[Changes of GABA immunoreactivity in aged rat cochlear nucleus].

OBJECTIVES: To detect the changes of gama-aminobutyric acid (GABA) immunoreactivity in the aged rat cochlear nucleus and to investigate its relationship with the initiation of presbycusis. METHODS: The cochlear nucleuses of adult (2-3 months) and aged (20-36 months) Wistar rats were stained with streptavidin/peroxidase (S-P) technique. GABA-positive neurons were counted with a computer-aid image analysis system. RESULTS: Most of the GABA-immunoreactive cell bodies were found in the superficial layers of the dorsal cochlear nucleus (DCN). The number of GABA-positive neurons decreased significantly in the DCN of the aged rat cochlear nucleus (P < 0.001). CONCLUSION: The reduction of GABA positive neurons in the aged rat cochlear nucleus may be related to presbycusis.

[Effects of gentamicin on survival and neuritogenesis of cultured mouse spiral ganglion cells].

The effect of gentamicin on survival and neuritogenesis of cultured spiral ganglion cell(SGC) from NIH mouse 1-5 days after birth was observed. Different doses (25, 50 and 100 mg/L) of gentamicin were add to the medium of SGC, and cultured for 14 days. After that, the specimen was fixed with 4% paraformaldehyde and stained with toluidine blue. The amount of survival SGC was counted, and the length of neurite was measured with micrometer under microscope. It was shown that the survival and length of neurite of SGC had no difference between Gentamicin treated groups and control groups, and there were also no difference among 3 gentamicin treated groups. It was suggested that gentamicin had no effect on survival and neuritogenesis of SGC. The effects of aminoglycoside antibiotics on SGC were probably secondary to the injury of Corti's Organ.

[The role of cytokeratin 7 in the differential diagnosis of primary ovarian carcinoma and metastatic ovarian carcinoma originated from the gastrointestinal tract].

OBJECTIVE: To assess the role of cytokeratin 7 monoclonal antibody in the differential diagnosis of primary ovarian carcinoma and metastatic ovarian carcinoma originated from the gastrointestinal tract. METHODS: Immunohistochemical study using cytokeratin 7 monoclonal antibody and ABC kit. RESULTS: All the 46 cases of primary ovarian carcinoma were CK 7 positive, while in the metastatic ovarian carcinoma of intestinal origin, all cases remained negative for CK7. Half of the 34 cases of metastatic ovarian carcinoma of gastric origin were CK 7 positive. The positive result of CK7 was significantly higher in the primary ovarian carcinoma than in each group of the metastatic ovarian carcinoma (P < 0.001). CONCLUSION: CK 7 is seemed to be a useful antibody in the differential diagnosis of ovarian carcinoma.

[Effect of HHV-6 on expression of M-CSF in patients with hematological disorders].

OBJECTIVE: To study the relationship between HHV-6 infection and abnormal expression of M-CSF in bone marrow mononucleated cells (MNC) from patients with hematological disorders. METHODS: M-CSF expression level were detected in 30 specimens of MNC from patients with hematological diseases by ABC immunocytochemical staining and reverse transcription polymerase chain reaction methods. RESULTS: The percentage and score of M-CSF antigen positive cells, and percentage of M-CSF mRNA expressing samples derived from HHV-6 infected group were 36.9% +/- 24.9%, 45.9 +/- 31.4 and 83.3%, respectively, which were significantly higher than those of control (20.3% +/- 19.4%, 25.3 +/- 24.2 and 50.0%, respectively) (P < 0.001). HHV-6 could promote M-CSF production to a large extent in MNC from leukemic patients than that from patients with benign hematological disorders. M-CSF existed mainly on membrane or in cytosol of granulocytes and monocytes. CONCLUSION: HHV-6 infection in vitro could promote M-CSF production in MNC from patients with hematological diseases.

[Experimental study on mechanism of xiukeyin in anti-bacterial translocation from gut].

OBJECTIVE: To provide a TCM therapeutic base for treatment of critical multiple organ failure (MOF) patients clinically, the mechanism of Xiukeyin (XKY, Shock Beverage) in anti-bacterial translocation from gut in rats was studied. METHODS: Based on the pathophysiology of MOF following severe injury, such as shock caused enterogenous-sepsis and bacterial translocation from gut, a hemorrhagic shock model of rat was established and used to determine the effect of XKY on anti-bacterial translocation. XKY was formulated by modifying Wenpi Decoction. RESULTS: The incidence of intestinal bacterial translocation to mesenteric lymph nodes, liver and spleen was lower in the model rats infused via gastrogavage with XKY (3/15) than that in the noninfused rats (11/13) (P < 0.01). The amount and species of intestinal flora in XKY infused or noninfused rats were not different statistically (P > 0.05). Histological examination showed that the intestinal edema was severer in the model group than that in the XKY treated group. CONCLUSIONS: XKY could inhibit the shock induced enterogenous bacterial translocation, the mechanism might be due to its protective action on intestinal mucosa. XKY showed no effect on the growth of intestinal bacteria.

