Molecular characterization of invasive and noninvasive Campylobacter jejuni and Campylobacter coli isolates.

Campylobacter jejuni is one of the most common causes of bacterial diarrhea worldwide and is the primary bacterial cause of food-borne illness. Adherence to and invasion of epithelial cells are the most important pathogenic mechanisms of Campylobacter diarrhea. Molecular characterization of invasive and noninvasive Campylobacter isolates from children with diarrhea and symptom-free children was performed by random amplified polymorphic DNA techniques (RAPD). A distinct RAPD profile with a DNA band of 1.6 kb was observed significantly more frequently among invasive (63%) than among noninvasive (16%) Campylobacter isolates (P = 0.000005). The 1.6-kb band was named the invasion-associated marker (IAM). Using specifically designed primers, a fragment of 518 bp of the iam locus was amplified in 85% of invasive and 20% of noninvasive strains (P = 0.0000000). Molecular typing with a PCR-restriction fragment length polymorphism assay which amplified the entire iam locus showed a HindIII restriction fragment polymorphism pattern associated mainly with invasive strains. Although cluster analysis of the RAPD fingerprinting showed genetic diversity among strains, two main clusters were identified. Cluster I comprised significantly more pathogenic and invasive isolates, while cluster II grouped the majority of nonpathogenic, noninvasive isolates. These data indicate that most of the invasive Campylobacter strains could be differentiated from noninvasive isolates by RAPD analysis and PCR using specific primers that amplify a fragment of the iam locus.

Soft lithography in biology and biochemistry.

Soft lithography, a set of techniques for microfabrication, is based on printing and molding using elastomeric stamps with the patterns of interest in basrelief. As a technique for fabricating microstructures for biological applications, soft lithography overcomes many of the shortcomings of photolithography. In particular, soft lithography offers the ability to control the molecular structure of surfaces and to pattern the complex molecules relevant to biology, to fabricate channel structures appropriate for microfluidics, and to pattern and manipulate cells. For the relatively large feature sizes used in biology (> or = 50 microns), production of prototype patterns and structures is convenient, inexpensive, and rapid. Self-assembled monolayers of alkanethiolates on gold are particularly easy to pattern by soft lithography, and they provide exquisite control over surface biochemistry.

Exaggerated neurogenic inflammation and substance P receptor upregulation in RSV-infected weanling rats.

Respiratory syncytial virus (RSV) infection in adult rats causes exaggerated inflammation after sensory nerve stimulation in the extrapulmonary, but not in the intrapulmonary airways. The goal of this study was to analyze neurogenic inflammation in weanling F-344 rats infected with RSV 18 +/- 2 d after birth. Five days after RSV inoculation, the extravasation of Evans blue-labeled albumin after nerve stimulation was significantly greater in the intrapulmonary airways of RSV-infected weanling rats than in pathogen-free control rats. In contrast, no difference was found in the extrapulmonary airways. The level of messenger RNA (mRNA) encoding the substance P (SP) receptor (neurokinin 1 [NK1]) increased fourfold in RSV-infected lungs, whereas mRNA encoding the VIPR1 receptor for the antiinflammatory vasoactive intestinal peptide (VIP) increased to a much lesser degree. mRNAs encoding the other neurokinin (NK2) and VIP (VIPR2) receptors were not affected by the virus. Selective inhibition of the NK1 receptor abolished neurogenic inflammation in RSV-infected intrapulmonary airways. Also, neurogenic inflammation and NK1 receptor upregulation in infected lungs were inhibited by prophylaxis with a monoclonal antibody against RSV. These data suggest that RSV lower respiratory tract infection makes the intrapulmonary airways of young rats abnormally susceptible to the proinflammatory effects of SP by selectively upregulating the expression of NK1 receptors.

Dosage effect of minor arginyl- and isoleucyl-tRNAs on protein synthesis in an Escherichia coli in vitro coupled transcription/translation system.

