Comparison of the gastric antisecretory and antiulcer potencies of telenzepine, pirenzepine, ranitidine and cimetidine in the rat.

In different rat models, the antisecretory and antiulcer effects of the M1-antimuscarinics telenzepine and pirenzepine, the nonselective antimuscarinic atropine, and the H2-blockers ranitidine and cimetidine were compared to each other. Intravenous telenzepine proved to be more potent in inhibiting gastric acid secretion in the Ghosh-Schild rat (carbachol-stimulated), the chronic fistula rat (basal secretion), or, both intravenously and orally, in the modified Shay rat, as compared to pirenzepine, cimetidine or ranitidine. After intravenous administration, only atropine was equally potent to telenzepine in all three models, but it was less potent than telenzepine after oral administration in the modified Shay rat. Gastric mucosal lesions induced by pylorus ligation plus acetylsalicyclic acid or acetylsalicyclic acid plus HCl were best inhibited by telenzepine and atropine, with pirenzepine, ranitidine and cimetidine being less potent, their relative potencies depending on the particular experimental model used. Thus, among the antiulcer drugs tested, telenzepine was the most potent one with respect to both antisecretory and antiulcer activity. Moreover, the duration of the antiulcer effect of telenzepine proved to be significantly longer than that of pirenzepine in the modified Shay rat.

Animals

[Follow-up and management of patients with incomplete cone biopsies for dysplasias (author's transl)].

Report of 86 cone biopsies for dysplasias of the cervix. In 11 of these cases the cone biopsy did not show a clearly dysplasia free margin. These patients were followed from 1 to 6 years without further treatment and none showed a suspicious or positive pap smear. In young patients who desire further pregnancies it appears to be justified to follow without further treatment even after incomplete cone biopsies for dysplasia. Two patients with complete removal of the dysplasia and a normal margin in the cone biopsy later showed positive pap smears. The importance of further follow-up of the cases is stressed.

Cinnabarinate synthase from baboon (Papio ursinus) liver. Identity with catalase.

The enzyme chinnabarinate synthase was purified from the nuclei of baboon liver. Two purified fractions were obtained that exhibited a typical haem protein absorption spectrum; a probable identity with catalase was demonstrated. It was confirmed that catalase in the presence of Mn2+ produces cinnabarinate from 3-hydroxyanthranilate. Doubt is expressed on the existence of a distinct cinnabarinate synthase enzyme.

3-Hydroxyanthranilic Acid

The effect of ascorbic acid supplementation on some parameters of the human immunological defence system.

We have investigated the effect of ascorbic acid (vitamin C) supplementation on some parameters of the human immune defence system in a group of 25 healthy, male university students. The subjects ingested 1 g ascorbic acid per day for a period of 75 days. Serum levels of IgA, IgG, IgM, C-3 complement component, cortisol and transcortin were measured before and after the ascorbic acid course. Corresponding measurements were performed on a control group of 20 healthy, male university students receiving no extra-dietary vitamin C. Our results showed that ascorbic acid supplementation caused a statistically significant increase in the serum levels of IgA, IgM and C-3 complement. Our study does not permit of conclusions regarding the mechanisms of action of ascorbic acid.

Adolescent

Clinical investigation of a very low dosage oral contraceptive combination: 0-75 mg lynestrenol and 0-0375 mg ethinyl oestradiol.

A trial was carried out in 3 clinics to test the efficacy of and cycle control obtained with a very low dosage oral contraceptive combination of lynesternol (0.75 mg) and ethinyl oestradiol (0.0375 mg). A total of 134 healthy, fertile women, ages ranging from under 19 years to over 40 years, participated and completed 1144 treatment cycles. No pregnancies occurred. Then length of the treatment cycle compared to the cycle length before treatment was unchanged in 79.3% of cases. Withdrawal bleeding occurred in 93.1% of all treatment cycles, and in 78.9% of cases the duration of the withdrawal bleeding corresponded to that of the bleeding before treatment. The proportion of irregular bleeding in the first treatment cycle was 51.4% but by the 12th treatment cycle this was reduced to 12.5%. Side-effects, when they did occur, were very slight; existing complaints clearly decreased during the treatment.
