[Distribution of beta-thalassemia CD17 mutant(A-->T) in Shanxi, Gansu and Xinjiang along the "Silk Road"].

OBJECTIVE: To explore the distribution of beta-thalassemia CD17 mutant along the "Silk Road". METHOD: Polymerase chain reaction in combination with dot-blot hybridization of allele-specific oligonucleotide probes(PCR/ASO) was used. RESULTS: Twenty-four cases of beta-thalassemia with CD17 mutant(A-->T) including 3 cases in Shanxi, 20 cases in Gansu and 1 in Xinjiang, were found among 85 probands of beta-thalassemia. CONCLUSION: According to distribution characteristics of beta-thalassemia CD17 along the "Silk Road" of China, the CD17 mutant of beta-thalassemia might probably originate from the "Silk Road" in Longxi district of Gansu.

[Gene identification and pedigree analysis of the double mutation heterozygote of beta-thalassemia[-28(A-->G).CD17(A-->T)/N]].

OBJECTIVE: To report the result of gene identification and pedigree analysis of the double mutation heterozygote of beta-thalassemia[-28(A-->G).CD17(A-->T)/N]. METHODS: Polymerase chain reaction in combination with dot-blot hybridization of allele-specific oligonucleotide(PCR/ASO) probes was used. RESULTS: Gene identification was carried out in four probands and 35 other family members. There were 22 patients with beta-thalassemia -28(A-->G) and CD17(A-->T) mutant gene. Based on the clinical features, the results of hematological and biochemical examinations and pedigree analysis, they proved to be the double mutation heterozygote of beta-thalassemia[-28(A-->G).CD17(A-->T)/N], i.e., double point mutations on the same chromosome. CONCLUSION: Double mutation heterozygote[-28(A-->G).CD17(A-->T)/N] is a new type of beta-thalassemia.

[Effect of antisense oligodeoxynucleotide (ASODN) to PML-RAR alpha fusion gene on acute promyelocytic leukemia cell line NB4].

OBJECTIVE: To study the effect of PML-RAR alpha expression on the proliferation and differentiation of NB4 cells, and explore the role of PML-RAR alpha fusion gene in the pathogenesis of acute promyelocytic leukemia (APL). METHODS: ASODN-mediated inactivation of PML-RAR alpha mRNA was measured by RT-PCR. The proliferation and differentiation of NB4 cells were determined by proliferation curve, morphology, membrane CD antigen and NBT test. NB4 cell cycle was analyzed by FACS. RESULTS: The inactivation of PML-RAR alpha mRNA by ASODN decreased the percentage of S phase cells (from 56% to 37%), inhibited the NB4 cell proliferation and induced NB4 cells to differentiate to morphologically and functionally mature granulocytes. CONCLUSION: PML-RAR alpha gene, as a molecular marker of APL, is possibly responsible for genesis of APL.

[Study of hemoglobinopathy in Kashi district of Xinjiang].

OBJECTIVE: To study the hemoglobinopathy in Kashi district of Xinjiang. METHODS: The abnormal hemoglobin carriers were investigated with microelectrophoresis. The propositus of thalassemia were tested with two steps of screening technique. The structural analysis of abnormal hemoglobin was carried out with finger print analysis technique. Gene identification of thalassemia was performed with PCR/ASO technique. RESULTS: The incidence of hemoglobinopathy and thalassemia were 0.8% and 3.07%, respectively. Both were higher than the average level in the population of Xinjiang and the whole country. Two kinds of variant of HbJ Tashikuergan [alpha 19(AB1)Ala-->Glu] and HbD Punjab[P121(GH4)Glu-->Gln] were found in the structural analysis of 8 cases of abnormal hemoglobin. Four kinds of mutation: CD8(-AA),CD8/9(+G),CDs41/42(-TTCT) and IVS-I-5(G-->C) were revealed at the gene identification of 10 propositus of beta-thalassemia. HbJ Tashikuergan was firstly reported in the world, and CD8(-AA) and CDs8/9(+G) were firstly discovered in China. CONCLUSION: The types and distribution of abnormal hemoglobin and thalassemia in Kashi district were different from those in other areas of our country, and also unsimilar to our neighbour countries.

