[Determination of trace zinc in milk using pulse nebulization atomic absorption spectrophotometry technique].

A rapid, diect, pulse nebulization method for the determination of trace zinc in milk by FAAS with model WYX-402A atomic absorption spectrometer is described. The influence of pulse nebulization volume, uptake rate of solution and other factors on the uptake signal has been discussed. The milk sample was diluted with 1.5% HCl solution. A 100 microL aliquot of the solution was then nebulied and the peak height of the transient output pulse was monitored. The result of the determination of the 1+4 diluted sample is in agreement with that of digested sample with HNO3-HClO4. The average recovery of 99.0% with relative standard deviation of 3.3% was achieved and the detection limit was 0.028 microg/mL. The method is simple, rapid, accurate and precise.

Diagnosis of mucosa-associated lymphoid tissue (MALT) lymphoma in gastroendoscopic biopsy specimens.

UNLABELLED: We evaluated the diagnostic criteria of gastric lymphoma in endoscopic biopsies. METHODS: Our study included cases diagnosed as lymphoma by gastroendoscopic biopsy between 1984 and 1994 that were available in the files of the Department of Gastroenterology, Chinese PLA General Hospital. RESULTS: Among 53,400 cases of gastroendoscopy, we found 1672 (3%) cases of malignant neoplasm that were diagnosed by endoscopic features and histological findings. Thirty-eight cases of primary gastric lymphoma included 22 men and 16 women 16 to 82 years old, with a median age of 47.7. The endoscopy showed the ulcerative type in 12 cases, diffuse infiltrating type in 11 cases, massive infiltrating type in 6 cases, and large mucosal folds type in 4 cases. Five cases had a mixed type pattern of involvement. Histologically, 34 cases showed mucosa-associated lymphoid tissue lymphoma (89.5%), 2 cases had lymphoblastic lymphoma, and 2 cases were unclassifiable because of crush artifact of neoplastic cells. The majority of cases of primary gastric lymphoma have morphologic and clinical features that justify their inclusion in the category of low-grade lymphoma of mucosa-associated lymphoid tissue (MALT).

Histopathological changes in hepatocellular carcinoma after hepatic arterial embolization.

Hepatic arterial embolization (HAE) is used to treat hepatocellular carcinoma (HCC), but little is known about the histopathological changes occurring after HAE. Thirty-nine cases of liver neoplasms treated with HAE and 11 cases of liver neoplasms after digital selective angiography (DSA) were analyzed histopathologically. The pathological type, histological grade, degree of necrosis, capsular changes, number of HAE, vessel injury, and lymphocytic infiltration were recorded. Six cases showed 100% necrosis, 14 cases had 30 to 95% necrosis, and 19 cases 0 to 5% necrosis after HAE. Eleven cases of DSA did not reveal histological features of necrosis. The necrosis was related to the pathological type of tumor, encapsulation, and degree of vascular injury and was independent of histological grade, number of HAE, or lymphocytic infiltration of the neoplasm. HAE is an effective therapy for advanced stage of HCC. The encapsulated HCC is a preferred indicator for HAE.

The m2 form of the Helicobacter pylori cytotoxin has cell type-specific vacuolating activity.

The Helicobacter pylori toxin VacA causes vacuolar degeneration in mammalian cell lines in vitro and plays a key role in peptic ulcer disease. Two alleles, m1 and m2, of the mid-region of the vacA gene have been described, and the m2 cytotoxin always has been described as inactive in the in vitro HeLa cell assay. However, the m2 allele is associated with peptic ulcer and is prevalent in populations in which peptic ulcer and gastric cancer have high incidence. In this paper, we show that, despite the absence of toxicity on HeLa cells, the m2 cytotoxin is able to induce vacuolization in primary gastric cells and in other cell lines such as RK-13. The absence of Hela cell activity is due to an inability to interact with the cell surface, suggesting a receptor-mediated interaction. This result is consistent with the observation that the m2 allele is found in a population that has a high prevalence of peptic ulcer disease and gastric cancer. VacA is the first bacterial toxin described for which the same active subunit can be delivered by different receptor binding domains.