In Escherichia coli mRNA, the arginine codons AGA/AGG and the isoleucine codon AUA are rarely used with frequencies of about 0.35% and 0.41%, respectively. Six genes with a different number of these codons were expressed in an E. coli in vitro coupled transcription/translation system, which contained either tRNA prepared from E. coli cells carrying a plasmid with argU and ileX genes encoding rare tRNAs (tRNA(arg)(AGA/AGG) and tRNA(ile)(AUA)), designated codon-plus tRNA, or normal tRNA from cells lacking the plasmid. Genes having a low number of the rare codons, such as genes encoding chloramphenicol acetyltransferase and anti-gp120 single-chain Fv (artificially constructed to remove rare codons), were expressed at similar levels using with both tRNA preparations. On the other hand, the use of codon-plus tRNA increased the expression levels of genes having a relatively large number of the rare codons, including genes encoding archaeal proteins, green fluorescent protein of jelly fish origin, and a single-chain Fv of mouse origin, by about 20% higher than that using normal tRNA.

[Adaptive moving averaging based estimation of single event-related potentials].

Event-related potentials (ERP) is pertinent to medical research and clinical diagnosis. Estimation of single event-related potentials (sERP) is the objective of ERP processing. A new technique, adaptive moving averaging based method for estimation of sERP, is presented. After analysis of the properties of background noise by crossing zero, the window length of moving averaging is adaptively set according to the maximum width of the impulse noise for each recorded raw data. The experiments are made with real recorded data and the results demonstrate that the performance of sERP estimation is excellent. So the method proposed is suitable to sERP processing.

[The effect of IFN-alpha 2b-thymus peptides combined therapy in chronic hepatitis B].

OBJECTIVE: To determine the effects of IFN-alpha 2b associated with thymus peptides in treatment of chronic hepatitis B. METHODS: A total of 52 individuals with chronic hepatitis B received IFN-alpha 2b(5 x 10(6) IU/ml qod) for 6 months and thymus peptides(20 mg qod) for 3 months. 50 patients received SMMC or other medicine such as WuWeizi, CuiPenCao served as control. There were no differences between two groups in sex, age, duration and seriousness of the disease. RESULTS: The primary outcome measure for efficacy was the percentage of HBV DNA/HBeAg negativity after 6 months therapy. Rate of HBV DNA negativity was 75% and that of HBeAg was 73.1%, the inhibitory activity of IFN-alpha 2b against HBV was significantly higher than that of the control. Except transient fever, leukopenia and thrombocytopenia appearing at the beginning of the therapy, no serious adverse events were observed. CONCLUSION: Potent inhibitory activity against HBV of IFN-alpha 2b is suggested and much works need to be done to improve the long-term effect of the therapy.

[Kinetics of interaction of H+ with constant and variable charge soils].

The kinetics of interaction of H+ ions with constant and variable charge soils were studied, among which changes of H+ concentration, positive surface charge and soluble Al with time and their influencing factors were investigated in detail. The results showed that the consumption processes of H+ ions in both constant and variable charge soils could be divided into fast and slow processes, but the manifestations and influencing factors were different for the two types of soils. The variations in positive surface charges for variable and constant charge soils, due to the difference in the contents of free Fe-oxides, are remarkably different and thus can not be modeled mathematically in the same manner for the two types of soils. The variations in soluble Al for both variable and constant charge soils were controlled by the components, solubility and solution kinetics of dominant clay minerals in soils, and could be described by both modified Elovich and two-constant rate equations mathematically.

[Detection of positive-strand of transfusion transmitted virus fragment in the liver of cryptogenic hepatitis].

