Effects of stimulus size and eccentricity on horizontal and vertical vergence.

We measured the gain and phase of horizontal and vertical vergences of five subjects as a function of stimulus area and position. Vergence eye movements were recorded by the scleral search coil method as subjects observed dichoptic displays oscillating in antiphase either from side to side or up and down with a peak-to-peak magnitude of 0.5 degree at either 0.1 Hz or 1.0 Hz. The stimulus was a central textured disc with diameter ranging from 0.75 degree to 65 degrees, or a peripheral annulus with outer diameter 65 degrees and inner diameter ranging from 5 degrees to 45 degrees. The remaining field was black. For horizontal vergence at both stimulus frequencies, gain and the phase lag were about the same for a 0.75 degree stimulus as for a 65 degrees central stimulus. For vertical vergence, mean gain increased and mean phase lag decreased with increasing diameter of the central stimulus up to approximately 20 degrees. Thus, the stimulus integration area is much smaller for horizontal vergence than for vertical vergence. The integration area for vertical vergence is similar to that for cyclovergence, as revealed in a previous study. For both types of vergence, response gains were higher and phase lags smaller at 0.1 Hz than at 1.0 Hz. Also, gain decreased and phase lag increased with increasing occlusion of the central region of the stimulus. Vergence gain was significantly higher for a 45 degrees central disc than for a peripheral annulus with the same area. Thus, the central retina has more power to evoke horizontal or vertical vergence than the same area in the periphery. We compare the results with similar data for cyclovergence and discuss their ecological implications.

RESEARCH: Projected Climate Change Effects on Winterkill in Shallow Lakes in the Northern United States.

/ Each winter, hundreds of ice-covered, shallow lakes in the northern United States are aerated to prevent winterkill, the death of fish due to oxygen depletion under the ice. How will the projected climate warming influence winterkill and the need to artificially aerate lakes? To answer this question, a deterministic, one-dimensional year-round water quality model, which simulates daily dissolved oxygen (DO) profiles and associated water temperatures as well as ice/snow covers on lakes, was applied. Past and projected climate scenarios were investigated. The lake parameters required as model input are surface area, maximum depth, and Secchi depth as a measure of radiation attenuation and trophic state. The model is driven by daily weather data. Weather records from 209 stations in the contiguous United States for the period 1961-1979 were used to represent past climate conditions. The projected climate change due to a doubling of atmospheric CO(2) was obtained from the output of the Canadian Climate Center General Circulation Model. To illustrate the effect of projected climate change on lake DO characteristics, we present herein DO information simulated, respectively, with inputs of past climate conditions (1961-1979) and with a projected 2 x CO(2) climate scenario, as well as differences of those values. Specific parameters obtained were minimum under-ice and lake bottom DO concentration in winter, duration of under-ice anoxic conditions (<0.1 mg/liter) and low DO conditions (<3 mg/liter), and percentage of anoxic and low DO lake volumes during the ice cover period. Under current climate conditions winterkill occurs typically in shallow eutrophic lakes of the northern contiguous United States. Climate warming is projected to eliminate winterkill in these lakes. This would be a positive effect of climate warming. Fish species under ice may still experience periods of stress and zero growth due to low DO (<3 mg/liter) conditions under projected climate warming.

Depth selectivity of vertical fusional mechanisms.

We measured the ability to fuse dichoptic images of a horizontal line alone or in the presence of a textured background with different vertical disparity. Nonius-line measurements of vertical vergence were also obtained. Diplopia thresholds and vertical vergence gains were much higher in response to an isolated vertically disparate line than to one with a zero vertical-disparity background. The effect of the background was maximum when it was coplanar with the target and decreased with increasing relative horizontal disparity. We conclude that vertical disparities are integrated over a restricted range of horizontal disparities to drive vertical vergence.

Alterations in retinoid X receptor-alpha expression contribute to cell-type dependent differences in thyroid hormone regulation of malic enzyme transcription.