Characterisation of a monoclonal antibody and its use to purify the cytotoxin of Helicobacter pylori.

The vacuolating cytotoxin (VacA) is a major virulence factor of Helicobacter pylori which is not yet well characterised and is difficult to obtain in large quantities. Here we describe the production of a monoclonal antibody that recognises the native but not the denatured form of VacA. The antibody can be efficiently used in affinity chromatography for one-step purification of large quantities of VacA from culture supernatants. Elution at acidic pH dissociates the oligomeric molecule into monomers that reanneal in a time-dependent fashion. The purified cytotoxin is able to bind, and to intoxicate HeLa cells.

Differential roles for bone morphogenetic protein (BMP) receptor type IB and IA in differentiation and specification of mesenchymal precursor cells to osteoblast and adipocyte lineages.

Cumulative evidence indicates that osteoblasts and adipocytes share a common mesenchymal precursor and that bone morphogenetic proteins (BMPs) can induce both osteoblast and adipocyte differentiation of this precursor. In the present study, we investigated the roles of BMP receptors in differentiation along these separate lineages using a well-characterized clonal cell line, 2T3, derived from the mouse calvariae. BMP-2 induced 2T3 cells to differentiate into mature osteoblasts or adipocytes depending upon culture conditions. To test the specific roles of the type IA and IB BMP receptor components, truncated and constitutively active type IA and IB BMP receptor cDNAs were stably expressed in these cells. Overexpression of truncated type IB BMP receptor (trBMPR-IB) in 2T3 cells completely blocked BMP-2-induced osteoblast differentiation and mineralized bone matrix formation. Expression of trBMPR-IB also blocked mRNA expression of the osteoblast specific transcription factor, Osf2/ Cbfa1, and the osteoblast differentiation-related genes, alkaline phosphatase (ALP) and osteocalcin (OC). BMP-2-induced ALP activity could be rescued by transfection of wild-type (wt) BMPR-IB into 2T3 clones containing trBMPR-IB. Expression of a constitutively active BMPR-IB (caBMPR-IB) induced formation of mineralized bone matrix by 2T3 cells without addition of BMP-2. In contrast, overexpression of trBMPR-IA blocked adipocyte differentiation and expression of caBMPR-IA induced adipocyte formation in 2T3 cells. Expression of the adipocyte differentiation-related genes, adipsin and PPARgamma, correlated with the distinct phenotypic changes found after overexpression of the appropriate mutant receptors. These results demonstrate that type IB and IA BMP receptors transmit different signals to bone-derived mesenchymal progenitors and play critical roles in both the specification and differentiation of osteoblasts and adipocytes.

Crystal structure of apo-cellular retinoic acid-binding protein type II (R111M) suggests a mechanism of ligand entry.

The crystal structure of unliganded mutant R111M of human cellular retinoic acid-binding protein type II (apo-CRABPII (R111M)) has been determined at 2.3 A and refined to a crystallographic R-factor of 0. 18. Although the mutant protein has lower affinity for all-trans-retinoic acid (RA) than the wild-type, it is properly folded, and its conformation is very similar to the wild-type. apo-CRABPII (R111M) crystallizes in space group P1 with two molecules in the unit cell. The two molecules have high structural similarity except that their alpha2 helices differ strikingly. Analyses of the molecular conformation and crystal packing environment suggest that one of the two molecules assumes a conformation compatible with RA entry. Three structural elements encompassing the opening of the binding pocket exhibit large conformational changes, when compared with holo-CRABPII, which include the alpha2 helix and the betaC-betaD and betaE-betaF hairpin loops. The alpha2 helix is unwound at its N terminus, which appears to be essential for the opening of the RA-binding pocket. Three arginine side-chains (29, 59, and 132) are found with their guanidino groups pointing into the RA-binding pocket. A three-step mechanism of RA entry has been proposed, addressing the opening of the RA entrance, the electrostatic potential that directs entry of RA into the binding pocket, and the intramolecular interactions that stabilize the RA.CRABPII complex via locking the three flexible structural elements when RA is bound.

