Older people with impaired mobility have specific loci of periventricular abnormality on MRI.

BACKGROUND: Recent investigations using MRI suggest that older persons with mobility impairment have a greater volume of abnormal cerebral white matter compared with persons with normal mobility, thus raising the possibility that those with impairment have lesions in areas critical for the control of mobility. OBJECTIVE: To utilize automated image analysis methods to localize the specific regions of abnormal white matter that distinguish subjects with lower mobility from subjects with higher mobility. METHODS: Tissue classification was performed on subjects' dual-echo long repetition time spin-echo MRI using computer algorithms operating on intensity criteria integrated with anatomic information. Statistical analysis of group differences was obtained after spatially normalizing each brain to a standard reference brain. RESULTS: Four discrete periventricular regions, including bilaterally symmetric frontal and bilateral occipitoparietal regions, were identified as being sensitive (frontal) or specific (occipitoparietal) in discriminating the subjects with lower mobility from subjects with higher mobility. The symmetry of these lesions in individual subjects suggested pathology other than arteriolar infarction. CONCLUSIONS: These results suggest that damage to discrete frontal and occipitoparietal periventricular white matter locations may be associated with a mobility disorder of aging.

Nonradioactive detection of retroviral-associated RNase H activity in a microplate-based, high-throughput format.

None of the available antiretroviral drugs that are currently used in the clinic to treat infection with HIV-1 is directed against the RNase H active site of the reverse transcriptase. Here we developed a nonradioactive, 96-well plate assay designed to be used for high-throughput screening of compounds capable of inhibiting the RNase H activity of HIV-1 reverse transcriptase. We employed a tRNA as substrate that was labeled with digoxygenin-modified reporter residues. The labeled tRNA was prehybridized with a DNA oligonucleotide that contained a single biotinylated residue at its 5'-terminus to ensure its attachment to streptavidin-coated microplates. The uncleaved, immobilized DNA/tRNA substrate was detected through the use of established ELISA protocols. Incubation with purified HIV-1 reverse transcriptase initiated RNase H degradation and caused a signal reduction to negligible background levels. In contrast, the signal intensity remained unaffected when using an RNase H deficient mutant enzyme. The assay was validated using the hydrazone derivative BBNH that was previously shown to inhibit RNase H degradation below concentrations of 10 microM.

Solid-phase microextraction of monocyclic aromatic amines using novel fibers coated with crown ether.

Three solid-phase microextraction (SPME) fibers prepared by the sol-gel method, containing hydroxydibenzo-14-crown-4 (OH-DB14C4), dihydroxy-substituted saturated urushiol crown ether (DHSU14C4) and 3,5-dibutyl-unsymmetry-dibenzo-14-crown-4-dihydroxy crown ether (DBUD14C4), respectively, were evaluated for the determination of aromatic amine (aniline, m-toluidine, N,N-diethylaniline, N-ethyl-m-toluidine, 3,4-dimethylaniline). The sol-gel-derived hydroxy-dibenzo14-crown-4-coated fiber has the best affinity for several aniline derivatives. Optimization was carried out for the determination of aromatic amines with SPME fibers. The linearity was from 0.11 to 29 microg/ml and detection limits varied from 0.17 to 0.98 ng/ml. Relative standard deviation (n=5) was found to be 3.23-6.20%. The coating proved to be very stable at high temperature (to 340 degrees C) and in different solvents (organic and inorganic). The method was applied to the determination of aromatic amines in wastewater samples from a pharmaceutical factory.

A proinflammatory role of IL-18 in the development of spontaneous autoimmune disease.

Serum from patients with systemic lupus erythematosus (SLE) contained significantly higher concentrations of IL-18 than normal individuals. MRL/lpr mice, which develop spontaneous lupus-like autoimmune disease, also had higher serum levels of IL-18 than wild-type MRL/++ mice. Daily injections of IL-18 or IL-18 plus IL-12 resulted in accelerated proteinuria, glomerulonephritis, vasculitis, and raised levels of proinflammatory cytokines in MRL/lpr mice. IL-18-treated MRL/lpr mice also developed a "butterfly" facial rash resembling clinical SLE. In contrast, MRL/lpr mice treated with IL-18 plus IL-12 did not develop a facial rash. The facial lesion in the IL-18-treated mice showed epidermal thickening with intense chronic inflammation accompanied by increased apoptosis, Ig deposition, and early systemic Th2 response compared with control or IL-12 plus IL-18-treated mice. These data therefore show that IL-18 is an important mediator of lupus-like disease and may thus be a novel target for therapeutic intervention of spontaneous autoimmune diseases.

