Chlorine-susceptible and chlorine-resistant type 021N bacteria occurring in bulking activated sludges.

Two filamentous bacteria causing bulking in two activated sludges were examined. Investigations using morphological features, staining techniques, and fluorescent in situ hybridization identified both filaments as type 021N. However, an examination of the effect of chlorine on the sludges revealed a chlorine-susceptible type 021N in one sludge and a chlorine-resistant type 021N in the other.

Genetic diversity among 3-chloroaniline- and aniline-degrading strains of the Comamonadaceae.

We examined the diversity of the plasmids and of the gene tdnQ, involved in the oxidative deamination of aniline, in five bacterial strains that are able to metabolize both aniline and 3-chloroaniline (3-CA). Three strains have been described and identified previously, i.e., Comamonas testosteroni I2 and Delftia acidovorans CA28 and BN3.1. Strains LME1 and B8c were isolated in this study from linuron-treated soil and from a wastewater treatment plant, respectively, and were both identified as D. acidovorans. Both Delftia and Comamonas belong to the family Comamonadaceae. All five strains possess a large plasmid of ca. 100 kb, but the plasmids from only four strains could be transferred to a recipient strain by selection on aniline or 3-CA as a sole source of carbon and/or nitrogen. Plasmid transfer experiments and Southern hybridization revealed that the plasmid of strain I2 was responsible for total aniline but not 3-CA degradation, while the plasmids of strains LME1 and B8c were responsible only for the oxidative deamination of aniline. Several transconjugant clones that had received the plasmid from strain CA28 showed different degradative capacities: all transconjugants could use aniline as a nitrogen source, while only some of the transconjugants could deaminate 3-CA. For all four plasmids, the IS1071 insertion sequence of Tn5271 was found to be located on a 1.4-kb restriction fragment, which also hybridized with the tdnQ probe. This result suggests the involvement of this insertion sequence element in the dissemination of aniline degradation genes in the environment. By use of specific primers for the tdnQ gene from Pseudomonas putida UCC22, the diversity of the PCR-amplified fragments in the five strains was examined by denaturing gradient gel electrophoresis (DGGE). With DGGE, three different clusters of the tdnQ fragment could be distinguished. Sequencing data showed that the tdnQ sequences of I2, LME1, B8c, and CA28 were very closely related, while the tdnQ sequences of BN3.1 and P. putida UCC22 were only about 83% identical to the other sequences. Northern hybridization revealed that the tdnQ gene is transcribed only in the presence of aniline and not when only 3-CA is present.

Optimization of a yeast estrogen screen and its applicability to study the release of estrogenic isoflavones from a soygerm powder.

Here we describe a redesigned protocol of the yeast estrogen screen developed by Routledge and Sumpter. The redesigned test comprises two steps. First, a large amount of yeast with estrogenic compounds is incubated for 24 hr. Subsequently, a mixture of cycloheximide and the chromogenic substrate chlorophenol red-beta-d-galactopyranoside (CPRG) is added. The cycloheximide stops protein synthesis and allows for an end-point measurement of beta-galactosidase activity generated during the first 24 hr. CPRG is converted to chlorophenol red and reflects beta-galactosidase activity, which is indicative of the estrogenic activity. The modifications shorten the duration of the assay at least 1 day and avoid interference of the estrogenic CPRG or chlorophenol red. The redesigned and the original protocol were used to study the estrogenic activity of bisphenol A, methoxychlor, p,p'-DDT, and isoflavones (genistein, daidzein, and glycitein). Bisphenol A, methoxychlor, and genistein triggered higher levels of beta-galactosidase activity in the redesigned protocol. Estrogenic activity of p,p'-DDT could only be demonstrated with the redesigned protocol. Glycitein and daidzein failed to give a response with both protocols. We also studied deconjugation of beta-glycosidic isoflavones present in soygerm powder. Treatment of the soygerm powder with beta-glycosidase released isoflavones. The estrogenic response of the samples was confirmed with the redesigned protocol and correlated with the amount of genistein present. The release of isoflavones under conditions prevailing in the intestines was studied. Bacterial beta-glycosidase present in the large intestine released isoflavones, and moderate estrogenic activity could be demonstrated.

New perspectives in anaerobic digestion.

The IWA specialised group on anaerobic digestion (AD) is one of the oldest working groups of the former IAWQ organisation. Despite the fact that anaerobic technology dates back more than 100 years, the technology is still under development, adapting novel treatment systems to the modern requirements. In fact, most advances were achieved during the last three decades, when high-rate reactor systems were developed and a profound insight was obtained in the microbiology of the anaerobic communities. This insight led to a better understanding of anaerobic treatment and, subsequently, to a broader application potential. The present "state-of-the-art" paper, which has been written by members of the AD management committee, reflects the latest achievements and sets future lines for further development.