Triiodothyronine (T3) stimulates a marked increase (> 40-fold) in transcription of the malic enzyme gene in chick embryo hepatocytes (CEH), but has no effect on malic enzyme transcription in chick embryo fibroblasts (CEF) that express nuclear T3 receptors (TR) at levels which are similar to those of CEH. Heterodimerization of the TR with other nuclear proteins is a potential mechanism for the regulation of T3 action. For example, heterodimers of retinoid X receptors (RXR) and TR bind to T3 response elements (T3RE) with higher affinity and modulate transcription more effectively than TR homodimers. In the present report, we investigated the role of RXR in mediating differences in T3 responsiveness of the malic enzyme gene between CEH and CEF. Data from gel mobility shift analyses demonstrated that endogenous TRs from CEH and CEF bind to the major T3RE of the malic enzyme gene primarily as heterodimers with RXR alpha or a protein highly related to RXR alpha. The total binding activity of RXR alpha/TR complexes in CEF was decreased relative to that observed in CEH. Cell-type dependent differences in RXR alpha/TR complex formation were greater in cells incubated in the presence of T3 because T3 treatment increased RXR alpha/TR binding activity in CEH but had no effect on RXR alpha/TR binding activity in CEF. Decreased RXR alpha/TR complex formation in CEF relative to CEH was associated with a reduction in the abundance of RXR alpha protein and RXR alpha mRNA in the former cell-type. Expression of exogenous RXR alpha in CEF increased the T3 responsiveness of the malic enzyme promoter by about 4-fold. In contrast, expression of exogenous RXR alpha in CEH had no effect on the regulation of malic enzyme transcription by T3. These observations support the hypothesis that alterations in RXR alpha expression contribute to cell-type dependent differences in T3 responsiveness of the malic enzyme gene.

NMR studies of structure and dynamics in fruit cuticle polyesters.

Cutin and suberin are support polymers involved in waterproofing the leaves and fruits of higher plants, regulating the flow of nutrients among various plant organs, and minimizing the deleterious impact of microbial pathogens. Despite the complexity and intractable nature of these plant biopolyesters, their molecular structure and development are amenable to study by suitable solid-state and solution-state NMR techniques. Interactions of tomato cutin with water were examined by solid-state 2H and 13C NMR, showing that water films enhance rapid segmental motions of the acyl chains and are associated with a fivefold increase in surface elasticity upon cutin hydration. The suberization of wounded potato tissues was studied by solid-state 13C NMR, revealing the likely phenylpropanoid structures that permit dense cross-linking of the suberin structure and their proximity to the cell-wall polysaccharides. Finally, two new approaches were developed to elucidate the molecular structures of these biopolymers: partial depolymerization followed by spectroscopic analysis of the soluble oligomers; and swelling of the intact materials followed by magic-angle spinning (MAS) NMR analysis.

Induction of cyclooxygenase-2 by overexpression of the human catalase gene in cerebral microvascular endothelial cells.

Prostaglandin (PG) formation by the inducible (type 2) cyclooxygenase (COX-2) and reactive oxygen species (ROS) have been proposed to play important roles in cerebrovascular pathological processes. To explore the relationship between ROS and COX-2 expression, adenovirus (Ad) vectors containing cDNA for human antioxidant enzymes including catalase (AdCAT:), copper/zinc superoxide dismutase (AdCu/ZnSOD), and manganese superoxide dismutase (AdMnSOD) were transferred into murine cerebral microvascular endothelial cells. AdCAT: (100 multiplicity of infection) infection increased the content and enzymatic activity of cellular Cat threefold and decreased the intracellular peroxide level. The expression of COX-2 mRNA and protein in cell lysates was up-regulated, and the amount of PGE(2) formed from exogenous arachidonic acid increased following AdCAT: infection in a dose-dependent manner, paralleling the expression of COX-2 protein. The AdCAT:-induced increase in PGE(2) formation was inhibited by NS-398, a selective inhibitor of COX-2 enzymatic activity. AdCAT: infection did not change the expression of the constitutive (type 1) COX protein. Although AdCu/ZnSOD and AdMnSOD infection increased the expression of superoxide dismutase proteins, COX-2 expression was not induced. An in vitro nuclear transcription assay indicated that overexpression of the Cat gene increases the transcription of the COX-2 gene. Furthermore, the stability of COX-2 mRNA induced by lipopolysaccharide was increased after AdCAT: gene transfer. These results indicate that AdCAT: gene transfer induces the transcriptional activation of the COX-2 gene and increases COX-2 mRNA stability. Therefore, peroxide may have regulatory effect on COX-2 function in the cerebral microcirculation.