A novel[2 + 3]cycloaddition reaction: singlet oxygen mediated formation of 1,3-dipole from iminodiacetic acid dimethyl ester and its addition to maleimides.

Sensitized photolysis of iminodiacetic acid methyl ester and maleimides follows a [2 + 3] cycloaddition pathway yielding pyrrolidine derivatives. This is similar to the photochemical reaction between C(60) and amines. A series of pyrrolidine derivatives are prepared by the method including multipyrrolidines from bis- and tris-maleimide starting materials. The yields range from 13% to 85%. The reaction is highly stereoselective. All the isolated products have the 1,3-dimethoxycarbonyl groups in the cis configuration. Various sensitizers may be used with slightly different yields. A plausible mechanism is proposed that involves the singlet oxygen abstraction of two alpha hydrogen atoms from the iminodiacetate and formation of a 1,3-dipole with a structure similar to the classical thermally generated 1,3-dipole.

Sum-frequency generation in chiral liquids near electronic resonance.

We demonstrate experimentally that visible-visible sum-frequency generation in the bulk of a chiral liquid is observable near electronic resonant transitions. Although the process is electric dipole allowed, it is rather weak because the orientational average over molecules effectively reduces the bulk chiral nonlinearity.

Motional effect in surface sum-frequency vibrational spectroscopy.

Fast orientational motion of molecules at a surface can affect the sum-frequency vibrational spectra of the surface. Calculation shows that the effect is significant if the molecular orientation varies over a broad range within the vibrational relaxation time. The stretch vibration of the free OH bonds at the vapor/water interface is used to illustrate the importance of the effect.

Surface vibrational spectroscopic study of surface melting of ice.

Surface melting on the (0001) face of hexagonal ice ( I(h)) was studied by sum-frequency vibrational spectroscopy in the OH stretch frequency range. The degree of orientational order of the dangling OH bonds at the surface was measured as a function of temperature. Disordering sets in around 200 K and increases dramatically with temperature. The results show that the disordered (quasiliquid) layer on ice is structurally different from normal liquid water.

Extraction and detection of arsenicals in seaweed via accelerated solvent extraction with ion chromatographic separation and ICP-MS detection.

An accelerated solvent extraction (ASE) device was evaluated as a semi-automated means of extracting arsenicals from ribbon kelp. The effect of the experimentally controllable ASE parameters (pressure, temperature, static time, and solvent composition) on the extraction efficiencies of arsenicals from seaweed was investigated. The extraction efficiencies for ribbon kelp (approximately 72.6%) using the ASE were fairly independent (< 7%) of pressure, static time and particle size after 3 ASE extraction cycles. The optimum extraction conditions for the ribbon kelp were obtained by using a 3 mL ASE cell, 30/70 (w/w) MeOH/H2O, 500 psi (1 psi = 7 KPa), ambient temperature, 1 min heat step, 1 min static step, 90% vol. flush, and a 120 s purge. Using these conditions, two other seaweed products produced extraction efficiencies of 25.6% and 50.5%. The inorganic species present in the extract represented 62.5% and 27.8% of the extracted arsenic. The speciation results indicated that both seaweed products contained 4 different arsenosugars, DMA (dimethylarsinic acid), and As(V). One seaweed product also contained As(III). Both of these seaweed products contained an arsenosugar whose molecular weight was determined to be 408 and its structure was tentatively identified using ion chromatography-electrospray ionization-mass spectrometry/mass spectrometry (IC-ESI-MS/MS).

Cloning, expression, and purification of histidine-tagged preS domains of hepatitis B virus.