[Dynamic effect of tifukang on brain proopiomelanocortin gene expression of exercise-induced fatigue rat].

OBJECTIVE: To study the mechanism of exercise-induced fatigue (EIF) and effect of Tifukang (TFK) from the gene transcription level. METHODS: In situ hybridization was adopted, using biotin-labelled proopiomelanocortin (POMC) cRNA probe and integrating with image pattern analysis, to observe the effect of TFK on changes of POMC mRNA expression in different areas of EIF rat's brain. RESULTS: TFK could significantly strengthen POMC mRNA expression in hippocampal CA1 and hypothalamus and inhibit obviously that in frontal cortex of chronic fatigue rats. In rats managed with long term (7 weeks in total) and high intensity exercise with the speed increased from 15 m/min gradually to 35 m/min, 20 to 25 min/d, after the final exercise, the POMC mRNA expression in frontal cortex and hippocampal CA1 decreased (the latter recovered at 30 minutes after ending exercise but the former was not recovered yet 3 hours after the ending of exercise), while that in hypthalamus increased. TFK could remarkably increase POMC gene expression in the above-mentioned three areas after the final exercise especially prominent at the time of 3 hours after the ending of exercise. CONCLUSION: The dynamic changes of POMC gene expression in different areas of EIF rat's brain were not the same, and TFK displayed an important dynamic regulating effect on it.

[Screening for the 1555G mutation in mitochondrial DNA in pedigrees with aminoglycoside antibiotic induced deafness].

OBJECTIVE: To identify the relationship between the 1555G mutation in mitochondrial DNA and aminoglycoside antibiotic induced deafness and provide theoretical evidence for establishing diagnostic method. METHODS: Blood samples were obtained from three pedigrees with aminoglycoside antibiotic induced deafness. DNA was extracted from the isolated leukocytes. The mitochondrial DNA fragments were amplified by PCR, 1555G mutation was detected by Alw26 I restriction endonuclease digestion, allele-specific oligonucleotide hybridization and DNA sequencing. RESULTS: Seven individuals from pedigrees A and C carried homoplasmic 1555G mutation, six individuals from pedigree B did not have 1555G mutation. CONCLUSION: 1555G mutation is not the only pathologic mitochondrial DNA mutation associated with aminoglycoside antibiotic induced deafness. This is the first finding of aminoglycoside antibiotic induced deafness pedigree without 1555G mutation.

[Jugular bulb anomaly].

OBJECTIVE: To differentiate and treat correctly the jugular bulb anomalies, as well as to avoid the unnecessarily enlarged operations. METHODS: Four cases of jugular bulb anunalies were reported and analysed. RESULTS: Two of 4 cases were misdiognosed as jugular glomus tumors; One was considered to be jugular bulb anomaly, but was done with an enlarged operation; another one was still accompanied with benign intracranial hypertension syndrome, was inappropriately treated. CONCLUSION: The jugular bulb anomaly was often misdiagnosed, therefore the anomaly should be differentiated from the jugular glomus tumor carefully.

[Application of three demensional reconstruction of CT in lateral cranial base surgery].

OBJECTIVE: To study the characteristics of 3D-CT of the lateral cranial base and its application in lateral cranial base surgery. METHODS: Five normal adult subjects and 10 with lateral cranial base lesions were scanned with Philips Tomoscan SR 7000 CT scanner. The scanning ranges extended from the top to the bottom of the temporal bone, including whole lesions, with bone window algorithm and contiguous serial 1.5 mm thin-cut scans in succession in axial plane. The 3D-CT reconstruction was processed in the Easy Viewing CT/MRI workstation. RESULTS: The 3D-CT reconstruction could clearly delineate the foraminan, fissures and important bony marks. The ranges of the lesions, the relationships between the lesions and the structures around them were demonstrated more clearly than 2D-CT. It was of important significance in differential diagnosis for jugular foramen lesions. 3D-CT could be used to imitate operation and design operative plan. CONCLUSION: 3D-CT was of important value in diagnosis of lateral cranial base tumors, and could be used to imitate operation and design operative plan.

