[Analysis of the EGG waveform of vocal polyps].

To study the effect of vocal polyps on vocal vibration, the authors analysed the EGG waveform of 201 cases of vocal polyps of various location and shape. The results showed that the EGG waveform in opening phase appeared a notch for vocal polyps with pedicle, and appeared steep for vocal polyps with a wide base. Among the polyps whose waveform appearing a notch in closing phase, most of their base located below vocal cord. Whether the polyps were located in the anterior, middle or posterior of the vocal cord and whether the polyps were unilateral or bilateral there are no effect on the EGG waveform. EGG was the potential technique for studying vocal function.

[Laryngeal papillomas in children].

A total of 31 cases of laryngeal papilloma in children in the 42 year period between 1952 and 1994 have been collected in this article. There are 18 males and 13 females, the medium age is 5. The shortest course was 3 days, and the average course was 10 months. Of these cases, 9 parts at the most involve pathological changes, and on average, above 3 parts. The rate of tracheotomies was approximatly 65 percent. The total cure rate and the initial cure rate were 80 percent and 67 percent respectively. As illustrated in this article, rapid development, a large affected area, a high rate of tracheotomy and cancer transformations constitute the marked clinical features of the disease. It is also noted that the disease classification should not be based on the number of operations. Furthermore, we believe it is necessary to locate tumors accurately and determine involved areas preoperatively by using modern diagnostic techniques, eliminating tumor tissue and resisting bleeding by adding laser radiation during operations, and to find and treat recurrent cases instantly through close post-operative follow-up.

[The relationship between interleukin-8 and airway hyperresponsiveness].

OBJECTIVE: To explore the relationship between interleukin-8 (IL-8) and airway hyperresponsiveness (AHR) of asthma. METHOD: IL-8 at a dose of 0.5 microgram/kg or 5 micrograms/kg was administered intranasally to guinea-pigs twice a week for 3 weeks. 24 hours after the last administration, airway responsiveness was measured as an overall index of airway response to increasing concentrations of inhaled histamine (25, 50, 100 and 200 micrograms/ml) and the numbers of different inflammatory cells in bronchoalveolar lavage fluid (BALF) was counted. RESULTS: The IL-8 treatment significantly enhanced airway responsiveness to histamine in a dose-dependent manner (P < 0.05 or 0.01) and induced a significant influx of neutrophils in BALF (P < 0.01), and there are many neutrophils within airway wall but not control animals treated with phosphate buffered saline (PBS). CONCLUSION: The IL-8 given into the airways can produce neutrophil inflammation of the airways and induce AHR, it may play an important role in asthma, especially in the development of AHR.

[Clinical analysis of huge intraocular foreign body extraction in seventeen cases].

OBJECTIVE: To discuss the definition of huge intraocular foreign body (IOFB), the selection of operation and the improvement of cure rate. METHODS: (1) The foreign body was localized by X-ray, and extracted through sclera; (2) direct localization by indirect ophthalmoscope; (3) extraction of IOFB in combination with pars plana vitrectomy (PPV); (4) extraction through anterior chamber. RESULTS: The IOFBs were extracted successfully in all cases. The foreign bodies of 13 eyes were extracted in combination with PPV (72.2%, secondary enucleation with coral porous hydroxyapatite implantation was performed on one case later), and extracted through sclera in 5 eyes. Post-operative visual acuity was improved in 6 eyes, not changed in 9 eyes and decreased in 3 eyes. CONCLUSION: IOFB extraction in combination with PPV is less damaging, accurate and safe, that reduces the complications and enhances the cure rate. It is worth to popularize.

[Reconstruction of the skin structure of the aging].

Based on the character of degeneration and change of the aging skin, reconstruction and repair of the skin structure are performed with a method of subdermal incision in order to increase the content of collagen protein and replace the degenerates collagen in the dermis. The animal experiment shows that the treated skin contracts by nearly thirty percent (P < 0.01) at 15 days after surgery. The epidermis becomes thicker and the dermis contains more fiberoblasts and collagen than that of preoperation. Seven patients were treated with the method. Afterwards their crow's feet deformity and the wrinkles of the nasolabial groove almost disappeared.

[Effects of the glucoprotein component of musk on the functions of rat polymorphonuclear leukocytes activated by fMLP in vitro].

