[Assay of Epstein-Barr virus in nasopharyngeal tissues and serum of patients with nasopharyngeal carcinoma].

OBJECTIVE: Epstein-Barr virus (EBV) is associated with nasopharyngeal carcinoma (NPC). Determination of EBV-VCA-IgA, EBV-EA-IgA in serum and assay of EBV DNA (PCR) in biopsy tissue has been used for diagnosis of NPC. This paper evaluated the value of the three determinations for diagnosis of NPC. METHOD: 146 patients were investigated. For each patient determination of EBV-VCA-IgA, EBV-EA-IgA in serum and EBV-DNA (PCR) in biopsy tissue were performed with double-blind studies, of 146 patients, 76 were NPC and 70 non nasopharyngeal carcinoma as control, identified with histopathological examination. The differences of the determinations between NPC and control were compared statistically with chi 2-test. RESULT: Of 76 NPC, the positive rates of EBV-DNA (PCR), EBV-VCA-IgA and EBV-EA-IgA were 90.8%, 75.0% and 26.3% respectivelly. For 70 control cases, the positive rates 2.9%, 38.6% and 2.9% respectivelly (P < 0.005). CONCLUSION: Our results suggested that assay of EBV-DNA (PCR) has better sensitivity and specificity than EBV-VCA-IgA and EBV-EA-IgA for diagnosis of NPC.

[Studies on identification of circumsporozoite protein genotyping of Plasmodium vivax].

OBJECTIVE: To develop a new method of genotyping circumsporozoite protein (CSP) gene for identification of field isolates of Plasmodium vivax. METHODS: Improved Chelex-100 ion-exchange method was used to extract DNA from blood filter paper samples, nested PCR and allele-specific PCR techniques, agarose gel electrophoresis analysis and dot/southern blotting-probe hybridization were employed for amplification, resolution and identification of the diagnostic fragments. RESULTS: Using the nest-allele-specific PCR assay reported here, small amounts of DNA extracted from a piece of blood filter paper sample were amplified which produced three different size ranges of diagnostic bands: 650-770 bp PV-species-specific band, 170-230 bp diagnostic band for temperate zone family and 588 bp band for PV type-2. The sizes and patterns of the bands produced by the reference strains were consistent with those of designed target sequences. Of 59 examined isolates from 6 provinces of China, 42 temperate zone family strains, 15 tropical zone family strains and two PV type-2 strains were identified. CONCLUSION: 1, Three genotype strains of P. vivax mentioned above could be identified by this method with only two rounds of PCR and without probe hybridization. 2, The preliminary results showed that PV type-1 including temperate zone family strains and tropical zone family strains as well as PV type-2 strains are present in China. In addition, another CSP genotype with both sequence characteristics of temperate zone and tropical zone family might also be present in China.

Lesion of the substantia nigra pars compacta downregulates striatal glutamate receptor subunit mRNA expression.

This is a study of the effect of the unilateral administration of dopamine (DA) in the pars compacta of the substantia nigra (SN) of the rat on striatal glutamate receptor subunit (GluR1, GluR2 and NMDAR1) gene expression determined by in situ hybridization. The location of the nigral lesion was determined by tyrosine hydroxylase (TH) immunohistochemistry and its extent by the striatal DA and 3,4-dihydroxyphenylacetic acid (DOPAC) concentrations. The DA-induced lesions produce significant bilateral reductions in the expression of GluR1 and NMDAR1 subunit mRNA in the medio-lateral striatum, whereas the expression of striatal GluR2 receptors was not changed. The reduction in GluR1 and NMDAR1 subunit mRNA may be the consequence of glutamatergic hyperactivity developed in the presence of a damaged nigro-striatal system and these may be associated with the genesis of some neurodegenerative diseases.

Difference in the in vivo influence of serotonin1A autoreceptors on serotonin release in prefrontal cortex and dorsal hippocampus of the same rat treated with fluoxetine.

