Concentration of serum lipids and aortic lesion size in female and male apo E-deficient mice fed docosahexaenoic acid.

Apolipoprotein (apo) E-deficient mice were fed an atherogenic diet with either 1% ethyl ester docosahexaenoic acid (DHA) or safflower oil (SO) as a source of linoleic acid for 8 week. Both genders fed DHA had higher proportions of eicosapentaenoic acid and DHA, and lower proportions of linoleic and arachidonic acids in the liver and serum phospholipids than those fed SO. Males fed DHA had greater liver weight and tended to have higher concentrations of serum lipids and liver cholesterol than those fed SO, and there were opposite trends in females. Dietary fats and gender led to no significant effect on lesion sizes in aortic arch and thoracic plus abdominal aorta. These results indicate that the interactive action of sex-related factor(s) with dietary polyunsaturated fatty acids is involved in metabolic changes of serum lipids in apoE-deficient mice, and addition of DHA, compared with addition of SO, is not effective to abolish the atherosclerosis in this animal model.

[The increased reactivity of isolated pulmonary artery rings from rats with hypoxia-induced pulmonary hypertension to activator of protein kinase C].

OBJECTIVE: To observe the reactivity of isolated pulmonary artery rings from rats with chronic hypoxic pulmonary hypertension (HPH) to protein kinase.C (PKC) activator. METHODS: Pulmonary artery rings from adult rats exposed to normobaric hypoxia for two weeks and normal rats were isolated for measurement of isometric contractions to PMA, the specific activator of PKC. Responses were examined as follows: (1) The maximal response (P(1)) to 0.5 micromol/L PMA and the time required to achieve a half-maximal response to 0.5 micromol/L PMA (t(1/2)); (2) Dose-response curve in response to PMA (0.01 - 10.0 micromol/L) and the dose of PMA that produced a contractile response as 50% of the maximal response (P(0)) to 80 mmol/L KCl (EC(50) KCl). RESULTS: P(1) of hypoxic group (34.3 +/- 2.4) P(0)% was greater than that of normal group [(26.2 +/- 2.9) P(0)%, n = 6, P < 0.05)], t(1/2) and EC(50) KCl of hypoxic group [(27 +/- 7) min, and (0.7 +/- 0.1) micromol/L] were smaller than those of normal group [(32 +/- 5) min, n = 6, P < 0.05, and (6.0 +/- 0.2) micromol/L, n = 12, P < 0.05]. CONCLUSIONS: The reactivity of pulmonary artery rings from hypoxic rats to PMA was enhanced, which indicates that protein kinase C may have an important role in the development of HPH.

The regulating effects of protein kinase C on the tone of guinea-pig trachea and human lobus bronchi.

The regulating effects of protein kinase C (PKC) on the tone of guinea-pig trachea and human lobus bronchi were investigated by measuring the tone of isolated tracheal and bronchial strips. The effects of PKC on the tone of guinea-pig tracheal and human lobus bronchi were observed and compared. The results showed that: (1) PKC activator PMA induced concentration-dependent relaxation in guinea-pig isolated trachea strips. This relaxation was completely ablated by the pretreatment with 5 x 10(-6) mol/L Ro31-8220 which is a PKC-specific inhibitor, but was not affected by the removal of epithelium (EP), or by the pretreatment with propranolol (beta-receptor blocker) or atropin (M-receptor blocker); (2) PMA led to concentration-dependent contraction in human lobus bronchi. This contractile response was completely depressed by the pretreatment with 5 x 10(-6) mol/L Ro31-8220 and was partly inhibited by 1 x 10(-5) mol/L isoptin (Ca(2+)-antagonist), but was not significantly affected by propranolol or atropin. It is concluded that PKC is involved in the regulation of airway smooth muscle tone. The regulating effects may vary in different animals.

Anomaly detection: eye movement patterns.

