Value of 8S/4S fractionation of estrogen receptors (ER) for prediction of response to hormonal manipulation in metastatic breast cancer.

Using sucrose gradient centrifugation, human breast cancer estrogen receptors appear in two molecular forms sedimenting at either 8S or 4S fractions. The sum of these two fractions has been valuable clinically in helping to predict response to hormonal therapies. It has been suggested that the 4S receptor may not be of predictive value and that the practice of using only dextran-coated charcoal methodology might thus overestimate the number of patients whose tumors might be hormonally dependent. The present study correlates clinical response to hormonal manipulations with 8S and 4S receptors as determined by sucrose density gradient centrifugation. Patients with only 4S positive receptors had a 56% response rate to hormonal manipulation compared with a 52% response rate in patients with positive 8S alone or with both 8S and 4S receptors. Although patient numbers are small in this and the other three published series addressing this question, our data do not confirm two previously published studies suggesting clinical importance of 8S/4S fractionation.

Interaction of sporozoites of Theileria parva with bovine lymphocytes in vitro. I. Early events after invasion.

Sporozoites of Theileria parva rapidly enter bovine lymphocytes by a mechanism of passive endocytosis that depends upon progressive circumferential binding of ligands on the parasite to receptors on the host-cell membrane. Within 10 min of entry, the micronemes of the sporozoite discharge their content and the enveloping host-cell membrane is lysed. The host cell responds within 30 min of invasion by polymerization of microtubules arrayed tangential to the sporozoite and converging upon the cytocentrum. Multivesicular bodies and lysosomes are generated and gather around the parasite but are ineffective in the absence of an endocytotic membrane with which they can fuse. Thus Theileria parva can be added to the category of obligate intracellular parasites that ensure their survival by lysis of the parasitophorous vacuole.

Plasmid-borne streptothricin resistance in gram-negative bacteria.

Plasmids, which code for a new type of drug resistance due to the acetylation of streptothricin and belonging to various incompatibility groups (I1, I2, W3, FII, S, X, and N) were identified in gram-negative bacteria. The gene for the acetyltransferase is closely linked to a gene for the streptomycin/spectinomycin adenylyltransferase AAD-3" on a novel type of a transposon, designated Tn1825. This is related to Tn7.

delta 5-3 beta-Hydroxysteroid dehydrogenase delta 4-5-isomerase activity and metabolism of dehydroepiandrosterone in rat preputial gland.

After intracardial injection of [1,2-3H]dehydroepiandrosterone ([3H]DHA) into female rats, [3H]DHA was found to accumulate and was metabolized in the preputial gland, but not in the diaphragm. The identified metabolites of [3H]DHA in the preputial gland were delta 4-androstenedione-3 alpha, 17 beta-diol. Cells were isolated from the preputial gland after treatment with trypsin and collagenase III, and centrifugation in Ficoll gradients. Activity of the enzyme complex delta 5-3 beta-hydroxysteroid dehydrogenase delta 4-5-isomerase (3 beta-HSD) responsible for transforming DHA into delta 4-androstenedione was found mainly in the 105,000 g pellet (microsomal fraction) of homogenates of the isolated cells. It used preferentially NAD over NADP as a coenzyme, with a pH optimum at 8.5. The apparent Km for DHA was 5.5 X 10(-5) M, and the Vmax was 1.72 nmol/min/mg microsomal protein. These findings indicate that DHA is preferentially taken up by the preputial gland where it undergoes metabolism to form more potent androgens, and suggest that DHA may have important androgenic influence on the preputial gland.

[Inflammatory brain diseases in clinical practice].

Over a period of twenty years all cases of inflammatory diseases treated in a neurological intensive-care unit were analysed in retrospective. For 90 cases of purulent bacterial meningitis, letality amounted to 30%. The need for agent-oriented, intensive early treatment is discussed. At a reduced incidence of tuberculosis, meningitis tuberculosa is generally neglected. In the majority of cases patients suffering from "encephalitic syndrome" make high demands on intensive-therapeutic concepts, including interdisciplinary cooperation, due to high complication-rates and lack of causal therapy. In 95 cases, letality also amounted to 30%.

[Morbidity rate in delirium tremens].

On evaluating an analysis of 105 cases of delirium tremens over a period of five years in a psychiatric base-unit, the following findings were obtained: Delirium tremens, particularly in conjunction with secondary diseases, constitutes a life-endangering medical emergency and requires intensive medical treatment. Letality was 8,57%. The morbidity-rate and the relapse-rate showed a clearly marked increase; the age of contracting the disease had its frequency peak between 30 and forty, i. e. earlier than in many other statistical surveys. Epileptic attacks as symptoms starting off or accompanying delirium occurred in almost one third of our delirium patients. The impact which these attacks have on the development in the course of process-like alcoholism was discussed.