Lysophospholipid growth factors in the initiation, progression, metastases, and management of ovarian cancer.

Levels of lysophosphatidic acid (LPA) and lysophosphatidylcholine (LPC) are elevated in the plasma and ascites of ovarian cancer patients, but not in most other tumor types. LPA increases cell proliferation, cell survival, resistance to cisplatin, cell shrinkage, and production of vascular endothelial growth factor, urokinase plasminogen activator, and LPA itself in ovarian cancer cells, but not in normal ovarian surface epithelial cells. PSP24 and members of the endothelial differentiation gene (EDG) family (EDG1, EDG2, EDG4, and EDG7) of G protein-coupled receptors mediate LPA signaling. Ovarian cancer cell lines do not express EDG1 mRNA, have variable EDG2 mRNA and protein levels, and frequently exhibit levels of EDG4 mRNA and protein, suggesting that EDG4 may contribute to the deleterious effects of LPA in ovarian cancer. In contrast, activation of the EDG2 LPA receptor on ovarian cancer cells may lead to apoptosis and counter the effects of other LPA receptors. Thus, the development of agonists and antagonists for the appropriate spectrum of LPA receptors may alter proliferation, apoptosis, or response to therapy of ovarian cancer cells. Indeed, over 60% of all current drugs target the G protein-coupled family of receptors, making the LPA receptor family a "drugable" target. LPC, although not as thoroughly studied, increases cellular proliferation and mediates multiple other functions through unique signaling pathways.

Effects of LPS and IL-6 on oxytocin receptor in non-pregnant and pregnant rat uterus.

PROBLEM: Little is known regarding the regulation of the timing of parturition. Recent evidence suggests an interaction between the immune system and uterine contractility in late gestation. METHOD: Pregnant rats were treated with LPS in vivo in attempts to establish a model of premature parturition induced by the pro-inflammatory response. Uterine explants were incubated in vitro to determine the effects of IL-6 on uterine synthesis of oxytocin (OT) and its receptor (OTR). RESULTS: LPS injection was quite toxic to pregnant rats and gave extremely variable results. In animals that delivered, there was a marked increase in the uterine concentrations of OTR and OTR mRNA. There was no consistent effect regarding the timing of parturition. IL-6 caused a significant increase in the concentration of OTR mRNA in uterine explants from pregnant rats but not in tissues from non-pregnant animals. CONCLUSION: Rat uterine concentrations of OTR are regulated by IL-6. Pro-inflammatory cytokines may stimulate uterine contractility in late gestation rat uterine tissues through a mechanism involving stimulation of OTR.

Conversion of epoxyeicosatrienoic acids (EETs) to chain-shortened epoxy fatty acids by human skin fibroblasts.