The symptom of a garden path in sentence processing is an important anomaly in the input string. This anomaly signals to the parser that an error has occurred, and provides cues for how to repair it. Anomaly detection is thus an important aspect of sentence processing. In the present study, we investigated how the parser responds to unambiguous sentences that contain syntactic anomalies and pragmatic anomalies, examining records of eye movement during reading. While sensitivity to the two kinds of anomaly was very rapid and essentially simultaneous, qualitative differences existed in the patterns of first-pass reading times and eye regressions. The results are compatible with the proposal that syntactic information and pragmatic information are used differently in garden-path recovery.

Dietary soy protein isolate, compared with casein, reduces atherosclerotic lesion area in apolipoprotein E-deficient mice.

The objective of this study was to compare the effects of dietary soy protein isolate and casein on atherosclerotic lesion development in apolipoprotein (apo) E-deficient mice. Male C57BL/6J apoE-deficient mice (9-10 wk old) in groups of 6-9 were used in a series of feeding studies. In the first experiment, mice were fed purified diets containing cholesterol (1 g/100 g) and cholate (0.25 g/100 g) for 6 wk; soy protein isolate or casein was used as the protein source. Although serum total cholesterol concentration did not differ between groups, the lesion area of the thoracic aorta in the soy protein isolate group was lower than that of the casein group (P < 0.01). In the second and third experiments, mice were fed the same purified diet as in Experiment 1, only without supplementation of cholesterol and cholate for 24 and 9 wk, respectively. In each of these two experiments, serum total cholesterol concentrations again did not differ between soy protein isolate- and casein-fed groups. Serum homocysteine concentrations did not differ between groups in Experiment 3. Dietary soy protein isolate, compared with casein, lowered the thoracic aorta lesion area (Experiment 2; P < 0.001) and the percentage of the aortic arch inner surface covered by lesions (P < 0.05). In the final experiment, mice were fed the cholesterol-free diets containing ethanol-extracted soy protein isolate or casein plus the soy protein ethanol extracts for 9 wk. There were no differences in serum total cholesterol concentration or thoracic aorta lesion areas between the two groups. These results indicate that the antiatherogenic effect of native soy protein isolate cannot be explained by its effect on serum lipids or homocysteine and suggest that both the protein component and the ethanol extracts of the soy protein isolate may contribute to the antiatherogenic effect of the native soy protein isolate.

Developmental expression and substrate specificities of alfalfa caffeic acid 3-O-methyltransferase and caffeoyl coenzyme A 3-O-methyltransferase in relation to lignification.

The biosynthesis of monolignols can potentially occur via two parallel pathways involving free acids or their coenzyme A (CoA) esters. Caffeic acid 3-O-methyltransferase (COMT) and caffeoyl CoA 3-O-methyltransferase (CCOMT) catalyze functionally identical reactions in these two pathways, resulting in the formation of mono- or dimethoxylated lignin precursors. The activities of the two enzymes increase from the first to the sixth internode in stems of alfalfa (Medicago sativa L.), preceding the deposition of lignin. Alfalfa CCOMT is highly similar at the amino acid sequence level to the CCOMT from parsley, although it contains a six-amino acid insertion near the N terminus. Transcripts encoding both COMT and CCOMT are primarily localized to vascular tissue in alfalfa stems. Alfalfa CCOMT expressed in Escherichia coli catalyzes O-methylation of caffeoyl and 5-hydroxyferuloyl CoA, with preference for caffeoyl CoA. It has low activity against the free acids. COMT expressed in E. coli is active against both caffeic and 5-hydroxyferulic acids, with preference for the latter compound. Surprisingly, very little extractable O-methyltransferase activity versus 5-hydroxyferuloyl CoA is present in alfalfa stem internodes, in which relative O-methyltransferase activity against 5-hy-droxyferulic acid increases with increasing maturity, correlating with increased lignin methoxyl content.

