Mechanism of inhibition of benzo[a]pyrene-induced forestomach cancer in mice by dietary curcumin.

Curcumin (diferuloylmethane), the major yellow pigment in turmeric, has been shown to inhibit benzo[a]pyrene (BaP)-induced forestomach cancer in mice through mechanism(s) not fully understood. It is well known that while cytochrome P4501A1 (CYP1A1) and epoxide hydrolase (EH) are important in the conversion of BaP to its activated form, (+)-anti-7,8-dihydroxy-9,10-oxy-7,8,9,10-tetrahydrobenzo[a]pyrene [(+)-anti-BaPDE], the detoxification of (+)-anti-BaPDE is accomplished by glutathione (GSH) S-transferases (GST). Therefore, it seems reasonable to postulate that curcumin may exert anti-carcinogenic activity either by inhibiting activation of BaP or (and) by enhancing the detoxification of (+)-anti-BaPDE. Administration p.o. of 2% curcumin in the diet to female A/J mice for 14 days, which has been shown to cause a significant inhibition in BaP-induced forestomach tumorigenesis, resulted in a modest but statistically significant reduction in hepatic ethoxyresorufin O-deethylase (EROD) activity, a reaction preferentially catalyzed by CYP1A1. While EROD activity could not be detected in the forestomach of either control or treated mice, curcumin feeding caused a statistically significant increase (approximately 2.3-fold) in hepatic EH and GST activities. Hepatic and forestomach GSH levels, and forestomach EH and GST activities were not affected by curcumin treatment. Even though the levels of various hepatic GST isoenzymes were significantly increased upon curcumin feeding, maximum induction was noticed for the pi class isoenzyme (mGSTP1-1), which among murine hepatic GSTs is highly efficient in the detoxification of (+)-anti-BaPDE. In conclusion, the results of the present study suggest that curcumin may inhibit BaP-induced forestomach cancer in mice by affecting both activation as well as inactivation pathways of BaP metabolism in the liver.

Specificities of human glutathione S-transferase isozymes toward anti-diol epoxides of methylchrysenes.

The specificities of human glutathione (GSH) S-transferase (GST) isozymes of class alpha (hGSTA1-1), mu (hGSTM1-1) and pi (hGSTP1-1), including the three allelic forms of hGSTP1-1 [hGSTP1-1(I104,A113), hGSTP1-1(V104,A113) and hGSTP1-1(V104,V113)], in catalyzing the GSH conjugation of anti-diol epoxide stereoisomers of 5-methylchrysene (anti-5-MeCDE) have been examined. The specific activities of human GSTs were significantly higher toward (+)-anti-5-MeCDE than toward the (-)-enantiomer of anti-5-MeCDE. All three variants of hGSTP1-1 were significantly more efficient than either hGSTA1-1 or hGSTM1-1 in GSH conjugation of (+)-anti-5-MeCDE. The catalytic efficiencies of hGSTP1-1 variants toward (+)-anti-5-MeCDE were in the order hGSTP1-1(I104,A113) > hGSTP1-1(V104,V113) > hGSTP1-1(V104,A113). The present study suggests that the I104,A113 allele, which is most frequent in human populations, may play a major role in the detoxification of (+)-anti-5-MeCDE. This may point to specificity, because previous studies from our laboratory have shown that the hGSTP1-1(V104,V113) isoform is significantly more efficient than the other two variants of hGSTP1-1 in catalyzing GSH conjugation of (+)-anti-7R,8S-dihydroxy-9S,10R-epoxy-7,8,9,10-tetrahydrobenzo[a]pyrene [(+)-anti-BPDE], the ultimate carcinogen of benzo[a]pyrene. Even though the mechanism of the differences in the activities of hGSTP1-1 variants toward anti-5-MeCDE versus anti-BPDE remains to be elucidated, it seems that the molecular configuration of the diol epoxide is an important determinant of the activity of hGSTP1-1 isoforms toward polycyclic aromatic hydrocarbon diol epoxides.

Functional analysis of the fibrinogen-related scabrous gene from Drosophila melanogaster identifies potential effector and stimulatory protein domains.

