IroN, a novel outer membrane siderophore receptor characteristic of Salmonella enterica.

Speciation in enterobacteria involved horizontal gene transfer. Therefore, analysis of genes acquired by horizontal transfer that are present in one species but not its close relatives is expected to give insights into how new bacterial species were formed. In this study we characterize iroN, a gene located downstream of the iroBC operon in the iroA locus of Salmonella enterica serotype Typhi. Like iroBC, the iroN gene is present in all phylogenetic lineages of S. enterica but is absent from closely related species such as Salmonella bongori or Escherichia coli. Comparison of the deduced amino acid sequence of iroN with other proteins suggested that this gene encodes an outer membrane siderophore receptor protein. Mutational analysis in S. enterica and expression in E. coli identified a 78-kDa outer membrane protein as the iroN gene product. When introduced into an E. coli fepA cir fiu aroB mutant on a cosmid, iroN mediated utilization of structurally related catecholate siderophores, including N-(2,3-dihydroxybenzoyl)-L-serine, myxochelin A, benzaldehyde-2,3-dihydroxybenzhydrazone, 2-N,6-N-bis(2,3-dihydroxybenzoyl)-L-lysine, 2-N,6-N-bis(2,3-dihydroxybenzoyl)-L-lysine amide, and enterochelin. These results suggest that the iroA locus functions in iron acquisition in S. enterica.

Clinical spectrum of HTLV-I in south Florida.

A total of 113 patients with infection due to human T-cell leukemia virus type 1 (HTLV-I) were evaluated at the University of Miami from January 1988 to March 1993. Forty patients were identified with adult T-cell leukemia/lymphoma (ATLL) and 63 with HTLV-I-associated myelopathy (HAM). Three had concomitant ATLL and HAM. Two HAM patients co-infected with human immunodeficiency virus type 1 (HIV-I) developed clonal lymphoproliferative disease during the study period. Patients with ATLL have a poor prognosis; multiple chemotherapy regimens including high-dose cytotoxic agents have been utilized with a small impact on survival. Most of our patients are currently treated with experimental regimens. Rheumatologic or autoimmune illnesses were identified, mostly in HAM patients, and a small number developed immunodeficiencies in the absence of other definable etiologic factors. Most of the patients were immigrants from areas of endemicity in the Caribbean basin, although many Americans were also recognized. HTLV-I/II infection was diagnosed serologically and typed as HTLV-I by polymerase chain reaction (PCR) or a modified Western blot when a DNA sample was not available. In 24 of 40 patients with ATLL, Southern blot hybridization performed on DNA extracted from peripheral blood lymphocytes or tumor tissue demonstrated clonal HTLV-I integration. In South Florida, ATLL and HAM are now seen frequently. Since HTLV-I infection is associated with a 4% lifetime risk of developing ATLL and an additional 0.25% lifetime risk for developing HAM, a large pool of asymptomatically infected individuals must exist here.(ABSTRACT TRUNCATED AT 250 WORDS)

[Venous digital subtraction arteriography of the pelvic and leg area with step displacement].

By combining a 40 cm image amplifier, a DSA equipment, and the technique of step displacement, it became possible to demonstrate transvenously the arterial flow, beginning with the descent of the tibia, in four stages, using only two series of examinations. Venous digital subtraction arteriography of the pelvis/leg region was performed in 140 patients. The results were compared partly with arterial digital and analogue arteriographies as well as with surgical findings.

A totally automated system for enzyme immunoassay of theophylline in serum.

We describe a procedure for enzyme immunoassay of theophylline (1,3-dimethylxanthine) in which all phases of the assay are totally automated in a Kinetic Analyzer (KA-150). This system permits assay of 75 10-mul samples per hour, with results available at 30-s intervals after initial sample preparation and preincubation. We compared results for 138 clinical samples by an ultraviolet method (x) and the present method (y). The slope of the comparison curve was 0.902, the y-intercept 0.402, and the correlation coefficient 0.984. The coefficient of variation for samples run in duplicate on the same day was 4.9%; it was 8.1% for samples run on different days. Specificity, sensitivity, simplicity, speed, and small reagent requirement all make this an attractive alternative to chromatographic procedures.
