[Study on inhibition and prevention of tumor and antioxidative effects of lithium carbonate in tumor bearing mice].

Two Kinds of tumor-bearing mice (hepatoma H22 and sarcoma S180) were administered with lithium carbonate (Li2CO3) for 17 or 10 days (advanced and simultaneous administration), in order to observe the effects of prevention and treatment of Li2CO3 on malignant tumor, as well as the relationship between Li2CO3 and lipid peroxidation in tumor-bearing mice. Meanwhile, we compared the toxic and side effects of cyclophosphamide (CP) with that of Li2CO3. The results showed that Li2CO3 had no significant toxic or side effects with the suggested doses. In the tests of inhibition and prevention of tumor, Li2CO3 could significantly inhibit the grouth of the two kinds of tumor, and increase the activity of superoxide dismutage (SOD) and decrease the contents of Malonyldialdehyde (MDA). In addition, Li2CO3 had no effect on the white blood cells (WBC) and decreased the micronucleus frequency (MNF) in bone marrow polychromatic erythrocytes (PCE), while CP had definite effect of decreasing the WBC and increasing the MNF in the tumor-bearing mice.

[Antenatal treatment of maternal-fetal Rh incompatibility hemolysis disease].

OBJECTIVE: To study the effective antenatal treatment of maternal-fetal Rh incompatibility hemolysis disease to improve fetal outcome. METHODS: Besides therapy of plasmapheresis, the immune globulin intravenous infusion (IVIg) was added for antenatal treatment of fetal hemolysis disease due to Rh incompatibility. Every 7-20 days 10 g IVIg was administered according to the titer of Rh antibody in maternal serum. RESULTS: 5 Rh negative pregnant women with poor obsterical history were treated with this protocol. Total IVIg infusion was 12 times with an average of 2.4 times per case, and there was no side effects. Plasmapheresis was used 18 times totally with an average of 3.6 per case. There was no fetal death. All the five newborns underwent exchange transfusion and IVIg therapy, and all were survival. CONCLUSIONS: IVIg may enter fetal blood circulation via placenta, and adjust fetal immune ability to reduce the degree of fetal hemolysis.

[Enhanced antitumor effect of suicide gene therapy by SCF, GM-CSF gene transfer in vivo and its immunological mechanism].

OBJECTIVE: To explore an efficient approach to enhance antitumor effect of suicide gene therapy by improving in vivo antigen presenting function and their immunological mechanisms. METHODS: The CT26 colon adenocarcinoma was treated with CD/5FC system combined with the mGM-CSF or/and mSCF gene transfection, and the antitumor effects were evaluated by the tumor growth, tumor-free mouse rate, splenic CTL activity and cytokine expression of tumor milieu. RESULTS: The tumor burden was reduced significantly and higher survival rate was obtained after the combined treatment. It was found that some cytokines(including mIL-4, mIL-2, mIFN-gamma and mTNF-alpha) were expressed in the tumor milieu after mGM-CSF or/and mSCF gene transfection following CD/5FC treatment but not in the control groups. The cytotoxic activity against CT26 cells of spleen cells of the treated mice was significantly incressed. CONCLUSION: The combination of mSCF or/and mGM-CSF gene transfection enhances the antitumor effect of the suicide gene therapy. Such an enhancement is associated with the induction of specific antitumor immune response and secretion of cytokines in tumor milieu.

[Study on the correlation between CT features and expression of p53 protein and PCNA in peripheral lung cancer].

OBJECTIVE: To investigate the correlation between CT features and expression of p53 protein and PCNA in peripheral lung cancer. METHODS: The expression of p53 protein and PCNA was examined by means of SP immunohistochemical technique in 32 cases of peripheral lung cancer proved by pathology; the relationship between the results and preoperative CT features was analyzed retrospectively. RESULTS: The expression of p53 protein and PCNA was correlated with tumor size, deep lobulation, frank cavitation, pleural indentation and mediastinal lymph node metastasis, but not with spiculation. CONCLUSION: The CT features of peripheral lung cancer with diameter more than 3 cm, deep lobulation, frank cavitation, pleural indentation or mediastinal lymph node metastasis indicate higher degree of malignancy and proliferative activity of tumor cells.

[Effect of the soluble immunocomplexes on the pathogenesis of hemorrhagic fever with renal syndrome].

OBJECTIVE: To determine the effect of the soluble immunocomplexes on the pathogenesis of hemorrhagic fever with renal syndrome (HFRS). METHODS: The soluble immunocomplex (IC) consisting of HFRS virus antigen and antibody was prepared previously in vitro, and was intravenously injected into BALB/c mice. The localization of IC in the tissues of mice was investigated by immunohistochemical method ABC. RESULTS: The results showed that 24 hours after injection, ICs were localized mostly in the blood vessels, such as the liver sinusoids, the splenic sinuses, the blood vessels of the mucosa and submucosa of the intestine, and the capillaries of the renal glomeruli, etc. 48 hours later or more, IC localized primarily on the parachymal cells, e.g., the hepatocytes, the epithelial cells of the renal tubules as well as some epithelial cells of the ducts and alveoli of some glands (submandular glands, pancreas). The cells degenerated in the areas with IC, especially in the hepatocytes and epithelial cells of renal tubules. These lesions were very similar to the pathological changes of the HFRS' patients, and were not found in patients injected with other IC. CONCLUSION: The soluble IC of HFRS can cause direct tissue injury in the HFRS.