The scabrous (sca) gene encodes a secreted dimeric glycoprotein with putative coiled-coil domains N-terminally and a C-terminal region related to the blood clot protein fibrinogen. Homozygous sca mutants have extra bristle organs and rough eyes. We describe a GAL4-based expression system for testing rescue of the sca mutant phenotype by altered SCA proteins and for misexpression. We find that deletion of the fibrinogen-related domain (FReD) greatly decreases SCA function, confirming the importance of this conserved region. SCA function could not be restored by FReDs from human fibrinogen chain genes. However, proteins lacking any FReD still showed some function in both rescue and misexpression experiments, suggesting that putative effector-binding regions lie outside this domain. Consistent with this, proteins expressing only the FReD had no rescuing activity but were recessive negative; i.e., they enhanced the phenotype of sca mutations but had no phenotype in the presence of a wild-type sca allele. This suggests that the FReD contributes to SCA function by binding to other components of the bristle determination pathway, increasing the activity of the linked N-terminal region.

Algebraic reconstruction for magnetic resonance imaging under B0 inhomogeneity.

In magnetic resonance imaging, spatial localization is usually achieved using Fourier encoding which is realized by applying a magnetic field gradient along the dimension of interest to create a linear correspondence between the resonance frequency and spatial location following the Larmor equation. In the presence of B0 inhomogeneities along this dimension, the linear mapping does not hold and spatial distortions arise in the acquired images. In this paper, the problem of image reconstruction under an inhomogeneous field is formulated as an inverse problem of a linear Fredholm equation of the first kind. The operators in these problems are estimated using field mapping and the k-space trajectory of the imaging sequence. Since such inverse problems are known to be ill-posed in general, robust solvers, singular value decomposition and conjugate gradient method, are employed to obtain corrected images that are optimal in the Frobenius norm sense. Based on this formulation, the choice of the imaging sequence for well-conditioned matrix operators is discussed, and it is shown that nonlinear k-space trajectories provide better results. The reconstruction technique is applied to sequences where the distortion is more severe along one of the image dimensions and the two-dimensional reconstruction problem becomes equivalent to a set of independent one-dimensional problems. Experimental results demonstrate the performance and stability of the algebraic reconstruction methods.

Repression of the A8L gene, encoding the early transcription factor 82-kilodalton subunit, inhibits morphogenesis of vaccinia virions.

The vaccinia virus early transcription factor (VETF) is a DNA binding protein comprised of 70- and 82-kDa subunits encoded by the D6R and A8L genes, respectively. A previous investigation suggested a novel role for the 70-kDa subunit in the morphogenesis of vaccinia virus particles. The principal objectives of the present study were to determine if the 82-kDa subunit of VETF is also required for morphogenesis and, if so, whether the block occurs before or after the incorporation of the genome into the assembling virus particle. To address these and other questions, we constructed and characterized a conditionally lethal recombinant vaccinia virus in which the A8L gene is stringently repressed by the Escherichia coli lac operator system. The amount of 82-kDa protein synthesized could be regulated by the amount of inducer: from undetectable to higher than normal levels. Virus replication, as determined by plaque formation or virus yield upon synchronous infection, was dependent on inducer. Nevertheless, de novo synthesis of the 82-kDa subunit was not required for viral early, intermediate, and late gene expression or DNA replication. Overexpression of the A8L gene alone, produced by high concentrations of inducer, inhibited viral late protein synthesis, whereas overexpression of the D6R gene alone or both VETF genes simultaneously had little inhibitory effect. Laser confocal fluorescence and quantitative electron microscopic analyses revealed that immature and DNA-containing intermediate stage particles accumulated in the absence of inducer, indicating that the A8L protein has a role in morphogenesis of the core and subsequent events.

A conserved p38 mitogen-activated protein kinase pathway regulates Drosophila immunity gene expression.

Accumulating evidence suggests that the insect and mammalian innate immune response is mediated by homologous regulatory components. Proinflammatory cytokines and bacterial lipopolysaccharide stimulate mammalian immunity by activating transcription factors such as NF-kappaB and AP-1. One of the responses evoked by these stimuli is the initiation of a kinase cascade that leads to the phosphorylation of p38 mitogen-activated protein (MAP) kinase on Thr and Tyr within the motif Thr-Gly-Tyr, which is located within subdomain VIII. We have investigated the possible involvement of the p38 MAP kinase pathway in the Drosophila immune response. Two genes that are highly homologous to the mammalian p38 MAP kinase were molecularly cloned and characterized. Furthermore, genes that encode two novel Drosophila MAP kinase kinases, D-MKK3 and D-MKK4, were identified. D-MKK3 is an efficient activator of both Drosophila p38 MAP kinases, while D-MKK4 is an activator of D-JNK but not D-p38. These data establish that Drosophila indeed possesses a conserved p38 MAP kinase signaling pathway. We have examined the role of the D-p38 MAP kinases in the regulation of insect immunity. The results revealed that one of the functions of D-p38 is to attenuate antimicrobial peptide gene expression following exposure to lipopolysaccharide.

