Effect of immunoglobulin G isotype on the infectivity of Chlamydia trachomatis in a mouse model of intravaginal infection.

It has been previously shown with an in vitro neutralization system that monoclonal antibodies (MAbs) to the major outer membrane protein (MOMP) of Chlamydia trachomatis, depending on the isotype of the MAb and the host cell used, can either neutralize or enhance the infectivity of this organism. MAbs to variable domain 4 (VD 4) of MOMP have been described that neutralize the infectivity of C. trachomatis when tested in a system in which either the host cell does not have detectable Fc gammaRIII receptors or complement is added to block the interaction of the MAb with the receptor. However, if Fc gammaRIII receptors are available, immunoglobulin G2b (IgG2b) MAbs to the VD 4 are able to enhance the infectivity of this pathogen. Two MAbs that recognize the sequence TLNPTIA in VD 4 of the MOMP but differ in isotype, E4 (IgG2b) and E21 (IgG1), were used to test whether in vivo the isotype of the MAb modulates the outcome of a vaginal infection in a murine model. A third MAb, CP33 (IgG2b), that recognizes the chlamydial lipopolysaccharide but does not neutralize infectivity of C. trachomatis, was also tested. Elementary bodies (EBs) of C. trachomatis, serovar E (BOUR), were pretreated with the three MAbs and were used to inoculate the vaginas of C3H/HeJ mice which had been pretreated with progesterone. Subsequently mice were monitored over a 5-week period with vaginal cultures. In the groups that were inoculated with EBs pretreated with MAbs directed to VD 4 of MOMP, there was a significant decrease (P < 0.05) in the number of mice infected. Only 30% of the mice were infected in the MAb E4-treated group, and 10% were infected in the MAb E21 group. This was in contrast to the groups inoculated with EBs pretreated with MAb CP33 and control untreated EBs, which resulted in 100 and 79% of the mice infected, respectively. Therefore, in this setting in which EBs were introduced in vivo coated with MAb, there was no enhancement of infection by IgG2b MAbs; rather, the results paralled the in vitro neutralization results, in which cells lacking Fc gammaRIII receptors were employed. Mice were also given the MAbs, as well as purified IgG as a control, by intraperitoneal injection before and after intravaginal inoculation with C. trachomatis. Despite relatively high levels of MAbs in serum and detectable levels of MAbs in the vagina at the time of infection, there was only modest protection in animals receiving MAb E21, with 60% of the mice infected in contrast to 90% of the mice receiving MAb E4, MAb CP33, and IgG. However, by the second week of infection compared to controls, there was a significant increase (P < 0.05) in the amount of chlamydiae recovered from the vaginas of mice that had received the two IgG2b MAbs, E4 and CP33. In summary, the presence of IgG2b MAbs directed to surface components of C. trachomatis at certain times during the course of infection may play a role in enhancing the infectivity of this pathogen.

The transcriptional integrator CREB-binding protein mediates positive cross talk between nuclear hormone receptors and the hematopoietic bZip protein p45/NF-E2.

Thyroid hormone (T3) and retinoic acid (RA) play important roles in erythropoiesis. We found that the hematopoietic cell-specific bZip protein p45/NF-E2 interacts with T3 receptor (TR) and RA receptor (RAR) but not retinoid X receptor. The interaction is between the DNA-binding domain of the nuclear receptor and the leucine zipper region of p45/NF-E2 but is markedly enhanced by cognate ligand. Remarkably, ligand-dependent transactivation by TR and RAR is markedly potentiated by p45/NF-E2. This effect of p45/NF-E2 is prevented by maf-like protein p18, which functions positively as a heterodimer with p45/NF-E2 on DNA. Potentiation of hormone action by p45/NF-E2 requires its activation domain, which interacts strongly with the multifaceted coactivator cyclic AMP response element protein-binding protein (CBP). The region of CBP which interacts with p45/NF-E2 is the same interaction domain that mediates inhibition of hormone-stimulated transcription by AP1 transcription factors. Overexpression of the bZip interaction domain of CBP specifically abolishes the positive cross talk between TR and p45/NF-E2. Thus, positive cross talk between p45/NF-E2 and nuclear hormone receptors requires direct protein-protein interactions between these factors and with CBP, whose integration of positive signals from two transactivation domains provides a novel mechanism for potentiation of hormone action in hematopoietic cells.

Clinical responses and platinum concentrations in tumors after intra-arterial and intravenous administration of cisplatin in the same patients with cervical cancer.