The preS domains of the hepatitis B virus are hydrophilic polypeptides that have been implicated, among other functions, in the binding of the virus to hepatocytes and in the induction of virus-neutralizing antibodies. A method of overproducing the preS domains of two different subtypes, adw and ayw, has been developed by adding a 6x His tag at the carboxy-terminal end of the polypeptides. Codons for the 6x His were added in reverse primers used to amplify the corresponding cDNAs. The polymerase chain reaction products were cloned into the expression vectors pET-3d (subtype ayw) and pT7-7 (subtype adw), under the control of the inducible bacteriophage T7 RNA polymerase promoter. Upon induction with isopropyl-beta-d-thiogalactopyranoside, proteins were overexpressed and purified by affinity chromatography on a Ni-nitrilotriacetic acid agarose column. This method yielded 20-40 mg of highly pure and very stable proteins per liter of cell culture. Circular dichroism and fluorescence spectroscopy of isolated preS-his-ayw and preS-his-adw, as well as their ability to bind polymerized human serum albumin, indicate that the 6x His tag does not modify the native-like conformation and, therefore, they may be considered as useful tools to study the function of these viral polypeptide regions.

Localization of the Drosophila MAGUK protein Polychaetoid is controlled by alternative splicing.

The Drosophila membrane-associated guanylate kinase (MAGUK) protein Polychaetoid (Pyd) is required for dorsal closure of the embryo, sensory organ patterning, and cell fate specification in the developing eye. We demonstrate that pyd is alternatively spliced resulting in two isoforms that differ by the presence or absence of exon 6. To determine the role of alternative splicing in Pyd function, we generated antibodies specific for each isoform. We find that the exon 6(+) form of Pyd is localized at adherens junctions of embryonic and imaginal epithelia, while the exon 6(-) form is distributed broadly along the lateral membrane. These results suggest that localization of Pyd is controlled by alternative splicing and raise the possibility that exon 6 represents a distinct protein-protein interaction domain.

Speciation and preservation of inorganic arsenic in drinking water sources using EDTA with IC separation and ICP-MS detection.

The native distribution of As(III) and As(v) in drinking water supplies can influence the treatment removal strategy. The stability of As(III) and As(v) in iron-rich drinking waters can be affected by the formation of Fe precipitates (Fe oxides and/or hydroxides designated by "FeOOH"). These precipitates (ppts) can form during the transport of the sample to the laboratory for arsenic speciation analysis. The analysis of the ppt indicates considerable loss of the aqueous arsenic species (As(aq)) to the solid phase "FeOOH" ppt. Studies of laboratory reagent water containing both As(III) and Fe(III) indicate that the resulting "FeOOH" ppt contained a mixture of As(III) and As(v) with near quantitative removal of the As(aq) in 18 hr. The corresponding aqueous fraction after filtration through a 0.45 microm filter was composed primarily of As(v). The formation of "FeOOH" ppt and the loss of As(aq) to the ppt can be virtually eliminated by the use of EDTA, which sequesters the FeIII). Reagent water fortified with Fe(III), As(III) and EDTA produced less than a 1 ppb change in the As(III)aq concentration over 16 d. The EDTA treatment was also tested on three well waters with different native As(III )/As(v) ratios. The native distribution of As(III)/As(v) was stabilized over a period of 10 d with a worst case conversion of As(III) to As(v) of 2 ppb over a 30 d period. All well waters not treated with EDTA had dramatic losses (a factor of 2-5) of As(aq) in less than 1 d. These results indicated that EDTA preservation treatment can be used to preserve As(aq) in waters where the predominant species is the reduced form [As(III)] or in waters which the predominant species is the oxidized form [As(v)]. This preliminary investigation of EDTA to preserve As species in Fe-rich waters indicates stability can be achieved for greater than 14 d.

IL-18 not required for IRBP peptide-induced EAU: studies in gene-deficient mice.

PURPOSE: Interleukin (IL)-18 has been described as a proinflammatory cytokine in rheumatoid arthritis and bacterial infectious diseases. The present study was designed to determine the role of IL-18 in a model of ocular experimental autoimmune uveitis (EAU). The initial studies were conducted to detect the expression of IL-18 in normal mouse eye tissue, and the later studies investigated induction of EAU in mice with an IL-18(-/-) phenotype. METHODS: IL-18 detection was performed by using 5-bromo-4-chloro-3-indoyl-ss--D-galactopyranoside (X-Gal) staining on frozen sections of eyes from mice (129/CD1, DBA1, and Balb/c), either of normal phenotype (+/+) or of deficiency (+/-, -/-) in the IL-18 gene which had been replaced by introduced genes including LacZ under the control of an IL-18 promotor. Severity of EAU was assessed in DBA1 and 129/CD1 wild-type (WT) or IL-18 knockout (KO) mice after immunization with the uveitogenic antigen: interphotoreceptor retinal binding protein (IRBP) peptide 161-180. Lymphocyte proliferation and cytokine production were also measured in WT and IL-18 KO DBA1 mice 15 days after immunization. RESULTS: IL-18 is constitutively expressed in the epithelial cells in iris, ciliary body, and retina. EAU-resistant mice (129/CD1) with an IL-18(-/-) phenotype remained resistant after immunization with IRBP peptide (P161-180). However, EAU-susceptible mice (DBA1) exhibited disease with similar histologic characteristics, despite a generalized reduction of interferon (IFN)-gamma and tumor necrosis factor (TNF)-alpha on an IL-18(-/-) phenotype. DBA1 IL-18(-/-) also demonstrated reduced IL-10 production. CONCLUSIONS: The IL-18 gene is not necessary for the initiation or pathogenesis of EAU induced by IRBP peptide 161-180. IL-18 is expressed in the epithelial cells in iris, ciliary body, and retina in the eyes, but its role in the eye remains undetermined.

