STR typing in a deficiency case of disputed paternity.

In a case of disputed paternity where the alleged father was dead and the children and their mothers were available for testing, we analyzed 23 conventional hemogenetic markers and six short tandem repeat loci. We deduced the possible types of the alleged father and calculated the probability of paternity. Conventional phenotypings did not provide conclusive proof of paternity. In contrast, typing of the short tandem repeat loci gave a combined probability of paternity of 0.9986, establishing paternity. The present study demonstrates the power of short tandem repeat typing as a tool for solving such a deficiency case.

[Phylogenetic relationships among six species of Macaca inferred from ribosomal DNA variations].

Restriction maps of ribosomal DNA repeating units of six species of Macaca (M. mulatta, M. facsicularis, M. arctoides, M. assamensis, M. thibetana, M. nemestrina) and two outgroup species, Rhinopithecus bieti and Hylobates leucogenys, were constructed with 15 restriction endonucleases and 28S, 18S rDNA probes cloned from human, M. arctoides shares a common restriction map with M. assamensis. Phylogenetic tree derived from the neighbor-joining was somewhat different that derived from the maximum likelihood methods using the NEIGHBOR and RESTML program in PHYLIP version 3.5c. However, the consensus of our phylogenetic trees supports the viewpoint that M. facsicularis and M. mulatta were maybe diverged first in the genus Macaca. Further studies using DNA sequencing method are required to resolve the phylogeny of the genus Macaca.

[Expression and significance of hepatitis B virus genes in human primary intrahepatic cholangiocarcinoma and its surrounding tissue].

In order to explore the significance of HBV in the pathogenesis of cholangiocarcinoma at molecular level, HBV DNA, X gene, pre-S gene, S gene and C gene in 40 cases of primary intrahepatic cholangiocarcinoma and its surrounding tissue were detected by in situ hybridization technique. The results showed that 33 cases (82.5%) were positive for HBV DNA, while 31 (77.5%), 26 (65.0%), 24 (60.0%) and 27 (67.5%) cases were positive for X gene, pre-S gene, S gene and C gene, respectively. The results of this study suggest that there exists a close relationship between human primary intrahepatic cholangiocarcinoma and HBV chronic persistent infection. The expression of X gene might play an important role in the pathogenesis of the primary intrahepatic cholangiocarcinoma.

[Endolaser photocoagulation in vitrectomy surgery].

OBJECTIVE: To verify the usefulness and effectiveness of endolaser photocoagulation in vitrectomy. METHODS: Endolaser photocoagulation was applied in 40 cases (45 eyes) of vitrectomy surgery. Indications for vitrectomy included vitreous hemorrhage 31 eyes (proliferative diabetic retinopathy 13, Eales's disease 13, retinal vein occlusion 4, and scleral penetrating injury with intraocular foreign body removal 1), complex retinal detachment 12 eyes, intraocular nonmagnetic foreign body removal and postoperative silicone oil tamponade 1 eye respectively. Treatment involved pan-retinal scatter therapy in 7 eyes, focal therapy in 22 eyes, sealing primary or iatrogenic retinal breaks in 14 eyes, draining retinotomy in 2 eyes, and encircling scleral buckle photocoagulation in 3 eyes. The follow-up times ranged from 2 to 18 months (average 9 months). RESULTS: Whitening of laser spots was not apparent in 6 eyes with retinal breaks following incomplete gas-fluid exchange. Recurrent retinal detachments occurred in 5 eyes postoperatively. CONCLUSION: Endolaser photocoagulation facilitates the performance and reduces the complications of vitrectomy surgery.

[Relationship between the express product of src gene (pp60c-src) and the initiation of gastric carcinoma].

The tissues of the fetal gastric epithelia, adult normal gastric epithelia, gastric epithelial lesions, gastric carcinoma and the tissues adjacent to carcinoma were studied immunohistochemically by using the specific monoclonal antibody of c-src gene express product (pp60c-src), MAb 327. The positive rates of pp60c-src in the fetal gastric epithelia, adult gastric epithelial inflammatory hyperplasia, intestinal glandular metaplastia and gastric carcinoma were 100.0% (12/12), 100.0% (30/30), 100.0% (16/16), 83.3% (40/48), respectively, all these pp60c-src positive rates were higher than that of adult normal gastric epithelia (60.0%, 12/20), a significant difference was noted statistically (P < 0.005). And there was statistical significant difference of pp60c-src express amount among different tissues (P < 0.005). The positive rates of pp60c-src in the intestinal and diffuse type gastric carcinomans were 100.0% (16/16) and 68.8% (22/32), respectively, a significant difference was found statistically (P < 0.025), the difference of pp60c-src express amount was significant (P < 0.05) but no significant differences of the positive rates and express amount of pp60c-src in the tissues adjacent to carcinoma of two types of carcinoma were found (P > 0.05). The results of this experiment indicated that the activation of c-src gene and increase of pp60c-src express were associated with the proliferation, turnover, transformation and malignant change of the gastric epithelial cells, with the initiation of the gastric carcinoma, and also with the differentiation and the histological types of the gastric carcinoma.