[Study on the methodology of potassium channel expression in Xenopus oocytes by messenger RNA from rat cochlear nucleus].

OBJECTIVE: To study the properties of voltage-dependent ion channels expressed in Xenopus oocytes by mRNA from rat cochlear nucleus and to lay a methodological foundation for studying the changes of ion channels in auditory centre with sensorineural hearing loss. METHODS: Poly (A)+ mRNAs isolated from the rat cochlear nucleus by acid guanidinum thiocyanate-phenol-chloroform (AGPC) method and Promega's new magnesphere technique were microinjected into Xenopus laevis oocytes to express functional ion channels. Voltage clamp technique was used to record the current of voltage-dependent ion channels. RESULTS: A voltage-dependent ion channel was detected from oocytes injected with rat cochlear nucleus mRNA. The maximum amplitude of the outward current was 364 +/- 42 nA (n = 16). It was identified as transient outward potassium channel. CONCLUSION: The model set up by the test may play an important role in studying the physiology, pathology and pharmacology of auditory centre.

[Nasal neuroendocrine carcinoma].

OBJECTIVE: To recognize and understand further nasal neuroendocrine carcinoma (NNC), improve the diagnosis and therapeutic effect. METHODS: Eight cases of the nasal neuroendocrine carcinomas (7 males, 1 female) were analyzed. Among them, 6 cases had tumors in left side of nose, 2 in right side. The average course was only 2.8 months, but the lesions were often extent. RESULTS: The patients were pathologically confirmed to be neuroendocrine carcinomas. 5 cases were of small cell type, 2 undifferentiated, 1 poorly differentiated. One patient died 14 months after treatment. 2 cases were lost for follow-up. The others were still in treatment. CONCLUSION: The nasal neuroendocrine carcinoma has rapid progress and poor prognosis. Clinically, combined treatment should be adopted, the effect of simple operation was poor.

[Stripping operation for auricular angioma].

OBJECTIVE: To improve the treatment of auricular angioma. METHOD: Twelve cases of auricular angioma were clinically analyzed and several treatments compared. RESULT: Eight of 12 cases were cured by the stripping operation, in which 7 cases had histories of recurrences with other methods. The lesions in 3 of 12 cases were controlled with the current treatments. The auricular in one case was totally excised due to the auricular fester, infection and necrosis of the cartilage. The cervico-facial trunk of facial nerve in one case during the stripping operation was surgically injured and sutured immediately by the end to end anastomosis. Facial paralysis disappeared half year after the operation. These cases were followed up from 8 months to 4 years. Only one case had local recurrence. CONCLUSION: Stripping operation can provide radical cure and is recommended for the treatment of auricular angioma.

[Isolation and observation of ciliated cells from guinea pig's nasal epithelium].

OBJECTIVE: To investigate nasal physiological function at a cellular level and to establish a method for isolating single ciliated cells from guinea pig's nasal epithelium. METHODS: The ciliated cells were isolated by means of mechanical dissociation after papain treatment and a differential interference contrast (DIC) microscope was used for 1000-fold observation (objective lens: 100-fold; eye piece: 10-fold). RESULTS: A total of 12,587 single ciliated cells could be isolated from nasal epithelium of each guinea pig. The criteria for good viability of isolated nasal ciliated cells include: (1) actively motile cilia; (2) a smooth cytoplasmic membrane; (3) cytoplasm in a state of translucence (birefringence); (4) cells neither swollen nor shrunk. With short term culture at room temperature, the isolated nasal ciliated cells retained a good viability for 2-9 hours or longer. The living ciliated cells showed swing motions of cilia with fast and slow phases at a rate of 51-119 times per minute. CONCLUSION: A method for isolating ciliated cells from guinea pig's nasal epithelium and a criteria for good viability of isolated ciliated cells have been established. Ciliary motion pattern and ciliary beating frequency were also studied.

[Extraction, amplification, recombination and sequencing of the mitochondrial DNA from celloidin embedded human temporal bone sections].

OBJECTIVE: To establish an analytical method of DNA extracted from human celloidin-embedded temporal bone sections by molecular biologic technique. METHODS: Polymerase chain reaction (PCR) with different primers, special amplification methods or coorperated with enzyme restricted reactions was adapted to detect several types of genetic mutations from 9 human celloidin-embedded temporal bone sections. The pGEM-T vector was used for the recombination and cloning of the amplified DNA fragment. RESULTS: The 135 bp fragments of normal mtDNA were detected from all of the 9 cases. The 316bp fragment related to mtDNA 4977bp deletion was also detected in two cases who suffered from presbycusis before death by the nested PCR. The result of sequencing confirmed the accuracy of PCR. CONCLUSION: The application of molecular biologic techniques in temporal bone research is important in improving the quality and value of retrospective study of ear diseases and has resulted in findings of the relevant mutant or pathogenetic genes in the ear diseases such as presbycusis, ototoxic deafness, otosclerosis and viral infections of the ear.

