Constitutive activation of different Jak tyrosine kinases in human T cell leukemia virus type 1 (HTLV-1) tax protein or virus-transformed cells.

HTLV-1 infection causes an adult T cell leukemia in humans. The viral encoded protein tax, is thought to play an important role in oncogenesis. Our previous data obtained from a tax transgenic mouse model revealed that tax transforms mouse fibroblasts but not thymocytes, despite comparable levels of tax expression in both tissues. Constitutive tyrosine phosphorylation of a 130-kD protein(s) was observed in the tax transformed fibroblast B line and in HTLV-1 transformed human lymphoid lines, but not in thymocytes from Thy-tax transgenic mice. Phosphotyrosine immunoprecipitation followed by Western blot analysis with a set of Jak kinase specific antibodies, identified p130 as Jak2 in the tax transformed mouse fibroblastic cell line and Jak3 in HTLV-1 transformed human T cell lines. Phosphorylation of Jak2 in tax transformed cells resulted from high expression of IL-6. Tyrosine phosphorylation of this protein could also be induced in Balb/c3T3 cells using a supernatant from the B line, which was associated with induction of cell proliferation. Both phosphorylation and proliferation were inhibited by IL-6 neutralizing antibodies. Constitutive phosphorylation of Jak kinases may facilitate tumor growth in both HTLV-1 infected human T cells and the transgenic mouse model.

Occupational Stress and Dysmenorrhea in Women Working in Cotton Textile Mills.

The authors conducted a cross-sectional study of 895 never-smoking female textile workers, aged 20-40 years, employed in three cotton textile mills in Anhui Province, China, to assess the association of self-reported occupational stress with dysmenorrhea. Stress was assessed by means of a questionnaire incorporated into a larger, general health survey of textile workers. Dysmenorrhea was defined as abdominal/pelvic pain during menses. Proportions of no/low, moderate, and high levels of occupationally-related emotional stress among all the subjects were 56%, 23%, and 21%, respectively. The overall prevalence of dysmenorrhea in the population was 59.7%. The adjusted odds ratios of dysmenorrhea for women who had moderate and high levels of occupational stress relative to those with low levels were 1.6 (95% CI:1.1-2.2) and 2.3 (95% CI:1.6-3.4), suggesting an exposure-response relationship. The estimated odds ratio based on assigned scores (0, 1, and 2 assigned for no/low, moderate, and high degrees of occupational stress) was 1.5 (95% CI:1.3-1.8). In the analyses stratified by the mills and the women's job titles, a positive association was found in all groups. The association remained significant (OR = 1.6, 95% CI:1.3-2.0) when the analysis was restricted to those women with only one reported livebirth pregnancy. The findings suggest that high degrees of occupational stress predispose female textile workers to dysmenorrhea.

Decreases in growth hormone receptor signal transduction contribute to the decline in insulin-like growth factor I gene expression with age.

Several investigations have clearly indicated that plasma concentrations of insulin-like growth factor I (IGF-I) decrease with age and contribute to the decrease in tissue function that is characteristic of aging animals and man. Plasma IGF-I is regulated by GH released from the pituitary gland, and although data demonstrate a decline in GH secretion with age, GH receptor (GHR) density in liver tissue has been reported to increase. In this study, the effects of aging on GHR signal transduction were assessed in hepatic tissue to determine whether alterations in the response to GH contribute to the decline in IGF-I. Liver slices from female C57BL/6 mice (10, 17, and 31 months old) were prepared in medium and stimulated with GH. Basal GHR binding increased more than 2-fold in 31-month-old animals compared to that in either 10- or 17-month-old animals (P < 0.01), whereas the Ka values were similar in the three age groups. However, GH (2 nM)-induced IGF-I gene expression decreased dramatically with age (P < 0.01). In 10-month-old animals, GH-induced phosphorylation of the GHR complex was maximal 10 min after the addition of hormone, whereas GH-induced MAP kinase activity was maximal at 15 min. GH-induced JAK2 kinase and GHR complex phosphorylation as well as MAP kinase activity were significantly lower in 31-month-old animals than in either the 10- or 17-month-old groups (P < 0.05). The results of this study demonstrate that GH induces phosphorylation of JAK2 and the GHR complex, activates MAP kinase, and increases the expression of IGF-I messenger RNA in liver. In 17-month-old animals, decreases in IGF-I gene expression were evident that were not directly associated with diminished GHR complex phosphorylation or MAP kinase activity. By 31 months, there was a decrease in IGF-I gene expression that was associated with a marked decline in JAK2 and GHR complex phosphorylation. These data suggest that the signal transduction pathway for GH is impaired with age and that these changes may contribute to the decline in IGF-I gene expression.