To investigate the effects of musk-1, a glucoprotein component isolated from the water extract of musk, on the release of superoxide anion as well as beta-glucuronidase and lysozyme of rat polymorphonuclear leukocytes activated by fMLP. An in vitro incubation system with rat polymorphonuclear leukocytes was used. Superoxide anion production was determined by cytochrome C reduction. beta-glucuronidase and lysozyme release was quantitated by enzyme reactions in which phenolphthaleinglucuronic acid and micrococcus lysodeikticus were as the substrates, respectively. In comparison with control, musk-1 at final concentrations of 1-100 micrograms/ml can increase superoxide anion production by 23.0%-83.6% and decrease beta-glucuronidase and lysozyme release by 7%-47% and 9%-22%, respectively, in rat polymorphonuclear leukocytes. It is concluded that Musk-1 can significantly affect the functions of rat polymorphonuclear leukocytes. Therefore, inhibition of lysosomal enzyme release might be considered as one of the mechanisms of anti-inflammatory role of musk.

[Partial deletion of mitochondrial DNA in mitochondrial encephalomyopathies].

OBJECTIVE: To characterize the deletions of mitochondrial DNA (mtDNA) in Chinese patients with Kearns-Sayre syndrome (KSS) and chronic progressive external ophthalmolegia (CPEO) and identify deletion mutations of mtDNA be the etiology of these diseases. METHODS: Patients and preparation of total DNA: Two patients with KSS and two with CPEO and ten with other mitochondrial myopathies or encephalomyopathies were determined by histological and biochemical assays. Total DNA was isolated from 100 mg to 150 mg of frozen muscle obtained by biopsy using the methods described by Zeviani et al. (1988). Preparation of mtDNA probes: MtDNA were isolated and purified according to Palva's method (1985). The mtDNA was linearized by digestion with the restriction endonuclease Pvu II and separated by electrophoresis through low melting agarose gel. The 16.5 kb DNA band was recovered from the gel and labeled with alpha-p32 dCTP by random primer labeling. Southern blot analysis: Portions of five micrograms total DNA obtained from the patients and controls were digested with Pvu II, EcoR I, Hind III And Xba I respectively. The digested DNA was separated by agarose gel electrophoresis and transferred to nitrocellulose membrane and then hybridized with P32 labeled mtDNA as previously described (Zhang, 1991). RESULTS: A large-scale deletions of mtDNA were identified in two patients with KSS and two patients with CEPO. The deletions ranged in size from 2.4 kb to 5.5 kb. The proportion of mutated mtDNA in each patients ranged from 54.6% to 84.6% of total mtDNA. No detectable deletions were found in mtDNA of ten patients with other mitochondrial myopathies or encepholomyopathies and normal controls. CONCLUSIONS: Deletions of mtDNA were found in all KSS and CEPO patients detected. These results supported The view point of the deletions are important causes of physical defect in KSS and CEPO.

[Use of appendix to restore a ureter: report of 2 cases].

In two cases the appendix was utilised as a substitute for part of the left ureter. Case one used the appendix as a conduit from the upper left ureter to the right one to treat the megaureter, and the other one used the appendix as a connection to join the two transected openings of the left ureter to treat the obstruction caused by the chronic ureteritis of the left ureter. Long-term observation after the operation showed satisfactory functions of kidneys. We suggested that the appendix is an autologus tissue whose active peristalsis, little mucous absorption along with its easy performance make this method highly valuable and practical.

[Immunohistochemical study of hepatitis C virus core antigen and HBxAg in liver cirrhosis and hepatocellular carcinoma tissues].

OBJECTIVE: To study the distribution and significance of hepatitis C virus core antigen in liver cirrhosis and hepatocellular carcinoma tissues. METHODS: Hepatitis C virus antigen and HBx-Ag were detected in liver cirrhosis (LC) and hepatocellular carcinoma (HCC) tissues with immunohistochemical methods. RESULTS: In some cases, not only was HCV core antigen positive stain distributed in the liver cells and nuclei of the cancer cell but it was also found in the cytoplasm. In different cases, it may be predominantly cytoplasm positive or nuclear positive or both. In liver cancer tissues, the HCV core antigen cytoplasmic positive cells were focally distributed and whereas the nuclear positive cells were diffusely distributed. The HCV core antigen nuclei positive cells were often observed in HCC tissues, but the cytoplasmic positive cells were often observed in the pericancerous liver tissue. The detection rates of HCV core antigen in LC, HCC and pericancerous liver tissues were 67.3% (66/98), 75.0% (78/104) and 48.1% (25/52), respectively. Statistical analysis suggested that: HCV core antigen nuclei positive rate in HCC be much higher than that in LC and pericancerous liver tissues (P < 0.01) and HCV core antigen nuclei positive rate be much higher than the cytoplasm positive rate in HCC tissues (P < 0.01). CONCLUSION: HCV, of which the infection being very common in LC and HCC of our country, may play an important role in the development of LC and HCC except HBV infection.