Transformation of dehydroepiandrosterone into dihydrotestosterone by isolated cells from rat preputial gland.

After the rat preputial gland was treated with collagenase and trypsin, five bands of cells were isolated by centrifugation in Ficoll gradients. Homogenates of the heavier cells (Bands IV and V) which contained less lipids, were more active than the homogenates of the lighter cells (Bands I, II and III) in transforming [1,2-3H]-dehydroepiandrosterone ([1,2-3H]-DHA) into [3H]-androstenedione and [3H]-testosterone and the latter into [3H]-dihydrotestosterone (DHT). In the presence of NAD, NADH and NADPH-generating system, [1,2-3H]-DHA was transformed into [3H]-DHT in 50-60% yield by homogenates of cells in Bands IV and V. DHT levels in the preputial gland were measured by radioimmunoassay. The levels in female rats reduced by 77% from 3.14 +/- 0.27 to 0.72 +/- 0.10 pg/mg tissue after adrenalectomy, and by 45% to 1.71 +/- 0.10 pg/mg tissue after ovariectomy. In male rats, the level reduced by 15% from 4.58 +/- 0.55 to 3.88 +/- 0.62 pg/mg tissue after adrenalectomy and by 40% to 2.74 +/- 0.21 pg/mg tissue after orchidectomy. These results demonstrated the transformation of DHA into DHT in the preputial gland of the rat, and that the adrenal is an important source of precursor steroid (DHA) for DHT formation in the preputial gland.

Immunochemical demonstration of human chorionic gonadotrophin (hCG) in tissue of breast carcinoma.

Immunoreactive human chorionic gonadotrophin (hCG) was found in 9 of 65 surgically removed malignant breast tumours. The concentrations found ranged from 5 to greater than 500 mIU hCG/g tumour. Of the nine, seven extracts were assayed for total protein content; when expressed as mIU/mg extract protein, the hCG concentrations ranged from 0.28 - > 18.2 mIU/mg protein. Comparison of oestrogen receptor and hCG presence in the tumours showed no strong correlation. All tumours in which hCG was detected were shown to be of ductal origin. The hCG was measured by a beta-chain specific radioimmunoassay which measures only hCG even in the presence of high levels of other gonadotrophin hormones. In further study of these specimens an immunoperoxidase staining technique was used to stain for hCG in formalin-fixed sections of the same tumours assayed by RIA. This technique confirmed hCG in the positive specimens examined (tissue was available for 7 of the 8 positives) and no significant staining in any negatives. The hCG was shown to be localized within the cytoplasm and on the surface of the malignant cells but not in surrounding normal cells. This intracellular localization demonstrates that the malignant cells may produce hCG, rather than stimulating the pituitary to do so.

Tissue immunoreactive prolactin hormone in breast cancer.

The prolactin contents of the cytosol extracts of 159 human mammary carcinomas was measured by specific radioimmunoassay. Of these extracts 157 contained measurable prolactin (detection limit, 7.8 ng/g tumor). The levels measured were in most cases greater than would be expected to be due to only serum contamination. The levels of prolactin did not correlate with either estrogen or progesterone receptor content of the tumors, or menopausal status of the patient.

Immunochemical demonstration of immunoreactive insulin in human breast cancer.

The immunoreactive insulin contents of the cytosol extracts of 57 human mammary carcinomas were measured by specific radioimmunoassay. All of these extracts contained measurable insulin (detection limit 1.3 ng/g tumor). In some cases (23) the levels were quite greater than the normal human serum levels. Immunoperoxidase technique revealed the presence of immunoreactive insulin within the cytoplasm of tumor cells in 6 cases.

[Clinical aspects and therapy of Landry's paralysis].

On the basis of an analysis of 56 acute cases of polyneuritis of the Landry type treated in a period of 15 years at a neuropsychiatric intensive care unit, an attempt is made to establish relations between spinal fluid findings, kind of the course of the disease, rate of development of paralyses and the prognosis. According to the results of the examinations, the spinal-fluid findings cannot be attributed a prognostic significance. It appears, however, remarkable that the group of patients with a high rate of spreading of the pareses showed both the highest mortality and full remission rates. On the basis of own experiences and compilation of intensive medical literature, the most frequent complications of the disease and its therapy problems are dealt with.
