[Separation and determination of metsulfuron, bensulfuron, acetochlor by high performance liquid chromatography].

In this paper a high performance liquid chromatographic method for the separation and determination of metsulfuron, bensulfuron and acetochlor in the composite herbicide "Nongjiayi" is described. The concentration of methanol in the mobile phase and the temperature of the column affected the k' factor and the separation of the three active ingredients. Impurities in the "Nongjiayi" did not interfere with the determination. Chromatographic conditions were Shim-Pack CLC-C8 (150 mm x 6 mm i.d., 5 microm), mobile phase of methanol-water (60:40, V/V) and UV detector at 240 nm. Quantification was carried out with the three external standards. The average recovery was 98.81% with RSD=0.80% for metsulfuron, 99.10% with RSD=0.78% for bensulfuron and 99.72% with RSD=0.69% for acetochlor. The linear range of this method was 0.10-0.40 microg (r=0.9994) for metsulfuron, 0.40-1.60 microg (r=0.9995) for bensulfuron and 10-40 microg (r=0.9987) for acetochlor. The method is accurate, rapid and reproducible and can be applied to quality control for the "Nongjiayi".

Regulation of neuronal and recombinant GABA(A) receptor ion channels by xenovulene A, a natural product isolated from Acremonium strictum.

Xenovulene A (XR368) is a natural product exhibiting little structural resemblance with classical benzodiazepines yet is able to displace high-affinity ligand binding to the benzodiazepine site of the gamma-aminobutyric acid (GABA)A receptor. We have characterized this compound and an associated congener (XR7009) by use of radioligand binding and electrophysiological methodologies with native neurons and the Xenopus oocyte expression system. Xenovulene A, and the more potent XR7009, inhibited [3H]flunitrazepam binding to rat forebrain with Ki values of 7 and 192 nM, and 1.7 and 42 nM, respectively, each site accounting for approximately 50% of the total specific binding. In cerebellar and spinal cord membranes, these ligands identified only single binding sites. These ligands demonstrated no intrinsic agonist activity at recombinant GABA(A) receptors comprising alpha1beta1gamma2S subunits expressed in Xenopus oocytes, yet at 1 microM both significantly potentiated the GABA-induced response and reduced the GABA EC50 from 10.9 (control) to 5.1 (Xenovulene A) or 2.7 microM (XR7009). The rank potency order for enhancement of the 10 microM GABA response is: XR7009 (EC50, 0.02 microM) > diazepam (0.03) > Xenovulene A (0.05) > flurazepam (0.17). The activity of XR368 and XR7009 was reduced by the benzodiazepine antagonist, flumazenil, and absent in receptors devoid of the gamma2 subunit. These agents exhibited receptor subtype selectivity because alpha3beta1gamma2S receptors were less sensitive to these compounds relative to alpha1 subunit-containing receptors, whereas alpha6beta1gamma2S receptors were completely insensitive. Potentiation of the response to GABA on native GABA(A) receptors in cortical neurons substantiates the profile of the novel structures of Xenovulene A and XR7009 as specific benzodiazepine agonists.

[Clinical analysis of 120 patients with fibromyalgia].

The clinical characteristics of 120 patients with fibromyalgia is reported. Most of the patients were 21-50 years old. More than 56% patients had three chief symptoms: generalized myalgia, abnormal sensations and easily fatigue. 49.2% of the patients complained multiple arthralgia; 29% with sleep disorder and 23% with local myospasm of lower limbs. The trigger point was regarded as an important physical sign. The etiology and differential diagnosis for fibromyalgia were discussed.

Interdigitation of bilayers from ether lipid analogs: (R)-PAF, (R)-Lyso-PAF and the antineoplastic (R)-ET-18-OMe.

The ether lipid analogs (R)-PAF, (R)-ET-18-OMe and (R)-Lyso-PAF can form bilayers. Using differential scanning calorimetry (DSC) and small angle X-ray diffraction, we have investigated bilayers formed entirely of these molecules. Small angle X-ray diffraction experiments strongly indicated that these bilayers are interdigitated at 98% relative humidity in the gel phase. The slight differences in the structures of the ether lipids affect their thermal behavior and packing properties. Of the three, (R)-Lyso-PAF shows the highest phase transition temperature, broadest phase transition peak and smallest enthalpy change. These characteristics may be attributed to hydrogen bonding of the -OH group at sn-2 position of its glycerol backbone to water or to other (R)-Lyso-PAF molecules. The interdigitated structure shows that these bilayers are similar to lipid bilayers and suggests that these molecules could easily insert themselves into membranes to reach their respective active sites or to perturb the membranes. The packing and thermal characteristics of (R)-Lyso-PAF may help explain its biological inactivity.