4 T-fMRI study of nonspatial shifting of selective attention: cerebellar and parietal contributions.

Regional blood oxygenation in the cerebellum and posterior cerebral cortices was monitored with functional magnetic resonance imaging (fMRI) at four Tesla while 16 normal subjects performed three tasks with identical visual stimulation: fixation; attention focused upon either stimulus shape or color and sustained during blocks of trials (sustained attention); and rapid, serial shifts in attention between stimulus shape or color within blocks of trials (shifting attention). The stimuli were displayed centrally for 100 ms followed by a central fixation mark for 900 ms. Each stimulus was either a circle or a square displayed in either red or green. Attention shifting required switching between color and shape information after each target detection and occurred on average once every three seconds. Subjects pressed a response key upon detecting the target; reaction time and response accuracy were recorded. Two protocols for T2*-weighted echo-planar imaging were optimized, one with a surface coil for the cerebellum alone and the other with a volume coil for imaging both cerebellum and posterior brain structures (parietal, occipital, and part of temporal cortices). Because fMRI of the cerebellum is particularly susceptible to cardiac and respiratory fluctuations, novel techniques were applied to isolate brain activation signals from physiological noise. Functional activation maps were generated for contrasts of 1) sustained attention to color minus fixation; 2) sustained attention to shape minus fixation; and 3) shifting attention minus sustained attention (to color and shape; i.e., summed across blocks of trials). Consistent with the ease of these tasks, subjects performed with >80% accuracy during both sustained attention and shifting attention. Analysis of variance did not show significant differences in false alarms or true hits across either attentional condition. A subgroup of subjects whose performance data were recorded during ten minutes of continuous practice did not show significant changes over time. Both contrasts between the conditions of sustained attention to color or to shape as compared with the fixation condition showed significant bilateral activation in occipital and inferior temporal regions (Brodmann areas 18, 19, and 37). The anterior medial cerebellum was also significantly activated ipsilateral to the finger used for responding. The principal comparison of interest, the contrast between the condition of shifting attention and the condition of sustained attention produced significant and reproducible activation: lateral cerebellar hemisphere (ansiform lobule: Crus I Anterior and Crus I Posterior; left Crus I Posterior); cerebellar folium; posterior superior parietal lobule (R and L); and cuneus and precuneus (R and L).

Real-time speech pattern element displays for interactive therapy.

New developments are presented and discussed for the interactive display and separate measurement of frication and nasality in addition to voice quality. Different clinical problems require different therapeutic and quantitative approaches and these are ideally supported by appropriate combinations of speech pattern elements. Particular use has been made of the laryngograph signal in order to solve the problems of analysis and so that the displays can be given both a sense of immediacy and be highly accurate.

Preventive effect of artemether on schistosome infection.

OBJECTIVE: To study the preventive effect of artemether (Art) in protecting the people from schistosome infection during flood fighting in schistosomiasis endemic area of Poyang Lake, Jiangxi Province. METHODS: From mid July to mid August in 1996, the water level in Poyang Lake rose due to torrential rains and 2 embankments, Zhedi and Jiangtongdi, which appeared in dangerous situation and were selected as the pilot spots. After those who went to fight against flood arrived at the pilots their sera were collected within 48 hours and were examined with indirect hemagglutination test (IHA), enzyme-linked immunosorbent assay (ELISA) and McAb-ELISA. Individuals with negative outcome in the 3 tests were then selected as the study subjects and were allocated randomly to the Art or the control group. The first dose of Art given to the individuals contacted with the infested water within 11-15 days was 6 mg/kg. If the individual continually contacted the infested water, the same dose of Art was given once every 15 days. After the individuals withdrew from the pilot, one more dose of Art was administered 7-15 days later. Placebo (starch) was given to individuals in the control group at the same period as in artemether group. Stool examinations were made in both groups 40-50 days after the last medication for evaluation of the preventive effect of artemether. Double blind method was used in the administration of both artemether and placebo. RESULTS: In Zhedi pilot, the individuals fought against flood for about 1 month. In Art group, 99 individuals receiving 3 doses of the drug completed the stool examination with egg-positive rate of 4% and no acute schistosomiasis was seen. In the control group, among 110 people who completed the observation, 44 were egg-positive with an infection rate of 40%, and 29 were identified as having acute schistosomiasis. In Jiangtondi, the studied individuals contacted the infested water for only about 4 hours. But in the control group 4 out of 102 individuals were egg-positive, while none of the 103 individuals in Art group receiving 2 doses of the drug showed schistosome infection. No apparent side effect was seen in the people treated with artemether. CONCLUSION: After oral Art was given to the people fighting against flood in schistosomiasis endemic area of Poyang Lake, it was shown that the oral Art has a promising effect on controlling acute schistosomiasis and reducing the infection rate.