Recent studies have demonstrated that antagonism of somatodendritic serotonin1A (5-HT1A) autoreceptors can potentiate the increase of extracellular 5-HT concentrations induced by selective serotonin reuptake inhibitors including fluoxetine. The present study was conducted to uncover any functional difference between the 5-HT1A autoreceptors located on the cell bodies of 5-HT neurons in the dorsal (DRN) and median (MRN) raphe nuclei. The investigational approach used in the present study was to detect extracellular 5-HT concentrations in two terminal areas, prefrontal cortex (Pfc) and dorsal hippocampus (Dhp), which are mainly innervated by the 5-HT neurons located in the DRN and MRN respectively. To avoid possible variation between individual animals a dual-probe microdialysis procedure was applied to determine 5-HT concentrations in both brain areas of the same rat. Fluoxetine (10 mg/kg, s.c.) alone produced a smaller increase in the extracellular 5-HT concentration in the Pfc than Dhp of the same rat (maximal 5-HT concentrations were 183% and 223% of the baseline values in Pfc and Dhp respectively). However, an antagonist of 5-HT1A receptors, WAY100635, subsequently injected (s.c.) at 1 mg/kg brought the 5-HT concentrations to similar levels in the Pfc (332%) and Dhp (308%). Since the 5-HT concentrations immediately before the injection of WAY100635 were lower in the Pfc (102%) than Dhp (186%), WAY100635 induced a larger 5-HT net increase in the Pfc (332%-102%=230%) than Dhp (308%-186%=122%). On the other hand, WAY100635 alone did not significantly change the extracellular 5-HT concentrations in both areas. Furthermore, extracellular concentrations of dopamine (DA) and two DA metabolites, 3,4-dihydroxyphenylacetic acid and homovanillic acid, in both areas were not altered by the administrations of fluoxetine and WAY100635. In conclusion, the present study demonstrated that the antagonist of 5-HT1A receptors, WAY100635, produced a more robust potentiation in the fluoxetine-induced 5-HT increases in the Pfc than Dhp. Since Pfc and Dhp are predominately innervated by 5-HT neurons located in the DRN and MRN respectively, this result may indicate a functional difference between the 5-HT1A autoreceptors located on the cell bodies of 5-HT neurons in the DRN and MRN.

Differential effects of olanzapine on the gene expression of superoxide dismutase and the low affinity nerve growth factor receptor.

Neuroanatomical studies of schizophrenia suggest that progressive neuropathological changes (such as neuronal atrophy and/or cell death) occur over the lifetime course of the disease. Early intervention with atypical neuroleptics has been shown to prevent progression of at least some symptoms, although the mechanisms by which neuroleptics may do this remain unknown. In this study, PC12 cells were used to determine the effects of the new atypical antipsychotic olanzapine on the gene expression of superoxide dismutase (SOD1) and the low affinity nerve growth factor receptor (p75). The results show that olanzapine increases SOD1 at concentrations of 10 and 100 microM after 48 hr of incubation in PC12 cultures. The treatment decreases p75 gene expression at concentrations 100 microM after 48 hr of incubation. Since both the upregulation of SOD1 mRNA and the antisense blockade of p75 mRNA have been associated with reduced cell death, our results suggest that olanzapine has neuroprotective potential and thus may be useful in preventing further neurodegeneration accompanying schizophrenia.

Microvessel density of hepatocellular carcinoma: its relationship with prognosis.