[A new way for inbred strain mice genetic monitoring and the discovery of sex-linkaging RAPD markers].

We used 21 10 bp random primers to amplify DNA for four colonies of BALB/ c mice, four individuals of C 57 BL mice, and four individuals of Kunming stock mice. The amplified band patterns were different between BALB/c and C 57 BL mice in the products of 13 primers, and 8 primer products showed difference between BALB/c and Kunming stock mice. These results indicated that we can easily distinguish different strains of mice by RAPD method. For the four colonies of BALB/c mice, the genetic background of Chengdu colony. Shanghai colony and Beijing colony were homogeneous at all RAPD markers, however, in Kunming colony, there were two BALB/c individuals were disclosed different amplified patterns by 4 primers, showing that these two Kunming BALB/c mice maybe suffer genetic contamination or mutation some time. An unexpected phenomenon discovered in this study is that sex specific amplification bands were amplified by primer OPG 2, OPE 4, OPE 9 in all male mice. Although an 1.2 kb band were disclosed by both OPG 2 and OPE 4 in male mice, cross RAPD and not hybridization showed that the two bands were not homologous.

[A rational treatment of stage I epithelial ovarian cancer].

OBJECTIVE: To evaluate the incidence of lymph node metastasis and the role of lymphadenectomy in stage I epithelial ovarian cancer. METHODS: Forty patients with stage I epithelial ovarian cancer treated from 1985 to 1990, were divided into two groups and retrospectively analyzed. First group of 20 patients were treated by routine surgery and cis-platinum based chemotherapy. Second group of 20 patients were treated by routine surgery and cis-platinum based chemotherapy plus retroperitoneal lymphadenectomy, and on the basis of with normal ovary and uterus preserved in the younger stage I a patients. A comparison was made between the five-year survival rates of the two groups. RESULTS: Four patients in the second group were found to have retroperitoneal lymph node metastasis and should be staged as II c postoperatively. In three of the four patients aortic lymph node metastasis were diagnosed. The chances of metastasis to the pelvic and to the aortic lymph nodes were nearly equal. There is a significant difference in the 5-year survival rates between the two groups (85% vs 100%, P < 0.05). Ten patients with their ovaries and uteri preserved are living and well. CONCLUSION: It is suggested that to obtain accurate FIGO staging and to improve survival and its quality, retroperitoneal lymphadenectomy should be performed in all patients with stage I eipthelial ovarian cancer, and younger patients with stage I a cancer may preserve their gestational functions if desired.

[The detection and studies on the change of H+ concentration in the regular points of the rabbit suffering from arrhythmia induced by aconitine].

The past experiment showed that there was a specific change of H+ concentration in Neiguan (PC 6) of the rabbit suffering from arrhythmia induced by aconitine. When arrhythmia appeared, th ecocentration of H+ in Neiguan (PC 6) increased obviously. The authors found that it happened not only in the regular points but also along the course of the Heart Meridian and Pericardium Meridian in the furthe study and research. This may mean that on the course of the corresponding meridian there is a specific change of H+ concentration when the Zang-fu organs are in dysfunctional. But the mechanism and biological significance of the change of H+ concentration should remain to be further studied.

[Preparation and characterization of human monoclonal antibody to breast carcinoma].