A study on visual evoked potential to show the relationship between the eyes and the twelve regular channels.

The visual electrophysiologic method was used to investigate the relationship between the eyes and the 12 regular channels. The values of the latent period and the amplitude of P100 wave were calculated and compared statistically. The results show that: 1) The eyes and the 12 regular channels are closely related, no matter whether the channels are classically associated with the eyes or not, while the control (non-channel)/ point has no such relationship; 2) the degrees of relationship between the eyes and the 12 regular channels vary significantly; and 3) acupuncture at the 12 regular channels induces different effects on the visual evoked potential that the Urinary Bladder Channel presents a facilitation effect whereas all the other channels chiefly show an inhibitory effect.

A comparative observation on treatment of heart failure due to diastolic insufficiency of the left ventricle--a report of 43 cases.

43 cases of heart failure due to diastolic insufficiency of the left ventricle were treated with TCM differential treatment as the treatment group, the total effective rate being 86.0%. Statistically significant difference (P < 0.05) in the therapeutic effects was found between the treatment group and control (25 cases) group. Improvement of the left ventricular diastole in the treatment group was more obvious than that of the control group (P < 0.01), while contractility of the left ventricle was also improved to a certain extent.

Lidocaine improving the cerebral protection by retrograde cerebral perfusion.

OBJECTIVE: To determine whether lidocaine can improve the cerebral protection provided by retrograde cerebral perfusion. METHODS: Sixteen mongrel dogs were placed on cardiopulmonary bypass and cooled to 20 degrees C. Retrograde cerebral perfusion was then carried out for 120 minutes, with the external jugular venous pressure kept at 3.33 kPa. Cardiopulmonary bypass was resumed, and the animals were rewarmed to 36 degrees C. The animals were divided into two groups. In the lidocaine group (n = 8), lidocaine was administrated continuously throughout the experiment. In the control group (n = 8), normal saline was given at the same rate. RESULTS: In both groups, cerebral tissue creatine phosphate and adenosine triphosphate concentrations and energy charge increased by the end of hypothermic cardiopulmonary bypass, decreased continuously during retrograde cerebral perfusion, and recovered gradually after resuming cardiopulmonary bypass. Nevertheless, they recovered to significantly higher levels in the lidocaine group than in the control group (creatine phosphate: 2.44 +/- 0.53 versus 1.61 +/- 0.49 mumol/g wet weight, P = 0.006; adenosine triphosphate: 0.71 +/- 0.18 versus 0.50 +/- 0.17 mumol/g wet weight, P = 0.029; energy charge: 0.59 +/- 0.10 versus 0.48 +/- 0.09, P = 0.044) by the end of the experiment. There was no significant difference between the two groups in the cerebral tissue water content (control group: 77.6% +/- 1.9%; lidocaine group: 77.6% +/- 1.3%). CONCLUSION: Continuous lidocaine infusion accelerates the recovery of cerebral tissue high energy phosphate contents after resuming cardiopulmonary bypass, but it has no effect on the formation of cerebral edema after retrograde cerebral perfusion.

[Clinical observations on treatment of endometriosis by tonifying kidney and removing blood stasis].

OBJECTIVE: To explore the mechanism of treating endometriosis by tonifying Kidney and removing blood stasis with Neiyixiao Recipe (NYXR). METHODS: One hundred and three patients of endometriosis were divided randomly into the NYXR group (58 cases, treated with NYXR) and the control group (45 cases, treated with danazol) and were reviewed for the improvement of clinical symptoms, serum level of FSH, LH, PRL, E2, P and T, humoral and cellular immunity (C3, C4, CD3, CD4, CD8), and plasma prostaglandin after treatment. RESULTS: There was no significant difference in effect between the NYXR group and the control group in relief of dysmenorrhea and degradation of elevated plasma prostaglandin (chi 2 = 0.71, P > 0.05). But there was significant difference between the two groups in curing infertility (chi 2 = 14.77, P < 0.01), and the effect of NYXR in regulating endocrine and immunity was significantly better than that of danazol. CONCLUSION: By tonifying Kidney and removing blood stasis, NYXR is multi-effective and being one of effective methods in clinical practice for treatment of endometriosis.