Clinical study on treatment of senile vascular dementia by acupuncture.

63 patients with senile vascular dementia were randomly divided into the treatment group (treated by acupuncture) and the control group (treated with piracetam). The authors observed the changes in the score of Hasegawa's dementia scale (HDS), p300, rheoencephalogram, topographic EEG, superoxide dismutase (SOD) activity in erythrocytes, and lipid peroxide (LPO) level in plasma before and after treatment. The statistical data showed that the total effective rate in the treatment group (80.6%) was significantly higher than that in the control group (25%), and the differences in the observed indexes before and after treatment were significant (P < 0.05 or P < 0.01) in the treatment group but not in the control group (P > 0.05), indicating that the acupuncture treatment was superior in immediate therapeutic effect on senile vascular dementia to drug treatment.

Application of autologous peripheral blood stem cell transplantation in children with malignant tumor.

OBJECTIVE: To investigate if low dose total body irradiation (TBI, 6.0-9.0 Gy) combined with intensified chemotherapy followed by autologous peripheral blood stem cell transplantation results in better survival in children with refractory leukemia or solid tumors. METHODS: Twenty-one children with malignant tumors were included in this study. There were 14 males and 7 females aged 3.5-12 years. Underlying disease included high-risk acute lymphoblastic leukemia (ALL, CR1 in 3 children and CR2 in 5 children), acute myeloblastic leukemia (AML, 9 children), non-Hodgkin's lymphoma stage IV (2 children), and neuroblastoma stage IV (2 children). The peripheral hematopoietic stem cells were collected six to eleven months after complete response, mobilized with high dose chemotherapy alone or combined with GM-CSF or G-CSF. The conditioning regimen consisted of chemotherapy with two to three combinations of the following drugs: cyclophosphamide, arabinosylcytosine, McNU, etopside, and ldarubicin on the basis of TBI (6.0-9.0 Gy). A mean of (1.8 +/- 0.5) x 10(8)/kg autologous mononuclear cells were transplanted. The patients were followed up after transplantation. RESULTS: Severe bone marrow suppression occurred in all patients around day +7. Peripheral white blood cell count decreased to 0 in all patients at day +4.8 +/- 2.9, and platelet count decreased to less than 20 x 10(9)/L at day +9.0 +/- 2.6. Successful engraftment was achieved in 21 patients, but four died of infection at day +17, +20, +31 and +67, respectively. Recovery of white blood cell (WBC) to 10 x 10(9)/L, absolute neutrophil count to 0.5 x 10(9)/L, platelet count to 20 x 10(9)/L occurred on 21 +/- 12, 26 +/- 13, and 27 +/- 10 days, respectively. During the follow up period, three patients relapsed at months, +1.5 years, and +2 years 10 months, respectively. One patient died of intracranial hemorrhage at +8 months. Thirteen patients had event-free survival for 2-12 years, with a mean of 6.7 +/- 3.4 years. CONCLUSION: Our preliminary data suggest that myeloablative therapy with low dose TBI (6.0-9.0 Gy) combined with intensified chemotherapy followed by autologous peripheral blood stem cell transplantation might be associated with favorable results in children with refractory leukemia or solid tumors.

[The epidemiology of irritable bowel syndrome and functional constipation of Guangzhou residents].

