltage electric burns ares dependent on many factors. One of the important factors is the change in coagulation. In this paper, a high-voltage electrical burn model was reproduced in rabbits for the purpose of investigating the coagulation mechanism. It was found that AT-III and PC showed elevation and declination, respectively, at 6h and 12h postburn. PAI:A and t-PA:A also showed obvious changes, while FDP showed continuous elevation at 6h up to 5d postburn. The findings probable provided the explanation of the occurrence of thrombosis at 6h to 12h postburn, and the second episode of thrombosis on the 5th day.

[An approach for PCR amplification of long DNA fragments].

The polymerase chain reaction (PCR) technique, evolved as a standard laboratory technique, has a major limitation: its inability to efficiently amplify fragments higher than 1.5 kb. In this paper, PCR conditions allowing the efficient amplification of long DNA fragments, especially a new proper buffer system, have been developed. The difference between the new PCR reaction buffer system and standard reaction system is that no KCl is included in the former. The results indicated that specificity and reproducibility for amplifying long DNA fragments by using new buffer system are satisfactory.

[Effect of indirect fluorescent antibody test for the diagnosis of amoebiasis].

Indirect fluorescent antibody test (IFAT) was performed by the method of [symbol: see text] (1971) with some slight modifications for the diagnosis of amoebiasis. The antigen used was prepared from 48-hour culture of Entamoeba histolytica (strains G2, G3, and G5) in LAS diphasic medium and only those amoebae clinging the culture were collected. The serum samples or blood drops on filter paper were positive by the improved IFAT in all of the 54 cases of amoebic liver abscess and the positive reactions of most cases were very marked. Of 23 cases of acute amoebic dysentery, 19 were positive, the positive rate being 82.6%. The results also show that 42 cases of other diseases and 40 healthy persons were all negative.

[An experimental model of electric injury by high voltage].

A standardized 10,000-volt, 2 + 0.15-A, 5-s electric burn rabbit model was established. Microscopic examination revealed that there was patchy cellular necrosis intermingled with viable cells in the muscle in the burn area. Grossly, there was a distinct interface between viable and nonviable tissues. The resulted injury correlated well with clinical features of electric burn.

Anastomotic false aneurysm following abdominal aortic aneurysmectomy and prosthetic grafting.

Anastomotic false aneurysm (AFA) of the aorta is a potentially lethal complication after prosthetic grafting. Nineteen aneurysms were encountered in 18 patients within a 30-year period (1960-1991). There were 10 men and 8 women, aged 27-80 years (mean 58 years). In 14 patients, the prostheses were made of silk, in 2 were PTFE, and in 1 each Dacron or silk-Dacron cross-weaved. Patients with an intact AFA had a pulsatile abdominal mass, abdominal pain, an occluded graft and peripheral emboli. Five patients were asymptomatic. Clinical onset of AFA varied from 2 weeks to 12 years (mean 5 years). The accurate rate of diagnosis of single plane angiography was 75% (3 of 4), and computed tomography 100% (ten of ten). Ultrasound was used only once and suggested an AFA. Four AFAs were less than 5cm in diameter. Five patients refused operation and died in 2 years from rupture. Operative mortality was 11% (1 of 9). Treatment was resection of AFA and replacement with a new graft. Life-long follow-up is required for patients with an aortic aneurysm. All ratroperitoneal AFAs should be resected, since the outcome of rupture is poor.

[The effect of Sindbis virus multiplication on gene expression of host cells].

Sindbis virus (SBV) infection mediated a rapid shutoff of host cellular gene expression (mRNA synthesis and protein synthesis); however the synthesis of cellular rRNA remained at the same level as the uninfected cells. Meanwhile a cellular protein P105 was shown to be enriched in the nuclear matrix. Actionmycin D treatment after virus infection resulted in an apparent reduce in the production of viral structural proteins and infectious virions. The results presented here not only demonstrated the complexity of SBV-mediated regulation of host gene expression, but also suggested SBV nonstructural protein nsP2 and capsid protein C were possibly involved in this process.
