Cytoplasmic expression of ribozyme in zebrafish using a T7 autogene system.

A cytoplasmic ribozyme expression system, based on codelivery of a ribozyme vector, a T7 autogene vector, and T7 RNA polymerase (RNAP), has been developed and used to generate a specific phenotype in zebrafish by targeting a no tail (ntl) mRNA. The expression of the no tail ribozyme sequence is under the control of a tandem of two promoters: The T7 promoter and an adenoviral va 1 (pol III) promoter. The coinjection of the ribozyme vector pT7vaRz, the T7 autogene vector pT7T7, and the T7 RNAP resulted in rapid synthesis of the ribozyme against the ntl mRNA in the cytoplasm of the injected zebrafish embryos, generating no tail phenotypes in up to 10-20% of the injected embryos. The phenotypic change rates have been found to be related to the concentrations of the plasmid vectors and T7 RNAP injected and to the ratios of the three injected components. This cytoplasmic ribozyme expression system may be useful for efficiently targeting other mRNA and for various biomedical applications. These potential applications may include rapid identification of biological functions of novel genes from zebrafish and humans based on partial gene sequence information and gene therapy of genetic and acquired diseases.

[Effects of five different polar extracts from Herba pogostemonis being gotten rid of volatile oil on gastrointestinal tract].

The effects of the five extracts from Herba pogostmonis being gotten rid of volatile oil on gastrointestinal tract were studied. The results showed that the five extracts could increase gastric secretion of acid and activity of pepsin, reduce the incidence of diarrhea induced by Folium sennae and relieve the gripping pain induced by abdominal administration of acetic acid. The extract of ethyl acetate could also inhibit the normal and neostigmine induced mice intestinal propellant movement. The effect of extract of ethyl acetate was consistent with clinical application. The above results indicated that the clinical effect of Herba pogostmonis may be comprehensive action of above five different polar extracts.

[Expression of neural cell adhesion molecule in murine livers with experimental autoimmune hepatitis].

OBJECTIVE: To investigate the expression of neural cell adhesion molecule (NCAM) in murine livers with experimental autoimmune hepatitis (EAH) and its relationship with histological grades. METHODS: To induce the EAH model, the syngeneic S-100 antigen emulsified in complete Freud's adjuvant was injected intraperitoneally to C57Bl/6 at day 1 and day 7. The mice were sacrificed at day 7, 14, and 21 after the first immunization. The expression of NCAM was observed using histoimmunochemistry and RT-PCR. RESULTS: The expression of NCAM increased gradually with aggravation of lymphocyte infiltration and hepatocyte injury. The administration of prednisolone decreased the histological grade, and inhibited the expression of NCAM. There was a correlation between the expression of NCAM and histological grade (r=0.71, P<0.01). CONCLUSIONS: NCAM provides a traffic signal for infiltration of lymphocytes in the liver, hence induces the injury of hepatocytes.

[The development and identification of animal osteoblasts from bone marrow culture].

This study was aimed to evaluate a modified method for animal osteoblast marrow culture on the basis of previously published method. The marrow mononuclear cells(MMNC) in femoral bone-marrow of SD rat were gained with Ficoll-Hapaque centrifuge, which were then cultured in 5 x 10(5) cells/cm3 on the 96-well culture plates with RPMI 1640 medium containing 30% horse serum in a humidified atmosphere of 5% carbon dioxide at 37 degree Celsius. The medium was changed every three days. Cultured cells were identified with inverted microscopy for observing the shape of living cells, and with Wright-Geimsa mixture stain and Alkaline phosphatase (ALP) stain. In order to avoid false positive reaction, we caused damage to the ALP in cultured cells and stained the cells with ALP again for comparison. The results of inverted microscopy showed that the small spherical mononuclear cells became polygonal, spindle-shaped or squamous successively from the fifth day onward. Wright-Geimsa staining showed the specific property of osteoblast. ALP stain positive reaction was between "2+" and "3+", then ALP stain negative reaction was noticed after the damage to ALP. These data demonstrated that MMNC in SD rat could become osteoblast in RPMI 1640 medium containing 30% horse serum. The modified osteoblast marrow culture method has the advantages of short time, less risk of contamination and higher efficiency.