Neurovirulence in feline immunodeficiency virus-infected neonatal cats is viral strain specific and dependent on systemic immune suppression.

Feline immunodeficiency virus (FIV) is a lentivirus that causes immune suppression and neurological disease in cats. Among animal viruses, individual viral strains have been shown to be neurovirulent, but the role of viral strain specificity among lentiviruses and its relationship to systemic immune suppression in the development of neurological disease remains uncertain. To determine the extent to which different FIV strains caused neurological disease, FIV V1CSF and Petaluma were compared in ex vivo assays and in vivo. Both viruses infected and replicated in macrophage and mixed glial cell cultures at similar levels, but V1CSF induced significantly greater neuronal death than Petaluma in a neurotoxicity assay. V1CSF-infected animals showed significant neurodevelopmental delay compared to the Petaluma-infected and uninfected animals. Magnetic resonance spectroscopy studies of frontal cortex revealed significantly reduced N-acetyl aspartate/creatine ratios in the V1CSF group compared to the other groups. Cyclosporin A treatment of Petaluma-infected animals caused neurodevelopmental delay and reduced N-acetyl aspartate/creatine ratios in the brain. Reduced CD4(+) and CD8(+) cell counts were observed in the V1CSF-infected group compared to the uninfected and Petaluma-infected groups. These findings suggest that neurodevelopmental delay and neuronal injury is FIV strain specific but that systemic immune suppression is also an important determinant of FIV-induced neurovirulence.

[Determination of lactitol in mice serum by high performance liquid chromatography].

An HPLC method for the determination of lactitol in mice serum was developed. A liquid chromatograph equipped with a refractive index detector and a Waters Sugar-Pakl column was used. The column temperature was maintained at 90 degrees C and the mobile phase was redistilled water. One milliliter of the serum sample was treated with 3.0 mL methanol. The precipitate was centrifuged for 15 min at 10,000 r/min. Two milliliters of the supernatant was dried on a water bath at 100 degrees C. The residue was dissolved with 1.0 mL water and 10 microL of which was injected. The average recovery was 98.6%. The detection limit was 0.1 microgram (S/N = 3.75). The linear range was 0-600 mg/L (r = 0.9992). Both the RSD's within day and between days were below 1.0% (n = 5).

Impaired brain development and reduced astrocyte response to injury in transgenic mice expressing IGF binding protein-1.

Numerous reports indicate that insulin-like growth factor-I (IGF-I) has a critical role in brain development, that it contributes to neuronal survival and that its activity is regulated by a family of IGF binding proteins (IGFBPs). In the present study, brain development was investigated in transgenic (Tg) mice that overexpress rat IGFBP-1 under the control of phosphoglycerate kinase promoter. Adult Tg mice had significantly decreased brain weight (299 +/- 17 vs. 499 +/- 11 mg), brain DNA content (684 +/- 8 vs. 810 +/- 31 microg) and brain protein (37 +/- 1 vs. 48 +/- 2 mg) compared with wild-type (Wt) mice and these deficits were already evident at birth. In Tg mice, myelin staining was generally reduced and there was a diminished thickness of the corpus callosum. The total area of hippocampus and dentate gyrus were significantly reduced by 55% and 72%, respectively. Bromodeoxyuridine labeling of proliferating cells in 3-day-old mice was markedly reduced by 41% in the dentate gyrus and by 19% in ventricular zones of Tg mice. Reactive astrogliosis reflected by morphology and glial fibrillary acidic protein expression of astrocytes in response to a mechanical lesion was substantially less in Tg compared with Wt mice. Mixed glial cell cultures from Tg animals were significantly less responsive to stimulation of proliferation by IGF-I than cultures from Wt mice. We conclude that overexpression of IGFBP-1 impairs brain development and reduces glial cell proliferation in response to injury.

Mammography underutilization among older women in Connecticut.

