Does whole-body hyperthermia therapy involve participation of the immune system?

The effect of micro-wave whole-body hyperthermia on tumor growth and lung metastasis counts was examined in adult BALB/c mice. The effective therapy with hyperthermia prolonging mean survival time of the animals and causing tumor regression resulted in concimitant depression of anti-tumor immunity measured in vivo. The beneficial effect of preimmunization of animals with killed tumor cells on subsequent challenge with live tumor cells was completely abrogated by hyperthermia treatment. It is postulated that this form of therapy is directly related only to heat sensitivity of tumor cells and that there is no profitable participation of the immune system in the anti-tumor effect of wholebody hyperthermia.

The search for an influence of whole-body microwave hyperthermia on anti-tumor immunity.

The effect of microwave whole-body hyperthermia was examined on the takes of tumor after i.v. administration of the tumor single cell suspension. It was shown that the longer the exposure to hyperthermia treatment the higher the number of lung nodules obtained by injection of the same dose of tumor cells. Also, the expression of contact hypersenstivity to oxazolone was strongly inhibited after hyperthermia treatment. It was documented that it is possible to transfer the diminished resistance to tumor with bone marrow, whereas impaired responsiveness to oxazolone was transferred with spleen cells or thymocytes.

Effect of whole-body microwave hyperthermia on delayed cutaneous hypersensitivity in tumor-bearing mice.

Tumor-bearing and normal BALB/c mice were examined for their responsiveness to contact-sensitizing agent oxazolone after whole-body microwave hyperthermia treatment. The effective therapy with hyperthermia prolonging mean survival time of the animals and causing tumors regression resulted in strongly pronounced hyporeactivity to oxazolone. The impaired responsiveness remained unchanged until death of the tumor-bearing animals. In the case of tumor-free animals delayed cutaneous response returned to normal values within 8-12 days after hyperthermia treatment. It is documented that the longer exposure to hyperthermia sessions the more pronounced depression of reactivity to oxazolone occurs.

The in vivo cytostatic effect induced by Propionibacterium granulosum on murine tumor cells.

The cytostatic action of Propionibacterium granulosum was studied in a mouse sarcoma in vivo. Kinetic analysis of tumor cells 28 days after tumor implantation and systemic immunotherapy showed that the cell cycle time was identical in both treated and untreated tumors. P. granulosum treatment resulted in a marked prolongation of the S phase and shortening of the G1 phase of the cell cycle. A pronounced drop in the number of labeled interphases and the reduction of the growth fraction were observed in tumors obtained from mice given injection of P. granulosum. Cloning efficiency of tumor cells from P. granulosum treated animals was quantitatively similar to that of control animals and only differences in the size of lung colonies were observed.

The correlation of susceptibility of different Propionibacterium strains to macrophage killing and antitumor activity.

The stimulation of the reticuloendothelial system and antitumor activity against sarcoma-L-1 of three different strains of Propionibacterium was investigated. The bacteria injected intraperitoneally with a single dose of 1 mg per mouse appeared to possess different effectiveness in both assays used. The same strains were examined for their susceptibility to phagocytosis and intracellular killing by murine peritoneal macrophages. The results obtained showed correlations between Propionibacterium resistance to degradation by phagocytic cells and their antitumor and stimulatory activities. It is concluded that the potency of antitumor and stimulatory activities of different strains of Propionibacterium are closely related to their cell wall structure.

Effect of hyperthermia on rabbit macrophages.

The effect of water-bath hyperthermia on rabbit peritoneal macrophages was studied in vitro. The cells were exposed to hyperthermia for 30 min to 4 hours and membrane transport of ions as measured by total and ouabain-inhibited 86Rb influx as well as membrane permeability for 86Rb and 51Cr-labelled intracellular proteins were investigated. Heat-treated macrophages were tested for their ability to phagocyte staphylococci and for reduction of nitroblue-tetrazolium. Moreover the effect of microwave whole-body hyperthermia on rabbit phogocytic cells was studied in vivo. Ion transport to macrophages was stimulated by both intensive (43 degrees C) and moderate (40 degrees C) hyperthermia. On the other hand exposition of the cells to 43 degrees C led to pronounced release of 86Rb and 51Cr from prelabelled cells. NBT reduction was generally decreased in macrophages exposed to 43 degrees C and increased in macrophages kept at 40 degrees C. Clearance of 32P-labelled staphylococci from peripheral blood of microwave-irradiated rabbits diminished when animals were exposed to microwave hyperthermia for f or 7 days (2 hours daily).

Action of three species of Propionibacteria (P. granulosum, P. acnes, P. avidum) on experimental tumor systems in mice.

The antitumor activity of three different species of Propionibacteria (P. granulosum, P. avidum and P. acnes) against murine Sarcoma 180 and Moloney-Sarcoma-Virus-induced tumor (MSV tumor) was investigated. All three Propionibacteria injected intraperitoneally or intratumorally in multiple doses of 1 mg per mouse appeared to be effective regarding the retardation of growth and the stimulation of regression of Sarcoma 180 in CFW mice. Moreover, the application of Propionibacteria significantly prolonged the survival of Sarcoma-180-bearing mice. No significant differences were found in antitumor effect against Sarcoma 180 between P. granulosum, P. Avidum and P. acnes, but in general, intratumoral injections of all three strains were more effective than intraperitoneal applications. All three strains of Propionibacteria were equally ineffective regarding the retardation of growth and the stimulation of regression of MSV tumors in adult NMRI mice, as assessed after intraperitoneal injections of P. granulosum, P. avidum or P. acnes (1 mg per mouse) 3 days prior to inoculation of virus particles.

