Giant discrete steps in metal-insulator transition in perovskite manganite wires.

Optical lithography is used to fabricate LPCMO wires starting from a single (La(5/8-0.3)Pr(0.3))Ca3/8MnO3 (LPCMO) film epitaxially grown on a LaAlO3(100) substrate. As the width of the wires is decreased, the resistivity of the LPCMO wires exhibits giant and ultrasharp steps upon varying temperature and magnetic field in the vicinity of the metal-insulator transition. The origin of the ultrasharp transitions is attributed to the effect of spatial confinement on the percolative transport in manganites.

[Comparison of IgA/VCA, IgA/EA, IgG/EA in immunoenzyme methods and ZEBRA ELISA in early diagnosis of nasopharyngeal carcinoma.].

BACKGROUND: To develop an ELISA method using Herpesvirus hominis type 4 (EBV) IgG/Zebra as capture antigen for large population screening. METHODS: The ELISA method used purified ZEBRA antigen to detect the IgG/ZEBRA antibody from serum in nasopharyngeal carcinoma (NPC) and normal healthy subjects. RESULTS: Of 288 NPC sera, 262 were detected positive, the sensitivity was 91%, while 5 of 96 normal sera were detected positive, the specificity was 94.8% and the results of NPC group and healthy group displayed significant difference (P less than 0.001). IgA/VCA, IgA/EA, IgG/EA in immunoenzyme methods and ZEBRA ELISA were compared during the NPC screening in two cities: Huizhou, Guangdong and Guiping, Guangxi, 5463 and 2017 samples respectively were tested and 5 earlier NPC patients were found. CONCLUSION: The results indicate that this method has high specificity and sensitivity, and can be used for large population screening to assist early phase NPC diagnosis.

The effect of the water content of acetonitrile on the electrochemistry of ferrocenyl thiol self-assembled monolayers.

A novel kind of ethylene-ferrocenyl dodecyl thiol bearing an electron-withdrawing pyridium group (FcCH=CH-Py(CH2)10SH) was synthesized and mixed with HOOCC(10)SH on an Au electrode to form mixed self-assembled monolayers (SAMs) in ethanol solution. The influence of the water content of acetonitrile on the electrochemistry of the SAMs was studied by cyclic voltammetry and ac voltammetry measurement. The results showed that the current decreased and the oxidation potential of the ferrocene group shifted negatively with increasing water content of acetonitrile.

Potential energy surface for activation of methane by Pt(+): a combined guided ion beam and DFT study.

A guided-ion beam tandem mass spectrometer is used to study the reactions of Pt(+) with methane, PtCH(2)(+) with H(2) and D(2), and collision-induced dissociation of PtCH(4)(+) and PtCH(2)(+) with Xe. These studies experimentally probe the potential energy surface for the activation of methane by Pt(+). For the reaction of Pt(+) with methane, dehydrogenation to form PtCH(2)(+) + H(2) is exothermic, efficient, and the only process observed at low energies. PtH(+), formed in a simple C-H bond cleavage, dominates the product spectrum at high energies. The observation of a PtH(2)(+) product provides evidence that methane activation proceeds via a (H)(2)PtCH(2)(+) intermediate. Modeling of the endothermic reaction cross sections yields the 0 K bond dissociation energies in eV (kJ/mol) of D(0)(Pt(+)-H) = 2.81 +/- 0.05 (271 +/- 5), D(0)(Pt(+)-2H) = 6.00 +/- 0.12 (579 +/- 12), D(0)(Pt(+)-C) = 5.43 +/- 0.05 (524 +/- 5), D(0)(Pt(+)-CH) = 5.56 +/- 0.10 (536 +/- 10), and D(0)(Pt(+)-CH(3)) = 2.67 +/- 0.08 (258 +/- 8). D(0)(Pt(+)-CH(2)) = 4.80 +/- 0.03 eV (463 +/- 3 kJ/mol) is determined by measuring the forward and reverse reaction rates for Pt(+) + CH(4) right harpoon over left harpoon PtCH(2)(+) + H(2) at thermal energy. We find extensive hydrogen scrambling in the reaction of PtCH(2)(+) with D(2). Collision-induced dissociation (CID) of PtCH(4)(+), identified as the H-Pt(+)-CH(3) intermediate, with Xe reveals a bond energy of 1.77 +/- 0.08 eV (171 +/- 8 kJ/mol) relative to Pt(+) + CH(4). The experimental thermochemistry is favorably compared with density functional theory calculations (B3LYP using several basis sets), which also establish the electronic structures of these species and provide insight into the reaction mechanism. Results for the reaction of Pt(+) with methane are compared with those for the analogous palladium system and the differences in reactivity and mechanism are discussed.