[Apoptosis-antagonizing protein Bcl-2 expression, distribution in non-Hodgkin's lymphoma and its clinical significance].

OBJECTIVE: Apoptosis-antagonizing protein Bcl-2 expression, distribution in non-Hodgkin's lymphoma (NHL) and its clinical significance were studied. METHODS: Four-stage peroxidase antiperoxidase complex (PAP) method was applied to comparatively investigate the expression and distribution of Bcl-2 protein in formalin-fixed, paraffin-embedded lymph node tissue comprising 12 cases of reactive follicular hyperplasia and 71 cases of NHL. RESULTS: (1) Bcl-2 protein was detected in 93.0% cases of NHL with T- or B-lymphocytic phenotype. (2) Bcl-2 protein was restricted in centrocytes but not in centroblasts in 8 of 9 cases of NHL stemed from germinal centers. (3) A significant increase in Bcl-2 protein expression was found in 5 of 10 cases of diffuse large B-lymphocytic NHL showing plasmacytoid differentiation. (4) In 7 cases of follicular NHL the neoplastic nodules were strongly labeled for Bcl-2 protein and only scattered positive cells were seen in its periphery; On the contrary, in reactive lymphoid follicles Bcl-2 protein was predominantly anchored in mantle zones whereas germinal center cells were unstained. CONCLUSION: (1) Bcl-2 protein is expressed in NHL of T- or B-cell origin. (2) Bcl-2 protein expression appears to be associated with differentiation stages of NHL. (3) Immunohistochemical staining for Bcl-2 protein conduces to differentiate between reactive and neoplastic follicles.

[The ionic mechanism of hyperpolarization induced by glucocorticoid and its modulatory effects in mammalian neurons].

This is the first report to investigate the rapid hyperpolarization by glucocorticoid in coeliac ganglion neurons of guinea pig in vitro. Hydrocortisone 21-hemisuccinate (F-suc) caused rapidly hyperpolarization in neurons; hyperpolarization persisted after the elimination of synaptic inputs and after suppression of protein synthesis with actinomycin D; F-suc also induced hyperpolarization when picrotoxin or Ca(2+)-free in perfusion solution; TEA and 4-AP can inhibit the hyperpolarization of F-suc. These results provided evidence that F-suc's hyperpolarization may be mediated by potassium channel in the membrane. Also F-suc suppressed the discharges of neurons; and modulated rapidly effects of neurons to excitatory or inhibitory amino acids (P < 0.05).

[Establishment of an apoptotic model induced by adriamycin in human hepatocellular carcinoma].

OBJECTIVE: Establishment of cell apoptotic model of human hepatocellular carcinoma. METHODS: Human cultured hepatocellular carcinoma cell line HCC-9204 was exposed in vitro to different concentrations of adriamycin for various lengths of time. Cell apoptosis was ascertained by cell morphology under transmission electron-microscope, DNA electrophoresis and flow cytometry. RESULTS: When HCC-9204 cells were treated with adriamycin at 20 mumol/L for 3 hr, cell apoptosis occurred. Typical apoptotic bodies appeared and DNA ladder could be demonstrated on DNA electrophoresis. Apoptotic peak was also shown by flow cytometry. Apoptotic cells accounted for 72.1% of the cell population. Cells in G1, S and G2 phase of cell cycle were 75.9%, 24.1% and 0%, respectively. Only 35.5% of adriamycin treated cells remained alive. CONCLUSION: The apoptosis of human cultured hepatocellular carcinoma cells, HCC-9204 cell line, was successfully induced by adriamycin. Establishment of this model will help further explore the molecular mechanism of hepatoma cell apoptosis.

[Extraction and sequencing of trace DNA from cornu Cervi pantotrichum].

The trace DNA extraction method was used to extract DNA from the blood, hair, Cornu cervi pantotrichum, hairy antler of the sika deer, Cervus nippon, and penis of Bulbalus bublis and Asinus vulgaris. A 307 bp fragment of mitochondrial DNA cytochrome b gene was amplified with primers L14841 and H15149 for these extractions. The purified PCR products were sequenced by the dideoxy method. DNA sequence obtained from the Cornu cervi pantotrichum is the same as that obtained from the fresh blood and hair of the sika deer, but the sequence of the so-called hairy antler is quite different, implying that it is not from the sika deer. In addition, the phylogenetic relationships among different species are congruent with the traditional known phylogeny.