Elevated interleukin-13 in patients with active lupus nephritis.

OBJECTIVE: To investigate the significance of interleukin-13 (IL-13) in patients with active lupus nephritis (LN). METHODS: Ten healthy volunteers and 16 patients with active LN were included in this study. The protein level of IL-13 in plasma was examined by enzyme linked immunosorbent assay (ELISA), and gene expression of IL-13 in peripheral blood mononuclear cells (PBMCs) by reverse transcription polymerase chain reaction (RT-PCR). Expression of IL-13 mRNA in renal tissue was studied by in situ hybridization (ISH) techniques. RESULTS: The level of IL-13 in plasma and the expression of IL-13 mRNA in PBMCs were significantly higher in LN patients than those in the controls (P < 0.001). Increased expression of IL-13 mRNA was detected in renal tissue of active LN patients compared to those in the controls (P < 0.001). Analysis of the linear correlation indicated that the level of IL-13 mRNA in the tubulointerstitial area in patients with active LN correlated with the concentration of serum creatinine (Scr), the glomerular activity index (GAI), the activity index of tubulointerstitium, and the level of serum C3 (P < 0.05 for each). CONCLUSION: The elevation of IL-13 may play an important role in the molecular pathogenesis of active LN.

Transgenic mice with overexpression of human scavenger receptor A on endothelial cells.

OBJECTIVES: To establish a new transgenic mouse model for determining the function and role of human scavenger receptor A (SR-A) in atherosclerosis in vivo. METHODS: Human scavenger receptor minigene-driven mouse tie-1 promoter was constructed and confirmed by endonuclease digestion and sequence analysis. Transgenic mice were generated via the microinjection method. PCR and Southern blot were used to screen the positive transgenic mice. RT-PCR and immunohistochemical analysis were used to detect the level and location of human SR-AI expression in transgenic mice. The activity of human SR-AI was determined by morphologic observation of aortic endothelial cells of transgenic mice under transmission electron microscopy. RESULTS: The electrophoresis assay showed the expected 4 fragments of 0.9 kb, 1.1 kb, 1.2 kb and 4.2 kb in the Sma I digest and 2 fragments of 0.8 kb and 6.7 kb in Bgl II digest of plasmids pTie-1/hSR-A. The fragment sequence of tie-1 promoter and human SR-A cDNA in plasmids pTie-1/hSR-A was correct and no ATG before the translation initiation sites of human SR-A was found by sequence analysis. 561 injected and surviving embryos with the purified human SR-A minigene were implanted into the oviducts of 19 ICR pseudopregnant mice. Among the 54 surviving pups from 13 foster mothers, 7 were identified by PCR and Southern blot analysis. The results of RT-PCR and immunohistochemical analysis showed human SR-A was specifically expressed on vascular endothelial cells of the aorta and renal artery, as well as hepatic sinusoidal endothelial cells in transgenic mice. Transmission electron microscope (TEM) of aorta of transgenic mice showed that a large number of vesicles, multivesicle bodies and swollen mitochondria filled the plasma of endothelial cells. CONCLUSIONS: A transgenic mouse model with overexpression of human SR-A in endothelial cells was successfully established. The transgene was integrated and transmitted into the chromosome of transgenic mice. Tie-1 promoter controlled the transgene to express in endothelial cells in mice. Pinocytic activity of aortic endothelial cells in transgenic mice was higher than that of C57BL/6J mice. Our studies will provide a new transgenic model for investigation of atherosclerosis and functions of human SR-A.

[Radiologic features of desmoplastic ameloblastoma].