Differential-pulse voltammetric determination of trace formaldehyde using magnetic microspheres and magnetic electrode.

A new type of magnetic polymer microsphere containing acylhydrazine groups on the surface was synthesized. They can be reacted with formaldehyde to produce an electroactive adduct. Reduction of these derivatives following aggregation on a magnetic electrode is possible and is effective in the indirect determination of formaldehyde. The experimental conditions and electrode structure are discussed. Under the optimum conditions, it was found that the peak potential (Ep) of formaldehyde is -1.01 V (vs. Ag/AgCl). Formaldehyde in the range 1-1000 micrograms l-1 can be determined. The detection limit for formaldehyde is 0.3 microgram l-1 and the relative standard deviation for the determination of 100 micrograms l-1 formaldehyde was 2.26%. The method was applied to the determination of formaldehyde in environmental samples with satisfactory results.

Muc1 mucins on the cell surface are adhesion sites for Pseudomonas aeruginosa.

Recently, we cloned and characterized a full-length cDNA of the hamster Muc1 gene, the expression of which appears to be associated with secretory cell differentiation (Park HR, Hyun SW, and Kim KC. Am J Respir Cell Mol Biol 15: 237-244, 1996). The role of Muc1 mucins in the airway, however, is unknown. In this study, we investigated whether cell surface mucins are adhesion sites for Pseudomonas aeruginosa. Chinese hamster ovary (CHO) cells not normally expressing Muc1 mucin were stably transfected with the hamster Muc1 cDNA, and binding to P. aeruginosa was examined. Our results showed that 1) stably transfected CHO cells expressed both Muc1 mRNA and Muc1 mucins based on Northern and Western blot analyses, 2) Muc1 mucins present on the cell surface were degraded by neutrophil elastase, and 3) expression of Muc1 mucins on the cell surface resulted in a significant increase in adhesion of P. aeruginosa that was completely abolished by either proteolytic cleavage with neutrophil elastase or deletion of the extracellular domain by mutation. We conclude that Muc1 mucins expressed on the surface of CHO cells serve as adhesion sites for P. aeruginosa, suggesting a possible role for these glycoproteins in the early stage of airway infection and providing a model system for studying epithelial cell responses to bacterial adhesion that leads to airway inflammation in general and cystic fibrosis in particular.

Effects of dexamethasone on Flt3 receptor expression and proliferation induced by recombinant human Flt3 ligand in malignant hematopoietic cells.

AIM: To investigate the effect of dexamethasone (DXM) on the expression of Flt3 receptor and the proliferation mediated by recombinant human Flt3 ligand (rhFL) in leukemia cells. METHODS: Eighteen malignant hematopoietic cell lines and 10 leukemia blasts from leukemia patients were examined by flow cytometry for the expression of Flt3 receptor before and after incubation with DXM 0.1 micromol/L for 24 h. The effect of DXM on the proliferation of malignant hematopoietic cells was measured by MTT assay. RESULTS: (1) Expression of the Flt3 receptor in malignant hematopoietic cell lines and leukemia blasts was widespread and extremely heterogeneous; (2) The presence of receptor on the surface of malignant hematopoietic cell didn't necessarily imply a significant ligand-induced response, at least in terms of proliferation. Conversely, some Flt3 receptor-negative malignant hematopoietic cells responded to rhFL; (3) DXM down-regulated the expression of Flt3 receptor and inhibited the proliferation induced by rhFL in some malignant hematopoietic cell lines and fresh leukemia cells. CONCLUSION: DXM may down-regulate the expression of Flt3 receptor on the surface of malignant hematopoietic cells and inhibit the proliferation induced by rhFL. A combination of rhFL and DXM may serve to control hematopoietic defects in malignant hematopoietic diseases.