Air pollution and unscheduled hospital outpatient and emergency room visits.

We conducted a time-series analysis of daily hospital visits and air pollution data to assess acute effects of air pollution on daily unscheduled outpatient visits to internal medicine, pediatric, and emergency departments in the No. 3 Affiliated Hospital of Beijing Medical University in Beijing, China. Sulfur dioxide was marginally significantly associated with total outpatient visits (beta = 41.5, SE = 24.2) and significantly associated with internal medicine (beta = 14.6, SE = 6.7), pediatric (beta = 12.7, SE = 3.7), and emergency room visits (beta = 6.8, SE = 2.7). Total suspended particulates (TSP) was a significant predictor for total outpatient (beta = 21.1, SE = 7.7) and pediatric visits (beta = 3.4, SE = 1.3) and a marginally significant predictor of internal medicine visits (beta = 4.2, SE = 2.2). In a season-specific analysis, SO2 was a significant predictor for total hospital outpatient visits in summer, although the mean daily SO2 concentration was only 17 micrograms/m3 (maximum = 51 micrograms/m3). In winter, SO2 was significantly associated with internal medicine, pediatric, and emergency room visits, and TSP was associated with total outpatient visits. This study suggests an exposure-response relationship between TSP and SO2 and hospital outpatient visits, both at high air pollution levels and at levels well below air quality standards recommended by the World Health Organization.

Comparing FEV1 in adults in two community-based studies.

Data collected in two large-scale longitudinal respiratory studies were analyzed to estimate age-related changes in FEV1 and to determine the between-study differences in change in FEV1 among nonsmoking participants. Two adult cohorts in the Netherlands were followed up for 21 years at 3-year intervals and a cohort of adults in 6 cities in the eastern United States was followed up for 6 years. The age distributions and follow-up rate differed for the two studies. The mean rates of decline in FEV1 for 50-year old male and female never smokers were 27.9 +/- 2.8 mL/yr and 22.1 +/- 0.9 mL/yr, respectively, in the Netherlands cohort. The estimates for men and women in the Six Cities cohort were 33% and 32% greater, respectively, than those for the Netherlands cohort (p < 0.01). The estimated rate of decline in FEV1 associated with each 1-year increase in age was 0.50 +/- 0.12 mL/yr for men and 0.61 +/- 0.08 mL/yr for women in the Netherlands cohort. This estimate was comparable for the Six Cities women and the Netherlands women but significantly different for the two cohorts of men. Inclusion of survey (period) effect in the regression model had only a trivial influence on accelerated rate of decline in FEV1 and did not alter the more rapid acceleration of decline in FEV1 among the Six Cities men. The present study also showed that use of a "prediction equation" for age adjustment to compare across population samples may produce a biased conclusion if the estimated rate of decline in FEV1 associated with age is different and the age distributions are different for the two populations. In addition, height adjustment for FEV1 was found to have a negligible influence on the longitudinal estimates of FEV1 decline in both study samples. In conclusion, substantial between study variation in decline in FEV1 occurs despite use of similar data analysis techniques. Differences in the initial age distributions and loss to follow-up appeared to be important factors in this analysis.

Additive effects of growth hormone and testosterone on lipolysis in adipocytes of hypophysectomized rats.

The effects of growth hormone (GH) and testosterone, alone or in combination, on the regulation of lipolysis in isolated adipocytes from hypophysectomized rats were investigated. Male Sprague-Dawley rats were hypophysectomized at 50 days of age. One week after operation, hormonal replacement therapy with L-thyroxine and hydrocortisone acetate was given to hypophysectomized rats. Groups of rats were treated with GH (1.33 mg/kg, daily), testosterone (10 mg/kg, once) alone or in combination. After one week of hormonal treatment, adipocytes were isolated from the pooled epididymal and perirenal fat pads and glycerol release after isoproterenol stimulation and 125I-cyanopindolol binding was measured. Hypophysectomy caused a marked decrease in basal and isoproterenol-stimulated lipolysis. There was no effect of testosterone treatment alone on lipolysis, but GH treatment resulted in an increase in isoproterenol-induced lipolysis but not to the levels observed in cells from control rats. Testosterone and GH in combination restored the lipolytic response to isoproterenol. Also 125I-cyanopindolol binding was decreased after hypophysectomy. Testosterone treatment alone and GH treatment alone increased the binding, while in combination the treatment had an additive effect. Affinity was not changed, but the effects seemed to be on receptor number, as determined by Scatchard analysis. Forskolin-stimulated cAMP accumulation in adipocytes was markedly reduced after hypophysectomy. Testosterone treatment alone had no effect. GH treatment alone increased forskolin-stimulated cAMP accumulation, although the level was lower than that found in control rats. The combined treatment resulted in a further increase to levels observed in adipocytes from control rats.(ABSTRACT TRUNCATED AT 250 WORDS)