Human carboxyhemoglobin at 2.2 A resolution: structure and solvent comparisons of R-state, R2-state and T-state hemoglobins.

The three-dimensional structure and associated solvent of human carboxyhemoglobin at 2.2 A resolution are compared with other R-state and T-state human hemoglobin structures. The crystal form is isomorphous with that of the 2.7 A structure of carboxyhemoglobin reported earlier [Baldwin (1980). J. Mol. Biol. 136, 103-128], whose coordinates were used as a starting model, and with the 2.2 A structure described in an earlier report [Derewenda et al. (1990). J. Mol. Biol. 211, 515-519]. During the course of the refinement, a natural mutation of the alpha-subunit, A53S, was discovered that forms a new crystal contact through a bridging water molecule. The protein structure shows a significant difference between the alpha and beta heme geometries, with Fe-C-O angles of 125 and 162 degrees, respectively. The carboxyhemoglobin is compared with other fully ligated R-state human hemoglobins [Baldwin (1980). J. Mol. Biol. 136, 103-128; Shaanan (1983). J. Mol. Biol. 195, 419-422] with the R2-state hemoglobin [Silva et al. (1992). J. Biol. Chem. 267, 17248-17256] and with T-state deoxyhemoglobin [Fronticelli et al. (1994). J. Biol. Chem. 269, 23965-23969]. The structure is similar to the earlier reported R-state structures, but there are differences in many side-chain conformations, the associated water structure and the presence and the position of a phosphate ion. The quaternary changes between the R-state carboxyhemoglobin and the R2-state and T-state structures are in general consistent with those reported in the earlier structures. The location of 238 water molecules and a phosphate ion in the carboxyhemoglobin structure allows the first comparison of the solvent structures of the R-state and T-state structures. Distinctive hydration patterns for each of the quaternary structures are observed, but a number of conserved water molecule binding sites are found that are independent of the conformational state of the protein.

Age-associated reductions in cardiac beta1- and beta2-adrenergic responses without changes in inhibitory G proteins or receptor kinases.

While an age-associated diminution in myocardial contractile response to beta-adrenergic receptor (beta-AR) stimulation has been widely demonstrated to occur in the context of increased levels of plasma catecholamines, some critical mechanisms that govern beta-AR signaling must still be examined in aged hearts. Specifically, the contribution of beta-AR subtypes (beta1 versus beta2) to the overall reduction in contractile response with aging is unknown. Additionally, whether G protein-coupled receptor kinases (GRKs), which mediate receptor desensitization, or adenylyl cyclase inhibitory G proteins (Gi) are increased with aging has not been examined. Both these inhibitory mechanisms are upregulated in chronic heart failure, a condition also associated with diminished beta-AR responsiveness and increased circulatory catecholamines. In this study, the contractile responses to both beta1-AR and beta2-AR stimulation were examined in rat ventricular myocytes of a broad age range (2, 8, and 24 mo). A marked age-associated depression in contractile response to both beta-AR subtype stimulation was observed. This was associated with a nonselective reduction in the density of both beta-AR subtypes and a reduction in membrane adenylyl cyclase response to both beta-AR subtype agonists, NaF or forskolin. However, the age-associated diminutions in contractile responses to either beta1-AR or beta2-AR stimulation were not rescued by inhibiting Gi with pertussis toxin treatment. Further, the abundance or activity of beta-adrenergic receptor kinase, GRK5, or Gi did not significantly change with aging. Thus, we conclude that the positive inotropic effects of both beta1- and beta2-AR stimulation are markedly decreased with aging in rat ventricular myocytes and this is accompanied by decreases in both beta-AR subtype densities and a reduction in membrane adenylate cyclase activity. Neither GRKs nor Gi proteins appear to contribute to the age-associated reduction in cardiac beta-AR responsiveness.