Epoxyeicosatrienoic acids (EETs), the eicosanoid biomediators synthesized from arachidonic acid by cytochrome P450 epoxygenases, are inactivated in many tissues by conversion to dihydroxyeicosatrienoic acids (DHETs). However, we find that human skin fibroblasts convert EETs mostly to chain-shortened epoxy-fatty acids and produce only small amounts of DHETs. Comparative studies with [5,6,8,9,11,12,14,15-(3)H]11,12-EET ([(3)H]11,12-EET) and [1-(14)C]11,12-EET demonstrated that chain-shortened metabolites are formed by removal of carbons from the carboxyl end of the EET. These metabolites accumulated primarily in the medium, but small amounts also were incorporated into the cell lipids. The most abundant 11, 12-EET product was 7,8-epoxyhexadecadienoic acid (7,8-epoxy-16:2), and two of the others that were identified are 9, 10-epoxyoctadecadienoic acid (9,10-epoxy-18:2) and 5, 6-epoxytetradecaenoic acid (5,6-epoxy-14:1). The main epoxy-fatty acid produced from 14,15-EET was 10,11-epoxyhexadecadienoic acid (10, 11-epoxy-16:2). [(3)H]8,9-EET was converted to a single metabolite with the chromatographic properties of a 16-carbon epoxy-fatty acid, but we were not able to identify this compound. Large amounts of the chain-shortened 11,12-EET metabolites were produced by long-chain acyl CoA dehydrogenase-deficient fibroblasts but not by Zellweger syndrome and acyl CoA oxidase-deficient fibroblasts. We conclude that the chain-shortened epoxy-fatty acids are produced primarily by peroxisomal beta-oxidation. This may serve as an alternate mechanism for EET inactivation and removal from the tissues. However, it is possible that the epoxy-fatty acid products may have metabolic or functional effects and that the purpose of the beta-oxidation pathway is to generate these products.

[The multimedia molecular biological image processing system].

With the use of computer image processing technology and molecular biological technology, we have developed a new multimedia molecular biological information processing system to detect the characteristic parameter of molecular biological electrophoresis image. The system can determine the macromolecules densities and molecular weight of the DNA amplified fragment or restriction endonuclease hydrolytic fragment protein and other macromolecules. The clinical use and biological application indicate that the system is useful to gene diagnosis and molecular biological researches.

Epidemiologic study of the irritable bowel syndrome in Beijing: stratified randomized study by cluster sampling.

OBJECTIVE: To explore the prevalence of irritable bowel syndrome (IBS) in Beijing and its risk factors. METHODS: Phase I: a screening for IBS in Beijing area according to symptoms using both Manning (modified including constipation) and Rome criteria. 2486 subjects were studied by cluster sampling of the inhabitant groups according to a stratified design of urban, suburban and rural areas, and sample size of each area studied was in proportion to the population of the area. Selection of the inhabitant groups was made by simple random sampling. Age of subjects enrolled in the study was 18-70 years. All subjects fulfilling the selection criteria were requested to fill in a questionnaire assisted by trained doctors or medical students during the visit to their families. Phase II: an aliquot of patients who fulfilled at least the Manning criteria were further selected according to their scoring series to undergo detail clinical examination in the hospital including laboratory examination, abdominal ultrasonography, colonoscopy or/and barium enema to exclude organic disease of the colon. Prevalence of IBS of the population was then adjusted by the rate of correct diagnosis during Phase II study. Study using Minnesota Multi-Personality Indices (MMPI) was done in some cases. Probable risk factors were explored by comparing their frequencies among IBS group and non-IBS group using chi 2 and logistic analysis of multifactors. RESULTS: The adjusted point prevalence of IBS in Beijing is 7.26% according to Manning criteria, and is 0.82% according to Rome criteria. There is a higher prevalence rate in city (10.50%) than in rural areas (6.14%) by stratified analysis (P < 0.001). Male to female ratio is 1:1.15. And IBS is more common in people aged between 18-40 years (51.6%), and among the intellectuals. Our study indicated that history of dysentery (OR 3.00), exposure to cool (OR 1.55) and ingestion of cold food and raw materials (OR 1.24) may be the most important risk factors (P < 0.001), and IBS patients may have a higher tendency of psychological abnormalities. CONCLUSION: IBS is a common disorder in Beijing and should be taken into consideration in the human welfare strategy.

[Evaluation of long-term community based intervention on cardiovascular diseases: 9-year follow up study of the mortality from the cohorts].

