Preparation and characterization of a set of monoclonal antibodies to TRAIL and TRAIL receptors DR4, DR5, DcR1, and DcR2.

The tumor necrosis factor-related apoptosis-inducing ligand (TRAIL/Apo 2L) is a novel cytotoxic ligand belonging to TNF superfamily. Among TRAIL receptors, death receptor 4 (DR4) and DR5 containing death domain (DD) in their cytoplasmic region mediate apoptosis-signaling upon TRAIL binding, while decoy receptor 1 (DcR1) and DcR2 with a truncated or non-functional DD play "decoy" role. The interaction of TRAIL and TRAIL receptors plays important roles both in immunoregulation and immune pathogenesis of some diseases. In this study, we raised hybridomas secreting monoclonal antibodies against TRAIL (FMU1.1, 1.2, 1.3), DR4 (FMU1.4), DR5 (FMU1.5, 1.6), DcR1 (FMU1.7) and DcR2 (FMU1.8, 1.9). These MAbs could be used for fluorescent staining and flow cytometry (FCM) analysis as well as immunohistochemistry (IHC). Moreover, FMU1.1, 1.3, 1.4 and 1.5 could be used as coating antibodies paring corresponding polyclonal antibodies to develop sandwich ELISAs to quantitate the soluble TRAIL (sTRAIL), sDR4 or sDR5 in serum samples respectively. In addition, cross-linking of DR4/DR5 by FMU1.4 or FMU1.5 MAbs could induce apoptosis of some DR4/DR5-expressing tumor cells. Thus, this set of monoclonal antibodies against TRAIL or TRAIL receptors may be useful in expression phenotypic and functional study of TRAIL and TRAIL receptors.

[Cataplasma of traditional Chinese medicine].

The TCM (traditional Chinese medicine) transdermal plaster (also known as "cataplasma") are flexible adhesive patches used for treatment of pain, resulted from arthritis, sprain and bruise, tendovaginitis, lumbar spine protrude, neuralgia, hyperosteogeny ache, abdominal discomfort and metastatic cancer, etc. Since the 1980's, investigators in China have used this modern patch delivery system for herbal drugs and obtained satisfactory results especially from the treatment of various types of pain associated with bone diseases, abdominal discomfort, and tumors, etc. The production of TCM cataplasma was successfully scaled up in early 90's and the commercial product line for an antirheumatic agent was first established in Shanghai by Leiyunshang Group. Thus far, a number of products in the form of TCM cataplasma became commercially available in the market, and clinical investigations with these products indicated that topically applicable herbal preparations, especially in the form of cataplasma, are preferred formulations with respect to the treatment comfort of the patient. Compared to the traditional preparations which utilize rubber and rosin as adhesives, cataplasma is advantageous in that the lipophilic and hydrophilic ingredients of the herbal extracts are solubilized and then "gellified" with the organic polymers, and that the drug matrix containing up to 40%-70% of water serves as a "drug reservoir" that will sustain the quick and continuous release of herbal ingredients over several days across the skin. While there are conventional remedies for palliation of pain and discomfort associated with bone diseases or cancers, administration of oral medicinal herbs combined with topical agents such as TCM cataplasma may significantly alleviate the symptoms and improve their quality of life. This article provides a review on three aspects, which include the process development, characteristics and developmental status of TCM cataplasma, and future development of such a technology.

Antidiabetic drugs of plant origin used in China: compositions, pharmacology, and hypoglycemic mechanisms.

The paper reviewed compositions and pharmacological effects of eight antidiabetic herbal drugs that have been approved by health regulatory agency for commercial use in China. Investigators attributed the hypoglycemic effect of these products to their ability to restore the functions of pancreatic tissues and cause an increase in insulin output, to inhibit the intestinal absorption of glucose, or to the facilitation of metabolites in insulin-dependent processes. Treatment with herbal drugs has an effect on protecting beta cells and smoothing out fluctuations in glucose levels. The use of these naturally derived agents in conjunction with conventional drug treatments such as an chemical agent or insulin permits the use of lower doses of the drug and/or decreased frequency of administration which decreases the side effects most commonly observed.