PURPOSE: To elucidate the relationship between angiogenesis and prognosis after curative resection of hepatocellular carcinoma (HCC). METHODS: An immunohistochemical study using anti-CD34 monoclonal antibody was carried out on surgical specimens from 78 HCC patients who had undergone curative resection; microvessel density (MVD) was counted and the overall survival and disease-free survival were analyzed retrospectively. RESULTS: Blood vessels in the tumor were strongly stained by anti-CD34 antibody, but not those in the surrounding liver parenchyma. There were three types of tumor vessels: capillary-like (n = 59), sinusoid-like (n = 16) and mixed-type (n = 3). The median MVD count was 100 per field. The HCC were designated as hypovascular (n = 36) with an MVD count below 100, and hypervascular (n = 42) with an MVD count of 100 or more per field. The 5-year survival and disease-free survival rates were 49.7% and 42.8% respectively, and statistical analysis showed that the MVD level was not correlated with tumor size, capsule status, Edmondson's grade, alpha-fetoprotein level, associated cirrhosis, gamma-glutamyltransferase, and serum HBsAg status. The sinusoid-like tumor vessels appeared more frequently in the more differentiated tumors (P<0.05). No statistical difference in overall and disease-free survival between different MVD levels and microvessel types was found. Tumor size was the only predicting factor in the entire series. In patients with small HCC (< or =5 cm, n = 40), 5-year survival and disease-free survival rates were 58.9% and 52.7% respectively, higher than the values in large HCC (39.8% and 32.0% respectively, P<0.05). The MVD level was an independent predicting factor of disease-free survival, 5-year disease-free survival in the hypovascular group (74.6%) being better than that in the hypervascular group (34.7%, P<0.05). CONCLUSIONS: The MVD level was not related to tumor size, capsule statuo, Edmondson's grade, alpha-fetoprotein level, associated cirrhosis, gamma-glutamyltransferase and serum HBsAg status. In the entire series, tumor size was the only factor influencing survival after curative resection. However, in patients with small HCC, the MVD level was an independent factor of disease-free survival. The pathological and clinical implications of different types of tumor vessels in HCC remain to be studied.

A murine model of IgE-mediated cow's milk hypersensitivity.

BACKGROUND: Cow's milk allergy (CMA) is one of the leading causes of food allergy in children. Understanding the mechanisms involved in the development of CMA has been hampered by the lack of suitable animal models. OBJECTIVE: We sought to develop a mouse model of IgE-mediated cow's milk hypersensitivity (CMH) that mimics the clinical features of immediate CMA in humans. METHODS: Three-week-old C3H/HeJ mice were sensitized by intragastric administration of cow's milk (CM) plus cholera toxin and boosted 5 times at weekly intervals. RESULTS: CM-specific IgE antibody levels were significantly increased at 3 weeks and peaked at 6 weeks after the initial feeding. Intragastric challenge with CM at week 6 elicited systemic anaphylaxis accompanied by vascular leakage, significantly increased plasma histamine, and increased intestinal permeability to casein. Histologic examination of intestinal tissue revealed marked vascular congestion, edema, and sloughing of enterocytes. The role of IgE in mediating CMH was confirmed by abrogation of passive cutaneous anaphylaxis reactions by heat inactivation of immune sera. Development of IgE-mediated CMH in this model is likely to be TH2 cell mediated because in vitro stimulation of spleen cells from mice allergic to CM induced significant increases in the levels of IL-4 and IL-5, but not IFN-gamma. CONCLUSION: This model should provide a useful tool for evaluating the immunopathogenic mechanisms involved in CMA and for exploring new therapeutic approaches.

Chronopharmacologic aspects of continuous AcSDKP infusion in mice.