An improved immunolabeling method for microtubular cytoskeleton in poplar (Populus nigra L.) free nuclear endosperm.

We investigated immunocytochemical staining of microtubular cytoskeleton of free nuclear endosperm, a tissue which is particularly difficult to fix. This tissue requires fixation for 45 hr to preserve the integrity of the microtubular network after paraformaldehyde based fixation. Low glutaraldehyde concentration in the fixative and the ethanol dehydration retains beta-tubulin antigenicity and the former improves preservation of tissue structure. An ethanol-free embedding method is recommended for immunocytochemical studies of ethanol sensitive target proteins.

Studies on kinetics of substrate utilization of hydrogen production from wastewater with immobilized cells of photosynthetic bacteria.

The kinetic characteristics of substrate utilization by immobilized cells of Rhodopseudomonas capsulata 386 and Rhodopseudomonas sp. D for H2 production was investigated. The results showed that substrate utilization did not proceed simultaneously with H2 evolution, H2 producing capacity of immobilized cells with agar was higher than that of alginate immobilized cells, but the appearance of maximum H2 producing activity was later than the last one. The kinetics of substrate utilization (glucose) by immobilized cells of strain D followed the first-order reaction, rate constant k value was 1.2 x 10(-2) L/h, analysis of macrokinetics indicated that substrate utilization by immobilized cells in H2 evolution process was governed by biochemical reaction other than diffusion transfer limitation, because of the Thiele modulus were 0.125 and 0.154 for immobilized cells with D = 3.6 mm and 4.4 mm particles, the corresponding effective factor were determined as 0.998 and 0.988, respectively. An immobilized cell bioreactor, fed by glucose and lactate, was employed for continuous H2 evolving system. It was found that hydrogen production were 0.659 L/d and 0.477 L/d in immobilized cells system of strain 386 and D when lactate used as H2 evolving substrate, while the volumetric H2 rate were all up to 1.0 L/L.d.

Expression of human plasminogen activator inhibitor-2 in the baculovirus expression system.

A cDNA coding for human plasminogen activator inhibitor-2 (PAI-2) was transferred into the genome of Autographa californica nuclear polyhedrosis virus adjacent to the polyhedrin promoter. Cells infected with recombinant virus synthesized a 43-kDa PAI-2 protein, the majority of which was secreted. The recombinant PAI-2 was identical to native PAI-2 purified from human placenta with respect to interaction with polyclonal antibody and inhibition of urokinase-type plasminogen activator.

Expression of functional insulin-like growth factor-1 receptor on lymphoid cell subsets of rats.

It has become evident that insulin-like growth factor-1 (IGF-1) acts as a growth factor for immune cells, yet the precise regulatory role of IGF-1 in the immune system is unknown. The aim of this study was to examine the distribution of IGF-1 receptors on rat lymphoid cells. A flow cytometric method was used, with a biotinylated and functionally active IGF-1 analogue, namely des(1-3)IGF-1, which binds well to IGF-1 receptor but poorly to IGF binding proteins, followed by phycoerythrin-conjugated streptavidin (PE-SA) staining. Our results showed that IGF-1 receptors were readily detectable on a wide variety of the immune cells, including T cells, B cells and monocytes, but the binding capacity for IGF-1 was monocytes > B cells > T cells, as determined by titration experiments. Furthermore, the level of expression on resting CD4+ T lymphocytes was greater than on CD8+ cells, and the concentration of biotin-des(1-3)IGF-1 required to demonstrate the binding to IGF-1 receptor on CD8+ cells (68 nmol/l) was 200-fold higher than for CD4+ cells (0.34 nmol/l), indicating that most of the IGF-1 receptor on CD8+ cells represented lower affinity sites. The level of IGF-1 receptor expression was increased several-fold after concanavalin A stimulation on both CD4+ and CD8+ T-cell subsets. Kinetic analysis of the expression of IGF-1 receptor and its association with interleukin-2 receptors (IL-2R) following activation showed a similar pattern, with no significant differences in the ratio of IGF-1 receptor: IL-2R per cell during the 3 days of cell culture. Our studies suggest that biological activities of IGF-1 include direct stimulation of immune cells, and that expression of IGF-1 receptor may have a role in regulation of T-cell function.