Conformation of the sebacyl beta1Lys82-beta2Lys82 crosslink in T-state human hemoglobin.

The crystal structure of human T state hemoglobin crosslinked with bis(3,5-dibromo-salicyl) sebacate has been determined at 1.9 A resolution. The final crystallographic R factor is 0.168 with root-mean-square deviations (RMSD) from ideal bond distance of 0.018 A. The 10-carbon sebacyl residue found in the beta cleft covalently links the two betaLys82 residues. The sebacyl residue assumes a zigzag conformation with cis amide bonds formed by the NZ atoms of betaLys82's and the sebacyl carbonyl oxygens. The atoms of the crosslink have an occupancy factor of 1.0 with an average temperature factor for all atoms of 34 A2. An RMSD of 0.27 for all CA's of the tetramer is observed when the crosslinked deoxyhemoglobin is compared with deoxyhemoglobin refined by using a similar protocol, 2HHD [Fronticelli et al. J. Biol. Chem. 269: 23965-23969, 1994]. Thus, no significant perturbations in the tertiary or quaternary structure are introduced by the presence of the sebacyl residue. However, the sebacyl residue does displace seven water molecules in the beta cleft and the conformations of the beta1Lys82 and beta2Lys82 are altered because of the crosslinking. The carbonyl oxygen that is part of the amide bond formed with the NZ of beta2Lys82 forms a hydrogen bond with side chain of beta2Asn139 that is in turn hydrogen-bonded to the side chain of beta2Arg104. A comparison of the observed conformation with that modeled [Bucci et al. Biochemistry 35:3418-3425, 1996] shows significant differences. The differences in the structures can be rationalized in terms of compensating changes in the estimated free-energy balance, based on differences in exposed surface areas and the observed shift in the side-chain hydrogen-bonding pattern involving beta2Arg104, beta2Asn139, and the associated sebacyl carbonyl group.

Alpha-subunit oxidation in T-state crystals of a sebacyl cross-linked human hemoglobin with unusual autoxidation properties.

In the crystal structure of human T-state hemoglobin with a sebacyl residue cross-linking the two beta-subunit Lys82,s (DecHb), the Fe atoms of the alpha-subunit hemes are found to be oxidized with a water molecule bound. The three-dimensional structure and heme geometries were compared to those of deoxyhemoglobin and other partially and fully oxidized hemoglobins [R. Liddington, Z. Derewenda, E. Dodson, R. Hubbard, G. Dodson, J. Mol. Biol. 228 (1992) 551]. The heme geometries of the alpha-subunits are consistent with those observed in oxidized structures. The proximal histidines of the alpha-subunits move toward the heme plane shifting the F-helix and FG-corner in a manner observed for partially oxidized human hemoglobin. This supports the hypothesis that these perturbations may precede the T- to R-state transition. Circular dichroism studies comparing DecHb and natural human hemoglobin in the deoxy and CO ligated forms confirm that the conformations of the deoxy forms are identical, but the ligated forms have slight differences in the solution structures. DecHb is found to be more resistant to autoxidation than natural hemoglobin. The time course of autoxidation of DecHb shows that it is virtually absent for the first 1500 min followed by a rapid increase. Thus, the discovery of the oxidation of the alpha-subunits in the deoxy-crystals is quite unexpected. The data confirm that ligation of the alpha-subunits precedes that of the beta-subunits. This may suggest a low ligand affinity of the alpha-diligated form of hemoglobin.

Effects of relative humidity and applied force on atomic force microscopy images of the filamentous phage fd.

The filamentous phage fd was studied by both contact- and tapping-mode atomic force microscopy under conditions of controlled variations in relative humidity and changes in the applied tip force. By spin-coating freshly cleaved mica with phage containing solutions having very low salt content followed by rapid humidity control, stable and reliable sample preparation was achieved. The apparent height of the phage varied by about 10-fold with a quadratic dependence on the stabilized relative humidity, extrapolating to 73% of the accepted X-ray diffraction-based height at 0% relative humidity. The variation in measured height with relative humidity largely reconciles previous widely varying atomic force microscopy estimates of this dimension for the filamentous phage. Our finding that contact-mode images of phage are more difficult to analyze than those acquired in tapping mode are consistent with previously published results on other biological specimens such as DNA.