[Control system for rotational gamma knife].

The features and composition of Rotational Gamma Knife are mainly introduced in this paper. The open architecture double PLCs and motion controller are used to fit special requirements of the Rotational Gamma Knife and thus to ensure its high reliability. In the end, safety measures which should be taken for patients, doctors and equipments are discussed.

[Study of the antiosteoporosis effects of bushenqianggu capsule].

OBJECTIVE: To study the antiosteoporosis effects of Bushenqianggu capsule. METHOD: Observing the pharmacological effects of Bushenqianggu capsule with blank and positive control, and then analyzing the data with biological statistics. RESULT: The testis and uterus weight index of "the kidney assessed" model mice, weight of thighbone and blood concentration of Ca2+ increased significantly. Meanwhile, the thighbone mass density of castrated rats was improved and the incidence of the thighbone fracture in the osteoporosis model rats induced by retinoic acid was reduced. Besides, it had analgesia and anti-inflammatory effects. CONCLUSION: Bushenqianggu capsule has a good effect on osteoporosis, which provides evidence for clinical use.

[Effects of recombinant human macrophage colony stimulating factor (rhM-CSF) on stromal cell derived during the time of mice colony formation unit-spleen (CFU-S) and its role on CD34+ cells expansion in vitro].

OBJECTIVE: To investigate the effects of stromal cell derived from mice spleen during in vitro expansion of CD34+ hematopoietic stem cells from umbilical cord blood. METHODS: Irradiated and non-irradiated mice at day 10, 12 and 14 were sacrificed and their spleens were taken for collecting spleen cells. The cells were stimulated with recombinant human macrophage colony stimulating factor(rhM-CSF) for collecting adherent spleen derived stroma cells. The effects of spleen derived stromal cells which treated with mitomycin served as a feeder layer, combined with different cytokines on in vitro expansion of purified cord blood CD34+ were assayed. Clonogenic assay and flow cytometry were employed to analyze phenotype of expanded cells and their ability to form different colony in vitro. RESULTS: 1) rhM-CSF efficiently stimulated spleen derived stromal cells to proliferate; 2) The mice stromal cells obtained during the time of colony formation-unit spleen (CFU-S) promoted the CD34+ cells to form colony in semisolid medium and, combined with cytokines, thereby efficiently expanded the hematopoietic cells from cord blood. After two weeks incubation, CD34 positive cells were 100 times more than original cells cultured and expanded cells still have the ability to form multi-lineage colony in vitro. CONCLUSION: rhM-CSF can stimulate the stromal cells derived from CFU-S to proliferate and support the survival and expansion of CD34+ cells separated from cord blood.

[Expression and purification of recombinant human interleukin-11 in Pichia pastoris].

OBJECTIVE: To express recombinant human interleukin-11 (rhIL-11) in methylotropic yeast Pichia pastoris. METHODS: By designing and synthesizing an artificial gene for IL-11, the expression vector pPICZ alpha-A-IL-11 was constructed and introduced into Pichia pastoris by linearized electroporation. The rhIL-11 protein was identified by ELISA and SDS-PAGE analysis. The bioactivity was analyzed by B9-11 cell line. A combination of liquid chromatography was developed to purify the rhIL-11 from ferment supernatant. RESULTS: The nucleotide sequence analysis indicated that the sequence of cloned artificial IL-11 gene accorded with that of designed; the secreted yield of rhIL-11 by yeast Pichia pastoris KM71-2424 in flask reached 60 mg/L. The biological activity of IL-11 in yeast supernatant and E. coli standard determined by B9-11 was 5.5 x 10(7) U/mg and 2.2 x 10(7) U/mg respectively. The rhIL-11 was purified to electrophoretic purity by a combination of liquid chromatography. CONCLUSION: The human IL-11 artificial gene was obtained and successfully expressed in the Pichia pastoris(KM71-2424). The biological activity of IL-11 in yeast supernatant was significantly higher than that of E. coli standard. The rhIL-11 was purified to electrophoretic purity.