[Clinical and experimental study on codonopsis pilosula oral liquor in treating coronary heart disease with blood stasis].

The effects of Codonopsis pilosula oral liquor (CPOL) on tissue-type plasminogen activator (t-PA) and plasminogen activator inhibitor (PAI) in the plasma of 25 patients of coronary heart disease with blood stasis were studied. It has been shown that those patients had a significant decrease in t-PA activity (P < 0.01) compared to healthy subjects. After 4 weeks of CPOL therapy, platelet aggregation significantly decreased (P < 0.05), there was no significant difference in t-PA and PAI. From the results shown above, it suggested that one of the effects of CPOL in influencing blood coagulation was its inhibition on platelet aggregation, but not through the elevating of fibrinolytic activity.

Comparison of four methods to generate immunoreactive fragments of a murine monoclonal antibody OC859 against human ovarian epithelial cancer antigen.

In the present study, four different proteases (pepsin, papain, bromelain and ficin) were screened with a murine monoclonal antibody OC859, in order to verify whether different digestion procedures could improve yield and stability of the F(ab')2 or Fab fragments. The yields of F(ab')2 or Fab fragments from digestion with pepsin, papain, bromelain and ficin were respectively 20.3 +/- 2.0%, 50.5 +/- 5.0%, 74.4 +/- 2.7% and 82.8 +/- 10.2% of the theoretical maximum. Immunoreactivity in a noncompetitive solid-phase radioimmunoassay (SPRIA) of the fragments generated by the four proteases were respectively 10 +/- 5%, 36 +/- 5%, 60 +/- 6% and 75 +/- 6% of the intact OC859 IgG. These results suggested that the fragmentation of OC859 with ficin gave a higher yield of superior immunoreactive fragments.

[Breast reconstruction by abdominis musculocutaneous flap].

Breast reconstruction was carried out with rectus abdominis musculocutaneous flap on 7 women (8 breasts) with defect of breast, and perfect result was obtained on all the patients. The flap which was normally excised in an abdominal lipectomy was used to cover the defect of breast. The rectus abdominis of the flap was used to fill the reconstractive breast and to carry the vascularization from the internal mammary artery. The advantages of the method are as follow: (1) the waist-line is reduced by closing the musculoaponeurtic defect; (2) The scar of the donor area is well hidden by the abdominoplasty; (3) The breast reconstruction need not use an implant. The disadvantage is a risk of postoperative herniation if the suture tears.

[Improvement of cerebral oxidative metabolism and cytochrome aa3 redox state by 7.5% hypertonic saline during hemorrhagic hypotension in dogs].

Our previous study reported that 20% (1.5 ml.kg-1 hypertonic saline could significantly improve the disturbances of cerebral oxidative metabolism induced by hemorrhagic hypotension in dogs. The aim of the present study is to evaluate if 7.5% hypertonic saline induces circulatory improvement. Ten dogs were randomly divided into two groups of 5 each resuscitated with either physiological saline as control or 7.5% hypertonic saline (4 ml.kg-1) after their mean arterial blood pressure decreased to 35 mmHg for 45 minutes by hemorrhage. The changes of cerebral tissue oxy- and deoxyhemoglobin, cerebral blood volume (total hemoglobin), and oxy-cytochrome aa3 were continuously monitored by near infrared spectroscopy throughout the experiment. The experimental result showed that oxy-hemoglobin, oxy-cytochrome aa3, and cerebral blood volume decreased but deoxyhemoglobin increased significantly 45 minutes after hemorrhage. Treatment with 7.5% hypertonic saline significantly restored these variables except for cerebral blood volume and all the animals survived to the end of experiment. But in the control group treated with the same dose of physiological saline, the above variables improved little compared with the baseline; and all the animals died before the end of 60 min experimental observation. Therefore we conclude that 7.5% hypertonic saline (4 ml.kg-1) can also effectively improve the disturbance of cerebral oxidative metabolism induced by hemorrhagic hypotension.