We evaluated 3 patients with advanced cervical cancer treated with cisplatin intra-arterially and intravenously. The dose of cisplatin was 50 mg/m2 in each infusion. Chemotherapy was repeated at 4-week intervals for three to four courses. The clinical response and the tumor concentration of platinum were evaluated in each course. All patients who received the intra-arterial infusion of cisplatin were judged to be responders, whereas none of them responded to the intravenous infusion. The platinum concentration in tumor tissue was significantly higher after intra-arterial infusion of cisplatin (1.97 +/- 0.04 vs. 2.86 +/- 0.10 microg/g). Although there were no apparent differences in side effects between intra-arterial and intravenous routes, 2 of 3 patients rejected an intra-arterial route. The present study suggests that intra-arterial administration of cisplatin may be useful in treating locally advanced cervical cancer.

Combination effect of granisetron and methylprednisolone for preventing emesis induced by cytotoxic agents.

To evaluate the combined effect of granisetron and methylprednisolone against acute emesis induced by cytotoxic agents, we investigated the clinical response and the urinary excretion of 5-HIAA in 85 patients with ovarian cancer who received the same anticancer chemotherapeutic regimen in a prospective randomized trial. Each patient received one of three different regimens (granisetron alone, methylprednisolone plus granisetron, and metoclopramide alone). The combination therapy of granisetron and methylprednisolone is effective for preventing acute emesis induced by cytotoxic chemotherapy.

Intrachondral microvasculature in the human fetal talus.

The intrachondral microvasculature of the growing talus of human was studied in 16 fetuses aged from 15 to 44 weeks of gestation, using interrupted serial sections and vascular injection of ink. The cartilage model of the talus was shown to be well vascularized throughout by cartilage canals. The cartilage canal contained blood vessels and connective tissue, with vessels originating from the perichondrial vessels. They were covered by a thick connective tissue wall that was continuous with the perichondrium. The functions of the cartilage canals were mainly to nourish the large masses of cartilage and to supply osteogenic tissue, which initiates the primary ossification center. As in the adult, the fetal talus was supplied with four to five main branches originating from the sinus tarsi and the tarsal canal; there were no anastomoses between the vessels of the adjacent cartilage canals and between the branches within the cartilage canal. This type of microvasculature is vulnerable to injury and, if impaired, may cause serious complications.

Differential screening of a subtracted cDNA library: a method to search for genes preferentially expressed in multiple tissues.

We have developed a new strategy for differential screening of genes that are expressed in two or more tissues, and have used it to identify genes that are preferentially expressed in both testis and ovary. In this approach, testis-specific cDNAs were first generated using the suppression subtractive hybridization technique, cloned and arrayed in microplates. The inserts from the subtracted testis-specific library were amplified by PCR and spotted on filters. The dot blots were screened by hybridization using either testis- or ovary-specific subtracted cDNA mixtures as probes. By screening about 2000 putative clones from the subtracted testis-specific library, we identified 14 candidate genes that hybridized to both cDNA probes. Northern blot analysis showed that 12 clones were preferentially or exclusively expressed in testis and ovary. Nucleotide sequence analysis indicated that three of the identified clones represented cDNAs from novel genes.

[The long-term effect of nifedipine (control release tablets) in patients with chronic obstructive pulmonary disease and cor pulmonale].

OBJECTIVE: To observe the long-term effect of nifedipine (control release tablets) in patients with chronic obstructive pulmonary disease and cor pulmonale. METHODS: A randomized, double blind, placebo-controlled design was made, two hundred and two patients (FEV1/FVC was 47.35% +/- 8.10%) were divided into two groups: nifedipine group (20 mg Bid) 102 cases, placebo group (one tablet Bid) 100 cases. RESULTS: The equilibrium test between both groups was comparable. Two-year follow-up rates of both groups were 94% and 89%, respectively. The comparision of the nifedipine to placebo group was as follows: in nifedipine group, the improvement of dyspnea, fatigue and exercise capacity showed significantly better results (P < 0.05); FEV1 and PaCO2 deterioration was unremarkable (P > 0.05); the pulmonary impedance plethysmogram B-Y1 that reflects pulmonary vascular compliance appeared significant improvement (P < 0.05) and the mortality of the disease that was the most important observation endpoint was declined (P = 0.051), but some electrocardiographic index reflected right ventricular load became worse (P < 0.05). CONCLUSIONS: Long-term taking controlled release-nifedipine is a safe, effective measure which might improve life quality and decrease mortality in patients with chronic obstructive pulmonary disease and chronic cor pulmonale, however the drug can not slow down or reverse the progress of this illness.

