[Mutation of p53 gene in human fetal gastric mucosal cells by sterigmatocystin in vitro].

Cell culture, flow cytometry and silver-staining PCR-SSCP methods were used to explore the effects of sterigmatocystin(ST) (1 mg/L and 3 mg/L) on carcinogenesis and mutation of tumor suppressor gene p53 in human fetal gastric mucosal cells in vitro. Four weeks after treated with ST, the cells showed vigorous growth and malignant transformation foci. Twenty-four weeks after ST treatment, the cells could form cellular colonies in soft agar(the mean colony number was 15 and 17 perdish for ST 1 mg/L and 3 mg/L groups respectively). Flow cytometric analysis showed that both proliferation indexes (PI) and the cellular DNA contents of ST treated cells were much higher than those of normal control. The DNA contents of ST treated cells were in DNA aneuploid range. Mutant p53 protein expression was also significantly higher in ST treated cells. Silver-staining PCR-SSCP analysis showed that abnormal electrophoretic migration bands could be seen at exon 8 of p53 gene in ST-treated groups 22 weeks after ST treatment, while no abnormal bands were found in control group. Thus, the results further confirmed the carcinogenic effects of ST on human fetal gastricmucosal cells.

[Characteristics of F-V curve configuration and its tangent time constants and its wave-speed mechanism in normal humans above 50 years old].

To understand the characteristics of F-V curve configuration and its tangent time constant (tau 1) in normal humans above 50 years old, the tau 1 at high, mid, low lung volumes (tau 175, tau 150, tau 125) were measured from F-V curves for 135 normal humans aged 50-84. The results showed that the convex type was the most frequent one (40%-75%) among the 4 types of F-V curve and its tau 1 characteristics were tau 175 > tau 150 < tau 125 and magnitude of tau 175 < 1s; on the other hand, the concave type was the least frequent one (0.18%). These suggested that the convex type and its tau 1 characteristics might be the changes in senility. Further, the mechanical mechanism for the convex type of F-V curve configuration was explored.

[Modulation of endothelin-1 on pulmonary surfactant synthesis in lung explants].

The effect of endothelin-1(ET-1) on pulmonary surfactant(PS) synthesis was studied in cultured lung explants without serum. The results showed that: 1. ET-1 increased [3H] choline incorporation with dose-dependence and time-dependence. 10(-10) mol.L-1 ET-1 increased the contents of total phospholipids, phosphatidylcholine and phosphatidylglcerol in lung tissues. The major membrane characteristic phospholipids, such as phosphatidylethanolamine, phosphatidylinostitol, phosphatidylserine and sphingomyelin, did not show changes. 2. BQ123, a ETA receptor selective antagonist, could decline the [3H] choline incorporation induced by ET-1(10(-12) and 10(-10) mol.L-1) (P < 0.01). 3. PMA, a protein kinase C(PKC) activator, increased [3H] choline incorporation into lung tissues (P < 0.01); the PKC inhibitor H7 decreased [3H] choline incorporation induced by ET-1(P < 0.01). The results demonstrated that ET-1 at physiological level can enhance the PS synthesis mediated by ETA receptor and PKC.

[Apoptosis of B lymphocytic leukemia induced by anticancer drugs and their cell cycle specificity].

OBJECTIVE: Evaluating the effect of apoptosis induced by anticancer drugs on human B lymphocytic leukemia cell lines (Nalm-6) and their cell cycle specificity. METHODS: Nalm-6 cells were treated with various anticancer drugs for 8-24 hours. Apoptotic cells and their cell cycle specificity were measured by using hypodiploid DNA-FCM and TdT assay + DNA staining. RESULTS: ADR, VP16, CPT, MTX, Ara-C could markedly induce apoptosis of Nalm-6 cells in S phase. CPM, PRD, 6MP were less capable of inducing apoptosis. CPM in high dose resulted in cell necrosis. PRD induced apoptosis in G1 phase, while 6MP induced apoptosis in G1 and S phase. The effect of CPM showed no marked cell cycle specificity. CONCLUSION: Hypodiploid DNA-FCM and TdT assay + DNA staining can be used to detect both tumor cell apoptosis and their cell cycle specificity which is helpful to predict prognosis and to design new chemotherapy regimen.

[The effects of nitric oxide synthase on secondary spinal cord injury: an experimental research].

OBJECTIVES: To investigate the effects of the changing amount of nitric oxide on secondary spinal cord injury and determine the role of nitric oxide during secondary spinal cord injury. METHODS: After close spinal cord modest injury (T9) by fluid-percussion model, rats were given saline, small dose of L-NAME (8 mg/kg) and large dose of L-NAME (40 mg/kg) respectively. The changes of hemodynamics, tissue constitution, and neural function were observed. RESULTS: We found small dose of L-NAME had little effects on the hemodynamics but could improve neural function. Whereas, large dose of L-NAME raised the MABP significantly, constricted pial arteriole (90 minutes, 10%), decreased regional spinal cord blood flow (SCBF) (90 minutes, 22%), augmented the dysfunction of neural system. Rats lost weight and the rate of death raised. CONCLUSIONS: These findings indicated that nitric oxide could have dual role during secondary spinal cord injury. Controlling local nitric oxide production suitably through changing the activity of nitric oxide synthase could improve tissue survival and benefit the recovery of neural function.