OBJECTIVE: To investigate the radiologic features of desmoplastic ameloblastoma. METHODS: The radiologic characteristics of 15 cases desmoplastic ameloblastoma which were diagnosed pathologically were analyzed retrospectively, and compared with solid or multicystic ameloblastoma. RESULTS: Desmoplastic ameloblastoma were mainly located in anterior and (or) premolar region of maxilla and mandible. There existed three features radiologically. 1. unilocular formation containing varying amounts of radiopaque islands or strands material. 2. multilocular destruction containing irregular or line-like radiopaque areas. 3. mixed destruction showing plxiform radiopaque material and unilocular change. Typical microscopic features were irregular and compressed epithelial islands or strands interspersed among dense fibrous connective tissue stroma. CONCLUSIONS: The desmopleatic ameloblastoma is a new type of ameloblastoma and exhibits some special characteristics radiologically and pathologically. It should be differentiated from osteofibroma and odontogenic myxoma.

[Immunohistochemical and ultrastructural study of basement membrane in oral verrucous carcinoma].

OBJECTIVE: To study the immunohistochemical alteration of basement membrane (BM) type IV collagen and laminin in oral verrucous carcinoma and its BM ultrastructural variations. METHODS: 16 cases of oral verrucous carcinoma (OVC), 10 cases of oral squamous cell carcinoma (OSCC) and 9 cases of oral mild to severe epithelial dysplasia (OMSD) were studied by using a immunohistochemical S-P method, and the results were analyzed by quantitative method. 3 cases of OVC were observed by TEM. RESULTS: The BM in OVC was thicker than in OSCC and OMSD. TEM found the basal lamina in some areas showed a marked reduplication. The BM in OVC was generally intact (13/16), whereas in OSCC it was mostly discontinuous (9/10), especially around the neoplasm front or the small cord consisted of a few cells, and mostly continuous in OMSD (6/9). There was a stromal inflammatory infiltration around tumor nests for all the oral lesions, but it was much heavier in OVC than that in OSCC and OMSD (P < 0.05). There was a positive correlation between intraepithelial lymphocytic infiltration and the BM continuity for OVC(P < 0.01). CONCLUSIONS: The more continuous BM and the heavier inflammatory infiltration in the connective tissue of OVC may be related to its biological behavior.

Alteration of surfactant proteins A and D in bronchoalveolar lavage fluid of Pneumocystis carinii pneumonia.

OBJECTIVE: To understand the interaction between surfactant proteins and pneumocystis carinii pneumonia (PCP), and the impact of corticosteriods on surfactant proteins. METHODS: We established rat models of PCP and bacterial pneumonia induced by subcutaneous injection of 25 mg cortisone acetate. At 8-12 wk, the bronchoalveolar lavage fluid (BALF) of rats was collected. Total nucleated cells of BALF were counted and differentiated, and the concentrations of surfactant protein A (SP-A) and surfactant protein D (SP-D) were measured by immunoblotting assay. The rats were divided into three immunosuppressive groups and a normal control group. Group I, normal control (n = 6), consisted of healthy SD rats; group II, negative control (n = 6), consisted of rats with cortisone acetate injection for over 8 wk without lung infection; group III, bacterial pneumonia (n = 11), rats were injected with cortisone acetate over 8 wk that resulted in bacterial pneumonia without other pathogens isolated; and group IV, PCP (n = 14), rats with injected cortisone acetate for 8-12 wk and developed PCP without other pathogens isolated. RESULTS: Our results indicated that the total cell count in BALF in the negative control group was lower than that in the normal control group (P < 0.001). During PCP infection, the total cell count and the percentage of polymorphonuclearcytes (PMNs) in BALF were significantly increased (P < 0.01), but were lower than those in the bacterial pneumonia group. The concentration of SP-A of BALF in PCP (45.1 +/- 22.1 micrograms/ml) was significantly increased in comparison with that in the negative control (16.2 +/- 9.9 micrograms/ml, P < 0.05) and bacterial pneumonia groups (6.2 +/- 5.6 micrograms/ml, P < 0.001). We also found that the relative content of SP-D was significantly higher in PCP (24,249 +/- 4780 grey values) than that in the negative control (13,384 +/- 2887 grey values, P < 0.001) and that in bacterial pneumonia (11,989 +/- 2750 grey values, P < 0.001). SP-A and SP-D were also higher in the moderate to heavy group of PCP than those seen in the mild group (P < 0.01, P < 0.001). SP-A and SP-D were higher in the negative control group than those in the normal control group, but there was no significant difference between the 2 groups. CONCLUSION: These results suggest that the concentrations of SP-A and SP-D in BALF are increased by pneumocystis carinii specific stimulation, but the alteration is not related to the corticosteriod usage.