With fusion of human heteromyeloma cells SHM-D33 and B lymphocytes obtained from axillary lymph nodes of a patient suffering from breast carcinoma, one hybridoma cell line which secretes human monoclonal antibody CM-1 to breast carcinoma was established. The hybridoma cells have been secreting human IgM (lambda light chain) stably for over 5 years. The CM-1 concentration of the supernatant of cell culture is 38.5 micrograms per ml and is the ascites that of 2 mg per ml. The reactivity of CM-1 to normal human tissues and several kinds of tumors was examined with immunohistochemical staining. The results showed that the CM-1 reacted strongly to breast carcinoma and weakly to myxoadenocarcinoma of the esophagus. CM-1 did not react to normal tissues or other kinds of tumors, such as breast fibroadenoma, medullary carcinoma of the thyroid, etc. After injection of 131I-CM-1 into abdominal cavity of the mice bearing breast carcinoma, the image of xenografted carcinoma can be obviously distinguished from the background of normal tissues. The inhibitory effect on growth of breast cancer xenograft in nude mice was 95% for CM-1-PYM conjugate and 58% for free drug. The above results showed that the hybridom cell line CM-1 stably secreting antibody reacted specificaly with breast carcinoma and it might be useful as a drug carrier in therapy of patients with breast carcinoma.

[Immunohistochemical study on morphine in human tissues from opiate associated death].

The morphine distribution in human tissues was studied by immunohistochemical method. Four cases of opiate associated death were examined. Morphine was demonstrated not only in some neuronal cytoplasma of cerebral cortex, hippocampus, basal ganglia thalamus, brainstem, and cerebellum, but also found in some nervous fibers and some capillary walls in the central nervous system. Besides, it was also found in the capsule and mesenchyma of heart, liver, spleen, lung, kidney, adrenal gland, pancreas, thymus, thyroidea and testis. Morphine was not seen in the parenchyma of these organs. It was suggested that the postmortem redistribution was not in the central nervous system, but in other organs. We considered that immunohistochemical staining of morphine is useful in the diagnosis of death from opiate addiction.

[Fast modulation of glutamate and GABA receptor--mediate electrophysiological responses by glucocorticoid].

Fast modulation by glucocorticoid on gluatmate and GABA receptor-mediated electrophysiological responses was investigated by extracellular iontophoresis and intracellular techniques in rat hypothalamic brain slices and guinea pig coeliac ganglion preparations respectively. The results were as follows: Glucocorticoid inhibited the glutamate receptor-mediated excitatory response and potentiated the GABA receptor-mediated inhibitory response within 5 min after perfusion. All these responses are reversible after removal of the glucocorticoid. Low Ca2+ high Mg2+ medium was ineffective to block the fast modulatory effects of glucocorticoid on the gluatmate and GABA receptor-mediated responses. It appears that the latter responses are mediated through some non-genomic mechanism without an involvement of synaptic circuitry.

[Application of CO2 enhanced ultrasound for two-stage operation of hepatic tumors].

The feasibility of CO2 enhanced ultrasound (CO2-EUS) was evaluated in two-stage operation patients with hepatic tumors. CO2-EUS was carried out in nine patients with indwelling catheter within hepatic artery for two-stage operation of liver cancer. CO2 microbubbles mixing by 5 ml of 5% NaHCO3 and 2.5 ml of 5% Vitamine C were injected into the indwelling catheter. The computed sonography of ACUSON 128XP/10 with a 3.5 MHz convex transducer was used for this study. The enhanced parenchyma of the liver obtained by CO2-EUS and the lasting time of enhancement was about 8 minutes. The hepatic tumors, after chemotherapeutic treatment via indwelling catheter, were variously enhanced by CO2 microbubbles. The enhanced sonogram of the tumors took the forms of hypoechoic, ring-like, or spotty enhanced pattern lasting for more than 30 minutes. The margin of the enhanced tumors was very clear in CO2-EUS. CO2-EUS detected five more lesions (size 1-3 cm) besides 9 lesions by conventional ultrasound. CO2-EUS was extremely useful in evaluating curative effects of tumor, increasing detection rates of small tumor, and improving thoroughness of two-stage operation.

[Pathological examination of 10 eyes with silicone oil injection].

OBJECTIVE: To find the histopathologic findings about 10 eyes of 10 patients with silicone oil injection. METHODS: The operative procedures included retinal detachment surgery, pars plana vitrectomy and silicone oil injection. The globes were enucleated 4 to 27 months after silicone oil injection. Paraffin sections were made for light microscopic examination. RESULTS: The retina showed severe degeneration, pre- and sub-retinal membranous fibrous tissue proliferation. The round empty vacuoles formed by silicone oil could be seen in the proliferative membrane. CONCLUSIONS: It is demonstrated that silicone oil has great toxic effect to the detached retina and it may stimulate the development of proliferative vitreoretinopathy.