[Effects of SDF-1 alpha and its receptor on regulation of hematopoiesis].

Hematopoietic stem/progenitor cell transplantation is one of main choices for treating patients with malignant tumor or heredity diseases in recently year. The homing of hematopoietic stem cells(HSC) is a committed step to ensure the bone marrow hematopoiesis reconstitution. Recent studies have showed that stromal cell derived factor-1 (SDF-1) and its receptor has important effects on proliferation, differentiation, homing of HSC to bone marrow and hematopoiesis reconstitution. The purpose of this review is to give a recent update informations on mechanism of hematopoiesis regulation mediated by SDF-1 and its receptor.

Total synthesis and antitumor activity of 12,13-desoxyepothilone F: an unexpected solvolysis problem at C15, mediated by remote substitution at C21.

A new epothilone analogue, 12,13-desoxyepothilone F (dEpoF, 21-hydroxy-12,13-desoxyepothilone B, 21-hydroxyepothilone D), was synthesized and evaluated for antitumor potential. A convergent strategy employed for the semipractical synthesis of 12,13-desoxyepothilone B (dEpoB) has been utilized to yield an amount of dEpoF sufficient for relevant biological studies. The results from an in vitro assay reveal that this new analogue is highly active against various tumor cell lines with a potency comparable to that of dEpoB. In particular, the growth of resistant tumor cells is inhibited by dEpoF at concentrations where paclitaxel (Taxol) is basically ineffective. A preliminary assessment of its in vivo activity is also promising. The new analogue, containing an additional hydroxyl group at C21, exhibits advantages over other epothilones in terms of water solubility, and can serve as a readily functionalizable handle to produce other useful compounds for pertinent biological studies.

Agonist anti-gp130 transducer monoclonal antibodies are human myeloma cell survival and growth factors.

We have previously reported obtaining two monoclonal antibodies (mAb) against the human gp130 interleukin-6 (IL-6) transducer which made possible the dimerization of gp130 and the activation of several IL-6-driven functions when used together. We report here that these mAb induce gp130-mediated signaling in human myeloma cells and support the survival and the long-term growth of five IL-6-dependent human myeloma cell lines. Their agonist activity is not affected by neutralizing antibodies to IL-6 or IL-6R. These mAb induce a transient proliferation of primary myeloma cells from most patients with multiple myeloma. Again, IL-6 inhibitors do not affect this agonist activity. By using highly purified primary myeloma cells, we found that these anti-gp130 mAb supported the long-term survival of primary myeloma cells from five patients with primary plasma cell leukemia but failed to induce their long-term growth. For patients with fulminant disease and secondary extramedullary proliferation, the antibodies supported a long-term survival and growth, and anti-gp130 mAb-dependent cell lines were obtained. For patients with medullary involvement only, a co-stimulatory signal is necessary, together with gp130 activation, to trigger cell survival and cycling. Leukemia (2000) 14, 188-197.

[Application of modified shuttle like incision in benign skin tumour in the maxillofacial region].

OBJECTIVE: To design a modified shuttle-like incision for benign skin tumours in the maxillofacial region. METHODS: Some merits of the modified shuttle like incision was verified through experiment upon Cadavers and accurate mathematical calculation. Then it was used in the resection of 24 patients with benign skin tumour in the maxillofacial region. RESULTS: If the radius of the tumours were separately 0.5 cm, 1.0 cm and 1.5 cm, the modified shuttle-like incision can save the skin area by 17.22%,15.42% and 16.98%; no(or little) tension was found in sewing. The sutural lines were levelly concealed in the kraissl's lines and no"cat ear"was found after sewing. After 1-7 years of following up, all of the scars of the 24 cases were appraised as good. CONCLUSION: The modified shuttle-like incision was better than the traditional one. It can be performed widely in the clinical operation on benign skin tumours in the maxillofacial region.