OBJECTIVES: The primary goals were to examine mammography use rates among older women in Connecticut and to determine if there was significant variation among different areas and racial groups in the state. The secondary goal was to examine what impact the initiation of Medicare reimbursement for mammography screening has had on mammography use. DESIGN: Statewide use rates were determined by retrospective Medicare Part B mammography claims analysis. Small area analysis methodology (SAA) was used to identify mammography rates for 23 hospital service areas (HSAs), representing all of the catchment areas for Connecticut's acute care hospitals. PARTICIPANTS: Female Medicare beneficiaries 65 years and older with Part B coverage residing in Connecticut during the study period. MEASUREMENTS: The main outcome (the use of at least one mammogram) was calculated for the calendar years 1991, 1992, and 1993. Mean annual use rates in 1993 were generated for the 23 HSAs and the different racial groups in Connecticut. To examine the effect that Medicare reimbursement for screening mammograms has had on mammography use, rates were calculated for women who met Medicare reimbursement criteria in 1991 through 1993. The rates in 1992 and 1993 were then compared with those in 1991, when the reimbursement program was first initiated. MAIN RESULTS: The mean statewide annual rates among women aged 65 years and older were 23.4% (1991), 24.5% (1992), and 24.9% (1993). The mammography use rates among black women 65 years and older were significantly lower than their white peers in 1991 (18.8% black vs 23.8% white, P < .001), 1992 (20.6% vs 24.7%, P < .001), and 1993 (22.0% vs 25.1%, P < .001). Significant variation was identified among hospital service areas (HSAs) within the state for each time interval studied. The use rates among women aged 65 years and older who were eligible for Medicare screening mammography reimbursement increased significantly from 14.6% in 1991, when Medicare reimbursement for screening mammograms was first initiated, to 18.9% in 1992 (P < .001). The rates in 1993 (17.4%) also increased from the baseline year 1991 (P < .001). However, the observed increases since 1991 have been limited in magnitude. CONCLUSIONS: Low mammography use persists among older women in Connecticut and, in particular, among older black women. The initiation of Medicare reimbursement for screening mammograms in 1991 has had some impact on mammography use although its effects are still limited. Through the use of small area analysis methodology, significant underutilization of mammography in localized areas of the state was identified. These findings have facilitated local outreach interventions. Additional research is needed to understand if health service barriers are contributing to the local variation in rates observed in this study.

[Experimental study prevention of reperfusion injury of skeletal muscle by local hypothermia and its clinical application].

Local hypothermia as a preventive method to reperfusion injury of skeletal muscles was studied. Sixteen Japanese rabbits were divided into four groups at random. Before the tourniquet was inflated, a cold gel pack was applied to the right hind leg of each rabbit for 15 minutes to produce local hypothermic condition, without application of tourniquet the left hind limb was under local hypothermic condition as a control. The duration of tourniquet ischemia was 4 hours, and then reperfusion for one and two hours in the A and B groups respectively; in the C and D groups the duration of ischemia was 5 hours, and reperfusion for one and two hours, respectively. The muscle temperature averaged 16.6 degrees C with a needle thermocouple in the hind limb under local hypothermia. The serum K+, LA, SOD, LPO were determined from bilateral femoral veins, and electron and light microscopic studies of sural muscles were done in the post-reperfusion period. It was found that the K+, LA, LPO were lower than that of the control groups (P < 0.01), but SOD was higher than that of the control group (P < 0.01). Electron and light microscopic studies showed sight but reversible damage of muscular structure with the possibility of in the hypothermic groups cell regeneration. Basing on this experimental results, this method was applied in 45 cases reparative and reconstructive surgery of limbs. The duration of application of tourniquet averaged 2 hours and 57 minutes, the longest being 4 hours and 31 minutes, when the muscle temperature had reduced to 22.4 degrees C. There were no postoperative complications associated with this technique. Local hypothermia appeared to be a safe and effective method of decreasing the reperfusion damage after ischemia.