The effect of bu shen sheng xue fang on beta-thalassemia at gene level.

Isoelectrofocusing electrophoresis, PCR-SSCP analysis with DNA direct sequencing and RT-PCR were used to analyze the constituents of hemoglobin, determine the pattern of globin gene mutation, and detect its mRNA transcription extent. The results indicated that Bu Shen Sheng Xue Fang (BSSXF) significantly increased the contents of hemoglobin (Hb) and fetal hemoglobin (HbF), and the chain ratio in patients with beta-thalassemia. The drug is markedly effective in heterozygous, but not effective in homozygous or genetic compound patients. It was suggested that the beta-globin gene defects were compensated by elevating the gamma/beta + gamma ratio, promoting the transcription and expression of gamma-globin gene in synthesizing HbF.

[Circulating ICAM-1 in sera of renal allograft recipients in monitoring of acute rejection].

OBJECTIVE: To study the significance of ICAM-1 in sera of renal allograft recipients in mornitoring of acute rejection. METHODS: The levels of circulating intercellular adhesion molecule-1 (cICAM-1) in sera of 87 renal transplantation recipients, including 29 patients with acute rejection and 8 patients with cyclosprine nephrotoxicity were measured by sandwich-ELISA. RESULTS: The levels of cICAM-1 were elevated transiently after transplantation. They decreased and reached normal levels within 2 weeks. In the acute rejection episode, the levels of cICAM-1 were significantly higher than those of patients with stable graft function and cyclosporine nephrotoxicity (P < 0.001). The increase of cICAM-1 was seen 1-3 days early than the diagnosis was made. On successful antirejection therapy, the levels of cICAM-1 decreased to normal, but in patients with steroid-resistant rejection, it did not decrease. CONCLUSION: The cICAM-1 in sera can be used to estimate the graft function, and it is also useful in prediction and differential diagnosis of acute rejection and prognosis.

[Detection of Epstein-Barr virus subtypes in Hodgkin's disease].

OBJECTIVE: To investigate Epstein-Barr virus (EBV) subtypes in Chinese patients with Hodgkin's disease (HD). METHODS: 113 cases of HD patients with EBV latent membrane protein 1 (LMP-1) and/or EBV encoded small RAN (EBER-1) positive and throat washings from 21 healthy persons were studied for EBV subtypes by polymerase chain reaction (PCR). RESULTS: 68 HD cases exhibited specific bands, in which 62 cases were EBV type A and 6 cases were EBV type B. 13 of the control washing gave rise to specific bands, of which 11 were EBV type A and 2 of EBV type B. CONCLUSION: The results showed that EBV type A is predominant in Chinese HD patients and healthy individuals.

[Fate of massive retrieved human bone allografts].

OBJECTIVES: To study retrieved massive human allograft specimen after they had been in situ for four to sixty-five months, and to investigate the changes union, bone replacement and degradation of articular cartilage. METHOD: BMP gene expression in the specimen was also studied with in situ hybridization, immunohistochemical methods, and the observations were correlated with the clinical and isotope scan date. RESULT: The union between the allograft and the host was achieved by the formation of an external callus derived from the cortex of the host. New bone replacement took place slowly. Analysis of the articular cartilage revealed evidence that some chondrocytes had survived in non bearing area of the articular. The expression of BMP in the surface of the cortex and united callus was positive 4 months after implantation. But it was negative in the center of cortex after it had been implanted for over 12 months. An external callus formed from the cortex of the host. Some chondrocytes survived in non-bearing area of the articular. Neovasculature penetrated into osteoarticular. CONCLUSION: Bone absorption may be the cause of collapse of articular surface. The results suggest that large allografts in humans are osteo-conductive rather than inductive. Incomplete repair of osteon may be related to slowed replacement of massive human allograft.