[Industrialization of medicinal plant tissue culture].

Compared with other countries, the industrialization of medicinal plant tissue culture is more important for our country because China is consuming and exporting the most amounts of herb materials in the world. Each year, many papers and patents are published on cell cultures of popular medicinal plants, such as Taxus sp., Catharanthus roseus, and Panax ginseng, and, meanwhile, the research on organ cultures of medicinal plants is increasing very quickly, which is deepening the study of medicinal plant tissue culture. During the past 30 years, Chinese scientists have cultured many medicinal plant cells, organs and hairy roots. In addition, the large-scale cultures have been tested on medicinal plants, such as Catharanthus roseus, Panax notoginseng, Anisodus acutangulus, Lithospermum erythrorhizon, and Taxus chinensis. However, the bioreactor size is not big enough for the commercial cultivation and we have not mastered the culture technique on a large scale. We should clearly understand the importance and great potential benefit of medicinal plant tissue culture and develop the tissue culture techniques for the modernization of TCM. To develop the technique that we have the property right, the pioneering spirit is needed in our research, and, meanwhile, it should be pointed out emphatically the collaboration is indispensable among scientists from different research fields.

[Expression and regulation of a novel soluble adhesive molecule sCD226 on human endothelial cells].

AIM: To explore the expression and regulation of a novel soluble adhesive molecule sCD226 on HUVECs. METHODS: The contents of sCD226 and NO in culture supernatant of LPS activated human umbilical vein endothelial cells (HUVECs) were detected by ELISA and Greiss method respectively, and linear correlativity of contents between sCD226 and NO was analysed. The influence of anti-TNF-alpha mAb and iNOS inhibitor on LPS-induced sCD226 production was observed by ELISA. RESULTS: (1)LPS might markedly stimulate sCD226 production from HUVECs in a dose- and time-dependent manner. The amount of sCD226 came to 188.5 microg/L when endothelial cells were stimulated by 100 mg/L LPS for 3 days. (2)The production of NO and sCD226 showed linear correlation when HUVECs were stimulated by 100 mg/L LPS.(3)LPS-induced sCD226 production in HUVECs was inhibited significantly by anti-TNF-alpha mAb and iNOS inhibitor. CONCLUSION: LPS-activated-HUVECs can secret sCD226 and the generation of sCD226 is closely correlated with the production of NO and TNF-alpha.

[Development and preliminary application of a double mAb sandwich ELISA for detecting soluble CD226].

AIM: To develop a double mAb sandwich ELISA for detecting sCD226 and apply it to clinical specimens. METHODS: The optimal concentrations of coating mAb and HRP-mAb, and the optimal diluents were determined. A double mAb sandwich ELISA then was established. The specificity, sentivity and stability of the method were identified. And clinical specimens were detected by this method. RESULTS: The optimal concentrations of coating mAb LeoA1 and HRP-mAb FMU3 were 2.5 mg/L and 1:400, respectively, and the optimal diluents for standard antigen and HRP-mAb were 1 g/L BSA 1 mL/L Tween20-PBS and 30 g/L PEG-PBS.The ELISA achieved satisfactory results in respect to specificity (no cross-reaction to human IgG, in block test blocking effect was does-dependent), sensitivity (the detected threshold was 110 ng/L) and stability (CV<10.2%). Statistical analysis showed that the serum sCD226 level in patients with rheumatoid arthritis (RA) was significantly higher than that in healthy adults. CONCLUSION: A double mAb sandwich ELISA with high specifity, and relative sensitivity for detection of sCD226 has been developed. It is shown to be an applicable method of detecting sCD226 in clinical specimens.

[Eukaryotic expression and application of human CD226 Ig fusion protein containing 3C protease-restricted site].