Inconsistent results characterized N-acetyl-Ser-Asp-Lys-Pro (AcSDKP or Goralatide) effects upon hematologic proliferation, possibly because its circadian organization had been overlooked. We investigated the circadian changes in AcSDKP disposition in plasma and in bone marrow during continuous infusion and AcSDKP effects upon the circadian rhythms in bone marrow granulomonocytic precursors (CFU-GM) and circulating blood cell counts. One hundred ninety-six male B6D2F1 mice received a constant infusion of AcSDKP (24 microg/ day) or 0.9% NaCl for 7 days, using an osmotic minipump. All mice were synchronized with an alternation of 12 hours of light and 12 hours of darkness for 3 weeks prior to study. Mice were sacrificed on the fifth or seventh infusional day at 3, 9, 15, or 21 hours after light onset (HALO) in order to assess plasma and bone marrow AcSDKP concentrations, CFU-GM, and/or circulating blood cell counts. In control mice, plasma and bone marrow AcSDKP concentrations displayed a circadian rhythm with a maximum level during the dark span, at 21 and 15 HALO respectively, while CFU-GM, leukocyte, lymphocyte, and monocyte counts peaked during early light. Continuous AcSDKP infusion increased fivefold mean plasma AcSDKP level at 3 or 9 HALO, thus inverted its physiologic rhythm and suppressed the CFU-GM peak that normally occurs at these times. This inhibition however, was indirect, because the rhythms in bone marrow AcSDKP concentration were similar with or without AcSDKP infusion. Conversely, mean leukocyte and lymphocyte counts were significantly reduced with AcSDKP infusion, while their circadian rhythms remained unaffected and were amplified. The results indicate that AcSDKP pharmacology displays circadian rhythmicity and warrant the exploration of chronopharmacologic schedules of AcSDKP delivery for further protecting bone marrow against chemotherapy insults.

The mouse alanine:glyoxylate aminotransferase gene (Agxt1): cloning, expression, and mapping to chromosome 1.

The human alanine:glyoxylate aminotransferase gene (AGXT) has been cloned and characterized in detail, and various mutant alleles have been shown to be responsible for primary hyperoxaluria type 1 (PH1). However, advances in understanding the basic mechanisms of this rare human disease have been hampered by the lack of a suitable animal model. Although several AGXT homologous genes have been cloned in a number of mammalian species, none of them allows the level of genetic experimentation that current methods provide for mouse embryo manipulation. Thus, we have carried out the molecular cloning and analysis of the mouse Agxt1 gene, as a necessary first step towards the generation of a mouse model for PH1. The full-length mouse Agxt1 cDNA is 1545 bp long, and encodes a 414 amino acid protein. Mouse Agxt1 is highly similar to its rat counterpart both at the nucleotide (91% identity) and the amino acid (92% identity) levels. Like its rat homologue, the larger mRNA species transcribed encodes a conserved amino terminal end characteristic of AGXT forms known to be targeted to the mitochondria. Mouse Agxt1 expression is restricted to the liver, and in vitro transfection of AGXT(-) cells with the cloned Agxt1 cDNA confers AGXT enzymatic activity. At the genomic level, mouse Agxt1 contains 11 exons, spanning 11 Kb, and it maps to the central portion of chromosome 1, a region of known synteny with human distal 2q, where AGXT has been previously mapped (2q36-37).

Effect of antiangiogenic agents on experimental animal models of hepatocellular carcinoma.

A new therapeutic strategy for treating metastasis in hepatocellular carcinoma (HCC) has entailed the use of antiangiogenic agents such as suramin, BB-94 (Batimastat), TNP-470, and carboxyamido-triazole (CAI, a synthetic inhibitor of non-excitable calcium channels that reversibly inhibits angiogenesis). These agents have been used to treat metastatic model of HCC in nude mouse (LCI-D20 mouse model). The results of these studies are summarized in this paper with emphasis on the inhibitory effects of the drugs on tumour growth, angiogenesis, invasion and metastasis in LCI-D20 mouse models. The results suggest that all of the agents used can significantly inhibit tumour growth, angiogenesis, invasion and metastasis of human HCC in nude mouse models, and may be candidates for the control of recurrence and metastasis after HCC resection.

Serum vascular endothelial growth factor is a predictor of invasion and metastasis in hepatocellular carcinoma.