OBJECTIVE: To investigate the prevalence rates of irritable bowel syndrome (IBS) and functional constipation in Guangzhou City. METHODS: A structured history using a questionnaire on gastrointestinal symptoms during the preceding one year was obtained from random sampling 2,892 residents, 1,006 consecutively patients who visited general practitioner and 495 consecutively patients who visited gastroenterologists. All the questionnaires were filled by trained doctors. RESULTS: The overall prevalence rates of irritable bowel syndrome and functional constipation in Guangzhou City were 5.6% and 3.0%. There were 10.1%, 34.3% and 5.4%, 4.8% visitors who complained IBS and functional constipation in the general internal outpatient department and gastrointestinal outpatient department. The female was predominant in both IBS and functional constipation. The ratio of male to female was 1:1.51 and 1:1.77. With the increase of age, the prevalence rate of functional constipation rose; but IBS did not. There were 23.9% IBS patients and 17.6% functional constipation patients who had ever seen a doctor for these diseases in the community. The percentages of IBS subclasses were as the following: diarrhea predominant: 63.2%; constipation predominant: 27.0%; others: 9.8%. CONCLUSION: The prevalence rates of IBS and functional constipation in Guangzhou City were lower than that in west countries.

[Sentinel lymph node biopsy in head and neck cancer patients].

OBJECTIVE: By performing sentinel lymph node (SLN) biopsy, its detecting and predictive value to the cervical metastasis in the head and neck cancer patients was studied. METHODS: Intraoperative SLN biopsy was done in 51 untreated head and neck cancer patients who all had no metastasis before operation. The lesions were: laryngeal cancer 23, pharyngeal cancer 6, thyroid cancer 20 and cancer of the tongue 2. Fifteen minutes before taking the SLN, local 0.2 to 0.5 ml 2% isosulfan blue dye was submucously injected 0.5 cm away from the upper, lower, right and left margin of the primary tumor. The frozen sections of SLN were compared with the routine pathological sections of the cervical lymph nodes. The metastatic relation between the SLN and the cervical nodes as well as SLN's predictive value to the cervical nodes metastasis were analyzed. RESULTS: SLN was successfully revealed in 48 (94%) of these 51 patients, averaging 2.5 nodes per patient. They were: laryngeal cancer 22, pharyngeal cancer 6, thyroid cancer 18 and lingual cancer 1. Eleven of these SLNs were positive, among whom two had lymph node metastasis in addition to the SLN. Two others were false negative. The positive predictive value of SLNs to the cervical lymph node metastasis was 85%. CONCLUSION: The predictive value of sentinel lymph node to the cervical metastasis is important in head and neck cancer patients.

[Autologous dendritic cells eliciting cytotoxicity of bone marrow cells against chronic granulocytic leukemia].

OBJECTIVE: To investigate the activity of bone marrow cells activated by autologous dendritic cells (DC) to mediate cytotoxicity against chronic granulocytic leukemia (CGL) cells. METHODS: DC were separated from bone marrow mononuclear cells (BMMNC) of two CGL patients in hematological remission and harvested after 3 days of culture in IMDM containing autologous plasma, rhGM-CSF and rhTNFalpha at 37 degrees C, 5% CO(2) humidified atmosphere. BMMNC obtained from the patients were divided into 3 groups to set up Dexter systems: the control group, rhIL-2 containing, and the third group having DC added at day 4. After 10 days of culture, non-adherent cells were harvested and the changes of immunological phenotype and the percentage of P210 positive cells were analyzed. The cytotoxicity were assayed with two-colour flow cytometry. The non-adherent cells from all the 3 systems served as effector cells, those from control system as target cells. RESULTS: The cytotoxic activity against target cells was greater in the DC-activated effector cells than that in rhIL-2-activated ones. The percentages of death cells in target cells were 63.12% versus 42.59% (case 1) and 61.60% versus 21.46% (case 2), respectively. In addition, there was a marked increase in the death cell percentage in the DC-activated effector cells themselves after incubation with target cells. This phenomenon was not found in the rhIL-2-activated effector cells. The percentage of P210 positive cells was significantly lower in non-adherent cells after 10 days of culture in Dexter system, comparing with that in non-cultured BMMNC. The least P210 positive cells were found in those cultured with DC and the less in those with rhIL-2. CONCLUSION: Autologous DC were able to activate bone marrow cells to generate cytotoxicity against CGL cells. Their effect was greater than that of rhIL-2. These activated bone marrow cells might mediate graft versus leukemia effect in vivo.