AIM: To construct, express and characterize the eukaryotic expression vector of encoding human CD226 (PTA1) extracellular region Ig fusion protein gene containing 3C protease-restricted site. METHODS: The gene fragment encoding extracellular region of human CD226 was cloned into the eukaryotic expression vector p-3C-Ig containing 3C protease-restricted site and human Ig Fc fragment gene. After sequencing, the vector was transfected into COS7 cells, and the expressed molecule was purified by affinity chromatography. Finally, the product was characterized by immunofluorescent staining and 3C protease digestion. RESULTS: After expression and purification, the Ig fusion protein could bind effectively to the CD226 ligand expressed on ECV304 cells. The Fc fragment could also be cut off by 3C protease, so that the CD226 extracellular fragment was obtained. CONCLUSION: The extracellular region of human CD226 Ig fusion protein with 3C protease-restricted site is expressed successfully in COS-7 cells, which lays the foundation for the structural and functional study of this molecule.

[Quality control of plant extract].

The current situation of plant extract in domestic and international market was analyzed in the paper. The quality control of 20 plant extracts which have reasonably good sales in USA market was compared and analyzed. The analysis of the quality control of six plant extracts indicated that there were two main reasons leading to the varied quality specifications among different suppliers. One reason was that the plant species utilized by different companies were different. The other reason was that the extraction processes were different among different production plants. Comparing with the significant international suppliers of plant extracts, the product quality of Chinese companies were not satisfactory. It was suggested that chromatography and chromatographic fingerprint techniques should be applied to improve the quality control standard of plant extract in our country.

[Studies on blood viscosity and external thrombus in patients with silicosis and silicosis complicated with tuberculosis].

OBJECTIVE: To explore the changes of blood viscosity and external thrombus in patients with silicosis and silicosis complicated with tuberculosis (TB). METHOD: Blood viscosity and external thrombus were measured in 288 patients with silicosis, 178 patients with silicosis complicated by TB and 150 healthy subjects. RESULTS: Blood viscosity and external thrombus value were significantly higher in the patients of silicosis and silicosis complicated with TB than in the healthy controls, except for patients of phase I of silicosis. Blood viscosity in the silicotics increased significantly with the advance of the disease, but no significant difference in external thrombus between patients in different phases. Apparent viscosity of whole blood significantly increased in the high-shear rate (200 s(-1)) and middle-shear rate (30 s(-1)) in patients of silicosis complicated with TB than in those without complication of TB at the same phases, but not seen in the low-shear rate (5 s(-1)) and in plasma viscosity, and the length and dried weight of external thrombus increased significantly too. There was no significant difference in blood viscosity and external thrombus between patients of silicosis at phase III and those of silicosis complicated with TB at the same phase. CONCLUSION: Blood in patients with silicosis appeared highly viscous and highly coagulant status. Blood viscosity and external thrombus value significantly increased with the advance of the disease, especially in the patients complicated with TB.

A new silymarin preparation based on solid dispersion technique.

Silymarin is a hepatoprotective agent that is poorly soluble in water. The present study describes a new preparation of solid dispersions in the form of "dripping pills" designed to enhance solubility. Dripping pills of silymarin were prepared at a 1:4 ratio by the traditional fusion method with the use of a mixture of silymarin and polyethylene glycol 6000 (PEG 6000). The prepared dripping pills were spherical and 3 to 4 mm in diameter, with an average weight of 30 mg per pill and with each pill containing 5 mg of silymarin. The dissolution rates of silymarin in dripping pill and of 3 other silymarin preparations, including Yiganling Film-Coating Tablet, Yiganling Sugar-Coating Tablet, and Legalon Capsule, were determined in pH 1.2 medium. The dissolution rate (T50) of the silymarin dripping pill was found to be significantly higher (by a factor of 7.5-11) than those of the other 3 preparations.

A pilot study of antitumor effect of gallium ethylenediaminetetramethylene phosphonate [Ga(III)-EDTMP] in tumor-bearing rats.