Synthesis and structure-activity relationships of potential anticancer agents: alkylcarbamates of 3-(9-acridinylamino)-5-hydroxymethylaniline.

A series of potential 9-anilinoacridine antitumor agents, 3-(9-acridinylamino)-5-hydroxymethylaniline (AHMA) derivatives with monosubstituent at C4' and disubstituents at C4' and C5' of the acridine ring and their alkylcarbamates, were synthesized for evaluation of their antitumor activity. A structure-activity relationship (SAR) study showed that the AHMA-alkylcarbamates were more potent than their corresponding parent AHMA compounds. In addition, the cytotoxicity of the AHMA-alkylcarbamate decreased with increasing length and size of the alkyl function. Among these compounds, AHMA-ethylcarbamate (18) and 4'-methyl-5'-dimethylaminoethylcarboxamido-AHMA-ethylcarb amate (34) possess potent cytotoxicity on the inhibition of human leukemic HL-60 cell growth in culture. Further in vivo studies of these compounds displayed significant anticancer therapeutic effects in mice bearing sarcoma 180, Lewis lung carcinoma, and P388 leukemia.

Induced expression of B7-1 on myeloma cells following retroviral gene transfer results in tumor-specific recognition by cytotoxic T cells.

The aim of this study was to evaluate whether tumor cells from patients with multiple myeloma activate allogeneic and autologous T cells. Results showed that myeloma cells expressed few B7-2 and no B7-1 in six cell lines and primary cells from 11 patients. They expressed substantial levels of HLA class I, CD40, and a set of adhesion molecules. In accordance with the low density of B7 molecules on these cells, they were poor allogeneic CD8+ T cell stimulators. Neither IFN-gamma plus TNF-alpha nor CD40 stimulation significantly induced B7-1 or up-regulated B7-2 on human myeloma cell line or primary myeloma cells from six of seven patients. However, such induction was found on autologous bone-marrow nontumoral cells and on autologous dendritic cells following CD40 stimulation. High B7-1 expression was stably obtained on human myeloma cell line using transduction with a B7-1 retrovirus, enabling these cells to stimulate allogeneic CD8+, though not CD4+, T cell proliferation. For one patient with advanced disease, B7-1 gene transfer made it possible to amplify autologous cytotoxic T cells that killed autologous myeloma cells in an HLA class I-restricted manner, but not autologous PHA blasts. These results suggest that B7-1 gene transfer could be a promising immunotherapeutic approach in multiple myeloma.

An agonist anti-human CD40 monoclonal antibody that induces dendritic cell formation and maturation and inhibits proliferation of a myeloma cell line.

CD40, a 48-50 KD cell membrane molecule, member of the nerve growth factor receptor and tumor necrosis factor receptor superfamily, is an important costimulatory molecule during the immune response. Anti-CD40 monoclonal antibody (MAb) has been shown earlier to costimulate with IgM or phorbol esters resting B cells to proliferate, differentiate, secrete immunoglobulins, and switch isotype. Here we report on an agonistic mouse anti-human CD40 MAb 5C11. The specificity of this MAb was verified by flow cytometry, Western blotting, and competition with anti-CD40 MAb 89. We studied the effects of MAb 5C11 on a multimyeloma cell line, XG2, that expresses the CD40 antigen strongly and found that this MAb caused the homotypic aggregation of XG2, strongly suppressed XG2 proliferation, and led to its apoptosis after 24 hr of treatment. Interestingly, MAb 5C11 also triggered the generation, proliferation, and maturation of dendritic cells from peripheral blood monocytes, either by itself or in combination with GM-CSF and IL-4.