Antineoplastic mechanism of Octreotide action in human hepatoma.

OBJECTIVES: To investigate whether apoptosis can be induced by Octreotide in human hepatoma cells in vitro and elucidate the antineoplastic mechanism of Octreotide in hepatoma. METHODS: A cultured human hepatoma cell line, BEL-7402, was exposed to Octreotide and apoptosis was evaluated by cytochemical staining (Hochesst 33,258), transmission electron microscopy, agarose gel electrophoresis and flow cytometry (FCM). RESULTS: After exposure to 0.2 microgram/ml Octreotide, apoptosis with nuclear chromatin condensation as well as fragmentation, cell shrinkage and the formation of apoptotic bodies was observed using cytochemical staining and transmission electron microscopy. A DNA ladder in agarose gel electrophoresis was also displayed. FCM showed that the apoptotic cell number rose with an increase in the concentration of Octreotide (0-2 micrograms/ml). There was a positive correlation between Octreotide concentration and apoptotic rate in BEL-7402 cells (r = 0.809, P < 0.05). CONCLUSION: Apoptosis in human hepatoma cells can be induced by Octreotide, which may be related to the mechanism of antineoplastic action of Octreotide in hepatoma.

The steroid 5alpha-reductase type II TA repeat polymorphism is not associated with risk of breast or ovarian cancer in Australian women.

The enzyme 5alpha-reductase type II (SRD5A2) converts testosterone to its more active form 5alpha-dihydroxytestosterone. The 3' untranslated region of the gene contains a (TA)(n) length polymorphism. The (TA)(9) allele has been reported to be associated with higher serum prostate-specific antigen levels in breast tumors and lower risk of relapse in breast cancer patients and more recently has also been reported to be linked to the codon 89 valine variant, which is itself associated with higher serum prostate-specific antigen levels in breast tumors and a more favorable breast cancer prognosis. We investigated whether the SRD5A2 (TA)(n) polymorphism was associated with risk of breast or ovarian cancer in Australian women by studying 946 breast cancer cases and 509 age-matched controls, and 544 ovarian cancer cases and 298 controls of similar age distribution. The (TA)(9) allele frequency was similar in breast cancer cases (0.110), breast cancer controls (0.125), ovarian cancer cases (0.106), and ovarian cancer controls (0.117). There was no difference in genotype distribution between breast cancer cases and controls (P = 0.5), ovarian cancer cases and controls (P = 0.7), or between the two control groups (P = 0.9). Genotypes containing at least one (TA)(9) allele were not significantly associated with risk of breast cancer overall (odds ratio, 0.86; 95% confidence interval, 0.67-1.12; P = 0.3) or in women stratified by age, menopausal status, or family history. Similarly, the (TA)(9) allele was not associated with risk of ovarian cancer (odds ratio, 0.87; 95% confidence interval, 0.61-1.23; P = 0.4) or with ovarian tumor behavior (invasive or low malignant potential), histology, stage, or grade. Given that this study had sufficient power to detect altered risks in the order of 1.4- to 1.7-fold, our results suggest that the SRD5A2 (TA)(9) allele is unlikely to be associated with moderate alterations in breast or ovarian cancer risk.

[Effect of photoperiod and temperature on fertility alteration of short photoperiod-low temperature induced male sterile rice].