The inhibitory effects of gallium ethylenediamine-N,N,N',N'-tetrakismethylene phosphonate [Ga(III)-EDTMP] was studied on a malignant tumor and metastatic bone lesion model induced with Walker carcinosarcoma 256 (WCS 256) in Wistar rats weighing 120 to 135 g. A water-soluble chelate, Ga(III)-EDTMP, was prepared for injection in a tumor-bearing model. Radiographic analysis at 14 days indicated that the bone invasion and osteolysis were markedly reduced in animals treated with Ga(III)-EDTMP. The calculated tumor inhibitory rates were 33.16+/-0.38% at a dosage of 5 mg/kg and 47.75+/-0.74% at 10 mg/kg. Biochemical markers such as serum calcium decreased by 23% (P<.05) and by 31% (P<.01) at dosages of 5 mg/kg and 10 mg/kg, respectively, whereas serum alkaline phosphatase (ALPase) activity decreased by 47% (P<.01) and 69% (P<.01) at the 2 dosages.

Preparation of indapamide slow-release pellets.

During the process of developing a slow-release formulation of indapamide, researchers created a drug-containing pellet coated with Eudragit RS100 (Rohm GMbH & Co. KG, Darmstadt, Germany) to control the rate at which the drug was released. The two main variables were the agglomerants used in the pellet preparation and the amount of Eudragit RS100 used to coat them. The optimal outcome was indicated by the greatest number of drug-containing pellets recovered through an 18- to 24-mesh sieve and a satisfactory 24-hour release curve. The kinetics of dissolution fit the Higuchi kinetics model. Stability tests of the drug pellets showed no notable changes in the rate of drug release, related substances (mean byproducts or impurities from interactions or decompositions), and drug content.

Metabonomic study on the biochemical profiles of a hydrocortisone-induced animal model.

This work describes the metabonomic study of a biochemical modification in vivo induced by high dose of hydrocortisone, which led to a unique pathologic condition similar to the 'kidney deficiency syndromes', an early stage of obesity and diabetes in traditional Chinese medicine. The methodology of the metabonomic approach consisted of GC/MS and multivariate statistical technique for the establishment of urine metabolic patterns of the treatment rats. In the study, 24-h urine was collected pre-dose and at days 1, 3, 7, and 10 post-dose after rats were injected with hydrocortisone at 1.5 mg/100 g. The acquired data were transferred into Matlab to be processed using principal components analysis (PCA). The results indicated that clear and consistent biochemical changes following hydrocortisone intervention under controlled conditions could be identified using chemometric analysis. The work suggests that this metabonomic approach could be used as a potentially powerful tool to investigate the biochemical changes of certain physiopathologic conditions such as metabolic syndrome, as an early diagnostic means.

Genetic and antigenic diversity in avian infectious bronchitis virus isolates of the 1940s.

In order to verify a commonly held assumption that only Massachusetts (Mass) serotype of infectious bronchitis virus (IBV) was prevalent in the United States between the 1930s (when IBV was first isolated) and the 1950s (when the use of commercial IBV vaccines began), we examined 40 IBV field isolates from the 1940s. Thirty-eight of those isolates were recognized as Mass serotype viruses based on their reactivity to Mass-specific monoclonal antibody (Mab) and neutralization by Mass-specific chicken serum. The remaining two isolates, N-M24 and N-M39, that did not react with Mass-specific Mab, resisted neutralization by Mass-specific chicken serum, and were neutralized only by homologous chicken antibody were identified as non-Mass IBV. When the first 900 nucleotides (nt) from the 5'-end of the spike (S1) glycoprotein gene and their deduced amino acid (aa) sequences were compared, the two non-Mass isolates differed from each other by 24% and 28%, respectively. In a similar comparison, the non-Mass viruses N-M24 and N-M39 differed from M28, a Mass-type isolate from the 1940s, by 21% and 22% (nt) and 28% and 27% (aa), respectively. These data indicate that antigenic and genetic diversity among IBV isolates existed even in the 1940s. Interestingly, when the N-terminal region of the S1 of M28 was compared to that of M41, a prototype Mass virus that has undergone countless number of in vivo and in vitro host passages, the two viruses differed by only 2% (nt) and 4% (aa). This finding suggests that frequent genetic changes are not inherent in all IBV genomes.