[Ultrasound bone measurement of the tibia: comparison with vertebral dual-energy X-ray absorptiometry and appendicular single photon absorptiometry].

OBJECTIVE: Ultrasonic techniques have been used to assess the mechanical properties of bone. Recently, a new ultrasonic bone instrument, which measured the SOS (speed of sound) at the tibial shaft, has been introduced (Sound Scan 2000, Myriad Ultrasound Systems Ltd.). The aim of this work was to investigate the correlation of tibial SOS with bone mineral density (BMD). METHODS: The tibial SOS was measured in 213 healthy women (aged 22-83 years) and 56 female primary osteoporosis with vertebral fractures (aged 53-82 years). Values of SOS were compared with BMD assessed by dual energy x-ray absorptiometry (DEXA) at spine and hip sites and single photon absorptiometry (SPA) at the distal forearm. RESULTS: Tibial SOS correlated more closely with the BMD of lumbar, hip and forearm (r = 0.387-0.928, P < 0.001). The tibia SOS values and the BMD values measured by the DEXA and SPA were negatively correlated to age and years since menopause. In particular, they were obviously lower in the group of 50 years old. The tibial SOS of vertebral fracture group was significantly lower than that of well-matched non-fracture-group (n = 104, aged 51-84 years). CONCLUSIONS: Tibial SOS was significantly correlated with BMD measured by DEXA or SPA respectively and able to discriminate patients with vertebral fracture from the age-matched controls.

[Simultaneous determinations of carbamate pesticides in cereal by high performance liquid chromatography with post-column fluorescence derivatization].

A liquid chromatographic method for the simultaneous determination of carbamate pesticides in cereal by Waters Carbamate Analysis System with post-column fluorescence derivatization is described. The cereal sample homogenate was extracted with acetone and water. The extract was partitioned between 5% sodium chloride solution and dichloromethane. The dichloromethane layer was concentrated to dryness. The residue was dissolved in n-hexane and extracted with acetonitrile (saturated with n-hexane). The acetonitrile layer was decreased with n-hexane (saturated with acetonitrile), then the acetonitrile layer was concentrated to dryness. The residue was dissolved in methanol. It's the test solution for HPLC. Nine carbamate pesticides and their three metabolites were separated by Waters Carbamate Column with gradient elution. Two steps underwent in the post-column system: hydrolysis by NaOH solution and the reaction of the product with o-phthalaldehyde and 2-mercaptoethanol. The resulting fluorescence derivative was detected at 445 nm (excitation at 339 nm). The selectivity, reproducibility and sensitivity of proposed method are better than those reported with other methods. The detection limits was 5 micrograms/kg of samples.

[Effects of acetylsalvianolic acid A on arachidonic acid metabolism in platelets].

Acetylsalvianolic acid A (ASAA) has been shown to be an antiplatelet compound. Our studies with RIA showed that: at the dosage of inhibiting platelet aggregation, ASAA was found to inhibit the formation of cyclooxygenase pathway metabolites TXB2 in platelets. In addition, ASAA was also shown to promote the formation of 6-keto-PGF1 alpha in rabbit aortic rings in vitro. The above results suggest that ASAA may inhibit the proaggregator TXA2 formation, promote mildly the antiaggregator PGI2 generation and probably, whereby, exert its antiplatelet activity.

[A study of color matching of maxillofacial prosthesis]

OBJECTIVE:The color difference between the patient's facial prosthesis and the surrounding skin have long puzzled both of dentist and patient.The purpose of the study was to introduce spectrophotometer into the color matching and set up a set of equations to increase the accuracy of the color matching,Referring to increase the accuracy of the color matching.Referring to color gamut,several sets of shade guide were made according to sixteen kinds of mixture-ratio of four pigments such as red,yellow,white and black,and every shade guide was measured.The mixture-ratio was related to the values of the CIE system by means of statistics,which resulted in the equations for color matching.In a word,use of the spectrophotometer may improve the accuracy of the the color matching of facial prosthesis.

Co-compartmentalization of MAP kinases and cytosolic phospholipase A2 at cytoplasmic arachidonate-rich lipid bodies.