[A simple nitric oxide exposure system for small animals].

A nitric oxide exposing system was designed for experimental research, which consist of equipments such as plexiglass chamber, blower, flowmeter, NO/N2 cylinder, pure O2 cylinder, NOx analyzer, and O2/CO2 monitor. The efficacy of the whole system has been verified through our practice as shown by the following results: measured NO inside the chamber were close to designed NO concentration, measured O2 concentrations in the chamber were similar to that of the atmosphere, and the highest nitrogen dioxide (NO2) and carbon dioxide (CO2) concentrations were lower than 3 ppm and 0.3% respectively. The experimental facility is simple in construction, easy to be operated and convenient for research on effects and toxicity of long-term inhaled nitric oxide in small animals.

[Clinical analysis in seventy-two patients with chronic thromboembolic pulmonary hypertension].

To improve the identification of chronic thromboembolic pulmonary hypertension. We restrospectively analysed clinical data of 72 patients with chronic thromboembolic pulmonary hypertension (CTEPH). The levels of arterial blood gases appeared to be decrease in PaO2 and PaCO2, and increase in P(A-a)O2. There were 80.3% of unsymmetrical pulmonary hyperlucencies on chest radiograph and 76.4% of right ventricular hypertrophy on electrocardiograph, 92.5% and 97.1% of right atrial and right ventricular enlargement, respectively. 11.1% of pericardial effusion was noted on echocardiograph, and 98.5% of rise in pulmonary arterial pressure calculated by Doppler. The mean pulmonary arterial pressure was 6.50 +/- 1.80 kPa (48.75 +/- 13.50 mmHg), in part, by right cardiac catheterazation. Both the incidences of pulmonary embolism were 100.0% on pulmonary angiography and on radionuclide lung perfusion scan. There were 43.1% of the history of deep venous disorders and 75.0% of the positive findings by radionuclide venography in the lower extremities, respectively. The misdiagnostic and the lost diagnosis rate of prehospitalization accounted for 90.3%. The understanding of clinical manifestations and laboratory findings of CTEPH is important to promote diagnostic sense and level of CTEPH.

[Limitations of the common pre-processing method in the signal-averaged electrocardiogram].

The isolation of the micro potentials from electrocardiogram using coherence averaging technique requires perfect signal alignment. The purpose of this paper was to assess the reliability of the maximum correlation coefficient (MCC) method by computer simulation. The ability of signal recognition, the impacts of correlation-window-width and threshold on accuracy of temporal alignment and on averaged results were analyzed. The results showed that the MCC method was totally insensitive to amplitude variation and relatively insensitive to the duration variation. In the process of signal alignment, the narrower correlation-window-width, the less sensitivity of the maximum correlation coefficient to the misalignment would be. With the threshold of 0.99, the window-width of both 40 and 100 ms induced the deformation on averaged result in time and frequency domain. The simulation results suggested that the deficiencies of MCC method could impose serious limitations on reliability of isolating the atrial and ventricular late potentials.

[Three-dimensional finite element stress analysis of magnetical retaining and bar retaining in overdenture implants].

The purpose of this study was to determine which type of denture retaining implants can produce better biomechanical effect on the bone tissue. Two mechanical models of magnetically retained complete overdenture supported by osseo-integrated implant and bar-retained complete over denture supported by osseo-integrated implant were made. The peak stress in mandibular peri-implant bone tissue of the models were studied under the same loading conditions vertically and horizontally by three-dimensional FEM stress analysis. The results were as follows: (1) Both types of dentures retention can produce significant stress effect on the peri-implant and other regions of the jaw bone and (2) Magnetical retention is more beneficial to maintenance both of peri-implant bone tissue and long-term success of denture supported by osseo-integrated implant than bar retention does.

[Resistant antibiotic analysis of methicillin-resistant staphylococcus].

Methicillin-resistant staphylococcus aureus is one of the important patholonic bacteria which cause nosocomial infection. In order to investigate the resistant antibiotic circumstances of this organism and hence provide foundations of prevention and treatment, we determined the resistant rates of 88 staphylococcus strains for methicillin and for other 14 kinds of commonly used antibiotics, and we determined the engendering beta-lactamase. The results showed that the isolation rate of methicillin-resistant staphylococcus aureus (MRSA) was 56%; the rate of methicillin-resistant staphylococcus coagulase-negative (MRSCoN) was 47.6%; the resistance rate and the multi-resistant rate of methicillin-resistant staphylococcus (MRS) for the 14 kinds of antibiotics were higher as compared with those of methicillin-sensitive (MSSA). Among all the tested strains, MRSA was the highest to engender beta-lactamase (92.9%); Methicillin-sensitive staphylococcus coagulase-negative (MSSCoN) was the lowest (39.4%). MRS was sensitive to vancomycin and furantoin. These suggest that when treating this bacterium, we should select these two antibiotics first.