Short photoperiod-low temperature induced male sterile rice is a new germplasm, whose fertility alteration characteristic is contrary to that of long photoperiod and high temperature induced male sterile rice. Compared with that of Yid1s sowed under natural conditions by stages, the fertility alteration of Yid1s was observed under simulated short photoperiod treatment. The results showed that the photoperiod-sensitive stage of Yid1s occurred from differentiation of rachis primordia (phase I) to secondary rachis-branch and spikelet primordia (phase III). The panicles induced by appropriate short daylight treatment at photoperiod-sensitive stage tended to be completely sterile under normal temperature, but the sterile effect was weakened under high temperature. The pollen fertility of Yid1s was reduced under high or low temperature. The temperature-sensitive stages of Yid1s based on the analysis of the effects of high and low temperature on fertility were basically identical, i.e., from pollen mother cell formation (phase V) to single nucleus (phase VII).

[Clinical analysis of tubulointerstitial lesions in IgA nephropathy].

OBJECTIVE: To analyze the characteristics of tubulointerstitial lesions (TIL) in IgA nephropathy (IgAN). METHODS: The pathologic and clinical materials of 609 IgAN patients were analyzed. RESULTS: (1) Prevalence characteristics of TIL: Among all the patients, these with mild TIL accounted for 47.1%, moderate TIL 21.7%, severe TIL was about 16.6% and these without TIL 14.6%. (2) Relationship between TIL and the clinical findings: With increase of the severity of TIL, hypertension became worse and the level of proteinuria increased gradually. On the contrary, serum albumin decreased and renal function degraded. (3) Relationship between TIL and the pathologic findings: With increase of the severity of TIL, the scores of vascular lesion, total glomerular lesion, hypercellularity and glomerulosclerosis increased gradually. Moreover, when the glomerular lesion and vascular lesion became worse, the severity of TIL also increased. The relationship with the crescents was different, the score of the presence of crescents was highest when TIL was moderate in severity; it would not increase when TIL was severe. CONCLUSION: With the increase of the severity of TIL, the condition of IgAN patients became worse. TIL may be one of the key factors for the prognosis of IgAN. TIL is closely related to the severity of glomerular and vascular lesions, and these lesions may further lead to the development of TIL.

[Effects of selenium on hepatocellular protooncogene c-myc, c-fos and c-jun expression induced by cadmium in rats].

OBJECTIVES: This study was conducted to explore effects of selenium on hepatocellular protooncogene c-myc, c-fos and c-jun expression induced by cadmium in rats. METHOD: Both cadmium and selenium were given to rats by i.p. and there were 5 SD rats in each group. Protooncogene c-myc, c-fos and c-jun expression in rat liver cells was measured with Northern Dot Hybridization. RESULTS: The results showed that cadmium chloride at doses of 5, 10 or 20 mumol/kg, significantly induced proto-oncogene c-myc, c-fos and c-jun expression, and when sodium selenite at the dose of 5 mumol/kg was given at the time, the effect of cadmium chloride on hepatocellular protooncogene c-myc, c-fos and c-jun expression was inhibited. CONCLUSION: Selenium at certain doses could inhibit hepatocellular protooncogene c-myc, c-fos and c-jun expression induced by cadmium in rats.

[The mandibular molar-area implantation of screw-typed dental implants].

OBJECTIVE: To study molar area implantation with screw-typed implant based on the specific anatomic properties of inferior alveolar nerve. METHODS: Depending on the degree of alveolar atrophy in mandibular molar area, the direct implantation, the buccal-to mandibular-canal implantation, and the repositioning inferior-alveolar-nerve implantation were employed. During the period of 1992 to 1998, a total of 22 cases of this kind with 45 dental implants were restored. The follow-up period was 5 years. RESULTS: 2 dental implants were lost respectively after 3 months of insertion and after 1 year of restoration. All the 43 others, however, still remained complete osseointegration, with no obvious crestal bone rebesorption and with excellent function. The 5-year accumulating survival rate of dental implants was 96%. CONCLUSIONS: These three surgical techniques, to a large extent, can solve the problems of mandibular molar area implantation. The key to success of each technique is the selection of patient and the selection of surgical project.

[Placental expression of epidermal growth factor receptor in pregnancy induced hypertension].