[Isolation of and intracellular calcium mobilization in vestibular hair cells of the patients with acoustic neuroma].

OBJECTIVE: To further understand the biological features of the human vestibular organ. METHODS: Vestibular hair cells were isolated from patients with acoustic neuroma during the surgery and intracellular free calcium concentration of the cells was monitored with calcium-sensitive dye Fura-2 and digital imaging microscopy. RESULTS: Isolated vestibular hair cells were mainly of type I and could survive about 2.5 hours in vitro. Perfusion with 150 mmol/L KCl, induced a depolarization, resulting in a temporary increase of 340 nm/380 nm ratio from 0.52 to 1.16 within 1 min. In Ca2+ free environment, 150 mmol/L KCl failed to induce intracellular calcium increase. In the presence of 2.5 mumol/L ionomycin, there was an irreversible increase in 340 nm/380 nm ratio. CONCLUSION: Human vestibular hair cell apparently possess voltage sensitive calcium channels. The method and observation aid in elucidating human vestibular biological features.

[Effect of basic fibroblast growth factor on cultured spiral ganglion cells following glutamate treatment].

OBJECTIVE: To understand the effects of basic fibroblast growth factor (bFGF) on survival and neurogenesis of the cultured spiral ganglion cell (SGC) of mouse after exposed to glutamate, and the effects of hFGF and glutamate on free intracellular calcium ion concentration. METHODS: After exposed to 2 x 10(-2) mol/L glutamate for 2 hours, the medium of SGC was replaced with the media containing 25, 50, 100 micrograms/L bFGF. Medium without bFGF was used as control. RESULTS: Twenty-four hours later, more cells in the control group showed signs of cell death than in bFGF groups. After 14 days, specimens were fixed in 4% paraformaldehyde and stained with toluidine blue. It was shown that the SGC in medium with bFGF exhibited more survival and longer neurites, and the effect was dose dependent (P < 0.01). Application of glutamate to the medium induced an increase to the the free intracellular calcium ion concentration in SGC, but bFGF had no such effect. CONCLUSION: Glutamate excitotoxicity was associated with free intracellular calcium ion concentration elevation in SGC, and bFGF could protect SGC against glutamate neurotoxicity.

[Expressions of fibronectin and laminin in the middle ear mucosa with experimental middle ear effusions: immunohistochemical study].

OBJECTIVE: To understand the role of two components of extracellular matrix: fibronectin (FN) and laminin (LN) in the proliferated mucosa of otitis media with effusions. METHODS: A model of otitis media with effusions was established by coagulating the orifices of the eustachian tube under an endoscope in 30 guinea pigs. The expressions of FN and LN in the proliferated mucosa from these ears were detected using Streptavidin peroxidase (SP) method. RESULTS: In 60 ears with the orifices of the eustachinan tube coagulated, 28 ears showed otitis media with effusions. The increased expressions of FN on the fibroblasts and LN on the basement membranes and neovasculations were noted in the ears with otitis media with effusions. CONCLUSION: FN and LN may involve in the proliferated response of middle ear mucosa with otitis media with effusions. It may play a certain role in the occurrence of middle ear mucosa proliferations and adhesions.

[90Y glass microspheres intraradiopraxis of advanced hepatic carcinoma].

OBJECTIVE: To evaluate intraradiopraxis with (90)Y glass microspheres for advanced hepatic carcinoma. METHOD: 28 patients were divided into two group: A treated by (90)Y and hepatic artery embolism and chemotherapy (HAEC) (n = 13), and B by (90)Y and hepatic artery embolism (HAE) (n = 15), and 15 cases (group C) only treated by HACE simultaneously were used as controls. The average age, course, and diameter of tumor were similar in all groups (P > 0.05). RESULT: The effective rate of the group A and B was 61.5% and 66.6% respectively, and that of the group C was only 33.3% (P < 0.001). The average survival time of the group A and B was 9.86 and 11.9 months respectively, and that of the group C was only 4.5 months (P < 0.001). CONCLUSION: The intraradiopraxis with (90)Y glass microsphere would be a valuable method for patients with hypo-leukocyte count, insensitive to chemotherapy or resistant to chemotherapy.