OBJECTIVE: To evaluate whether transfusion transmitted virus (TTV) replicates in the liver and to analyze the relationship between TTV and cryptogenic hepatitis. METHODS: A 3.2 kb TTV fragment was detected by nested polymerase chain reaction (PCR) in 31 serum samples of patients with cryptogenic hepatitis who came from a school where was in an outbreak area of cryptogenic hepatitis during 1996 and in 30 healthy individuals. A hybridization/nuclease protection assay was used to detect positive-strand TTV fragment from the liver specimens of the 7 patients. RESULTS: TTV DNA was detected in serum samples from 30 of 31 (96.9%) patients and from 18 of 30 (60%) healthy individuals, respectively. There was significant difference in the prevalence of TTV infection between the two groups. Based on hybridization/ nuclease protection assays, we detected positive-strand TTV fragment in all of 7 liver specimens of the patients. CONCLUSIONS: TTV is related possibly to cryptogenic hepatitis. TTV can replicate in the liver. Although TTV only causes mild liver damage, a few patients will suffer from chronic hepatitis.

[Expression of cytokines in acute heart transplantation rejection].

OBJECTIVE: To observe the expression and changes of local cytokines network in rat heart transplantation so as to determine the role of cytokines in acute rejection of rat heart transplantation. METHODS: The Wistar rats (receptor) were divided into four groups: group A(control, n = 12), group B(IL-2 Mab treated, n = 12), group C [Cyclosporine A (CsA) treated, n = 12] and group D(IL-2Mab + CsA treated, n = 12). Hearts from SD rats were transplanted into a cervical location in Wistar. The local expression of IL-1 beta, IL-2, CD25, IL-4, IL-5, IL-6, IL-10, TNF alpha, IFN gamma was examined at 1, 3, 5, 7, 9, 11, 14 days after operation by reverse transcription polymerase chain reaction (RT-PCR). RESULTS: Allografts survival time was (8.3 +/- 1.7) days in group A, (29.2 +/- 7.1) days in group B, (26.4 +/- 5.7) days in group C, (55.0 +/- 10.6) days in group D. The expression of IL-1 beta, IL-4, IL-10, IFN gamma was up-regulated, and the expression of IL-2, CD25, IL-5, IL-6, TNF alpha was significantly inhibited in group A. The expression of IL-1 beta, IL-5, IL-6, IL-10, IFN gamma was up-regulated, and the expression of IL-2, IL-4, TNF alpha was significantly down-regulated in group B. The expression of IL-1 beta, IL-2, CD25, IL-5, TNF alpha, IFN gamma was up-regulated, and the expression of IL-4, IL-6, Il-10 was significantly down-regulated in group C. The expression of IL-4, IL-5, IL-6, IL-10 was up-regulated, and the expression of IL-1 beta, IL-2, CD25, IFN gamma, TNF alpha was significantly down-regulated in group D. CONCLUSIONS: Cytokines play an important role in the development of acute transplantational rejection. Different cytokines play different roles in different local environments; the main principle possibly is immune deviation.

[Transportation and transformation of 14C-phenanthrene in closed chamber (nutrient solution-lava-plant-air) system].

The transportation and transformation of 14C-phenanthrene in a closed 'plant-lava-nutrient solution-air' chamber system was studied by using radioactivity technology. The results showed that in this closed chamber system, phenanthrene was degraded fast. The radioactivity of 14C left at 23d in the nutrient solution was only 25% of applied. At the end of experiment (46d), the distribution sequence of 14C activity in the components of closed chamber system was root (38.55%) > volatile organic compounds (VOCs, 17.68%) > lava (14.35%) > CO2 (11.42%) > stem (2%). 14C-activities in plant tissue were combined with the tissue, and existed in the forms of lava-bound(root 4.68%; stem and leaves 0.68%) and polar metabolites (root 23.14%; stem 0.78%).

[A study on the pathogenesis of Streptococcus mitis exotoxin].