[Antifertile effect of immunization with anti-idiotypic antibodies to porcine zona pellucida].

OBJECTIVE: To evaluate the antifertile effect of anti-idiotypic antibodies to PZP. METHODS: The New Zealand rabbits were immunized by 17D3 monoclonal antibody against PZP to produce anti-idiotypic antibodies (Ab2) which were then purified by immuno-affinity chromatography. RESULTS: The mice immunized with Ab2 showed decrease of pregnancy rate and a statistically significant reduction in the number of embryos as compared to controls (P < 0.05). Histological examination of ovaries showed that Ab2 interferes only with late follicular development. CONCLUSIONS: Ab2 to 17D3mAb could alleviate ovarian damage and suppress partial fertility. Clearly, further research is required to screen epitope of inner image of Ab2 and to express it by gene engineering.

[The use of bi-pedical rotatory door muscle-skin flap reconstruction in extended partial laryngectomy for late (T3 and T4) glottic cancer].

The purpose is to radical treatment for late glottic cancer by surgery, to restore the essential function of the larynx, 62 patients of late (T3 and T4) glottic cancer were treated by extended vertical partial and subtotal laryngectomy. At the same time, an appropriate method of reconstruction of laryngeal function by bi-pedical rotatory door muscle-skin flap was presented. The decannulation rate was 87.1% and 85.0% cases enjoyed satisfactory voice. All cases resumed normal mouth-food-taking. The conclusion is that selective treated with extended partial laryngectomy is effective for T3 and T4 glottic cancer.

[Speech reconstruction after laryngectomy by using modified Pearson's operation].

Since 1990, 16 patients received speech reconstruction after laryngectomy by using modified Pearson's operation. Among them, T3, T4 laryngeal cancer 12 cases, T4 hypopharyngeal cancer 4 case, 14 were male and 2 were female, the eldest was 73 years old and the youngest 51 years old, and the average age was 58 years. In the operation, the tracheo-pharyngeal speech tube were sewn up with mucosa remained at laryngopharynx. During 1-5 years follow up, the speech function and swallow function were satisfactory. The 3 and 5-year survival rates were 77.78% and 60.00% respectively. They obtained a good long-term speech function except one. It showed that speech reconstruction with the modified Pearson's operation has important value to advanced pharyngeal and laryngeal carcinoma.

[Determination of dioxane in alkyl alcohol polyoxyethylene ether].

In this paper, dioxane in alkyl alcohol polyoxyethylene ether has been determined by headspace gas chromatography. The standard curve and headspace detection conditions are given. Correlation coefficient is 0.9989. The recoveries of dioxane ranged from 91.6% to 97.6%. The relative standard deviations were less than 2.5%. The limit of determination was 20 micrograms/L. The method is simple, fast and accurate with higher sensitivity, less interference and better reproducibility.

[Effects of Yunqitang on both esophageal mucosal morphology and esophageal motility in reflux esophagitis patients].

OBJECTIVE: To observe the effects of Yunqitang (YQT) on both esophageal mucosal morphology and esophageal motility in patients with reflux esophagitis (RE). METHODS: According to Syndrome Differentiation of TCM, 42 RE patients were divided into three groups: Disharmony of Liver and Stomach (D) group, Deficiency-Cold of Spleen and Stomach (DC) group, Heat Syndrome caused by depression of Liver Qi (H) group. No. I, II, III of YQT were taken respectively for 4 weeks. Before and after treatment scores of typical symptoms were collected, gastroscope and esophageal motility were measured. RESULTS: (1) The symptom remission rate was 81.1%, there were significant differences between the group DC with group D and Group H (P < 0.01). (2) The esophageal mucosal healing rate was 61.9%, the effective rate was 90.5%, and the ineffective rate 9.5%. There weren't significant difference of effective rates among the three groups (P < 0.05). (3) The changes of esophageal motility: lower esophageal sphictor pressure (LESP), average peristaltic pressure (APP) of group D were higher (P < 0.05), LESP, gastro-esophageal barrier pressure (GEBP) and peristaltic conduct speed (PCS) of group DC were remarkably higher (P < 0.05), GEBP of group H was improved (P < 0.05). CONCLUSIONS: YQT has a good therapeutical effect, it's not only resolving reflux symptoms, healing esophageal mucosa, but also improving esophageal motile function.

[Effects of extremity cuffs against physiological changes induced by head-down bedrest].

-6 degrees head down bedrest (HDBR) 10d was used to simulate weightlessness. 12 volunteers were randomed into control group and cuffs group, with 6 in each group. Inflated cuffs on the upper part of thighs and arms were performed 12 h/d in cuffs group during bedrest. No countermeasures were applied in the control group. During head up tilt (HUT) +75 degrees 20 min orthostatic tolerance test after HDBR, 5 in control group became presyncopal, while only one in cuffs group. The study indicates that the use of inflated cuffs provides a protective effect against the cardiovascular deconditioning after bedrest.