[Brain cell apoptosis after cerebral hypoxia-ischemia in neonatal rat].

OBJECTIVE: To investigate action of apoptosis in mechanisms of neonatal hypoxia-ischemia (HIE). METHODS: Using HE staining, electronmicroscope, and terminal deoxynuleotidyl transferase-mediated dUTP nick end labeling (TUNEL) technique, we observed the histological features, time course and density of apoptotic cells and compared the ipsilateral hemisphere after permanent ischemia 1, 4, 18, 24, 40, 72 h, 7 days after 3 h hypoxia with that of sham-group. RESULTS: Neurons in the cortex, hippocampus, thalamus exhibited cells shrink, chromatin condensation, apoptosis bodys under microscope, as demonstrated by in situ labeling of DNA breaks. The peak for apoptosis was shown at 18 h in the cortex. CONCLUSION: In the HIE, apoptosis appears earlier than necrosis, and both of the different cell death forms may exist induced dose relativity.

[Facial nerve regeneration through autologous non-nerve grafts].

This study investigated the rabbits facial nerve regeneration over a 8 mm transectional gap of the bucal branches through autologous degenerated skeletal muscle and autologous vein respectively. At 20 weeks following the implantation of the mentioned non-neural materials, all the nerve defects of the experimented animals were repaired. In the electrophysiologic indices and morphometeric analysis, the differences between the respective experimental group and the nerve autografts group had no statistical significance. The results indicated that the non-neural tissues could guide facial nerve regenerating effectively, which suggested that they can be clinically practicable.

[Microbiology of acute pharyngitis and acute tonsillitis and treatment with Taileqi troches].

A qualitative analysis of microflora from pharynx of 100 cases of acute pharyngitis and acute tonsillitis was performed. These patients were randomly divided into two groups so as to assess treatment effect of Taileqi troches vs Amoxycillin. Results showed that beta-hemolytic streptococcus, Klebsiella pneumoniae, staphylococcus aureus were the most common organisms causing diseases. The predominant anaerobic organisms were bacteroides melaninogenicus, eptostrep. These findings suggest that anaerobic bacteria may play a pathogenic role in infection of the pharynx. The effective rate of treatment with troche alone were 86 percent, roughly consistent with Amoxycillin group. No adverse effect was observed.

[Pulmonary complications occurring after allogeneic bone marrow transplantation].

OBJECTIVE: To explore the risk factors for and pathogenesis of pulmonary complications (PC) occurred after allogeneic bone marrow transplantation (allo-BMT). METHODS: The PC in 185 patients undergone allo-BMT were analyzed. RESULTS: Ninety-three PC episodes were observed in 89 patients and most of them were due to infections, including bacterial pneumonia (n = 27), interstitial pneumonia (n = 7), pulmonary fungus disease (n = 16), tuberculosis (n = 4), obstructive lung disease (n = 2), pulmonary edema (n = 2), lung abscess (n = 1) and 34 episodes caused by two or more pathogens. The overall mortality for PC was 12.43% (23/185). CONCLUSION: The risk factors for PC occurred after allo-BMT were not related to age and sex of recipients, bone marrow status before BMT, conditioning regimen and pulmonary function. Graft-versus-host-disease significantly increased the morbidity and mortality of PC after allo-BMT.

Phrenic nerve conduction and diaphragmatic motor evoked potentials: evaluation of respiratory dysfunction.

OBJECTIVE: To investigate preliminarily the value of phrenic nerve conduction (PNC) and diaphragmatic motor evoked potentials (MEPs) in the evaluation of various respiratory dysfunction (RDF). METHODS: Thirty-four patients with various RDF, (19 patients with neurogenical diseases and 15 patients with respiratory disorders) were investigated. Fifty healthy volunteers served as controls. The phrenic nerve was cutaneously stimulated by electrical pulse current at the midpoint of the posterior border of the sternomastoid muscle, and the diaphragmatic muscle compound action potentials (DCAP) were recorded between the 7th and 8th intercostal space and xiphoid process. When the magnetic transcranial stimulation (MTS) of the cortex was given, the recordings were made under the condition of maximal deep inspiration. RESULTS: All patients with myopathies had normal PNC. The patients with Guillain Barre syndrome (GBS), hereditary motor and sensory neuropathy (HMSN) and myasthenic crisis had abnormal PNC. The findings in PNC studies remarkably correlated with RDF, while serial examinations were performed in the patients with GBS and myasthenia gravis (MG). In 7 patients with sleep apnea syndrome (SAS), 4 had abnormal PNC, and 2 of 3 patients with chronic obstructive pulmonary diseases (COPD), and 1 of 5 patients with chest tightness or breathlessness on the supine position showed decreased amplitude. When MEPs were recorded, 3 of 5 patients showed abnormal SAS (1 had no response, 2 lower amplitude). Three patients with COPD had normal MEP. CONCLUSIONS: PNC studies could not only evaluate neuromuscular RDF and predict the outcome of diseases, but also supply additional information about diaphragmatic dysfunction for the RDF caused by respiratory disorders. The results of PNC and diaphragmatic MEP may differentiate the types of SAS.