Long-term culture of lymphohematopoietic stem cells.

Pluripotent hematopoietic stem cells (PHSCs) show self-renewal and give rise to all blood cell types. The extremely low number of these cells in primary hematopoietic organs and the lack of culture systems that support proliferation of undifferentiated PHSCs have precluded the study of both the biology of these cells and their clinical application. We describe here cell lines and clones derived from PHSCs that were established from hematopoietic cells from the fetal liver or bone marrow of normal and p53-deficient mice with a combination of four growth factors. Most cell lines were Sca-1+, c-Kit+, PgP-1+, HSA+, and Lin- (B-220-, Joro 75-, 8C5-, F4/80-, CD4-, CD8-, CD3-, IgM-, and TER 119-negative) and expressed three new surface markers: Joro 177, Joro 184, and Joro 96. They did not synthesize RNA transcripts for several genes expressed at early stages of lymphocyte and myeloid/erythroid cell development. The clones were able to generate lymphoid, myeloid, and erythroid hematopoietic cells and to reconstitute the hematopoietic system of irradiated mice for a long time. The availability of lymphohematopoietic stem cell lines should facilitate the analysis of the molecular mechanisms that control self-renewal and differentiation and the development of efficient protocols for somatic gene therapy.

Structural similarity between TAFs and the heterotetrameric core of the histone octamer.

A complex of two TFIID TATA box-binding protein-associated factors (TA FIIs) is described at 2.0A resolution. The amino-terminal portions of dTAFII42 and dTAFII62 from Drosophila adopt the canonical histone fold, consisting of two short alpha-helices flanking a long central alpha-helix. Like histones H3 and H4, dTAFII42 and dTAFII62 form an intimate heterodimer by extensive hydrophobic contacts between the paired molecules. In solution and in the crystalline state, the dTAFII42/dTAFII62 complex exists as a heterotetramer, resembling the (H3/H4)2 heterotetrameric core of the histone octamer, suggesting that TFIID contains a histone octamer-like substructure.

A histone octamer-like structure within TFIID.

The general transcription factor TFIID nucleates initiation complex formation through direct core promoter binding, commits promoters within chromatin to transcription, and mediates the action of transcriptional activators, a phenomenon that may correlate with enhanced TFIID recruitment or conformational changes in TFIID-promoter complexes. Molecular studies of the multiprotein TFIID complex have identified a primary TATA binding subunit (TBP), TBP-associated factors (TAFs) that interact with and mediate the function of activators and intersubunit interactions but have yielded relatively little insight into the structural organization of the complex or the actual mechanism of transcriptional activation. Here we present biochemical evidence for the structural relevance of histone homologies in the human TFIID subunits hTAF80, hTAF31 and hTAF20/15. Together with analyses of native TFIID complexes and accompanying crystallographic studies, the results suggest that there is a histone octamer-like TAF complex within TFIID.

NMDA receptor-dependent LTD in different subfields of hippocampus in vivo and in vitro.

In simulations with artificial neural networks, efficient information processing and storage has been shown to require that the strength of connections between network elements has the capacity to both increase and decrease in a use-dependent manner. In contrast to long-term potentiation (LTP) of excitatory synaptic transmission, activity-dependent long-term depression (LTD) has been difficult to demonstrate in forebrain in vivo. Theoretical arguments indicate that coincidence of presynaptic excitation and low-magnitude postsynaptic activation are the necessary prerequisites for LTD induction. Here we report that stimulation paradigms which cause 1) sufficient excitation to result in NMDA receptor activation and simultaneously 2) attenuate the level of postsynaptic activation by recruitment of GABAA receptor-mediated inhibition consistently produce LTD of commissural input to area CA1 in the hippocampus of anesthetized adult rats, and of the perforant path input to the dentate gyrus in the hippocampus of anesthetized and unanesthetized adult rabbits. A functionally similar pre- and postsynaptic activation pattern applied to the hippocampal slice preparation by injecting hyperpolarizing current into the postsynaptic cell during NMDA receptor-mediated excitation also was effective in consistently inducing LTD. Results of studies in vitro show that Ca2+ influx through the NMDA channel is necessary for the induction of LTD, and moreover, that NMDA receptors also participate in the expression of LTD. Our findings demonstrate a general mechanism for the implementation of a theoretically derived learning rule in adult forebrain in vivo and in vitro and provide justification for the inclusion of use-dependent decreases of connection weights in formal models of cognitive processing.