Tumor progression is angiogenesis dependent, and vascular endothelial growth factor (VEGF) is a key growth factor in this process. sVEGF concentrations in 44 patients with hepatocellular carcinoma (HCC) and 5 with benign liver lesions were determined with an enzyme-link immunoadsorbent assay system (ELISA). Reverse transcript-polymerase chain reaction (RT-PCR) was carried out on surgical specimens of 51 patients with HCC. The relative levels of VEGF mRNA expression were measured by determining a ratio between PCR products of VEGF and the endogenous internal standard gene b-actin. UEA-1 was histochemically used to count microvascularity in tumor tissue. Elevated sVEGF concentrations were found in patients with HCC (172.84+/-111.75 pg/ml) as compared to individuals with benign liver lesions (95.74+/-36.20 pg/ml, P<.05). Of 44 cases with HCC, sVEGF concentrations in the patients with PV-emboli or with poor-encapsulated tumors were significantly higher than in those without PV-emboli or with well-encapsulated tumors (P<0.05). The expression levels of VEGF mRNA in tumors with PV-emboli and in poor-encapsulated tumors were higher than in those without PV-emboli and in well-encapsulated tumors (P<0.05). Microvascular density in HCC tissues was significantly correlated with the expression levels of VEGF mRNA (P<0.01; r=0.7). Circulating VEGF was derived from HCC tissue. sVEGF concentrations could be a new marker for predicting the angiogenesis, invasion and metastasis of HCC.

Docetaxel chronopharmacology in mice.

Docetaxel tolerance and antitumor efficacy could be enhanced if drug administration was adapted to circadian rhythms. This hypothesis was investigated in seven experiments involving a total of 626 male B6D2F1 mice, synchronized with an alternation of 12 h of light and 12 h of darkness (12:12), after i.v. administration of docetaxel. In experiment (Exp) 1, the drug was given once a week (wk) for 6 wks (20 mg/kg/wk) or for 5 wks (30 mg/kg/wk) at one of six circadian times, during light when mice were resting [3, 7, or 11 hours after light onset (HALO)], or during darkness, when mice were active (15, 19, or 23 HALO). Endpoints were survival and body weight change. In Exp 2 and 3, docetaxel (30 mg/kg/wk) was administered twice, 1 wk apart, at one of four circadian stages (7, 11, 19, or 23 HALO). Endpoints were hematological and intestinal toxicities. In Exp 4, circadian changes in cell cycle phase distribution and BCL-2 immunofluorescence were investigated in bone marrow as possible mechanisms of docetaxel tolerability rhythm. In Exp 5 to 7, docetaxel was administered to mice bearing measurable P03 pancreatic adenocarcinoma (270-370 mg), with tumor weight and survival as endpoints. Mice from Exp 5 and 6 received a weekly schedule of docetaxel at one of six circadian stages (20 or 30 mg/kg/wk at 3, 7, 11, 15, 19, or 23 HALO). In Exp 7, docetaxel (30 mg/kg) was given every 2 days (day 1, 3, 5 schedule) at 7, 11, 19, or 23 HALO. Docetaxel dosing in the second half of darkness (19 or 23 HALO) resulted in significantly worse toxicity than its administration during the light span (3, 7, or 11 HALO). The survival rate ranged from 56.3% in the mice treated at 23 HALO to 93.8 or 87.5% in those injected at 3 or 11 HALO, respectively (Exp 1, P < 0.01). Granulocytopenia at nadir was -49 +/- 14% at 7 HALO compared with -84 +/- 3% at 19 HALO (Exp 2 and 3, P < 0.029), and severe jejunal mucosa necrosis occurred in 5 of 8 mice treated at 23 HALO as opposed to 2 of 18 receiving docetaxel at 7, 11, or 19 HALO (Exp 2 and 3, P < 0.02). The time of least docetaxel toxicity corresponded to the circadian nadir in S or G2-M phase and to the circadian maximum in BCL-2 immunofluorescence in bone marrow. Docetaxel increased the median survival of tumor-bearing mice in a dose-dependent manner (controls: 24 days; 20 mg/kg weekly, 33 days; 30 mg/kg weekly or day 1, 3, 5 schedule, 44 or 46 days, respectively; Exp 5-7). Survival curves of treated mice differed significantly according to dosing time for each dose and schedule (P from log rank <0.003 to P < 0.03). In Exp 5 and 6, the percentage of increase in life span was largest if docetaxel was administered weekly at 7 HALO (20 mg/kg, 220%; 30 mg/kg, 372%) and lowest after docetaxel dosing at 19 HALO (80% with 20 mg/kg) or at 15 HALO (78% with 30 mg/kg). In Exp 7, (day 1, 3, 5 schedule), docetaxel was most active at 11 HALO (percentage increase in life span, 390%) and least active at 23 HALO (210%). Docetaxel tolerability and antitumor efficacy were simultaneously enhanced by drug dosing in the light span, when mice were resting. Mechanisms underlying the tolerability rhythm likely involved the circadian organization of cell cycle regulation. Docetaxel therapeutic index may be improved with an administration at night in cancer patients, when fewest bone marrow cells are in S or G2-M phase.