Androgen receptor in primary hepatocellular carcinoma and its clinical significance.

OBJECTIVE: To assess retrospectively the clinical value of androgen receptor (AR) levels in primary hepatocellular carcinoma (HCC) as a prognostic factor of the disease. METHODS: Fresh HCC tissue and the surrounding liver tissue were obtained surgically from 32 patients with HCC, and preserved in liquid nitrogen. The levels of AR in all specimens were determined by radio-ligand binding assay. RESULTS: The median level of AR was 42.8 fmol/mg protein in the cancerous tissue and 48.3 fmol/mg protein in the surrounding non-cancerous liver tissue. The overall survival rate of the patients with AR < 30 fmol/mg protein in either HCC or the non-cancerous liver was significantly higher than that of the patients with AR > or = 30 fmol/mg protein (P < 0.05 and P < 0.01, respectively). The relative risk on prognosis was 3.27 (P < 0.01) for AR level in HCC and 6.06 (P < 0.001) for AR level in the non-cancerous tissue. The main prognostic factors except the tumor size were not different between the group with higher AR level and that with lower AR level in HCC. The AR level in HCC had a positive correlation with the tumor size (r = 0.44, P < 0.05). CONCLUSIONS: AR can be detected in HCC and the AR status might be a prognostic parameter that provides additional predictive information on the survival. Different AR status might define a real difference of biological characteristics between HCCs.

[Evaluation of accuracy of Healthdyne Nightwatch to monitor sleep breathing disorder].

OBJECTIVE: To study the advantage of computerized portable polysomnograph (Healthdyne Night Watch, NW) compared to standard paper scoring polysomnograph (PSG). METHOD: NW was used to monitor the same patient with sleep apnea syndrome at the same night and compared with standard PSG. RESULT: Apnea and hypopnea index (AHI) recorded by PSG was 28.6 +/- 6.9 versus 28.7 +/- 7.0 recorded by NW. Mean lowest SaO2 recorded by PSG was 73.5% +/- 2.8% versus 74.5% +/- 3.2% recorded by NW. Using a PSG value of AHI > or = 5 and > or = 10 as the diagnostic criteria, the diagnostic agreement of NW was 93.7% and 95.3%, respectively. CONCLUSION: The computerized portable polysomnograph (NW) saves time and labour. It could be used in patient's home or ward and could replace PSG in most situations.

[Auto CPAP Set system for diagnosis and therapy of sleep breathing disorders].

OBJECTIVE: To compare the Auto CPAP Set system with standard polysomnography (PSG) in diagnosis and therapy of sleep breathing disorders. METHOD: Two studies were completed. The Auto CPAP Set system was compared with a simultaneously obtained PSG in eleven patients with sleep breathing disorders. Twenty-three additional patients with obstructive sleep apnea syndrome (OSAS) were treated by Auto CPAP Set. Student's t test was employed to evaluate statistical significance. RESULT: No differences were found between these two systems in AHI, SaO2 and the longest apnea duration (P > 0.05), and the Auto CPAP Set system had a sensitivity of 70% and a specificity 100% for diagnosis of OSAS. AHI decreased and SaO2 increased significantly in OSAS patients after treatment. CONCLUSION: The Auto CPAP Set system provides an accurate and simple method for diagnosis and therapy of sleep breathing disorders, and the time required for analysis of test results was shorter as compared with PSG.

[Autonomic nervous activities in patients with obstructive sleep apnea before and during continuous positive airway pressure treatment].

OBJECTIVE: Heart rate power spectral analysis (HRPSA) can provide information of sympathetic and vagus activities. The aim of the study was to investigate nocturnal variations of autonomic nervous system (ANS) before and during continuous positive airway pressure (CPAP) treatment in patients with OSA. METHOD: 56 subjects with severe OSA were recruited to undergo 7-hour nocturnal polysomnography and ECG monitoring in sleep lab before and during CPAP treatment. 30 subjects without OSA were included as controls. Subjects were excluded if they were receiving medications known to influence ANS activities. ANS was evaluated by HRPSA. RESULT: In addition to repeated sleep apneas and oxygen desaturations, mean areas of LF (1 383 +/- 305) and HF (662 +/- 190) in OSA subjects were significantly higher than those in the controls (LF 125 +/- 64, P < 0.001; HF 163 +/- 78, P < 0.001). With effective CPAP treatment, LF (221 +/- 81) and HF (221 +/- 108) decreased remarkably (P < 0.001, separately) and reached the levels of controls (P > 0.05, separately). CONCLUSION: The results suggested that nocturnal ANS activities are abnormal in patients with OSA, especially the enhancement of sympathetic nervous activity. Fluctuation of ANS induced by apnea and oxygen desaturation may explain the association between OSA and cardiovascular diseases. CPAP treatment is effective in improving ANS activities by way of eliminating apnea and reversing hypoxemia.