OBJECTIVE: To study the isolation, purification and pathogenesis of Streptococcus mitis pyrogenic exotoxin causing toxic shock syndrome. METHODS: Streptococcus mitis isolated from patients' throat were shaking cultivated. After being centrifuged, the supernatant fluid of the culture was precipitated with 20%, 40%, 60%, 80% (NH(4))(2)SO(4) respectively and the fast protein liquid chromatography(FPLC) was used for the final step of purification. Rabbits receiving subcutaneous injection with respective purified proteins were monitored daily for fever. The ability of the purified proteins to enhance the susceptibility of the rabbits to lethal Escherichia coli endotoxin shock is recorded, when the endotoxin was injected intravenously 4 hours after administration of 10 microg Streptococcus mitis exotoxin. RESULTS: Only the protein precipitated by 20% (NH(4))(2)SO(4) (molecular weight is 34,000) from culture supernatant fluid was pyrogenic for rabbits (average temperature increase near 1 degrees C), and it can also cause the proliferation of rabbit splenocytes (mitogenicity). All the animals receiving subcutaneous injection of exotoxin containing purified proteins precipitated with higher concentrations of (NH(4))(2)SO(4) died within 16 approximately 29 hours after intravenous injection of the Escherichia coli endotoxin, demonstrating the enhanced susceptibility of the animals to lethal endotoxin shock. The control rabbits displayed none of these effects. CONCLUSION: Streptococcus mitis exotoxin is a novel streptococcal pyrogenic exotoxin.

[Study on variation and interrelations of characteristics at ginseng germplasms].

The variation and interrelations of some characteristics of Panax ginseng germplasms(lines), which were five selfmating generations of some single plants were studied. Methods of multivariate analysis, such as correlation, regression, or clustering, and coefficient of variation were used. The results were of benefit to the breeding and standardized planting of Panax ginseng.

[Effect of toluene on urease activity of Lou soil].

Urease activity affected by toluene studying results showed as follows: Toluene had little action on Jackbean urease activity, but can remarkably increase soil urease activity in lower toluene concentrations and shorter time, the increasing variation was 1.48-3.96 times than without toluene treatment in seven soil samples tested. The variation was less than 3.98% when sterilized soil samples adsorbed the Jackbean urease. It may be because toluene can kill soil microorganism, release urease within cell and subsequently urease fixed into soil organic-inorganic colloidal. Different soil pH and microbiology can lead to vary differently for urease activity.

[The study of countermeasures on measles control in infants].

OBJECTIVE: To understand: a) measles antibody levels in purpura and their newborns, and the relation between them, b) maternal transferred measles antibody level of different age group infants, c) immunization rates of success in 6 and 8 month old infants, d) to study measles control countermeasures on infants. METHOD: To test measles IgG antibody levels of infant's blood using ELISA method. RESULTS: Antibody levels of measles in 58 pairs of mother and their newborns, 51 pairs were the same, while 7 pairs were different. Measles antibody levels in newborns was not high and reduced to 50% in 3 months, 20% in 6 months, and 15% in 8 months. The antibody levels in 6 and 8 month olds were low, nearly no protection to children. It was found that rates of immunization success and the antibody distribution were not statistically different between 6 and 8 months olds after measles vaccination. CONCLUSION: It was found that the measles antibody level in most matured women was low. Since mother's measles antibody level has close relation to their newborns, it seems that the newborn's antibody level can be improved by vaccination to matured women. In order to reduce morbidity of infant measles, the age of first vaccination of measles vaccine should be changed from 8 months old to 6 months old in some regions.

[A study on relationship between blood lead level in children and fetal umbilical cord].

OBJECTIVE: To study the relationship between blood lead level in children and fetus. METHODS: Blood specimens were collected from the fingers of children aged one to five years and from umbilical cord when the babies were born, to determine their lead level with atomic absorption spectrophotometry. RESULTS: The average blood lead level was (0.42 +/- 0.15) micromol/L in 1 122 children. Blood lead level in 27.54% of the children was above >/= 0.483 micromol/L. The average umbilical blood lead level was (0.16 +/- 0.12) micromol/L in 127 neonates. Blood lead level in 0.79% of the new-born babies were greater than >/= 0.483 micromol/L. CONCLUSION: Higher blood lead level in children was mainly caused by environmental pollution and unhealthy life behaviors. Background blood lead in fetuses has already reached a higher level, so prevention from lead poisoning in children should begin during pregnancy.