Amantadine increases aromatic L-amino acid decarboxylase mRNA in PC12 cells.

Amantadine is an antiviral agent that was unexpectedly found to cause symptomatic improvement in patients with Parkinsonism, although its mechanism of action remains to be elucidated. Aromatic L-amino acid decarboxylase (AADC) is a regulated enzyme that catalyzes the decarboxylation of 3,4-dihydroxyphenylalanine (L-Dopa). It may be especially important during L-Dopa therapy in Parkinsonism, during which it may be rate-limiting for the production of dopamine. This study reports the effects of amantadine on the gene expression of AADC in PC12 cells. It shows that amantadine induces AADC gene expression at concentrations of 10 and 100 microM after 24 hr of incubation. The results suggest that the stimulation of AADC mRNA by amantadine may be one of its effects on dopamine metabolism that may have relevance for potentiation of L-Dopa therapy in Parkinsonism.

L-deprenyl potentiates NGF-induced changes in superoxide dismutase mRNA in PC12 cells.

L-deprenyl protects neurons in a number of in vivo and in vitro models and it has been postulated that it ameliorates some neurodegenerative disorders. Superoxide dismutase (SOD) is one of the enzymes responsible for the inactivation of oxygen free radicals, and one of the mechanisms of the effect of L-deprenyl is thought to act by induction of SOD. In this study, PC12 cells were used to study the effect of L-deprenyl on gene regulation of SOD and its interaction with nerve growth factor (NGF). The results show that NGF induces SOD mRNA in a dose-dependent manner and that a similar effect was produced by L-deprenyl. In addition, L-deprenyl potentiates NGF effects. This study demonstrates that both L-deprenyl and NGF may be involved in common antioxidative mechanisms and that L-deprenyl may interact with neurotrophic factors.

Induction of pulmonary allergic responses by antigen-specific Th2 cells.

The development of pulmonary allergic responses was examined in mice following pulmonary transfer of Ag (conalbumin)-specific Th2 cells. The levels of serum-specific IgE, cellular infiltrates, airway mucus goblet cells, and airway responsiveness were analyzed and compared with those in Ag-sensitized and -challenged mice. Pulmonary transfer of the conalbumin-specific Th2 clone (D10) induced, in an Ag-specific manner, high levels of the Th2 cytokines IL-4 and IL-5 in the bronchoalveolar lavage fluids and mucosal eosinophils, concomitant with an increase in airway responsiveness. The D10 cell-induced responses were seen in the absence of serum specific IgE. In the presence of Ag, the transferred D10 cells not only remained in the lungs, but also increased in number 72 h post-cell transfer. Although significantly higher levels of IL-4 and IL-5 in the bronchoalveolar lavage fluids were found in D10-transferred mice, the levels of pulmonary eosinophilia, mucus goblet cells, and airway responsiveness were significantly lower than those in Ag-sensitized and -challenged mice. These results demonstrate that although Ag-specific activation of Th2 cells at mucosal sites is able to mediate the recruitment of eosinophils and the subsequent induction of airway hyper-responsiveness, the more severe pulmonary allergic responses were observed only in mice sensitized and challenged with Ag.