OBJECTIVE: To evaluate the effect of long-term intervention on all-causes mortality and the mortality of cardiovascular diseases. METHODS: In May 1987 in Beijing, 2 geographically separated community with a registered population of about 10 000 each were selected as either intervention or control community. In each community, a cohort containing 2 700 subjects (>/= 35 years) free of stroke was sampled. A baseline survey was administered to screen the eligible subjects for intervention. Death information from the cohorts was collected. RESULTS: By the end of Dec. 31, 1995, the 9-year cumulative all-causes mortalities were 3.51% (303/2 861) in the intervention cohort and 4.71% (390/2 760) in the control cohort. Cardiovascular diseases and neoplasm were the first and second leading causes of death in the cohorts. Both the all-causes mortality and the mortality of cardiovascular diseases were significantly lower in the intervention cohort than in the control group. The risk for dying of all-causes death was reduced by 22% (RR = 0.78, 95% CI: 0.67 approximately 0.91) and the risk for dying of cardiovascular diseases was reduced by 24% (RR = 0.76, 95% CI: 0.57 approximately 0.96) in the intervention cohort. CONCLUSION: Long-term community intervention for the risk factors of cardiovascular diseases can effectively reduce the mortality of cardiovascular diseases as well as all-causes mortality.

[Association of tumor necrosis factor microsatellite polymorphism with incidence and outcome of severe postoperative sepsis].

OBJECTIVE: To investigate whether tumor necrosis factor (TNF) microsatellite polymorphism within the TNF locus is associated with the incidence and outcome of severe postoperative sepsis. METHODS: 122 patients with severe postoperative sepsis were included in this study; 138 local ethnically matched healthy individuals served as controls. Microsatellite TNFc polymorphism within the TNF locus was typed using polymerase chain reaction (PCR) followed by polyacrylamide gel electrophoresis with silver staining. RESULTS: Microsatellite TNFc polymorphism had two alleles (TNFc1 and TNFc2) and three kinds of genotype (homozygotes TNFc1c1 and TNFc2c2, and heterozygote TNFc1c2). The genotype distribution fit Hardy-Weinberg equilibrium. The frequency of TNFc1 microsatellite allele was significantly higher in patients with severe sepsis (79%) than in healthy individuals (71%) (P < 0.05). The frequency of heterozygote TNFc1c2 was significantly higher in non-surviving patients (46%) with severe sepsis than in survivors (27%) (P < 0.05). CONCLUSIONS: TNFc microsatellite polymorphism is significantly associated with the incidence and outcome of severe postoperative sepsis.

[The relationship of expression of CD44v6 with metastasis and prognosis in breast cancer].

OBJECTIVE: To study the relationship of expression of CD44v6 with metastasis and prognosis in breast cancer. METHODS: The expression of CD44v6 protein in 78 paraffin and 37 fresh specimens of breast carcinomas was detected with immunohistochemistry (IHC). The expression of CD44v6 mRNA in 37 fresh specimens of breast carcinoma was detected with RT-PCR. The correlation of tumor size, TNM stage, status of axillary lymph node, tissue grade, prognosis and CD44v6 expression in breast cancer was analyzed to compare the sensitivity of the two methods. RESULTS: The expression of CD44v6 in breast cancer was correlated to metastasis of the axillary lymph node, tumor size and TNM stage. It was an important factor affecting survival of patients with breast cancer. Positive expressive rates of CD44v6 protein and mRNA were 70.3% and 59.5% respectively in 37 specimens of fresh carcinoma tissues (P > 0.05). CONCLUSIONS: The expression of CD44v6 is closely related to metastasis and prognosis in breast cancer, and the sensitivity to detect CD44v6 expression shows no significant difference between IHC and RT-PCR.

Patients with IBS seemed to have a higher tendency of abnormal personalities. CONCLUSION: IBS is a common disorder in Beijing which deserves greater care.