Two phenol sulfotransferase species from one cDNA: nature of the differences.

A phenol sulfotransferase from rat liver (EC 2.8.2.9), expressed in Escherichia coli from a single cDNA, was purified as two separable but catalytically active proteins. The proteins appeared to be identical to each other and to the natural liver sulfotransferase by comparison of their amino acid constitution, amino-terminal end group, and interaction with a polyclonal antibody raised against the liver enzyme. Each of the recombinant forms, alpha and beta, catalyzed the sulfuryl group transfer from 4-nitrophenylsulfate to an acceptor phenol, a reaction in which 3'-phospho-adenosine 5'-phosphate (PAP) is a necessary intermediate. Only form beta, however, catalyzes the physiological transfer of a sulfuryl group from 3'-phosphoadenosine 5'-phosphosulfate (PAPS) to the free phenol. Evidence is presented that sulfotransferase alpha, but not beta, has 1 mol of PAP tightly bound per enzyme dimer. The ability to utilize PAPS as a sulfate donor could be altered: form alpha could be treated and purified as form beta to acquire the ability to use PAPS, whereas form beta was treated by extended incubation with PAP, lost its ability to use PAPS, and was purified as form alpha.

Tetraploid or near-tetraploid clones characterized by two 8;21 translocations and other chromosomal abnormalities in two patients with acute myeloblastic leukemia.

Acute myeloblastic leukemia (AML) with tetraploidy is rare. There have been only six such cases studied with banding techniques in the literature. Two were diagnosed as having AML-M2 and found to have similar near-tetraploid karyotypes with t(8;21) and missing Y chromosomes. We report two further cases of AML with tetraploid or near-tetraploid clones characterized by two t(8;21) and other chromosomal changes. Their cytogenetic findings were compatible with the diagnosis of AML-M2. Giant and bizarre blasts were seen on bone marrow (BM) smears from both cases. Immunologically, the blasts express CD2, CD15, and HLA-DR in case 1 and CD2 and CD65 in case 2. Cytogenetic studies on BM cells at diagnosis revealed that both cases had three related abnormal clones besides a normal one: 46,XY,t(8;21) (2%)/46,idem,add(7)(q31)(6.8%)/92, idem x 2 (80.6%) for case 1; and 46,XX,t(8;21)(13.4%)/47, idem,+4 (46.3%)/94,idem x 2 (39.1%) for case 2. Flow cytometric analysis displayed two cell populations in the former: one was in the diploid range and the other was in the tetraploid range. The patients did not obtain complete remissions and survived four and six months, respectively. These results indicate that tetraploid or near-tetraploid clones are secondary events which are associated with t(8;21) leukemia and may be associated with poor prognostic significance.

Lack of mutation in tumour-suppressor gene p53 in gestational trophoblastic tumours.

The objectives of this study were to better our understanding of the carcinogenesis of gestational trophoblastic tumours and to investigate the possible presence of mutational alteration of the p53 tumour-suppressor gene in these tumours. Amplification-based direct DNA sequencing was performed on 14 hydatidiform moles, six invasive moles, eight choriocarcinomas and ten normal early placental tissues. No mutation in exons 5-8 was detected in any of these 38 tissue specimens. These results suggest that a mutation in p53 tumour suppressor either does not exist or is a very rare event in gestational trophoblastic tumours. The gestational trophoblastic tumours probably involve a tumour-suppressor gene other than p53 gene or may follow a completely different pathway to their malignant phenotype.

Karyotype conversion in two patients with chronic myeloid leukaemia after busulphan-induced marrow hypoplasia.

SUMMARY: We report two patients with chronic myeloid leukaemia (CML) developing hypoplasia and karyotype conversion after conventional busulphan therapy. Initially, the percentage of Ph-positive metaphases in marrow for both patients was 100%, which steadily diminished up to a complete disappearance in case 1 and decreased dramatically in case 2 following hypoplasia. Thereafter Southern blot and RT-PCR assays revealed no abnormalities. Both patients have survived 9 years and remained in good clinical and haematological remission without any treatment until recently. We believe that the high sensitivity to busulphan therapy result in hypoplasia and karyotype conversion, which contributed to prolonged survival.