HACCP (Hazard Analysis and Critical Control Points) to guarantee safe water reuse and drinking water production--a case study.

To obtain a sustainable water catchment in the dune area of the Flemish west coast, the integration of treated domestic wastewater in the existing potable water production process is planned. The hygienic hazards associated with the introduction of treated domestic wastewater into the water cycle are well recognised. Therefore, the concept of HACCP (Hazard Analysis and Critical Control Points) was used to guarantee hygienically safe drinking water production. Taking into account the literature data on the removal efficiencies of the proposed advanced treatment steps with regard to enteric viruses and protozoa and after setting high quality limits based on the recent progress in quantitative risk assessment, the critical control points (CCPs) and points of attention (POAs) were identified. Based on the HACCP analysis a specific monitoring strategy was developed which focused on the control of these CCPs and POAs.

Probiotic removal of herbicides with endocrine disrupting potential from aqueous matrices.

Water dosed with 50 mg/L of 2,4-D and atrazine was treated to remove the herbicides which both are reported to have endocrine disrupting potential. Both chemicals could be removed effectively with activated carbon. Yet, traces of endocrine disrupter remained in the water and are able to enter the food chain. The further biological elimination of these compounds was investigated. Milk alone did not decrease the amount of chemicals present in the aqueous supernatant. Yet, the probiotic products Bifidus yoghurt (GB), Actimel (Danone) and Yakult (Yakult Honsha) appeared to bind a substantial amount of the endocrine disrupting chemicals to the particulate fraction and thus render them potentially less bio-available in the aqueous phase.

Efficient control of activated sludge filamentous bulking by means of a multi-component additive.

A multi-component additive formulated for a more efficient control of activated sludge filamentous bulking was evaluated at a full-scale treatment plant experiencing a severe filamentous bulking. It was found that, besides offering an immediate improvement of sludge settling, the multi-component additive was able to eliminate the filamentous bacteria causing the bulking. Hence, contrarily to ordinary additives, this novel additive yielded immediate as well as long-term improvement of sludge sedimentation upon a few additions.

Use of a nitrifying culture to shorten the activation time of biofilters for the removal of ammonium and nitrite in freshwater aquaria.

The removal of ammonia (NH3) through nitrification in intensive aquaculture systems is an important process as the total ammonia nitrogen (TAN, compromising NH4+ and NH3) concentration often is the key limiting water quality parameter in these intensive aquaculture systems. In this study, the performance characteristics of a suspension of nitrifying cells (named ABIL) have been explored This aqueous suspension contains a highly active, nitrifying microbial consortium and is stable for several months when preserved at 4 degrees C. Tests were performed in freshwater at lab scale (70 L, 20 - 24 degrees C). Results showed that the application of the consortium at a dose of 5 mg Volatile Suspended Solids (VSS) L(-1) assures a total removal of ammonium and nitrite species from 10 mg N L(-1) to below the detection limit within a period of four days. Experimentally, at a substrate level of 10 mg TAN L(-1), a rate of biological ammonium and nitrite conversion of the order of 0.3 - 0.5 g TAN g(-1) VSS(-1) d(-1) could be achieved by the consortium in the freshwater aquaria systems tested Provided adequate aeration and dissolved oxygen levels of 6 mg per L or more, no important intermediary nitrite concentrations were noticed Only a small amount of TAN was not found back as nitrate and might have been lost due to ammonia stripping After 12 months preservation of the inoculum at 4 degrees C, no important decrease in ammonium removal activity and only a minor decrease in the nitrite removal rate of the consortium were noticed.

Assessment of the fouling potential of wastewater treatment plant effluent using a modified capillary suction time measurement.

A new technique was developed to measure membrane fouling, which is derived from the capillary suction time (CST)--a parameter often used to indicate the dewaterability of sludge--and which is called the modified CST (CSTm). The results indicate a good repeatability and reproducibility of the test. Spiking tests with suspended solids and humic acid, which have a high fouling potential, resulted in a significant rise of the CSTm value, indicating a sensitive assessment of these fouling components. However, the CSTm measurement is mainly indicative for the colloidal fouling of the sample.

First steps in microbiological calcium removal from calcium-rich industrial wastewater.

A novel application for the process of microbial carbonate precipitation (MCP) was developed. It facilitates the removal of soluble calcium from calcium-rich industrial wastewater. It was shown that via the urea hydrolysis pathway, mediated by autochthonous bacteria, calcium removal exceeding 90% could be maintained in a semi-continuous reactor system.