[Effect of +Gz-induced cerebral ischemia on the distribution of nitric oxide synthase in rabbit brain].

In order to study the effect of repeated short termed + Gz -induced cerebral ischemia on rabbit brain, rabbits were exposed to +4.0 Gz for 3 times (with 30 min interval in between) which caused the arterial pressure at eye level dropped to 0 kPa and lasted for 30 s. Changes of distribution of the nitric oxide synthase(NOS) positive neurons in rabbit brain were observed using the NADPH-diaphorase histochemical reaction. The results showed that NOS positive neurons in the parietal cortex, piriform cortex, hippocampus and dentate gyrus increased by 167%, 126%, 321%, 172% (P < 0.01) respectively, immediately after repeated + Gz exposures, as compared with those of controls. It still remained significantly more than that in controls (P < 0.01) 1 h and 6 h after exposure. It suggests that NO may have some effect on the neurons during cerebral ischemia.

[Changes of plasma endocrine hormone in pilots under Coriolis acceleration].

Plasma endocrine hormones were studied in both 24 motion sickness (orthostatic intolerance) and healthy pilots. Coriolis acceleration of 3.75, 5.00 and 6.25 pi 2 cm/s2 were given with intervals of 3-4 min AT-II, insulin, cortisol, Aldosterone and gastrin were determined by radioimmunoassay. It was found that aldosterone, AT-II, gastrin increased with increase of coriolis acceleration in all pilots. (P < 0.05), but cortisol and insulin only increased in healthy pilots (P < 0.05). It suggests excitation of the autonomic nervous system might be insufficient in orthostatic intolerant pilots and that determination of endocrine hormones may be useful in the evaluation of autonomic nervous activities.

Relationship between apoptosis of brain cells and free radical scavenging ability in rats during acute intermittent hypoxia.

It has been found that plateau hypoxia could induce apoptosis in rats' brain, but the mechanism was not clear. In order to study it further, we utilized acute intermittent hypoxia model to investigate reduced gliutathione (GSH), oxidized gliutathione(GSSG), super-oxide dismutase (SOD) and programmed cell death (PCD) or apoptosis (AP) in rats' brain. The results showed that with the increase of hypoxic exposure duration, apoptotic cells increased, the ratio of GSH/GSSG and the activity of SOD dropped. Regression analysis showed that there was a negative power relationship between the number of apoptotic cells and the ratio of GSH/GSSG or between the number of apoptotic cells and the activity of SOD in a limited range. It was concluded that there would be an intrinsic relationship between apoptosis and free radical scavenging ability in the organ.

[Effect of repeated cerebral ischemia induced by lower body negative pressure on neuronal morphology and ions contents, ATPase activity of brain tissue in rats].

To investigate the effect of repeated + Gz-induced loss of consciousness on the brain and its mechanism, we observed the changes of ions contents, ATPase activity of brain tissues and neuronal morphology after repeated cerebral ischemia induced by lower body negative pressure(LBNP) in rats. Thirty male Sprague-Dawley rats were divided into 3 groups randomly. Rats were anesthetized and exposed to LBNP of -4 kPa at a rate of 0.67 kPa/s. The pressure rapidly returned to control level 2 min after EEG becomes isoelectric. Cortical Na(+)-K(+)-ATPase activity decreased significantly lh after one LBNP exposure (P < 0.05), While Na+, K+ and water contents of brain tissues showed a tendency to increase (P > 0.05). Na(+)-K(+)-ATPase activity decreased significantly while Na+, K+ and water contents increased significantly 1 h after three LBNP exposures, and ischemic changes were found in a few neurons. It suggested that three cerebral ischemia exposure induced by LBNP can result in disturbance of ion homeostasis and neuronal damage.

[Effect of guchijianzhou decoction on antioxidation in aged mice].

OBJECTIVE: To observe the antioxidation effect of Guchijianzhou decoction in aged mice. METHODS: The Guchijianzhou decoction was given to aged mice by gastric intubation and compared with a group of young mice and a group of aged mice giving tape water. The LPO, SOD and CAT level were examined. RESULTS: There were significant differences as the experimental group was campared with the control groups. CONCLUSION: The results show that Guchijianzhou decoction is effective in antioxidation and against danage by free radical. It can reinforce antibacterial function and is benificial in the restoration of periodontal health.

[Mechanism of preventing the recurrence of bladder carcinoma by intravesical instillation of BCG plus IL-2].

OBJECTIVE: To study the mechanism of preventing the recurrence of bladder carcinoma by intravesical instillation of BCG plus IL-2. METHOD: Intravesical instillation of BCG or BCG plus IL-2 were carried out in the bladder transitional cell carcinoma patients undergoing surgical operation (16, 19 cases respectively). RESULT: They were followed up for 14-22 months. The recurrence rates were 31.25% and 21.05% respectively. At 6 weeks after the instillation of IL-2 plus BCG, the peripheral blood NKCF activity was increased significantly and there was a significant positive correlation between the activities of NKCF and IL-2. CONCLUSION: The intravesical instillation of IL-2 plus BCG is clearly superior to only BCG in preventing recurrence of bladder carcinoma. There may be immuno-enhancement and immuno-synergism.