[Damage-loss relationships and integrated control of 3rd instar larvae of Actias selene nigpoana Felder in planting area of Cornus officinalis Sieb. et Zucc].

In the present paper, the damage-loss model of Actias selene ningpoana to Cornus officinalis was tested and the results indicated that the yield loss rates obeyed the equation Y = 100 - EXP(4.6042 - 0.0315X). The economic threshold of the 2hd and 3rd instar larvae of A. selene nigpoana was then determined as 22 and 8 insects per tree respecitively. Suggestions for integrated control have been made based on the research of activities of A. selene ningpoana in the forest.

Detection of human herpesvirus 6(HHV-6) DNA in salivary glands by the polymerase chain reaction.

To assess the presence of HHV-6-Specific DNA in human salivary glands, eighteen specimens of salivary gland tissue were investigated using the polymerase chain reaction. Eight of nine parotid glands, five of seven submandibular glands and one of two sublingual glands were found to have amplification of the HHV-6-specific sequence. The findings suggest that salivary gland tissue is one of the potential sites for HHV-6 persistence following primary infection and that saliva is a vehicle for transmission of the virus.

[Application of ultrasound biomicroscopy in diagnosis of anterior segment vitreoretinal disorders].

PURPOSE: To investigate the diagnostic value of ultrasound biomicroscopy(UBM) in anterior segment vitreoretinal disorders(ASVD). METHODS: 60 eyes of 58 sequential patients were examined with UBM. The ultrasound biomicroscopic images were compared with the results of clinic examination, B-scan ultrasonography and the finding during surgery to evaluate the reliability of UBM for ASVD. RESULTS: UBM allows structural details of the conjunctiva, rectus muscle, sclera, uvea, retina and vitreous to be visualized at microscopic resolution in living eye. The area of UBM may cover from cornea to vitreous base, partially to the equator of eye. A variety of the changes of ASVD were imaged in 60 eyes, including retinal detachment in 43 eyes, retinal circumferential contraction in 29 eyes, retinal anterior displacement in 24 eyes, vitreous opacities or fibrosis in 57 eyes, ciliary orchoroidal detachment in 26 eyes, ciliary nonpigmented epithelial detachment in 6 eyes, proliferative tissue at the sclerotomy site in 9 eyes, retinal neoplasm in 2 eyes, ciliary cyst in 6 eyes, proliferation of peripheral silicone oil in 8 eyes, silicone oil emulsification in 7 eyes and vitreous foreign body in 2 eyes, respectively. CONCLUSIONS: Our studies showed the feasibility of UBM for ASVD not visible by current techniques UBM can be used for the diagnosis, differential diagnosis and the instructive treatments of ASVD.

[Studies on the distribution and the control decision of white grub population in field of Paeonia lactiflora].

The present paper showed the result of accumulated distribution pattern of the mixed population of white grubs [Holotrichia oblita (Faldermann)] in the field. The x2-test indicated that the numbers of grub were 2.5-8 per squar-meter, which agreed to negative binomial distribution. The accepted and refused lines of sequential sampling were calculated in different occurrence degree.

[A strain of A/PR/8/34 (H1N1)-like virus isolated from men].

An influenza virus was isolated in May, 1991 from a child with acute respiratory disease. The identification results with serologic method have revealed that the isolate was A/PR/8/34 (H1N1)-like virus. The analyses of nucleotide sequences in HA1 domain showed that there were 12 different positions on nucleotide sequences between the isolate and the A/PR/8/34 (H1N1) virus. These differences induced substitutions of 4 amino acids in HA1 protein molecule of isolate as compared with that of A/PR/8/34 (H1N1) virus. The oligonucleotide analyses of viral genome revealed that there were 11 different points between isolate and A/PR/8/34 (H1N1) virus, e. g. 2 deleted and 9 inserted in genome of isolate as compared with A/PR/8/34 (H1N1) strain. Therefore, the isolate possibly derived from A/PR/8/34 (H1N1) virus by laboratory contamination could be ruled out. However, where did A/Guangdong/6/91 (H1N1) virus come from? This question will be settled by further study.