[Effects of NM23/NDPK in signal transduction of metastatic mouse forestomach carcinoma cells].

OBJECTIVE: To understand the role of NM23/NDPK in the transmembrane signal transduction proccess in tumor metastasis. METHODS: The NDPK activity, IP3 contents, the invasive potentials of the cells of mouse forestomach carcinoma(MFC) substrains with different metastatic potentials and the effect of LN and EGF were observed. RESULTS: Both the NDPK activity and the contents of IP3 of the cells of the subclone with lower metastatic potential was higher than that with higher metastatic potential. Also, the cells with lower metastatic potential had lower invasive potential and higher responsibility to the challenge of EGF compared with that higher metastatic potential. However, no different effect of LN on the cells of those subclones was found. CONCLUSIONS: NDPK might regulate the metastasis of FMC cells by impacting the G protein efficacy with the selection of exotic signal ligands.

[Enantiomeric high performance liquid chromatographic separation of epinephrine and analogous using chiral mobile phase additives].

The enantiomeric resolution of epinephrine, isoproterenol, ephedrine was studied by reversed phase HPLC. beta-CD, DM-beta-CD, TM-beta-CD were used as chiral mobile phase additives. The effects of the concentration of DM-beta-CD and the concentration of methanol on resolution of D,L-epinephrine were investigated. The results showed: D,L-epinephrine and D,L-isoproterenol can be separated by beta-CD, DM-beta-CD, TM-beta-CD, but D,L-ephedrine can't. Hydrogen bonding plays an important role in the chiral recognition mechanism. In the separation of isoproterenol, the enantioselectivity of TM-beta-CD was better than those of beta-CD and DM-beta-CD. As the concentration of DM-beta-CD changed from 0.04 mmol/L to 1.00 mmol/L, the resolution varied slightly. The experiment of the effect of methanol concentration on Rs showed: when the ratio of methanol/water was 40/60, the Rs reached the maximum. The coefficients of variation of retention time of D-epinephrine and L-epinephrine were 1.3% and 1.4% respectively. It is demonstrated that the chromatographic systems with a dynamically-generated stationary phase with methylated-beta-cyclodextrin are versatile tools for enantiomer separation. This technique leads to column with excellent time stability and good reproducibility of the enantio-selectivity.

[Quality control for huoxiang-zhengqi liquid].

The Rhizoma Atractylodis, Cortex Magnoliae Officinalis and Pericarpium Citri Reticulatae in Huoxiang-zhengqi Liquid were identified by TLC. The total contents of magnolol and honokiol were determined by HPLC to be no less than 1.9 mg/ml, with an average recovery of 104.4% and 103.3%, RSD 1.23% and 0.91% respectively.

[Studies on the crystal structure of ivermectin (H2B1a)].

The crystal structure of ivermectin (H2B1a) was determined by X-ray diffraction. Ivermectin was found to be composed of more than 80% H2B1a and less than 20% H2B1b. The crystal of H2B1a belongs to the space group C2 with a = 40.736(6)A, b = 9.295(3)A, c = 14.966(3)A, beta = 106.93(1) degree. After structure refinement using least-squares method, the final R factor of H2B1a is 0.0658. The space configuration of H2B1a is a long chain along the C-axis. The molecular aggregates are extended into pairs of long chains parallel to each other, some distance apart. The distance between chains is the same, but their direction is opposite. This structure of H2B1a is similar to some biomacromolecules.

[Synthesis and application of novel acrylic resin as enteric coating].

The pH sensitive two- and three-component acrylic resins were synthesized by radical solution copolymerization of methyl methacrylate (MMA), methacrylic acid (MAA) and/or butyl acrylate (BA) and their acid values were measured by acid-base titration. The dissolution of naproxen pellets coated with the synthesized resins in dissolution medium of different pH value was studied. The results show that the acrylic resin with different acid value can be prepared by adjusting the monomer ratios (MMA:MAA or MMA:MAA:BA). The smaller the acid value of the resin is, the higher pH sensitive value it has. The release of naproxen from pellets coated with these resins is greatly influenced by the pH value of the dissolution medium and the pH sensitive value of the coating resin. The three-component resins are plastic and can be used as coating material without adding plastisizers.