OBJECTIVE: To clarify the expression of epidermal growth factor receptor (EGFR) in placenta from women with pregnancy induced hypertension (PIH). METHODS: EGFR of placenta was determined by immunohistological method and compared between normal pregnancies (34 cases) and pregnancies accompanied by PIH (30 cases). RESULTS: Positive immunostain of EGFR was located on cytoplasm, and membrane of syncytiotrophoblast and a small part of cytotrophoblast. Compared with that of normal pregnancy, the expression of EGFR in placenta from pregnancies complicated with PIH decreased significantly (P < 0.05). No significant differences of EGFR expression in placenta were found among women with mild, moderate and severe PIH (P > 0.5). No significant correlation was found between expression of EGFR and neonatal birthweight, placental weight (P > 0.01, for both). CONCLUSION: Decreased expression of placental EGFR was found in women with PIH, and that may play a role in the pathogenesis of PIH.

[The CPBC types used for clinical choice and observation on their microstructure].

UNLABELLED: According to the primary properties of calcium phosphate bone cement (CPBCs), we have chosen certain types of them for use in cranioplasty and have observed the microstructure of their set bodies by means of SEM. Five kinds of CPBCs powder have been prepared, including: octacalcium phosphate precipitated hydroxyapatite (OCP-PHA) type, octacalcium phosphate hydroxyapatite (OCP-HA) type, octacalcium phosphate (OCP) type, calcium deficient hydroxyapatite (CDHA) type and hydroxyapatite (HA) type. The mixing liquids were deionized water and 0.25 M Na2HPO4/NaH2PO4 buffer solution. The setting time was measured by Gillmore method. The compressive strengths were measured using the TS-14 automatically measured instrument-II of single granule's compressive strength. The CPBC types used for clinical cranioplasty were defined according to Ginebra's Criteria of the CPBCs, and the ultrastructure of the set-bodies of the defined CPBC type was observed by SEM before and after its immersion in Ringer's Solution. RESULT: Two out of five CPBCs, OCP-PHA-CPBC and CDHA-CPBC were selected for clinical cranioplasty. Before and after they were immersed in Ringer's Sol., the SEM found their set-bodies to be structurally porous and to dissolve with the increase of immension time. These data indicate that OCP-PHA-CPBC and CDHA-CPBC are sufficient for the reconstruction of non-stress-bearing bone, and the porous structure of their set-bodies is advantageous to fibroplasty or fibrovasculization in their set bodies.

[Clinical and pathologic analysis of Sjögren's syndrome with renal impairment: a report of 84 cases].

OBJECTIVE: To further study the renal damage of Sjögren's syndrome (SS) and its clinical and pathologic characteristics with biopsy and recent technology. METHODS: 84 patients of SS with renal impairment from 1993 to 1999 were analyzed by routine, immunoassay, tubular function and biopsy examination. RESULTS: 55 of the 84 cases presented with renal tubular acidosis (RTA), 5 with diabetes insipidus and 3 with hypokalemic paralysis. Glomerulopathy occurred in 22 cases (nephrotic syndrome 12, glomerulonephritis 10) and mild renal failure (RF) was found in 14. 69.1% of the patients had hypergamma-globulinemia. 64.3% and 44.1% of the patients showed positive anti SS-A and anti SS-B. In 37 renal biopsy specimens 21 showed chronic interstitial nephritis (CIN) with extensive lymphoplasmic cell infiltration and tubular atrophy. 10 of the 37 specimens revealed lupus nephritis (LN, type III and IV) and 5 mesangial proliferative glomerulonephritis (MsPGN). With immunofluorescence tests, no positive findings were seen in most of the specimens, but deposition of IgA, IgM or C(3) was seen in some patients. IgG deposits in the interstitial lymphoplasmic cells were found in 1 patient. 25 patients were treated with prednisone combined with cytoxan (CTX). In 14 patients with renal failure, serum creatinine level returned to normal after treatment. CONCLUSION: Renal impairment may be the presenting or predominant feature in SS. The major clinical manifestations are RTA and GN. Treatment with prednisone may decrease the infiltration of lymphoplasmic cells in the interstitium, improve the renal function and correct RTA.