[Effects of ACM sequential chemotherapy on ovarian function in trophoblastic tumors].

OBJECTIVE: To study the changes of ovarian function during and after ACM sequential chemotherapy on trophoblastic tumors. METHODS: 17 patients with trophoblastic tumors, received totally 48 cycles of ACM chemotherapy. Of these, 7 underwent hysterotomy and focal ectomy and ovarian biopsy. Changes of menstruation and BBT were observed. beta-hCG, E2, FSH and LH were assayed and histological and immunohistochemical studies in ovaries biopsied were made. RESULTS: Amenorrhea and anovulatory BBT were predominant at the onset of chemotherapy and high level of beta-hCG. Following chemotherapy and decrease of beta-hCG, menstruation recovered with less amount and BBT transformed into ovulatory types with shorter luteal phase. The E2 level appeared to be lower and the FSH and LH level higher. The different kinds of follicles were decreased, but estrogen receptors unchanged. Follow-up showed that ovarian function of 12 of the 17 patients returned to normal in one year. CONCLUSION: ACM chemotherapy suppresses ovarian function mildly and temporarily. Trophoblastic tumor also affects ovarian function. ACM chemotherapy seems to be suitable for young patients with trophoblastic tumors who desire child-bearing.

[Study on cellular proliferation activity of placental site trophoblastic tumor].

OBJECTIVE: To study the relationship among the cellular proliferation activity, benign clinical process and pathologic characteristics of placental site trophoblastic tumor (PSTT). METHODS: Paraffin-embedded blocks from ten patients with PSTT were reexamined by mitotic count, argyrophilic nucleolar organizer regions (AgNOR) staining and flow cytometric DNA analysis. Five cases of hydatidiform mole (HM) and 5 choriocarcinoma (CC) were chosen as controls. RESULTS: Mean mitotic figures of PSTT were 1.3 (0-3) per 10 high power field, while those of HM and CC were 0.8 (0-2) and 2.2 (1-4) respectively. AgNOR number of PSTT was 2.70 +/- 0.55 per cell, while those of HM and CC were 1.96 +/- 0.38 and 4.50 +/- 0.73 respectively. Flow cytometric DNA content revealed that PSTT had DNA index (DI) of 1.10, S phase of 16.7% and proliferating index (PI) of 26.6%. Eight of 10 Cases were followed up, 7 are alive and 1 died of primary lung cancer. CONCLUSION: PSTT has relative low cellular proliferation activity. The good clinical procedure and benign pathological feature of PSTT may be associated with its diploidy DNA and low cellular proliferation activity.

[A legionella pneumophila strain serologically cross-reacting with thirteen serogroups of this species].

A bacterial strain, designated as Jin-1, was isolated from a water sample taken from the Jinyang Lake located in the suburbs of Taiyuan City, Shanxi Province. Its morphologic and tinctorial properties, nutritional requirements, growth characteristics, biochemical reactions, as well as results of DNA and peptide analysis overall met the criteria for identification of Legionella pneumophila. In serogrouping, extensive cross-reactions between Jin-1 and the type strains of L. pneumophila serogroup 1 through 14 were observed in both slide and tube agglutination tests mediated by the IgM antibodies. However, Jin-1 revealed the L. pneumophila serogroup 5 specificity in IFA, ELISA and dot-ELISA, all mediated by the IgG antibodies. Jin-1 was finally identified as a strain of L. pneumophila serogroup 5 with the more complex antigenic composition than the ATCC type strain of this serogroup. The taxonomic conclusion has been confirmed by CDC. A Legionella pneumophila strain which serologically cross-reacting with so many serogroups of this species has never been reported in the published articles. It is presumed that the cross-reacting antigen of Jin-1 belongs to the thymus-independent antigen by nature.

[Detection of konjac glucomannan in seven Amorphophallus Blume species].

Seven Amorphophallus species were detected for konjac glucomananam (KGM). It has been found out that A. albus, A. konjac, A. yuloensis, A. xemengensis and A. dunnii are of higher contents of KGM (30%), but A. sinensis and A. yunnanensis are not as further comfirmed by TLC analysis. It follows that A. albus, A. konjac, A. yuloensis, A. xemengensis and A. dunnii are high quality resources of KGM, but A. sinensis and A. yunanensis cannot be used as substitutes for the above-cited five species.