Lipid bodies are inducible lipid domains abundantly present in leukocytes engaged in inflammation. They are rich in esterified arachidonate and are also potential sites for eicosanoid-forming enzyme localization. It is therefore of interest to know whether arachidonate-releasing cytosolic phospholipase A2 (cPLA2) localizes at lipid bodies. Here, we present evidence that cPLA2 and its activating protein kinases, mitogen-activated protein (MAP) kinases, co-localize at lipid bodies. U937 cells express high levels of cPLA2 and contain numerous cytoplasmic lipid bodies. Using double-labeling immunocytochemistry we demonstrated punctate cytoplasmic localizations of both cPLA2 and MAP kinases in U937 cells that were perfectly concordant with fluorescent fatty-acid-labeled lipid bodies. The co-localization of cPLA2 and MAP kinases at lipid bodies was confirmed by subcellular fractionation and immunoblot. Lipid body fractions free of cytosol and other organelles contained significant amounts of [14C]arachidonate-labeled phosphatidylcholine and cPLA2 enzymatic activities. Immunoblotting with specific antibodies identified cPLA2 as well as MAP kinases, including ERK1, ERK2, p85, and p38, in lipid bodies. The co-compartmentalization within arachidonate-rich lipid bodies of cPLA2 and its potentially activating protein kinases suggests that lipid bodies may be structurally distinct intracellular sites active in extracellular ligand-induced arachidonate release and eicosanoid formation.

[The types and distributive features of gene mutation on beta-thalassemia in three districts in northwest China].

OBJECTIVE: To sum up the results of gene analyses on beta-Thalassemia in three districts in northwest China and discuss the types and distributive features of the mutation. METHODS: Polymerase china reaction in combination with dot-blot hybridization of allele-specific oligonucleutide probes(PCR-ASO). RESULTS: In the gene analysis carried out in 85 probands with beta-Thalassemia, twelve types of gene mutation were identified from five nationalities. CD8(-AA), CDs8-9(+G), CDs27-28(+C) heterozygote and [-28(A-->uG). CD17(A-->uT)/N] double heterozygote were firstly assayed in these districts besides the common types in Chinese, of which, [-28(A-->uG). CD17(A-->uT)/N] as double gene mutation in the same chromosome is rare. CONCLUSION: There are significant differences in the distribution and nationality features of gene mutation types for beta-Thalassemia in the three districts, so the three districts as a whole is an area with special distributive features of the disease.

RRR-alpha-tocopheryl succinate induction of prolonged activation of c-jun amino-terminal kinase and c-jun during induction of apoptosis in human MDA-MB-435 breast cancer cells.

We have demonstrated that RRR-alpha-tocopheryl succinate (10 microg/mL vitamin E succinate (VES) treatment of estrogen receptor-negative MDA-MB-435 human breast cancer cells induces 9, 19, 51, and 72% apoptotic cells on days 1-4, respectively, after treatment, which involves transforming growth factor-beta signaling. Here, we show that VES-triggered apoptosis of MDA-MB-435 cells induced prolonged elevated expression of c-jun mRNA and protein (neither of which was caused by major increases in stability) and also induced enhanced activator protein-1 (AP-1) binding to the consensus DNA oligomer. Furthermore, VES treatments resulted in increased AP-1 transactivation activity, as measured with an AP-1 promoter/luciferase reporter construct and by the measurement of increased mRNA expression of the AP-1-dependent endogenous gene collagenase. Evidence of VES-induced involvement of the c-jun amino-terminal kinase in these AP-1-dependent events was suggested by data showing prolonged activity of this kinase, as measured by a kinase assay using glutathione S-transferase-c-jun as the substrate. The c-jun-dependent transcriptional activity was verified by cotransfection of a chimeric transcription factor having a galactose 4 DNA-binding domain coupled with the transactivation domain of c-jun plus the reporter plasmid 5X GAL4-luciferase. MDA-MB-435 cells infected with an adenovirus expression vector containing the TAM-67 sequence for dominant/negative-acting mutant c-jun or transiently transfected with c-jun antisense exhibited a 50-77% reduction in VES-mediated apoptosis as compared with control adenovirus-infected or control sense oligomer-transfected cells.