Effect of three strains of propionibacteria (P. granulosum, P. avidum, P. acnes) and cell-wall preparations on lymphocytes and macrophages.

The effect of the strains of Propionibacteria (P. granulosum, P. avidum, P. acnes) on functional and metabolic activities of mouse lymphocytes and rabbit macrophages was examined in vivo and in vitro. In some of the experiments bacterial cell walls isolated from the above bacteria were tested and compared with the activity of whole bacteria. In isolated lymph-node lymphocytes influx of 86Rb and its inhibition by ouabaine, release of 86Rb and 51Cr from prelabelled cells, as well as spontaneous and PHA-stimulated incorporation of 3H-uridine were tested after treatment with 10 or 50 microgram/ml of Propionibacteria (whole cells or cell walls). Phagocytosis of 32P-labelled Staphylococci and reduction of Nitro-BT were examined in isolated rabbit peritoneal macrophages treated with the above concentrations of Propionibacteria. Clearance of 32P-labelled Staphylococci from peripheral blood in rabbits treated with 5 mg per kg body weight of Propionibacteria was also studied. All three strains of Propionibacteriae stimulated 86Rb influx into lymphocytes, the effect being observed only in concentrations leading to parallel increase of 86Rb and 51Cr release from prelabelled cells. No significant differences in activity between the three tested strains of bacteriae were found. Propionibacteria of a dosage of 50 microgram/ml significantly increased spontaneous and PHA-stimulated incorporation of 3H-uridine into lymphocytes. All three strains of Propionibacteria markedly stimulated both phagocytosis and intracellular digestion (as measured by the ability to reduce Nitro-BT) in isolated peritoneal macrophages. This was accompanied by faster clearance of Staphylococci from peripheral blood of rabbits treated with 5 mg per kg body weight of Propionibacteriae. The obtained results showed direct stimulation of lymphocytic and macrophagic functional activity by whole cells and isolated cell walls of all three tested strains of Propionibacteria.

Recovery of hemopoiesis in lethally irradiated mice after treatment with propionibacteria.

Three investigated strains of propionibacteria (P. acnes, P. granulosum and P. avidum) injected intraperitoneally in doses of 1.5 mg per mouse, significantly prolonged survival of lethally (8.50 R) irradiated mice. P. granulosum, being the most effective in increasing survival of irradiated mice was chosen for the studies on the activity of bone-marrow pluripotent stem cells. The number of endogenous spleen colonies was significantly increased in mice injected with P. granulosum 3 days prior to or 4 hours after irradiation with 650 R. However, injection of P. granulosum to donors prior to bone-marrow transplantation significantly decreased the number of exogenous spleen colonies in recipients irradiated with 850 R. These results, together with the observed increase in number of CFU-S in peripheral blood and elevated relative spleen weight after injection of P. granulosum, suggest that propionibacteria stimulate migration of CFU-S from bone-marrow via peripheral blood into spleen. Stimulation of CFU-S proliferation and differentiation is also taken into account.

Selective display of histamine receptors on lymphocytes.

Histamine, acting on histamine type 2 receptors, increases intracellular cyclic adenosine monophosphate (AMP) and thus modulates the immunologic functions of lymphocytes. Lymphocyte cyclic AMP levels were used to follow the development of histamine receptors. The B lymphocytes have no functional histamine receptors. As T lymphocytes "mature" in immunologic function--from thymocytes to cortisone-resistant thymocytes to splenic T lymphocytes--their response to histamine increases. The response of these subpopulations of lymphocytes to isoproterenol is the inverse of the histamine response. It is suggested that the changing display of histamine receptors plays an important part in the control of immunologic responses.

Local immunotherapy of stomach and intestinal carcinoma by Propionibacterium granulosum.

The effect of cell suspension of Propionibacterium granulosum strain KP-45 injected intratumorally was estimated in patients with advanced malignant neoplastic disease of the upper and lower part of the digestive tract. These patients received no other anticancer treatment, and results of therapy with P. granulosum were compared with a control group of nontreated patients. In the course of immunotherapy, general clinical state, dynamics of tumor growth as judged by endoscopy and morphologic, biochemical and immunologic parameters of the peripheral blood, were evaluated. Treatment with KP-45 preparation usually resulted in partial regression of tumor growth, accompanied by improvement of the clinical state of these patients, as well as reappearance of normal values of blood picture and biochemical parameters. Enhancement of cellular immune response, evaluated by increased number of peripheral blood T lymphocytes and increased skin reactivity to PHA and PPD, was observed. No serious side effects or complications of local immunotherapy were noted. No patients who had immunotherapy suffered from bacterial infection; this was not the case in the control group.