[The preparation of collagen burn pellicle of compound sulfadiazine silver and assessment of its efficacy in an animal experiment on deep partial thickness burn wound].

OBJECTIVE: To prepare the collagen burn pellicle of compound sulfadiazine sliver and observe its therapeutic effect on deep partial thickness burn wound. METHODS: The initator method was adopted for preparing the collagen burn pellicle of compound sulfadiazine sliver. A model of deep partial thickness burn wound was established in 84 SD rats for the observation of the effect of collagen burn pellicle of compound sulfadiazine sliver on burn wound healing. RESULTS: The collagen burn pellicle of compound sulfadiazine sliver enhanced the coming off of necrotic tissues and the healing of burn wound. The hydroxyproline content of burn wound was higher in the experiment group than that in the comparison groups, (P < 0.05). The percentage of G0/G1-phase in full skin cells of burn wound at 5, 7, 10 and 14 days after burn was lower than that in the comparison groups (P < 0.05). The percentage of S-phase at 5, 7 and 14 days was higher in the experiment group than that in the comparison groups (P < 0.05). The water content in full skin cells of burn wound at 24, 36 and 48 hours after burn was significantly lower in the experiment group than that the comparison groups (P < 0.05). CONCLUSION: The collagen burn pellicle of compound sulfadiazine sliver can enhance wound healing in the management of deep partial thickness burn wound.

[A report of twice outbreak epidemic of acute tonsillitis and acute pharyngitis].

OBJECTIVE: To discuss the pathogenic factors, prevention and treatment of fulminant epidemic of acute tonsillitis and acute pharyngitis. METHOD: We investigated and analysed the cases of twice fulminant epidemic of acute tonsillitis and acute pharyngitis. RESULT: In the first time, 91 patients among 518 persons were attacted, the sick rate was 17.6%. In the second time, 39 among 295 persons were attacted, the sick rate was 13.2%. The main symptoms were pharyngodynia and fever. Bacterial culture of throat swab of 20 cases of the first time and 17 cases of the second time were carried out. The positive rate of beta-hemolytic streptococcus was 82.5% and 85.3% respectively. CONCLUSION: Environment and individual health condition are important factors in the fulminant epidemic of acute tonsillitis and acute pharyngitis.

[The application of mastoid cavity plombage in open method tympanoplasty].

OBJECTIVE: The role mastoid cavity plombage played in restoring physiological function of external meatus in open method tympanoplasty was studied. METHOD: 85 cases of cholesteatoma otitis media were performed open method tympanoplasty with the broken bits of their own mastoid process cortex, homological costal cartilage or U shape periosteum connective tissue valve. And plombage was performed at the same time. The reconstruction of ossicular chain (type 0 tympanoplasty): 10 ears did not undergo reconstruction due to increase ment of bone conduction threshold or were in preparation for the second phase operation; 40 ears underwent type III tympanoplasty; 35 ears underwent type IV tympanoplasty. RESULT: The total improvement rate of hearing was 76%, among which 80% were performed type III tympanoplasty and 71.4% underwent type IV tympanoplasty. After operation, the time waited till dry-ears was 19.8 +/- 65.31 days and the dry-ears rate was 97.6% (83/85). Six months after operation, 89.4% (76/85) had smooth external meatus and fine self-cleaning function and no recurrence was found. CONCLUSION: As to cholesteatoma otitis media that could not be performed closed method tympanoplasty, U shape periosteum connective tissue valve and mastoid process cortex were used. Mastoid cavity plombage with homological costal cartilage could effectively overcome the shortcomings of destruction of physiological structure of external meatus caused by open method tympanoplasty, the self-cleaning function of skins of external meatus was remained and the life quality of patients after operation was increased.