Morbidity and mortality of early postoperative intraperitoneal chemotherapy as adjuvant therapy for gastric cancer.

Intraperitoneal chemotherapy (IC) is emerging as a valuable adjuvant therapeutic modality in patients with gastric cancer. The purpose of this study was to assess morbidity and mortality of early postoperative IC (EPIC) in gastric cancer patients. Two hundred forty-eight gastric cancer patients thought to have resectable cancer were randomized intraoperatively to receive EPIC with mitomycin C on postoperative day 1 and 5-fluorouracil on postoperative days 2 to 5 versus surgery only. Sixty-four patients who were stage IV at histopathologic examination remain in the analysis. Morbidity and mortality were compared using Fisher's exact test. All patients completed the therapy. In the study group, overall morbidity was higher than in the control group (28.8% versus 20.3%, respectively), although the difference was not significant (P = 0.121). Intra-abdominal sepsis without anastomotic leak (P = 0.008) and bleeding (P = 0.002) occurred significantly more often in the study group. Also, 37.6 per cent of patients who received EPIC experienced a variety of minor complications attributable to EPIC. Postoperative mortality was higher in the study group (5.6%) than in controls (0.8%), but not significantly (P = 0.299). Patients treated with EPIC stayed in the hospital an average of 4 days longer (P = 0.002); in patients with morbidity, however, there was no difference with the control group. A period analysis of the morbidity demonstrated that it followed the pattern of a learning curve. Surgery with EPIC tended to increase the postoperative morbidity and mortality. The therapy-associated risk must be justified by a significant improvement in survival of treated patients with stage III disease. Selective application of perioperative IC may be indicated.

Reduced anti-allodynic effect of the adenosine A1-receptor agonist R-phenylisopropyladenosine on repeated intrathecal administration and lack of cross-tolerance with morphine in a rat model of central pain.

UNLABELLED: We examined the development of tolerance to the antiallodynic effect of chronic intrathecal (IT) administration of the adenosine analog R-phenylisopropyladenosine (R-PIA) in a rat model of central pain after ischemic spinal cord injury. After 10 days of IT R-PIA treatment, the effect of IT morphine was also assessed to examine whether cross-tolerance between R-PIA and morphine was present. IT R-PIA completely alleviated allodynia-like behaviors to mechanical and cold stimuli in spinally injured rats. The anti-allodynic effect of R-PIA was maintained for 6-7 days with twice-daily administration and was reduced thereafter, particularly with respect to cold allodynia. IT morphine alleviated mechanical and cold allodynia in rats rendered tolerant to R-PIA to a degree comparable to that in R-PIA-naive (control) rats, which indicates that the anti-allodynic property of R-PIA is independent of the mechanisms by which morphine acts. The possibility of using agonists of adenosine receptors in treating refractory pain in patients with spinal cord injury is discussed. IMPLICATIONS: There is often no satisfactory treatment for chronic pain after spinal cord injury. Our study suggests such pain can be treated with a spinal injection of R-phenylisopropyladenosine in rats. Reduced effect to R-phenylisopropyladenosine was noted with repeated administrations. However, there was no cross-tolerance to morphine.

Eosinophil lipid bodies: specific, inducible intracellular sites for enhanced eicosanoid formation.

The specific intracellular sites at which enzymes act to generate arachidonate-derived eicosanoid mediators of inflammation are uncertain. We evaluated the formation and function of cytoplasmic lipid bodies. Lipid body formation in eosinophils was a rapidly (<1 h) inducible response which was platelet-activating factor (PAF) receptor-mediated, involved signaling through protein kinase C, and required new protein synthesis. In intact and enucleated eosinophils, the PAF-induced increases in lipid body numbers correlated with enhanced production of both lipoxygenase- and cyclooxygenase-derived eicosanoids. All principal eosinophil eicosanoid-forming enzymes, 5-lipoxygenase, leukotriene C4 synthase, and cyclooxygenase, were immunolocalized to native as well as newly induced lipid bodies in intact and enucleated eosinophils. Thus, lipid bodies are structurally distinct, inducible, nonnuclear sites for enhanced synthesis of paracrine eicosanoid mediators of inflammation.
