[Refraction of the dog eye].

The refraction of 50 eyes of nonsedated dogs was measured by retinoscopy. The slightly hyperopic to middle-grade myopic refraction found in earlier studies was seen in individual cases; however the mean value of 0.5 D is close to emmetropy. The differences in the results of measurements made under non-influenced accommodation and cycloplegia were not significant. Keratometry with the Javal ophthalmometer showed a mean cornea radius of 8.77 mm. A average axial length of 20.9 mm was determined by sonography. In some instances the parameters examined varied considerably.

[Toxicologic studies with thymostimulin].

Thymostimulin (Tp-1 Serono) was tested toxicological in rats and monkeys during 6 weeks of repeated administration. Corresponding to the therapeutic use intended in man the substance was given to rats and monkeys intramuscularly in dosages of 10 or 100 mg/kg b.w. and due to technical reasons subcutaneously in a dosage of 1000 mg/kg b.w. No substance-related effects, either systemic or local, were observed in the monkeys. After treatment with the high dose level (1000 mg/kg b.w. s.c.) the rats exhibited inflammatory changes in the region of the injection sites.

[Topical treatment of acne vulgaris with erythromycin and benzoylperoxide (author's transl)].

16 healthy persons were treated for 7 weeks with a 1% erythromycin containing ointment. Another 25 persons (among them 10 acne patients) were treated during the same period of time with a combination therapy consisting of erythromycin (1% in an unspecific ointment base) and benzoylperoxide (5% in a gel base). The combined therapy reduces the micrococcaceae in the pilosebaceous ducts significantly. On the other side the therapy consisting only of erythromycin causes only initially a decrease and then an increase in the number of micrococcaceae. Induction of resistance to erythromycin is in both cases considerable. In both cases a reduction of propionibacteria which are most important for the pathogenesis of acne must be assumed. Because of the probable pathogenetic role of staphylococci in acne vulgaris an additional improvement of the therapeutical results can be expected in the case of combination therapy. In vitro experiments demonstrate further that an erythromycin treatment leads to considerable resistance induction in S. aureus, regardless of whether it is applied alone or in combination with benzoyl peroxide.

[Pharmacology of N-(2-imidazolin-2-yl)-N-(4-indanyl)amino (indanazoline), a new vasoconstrictive imidazoline compound].

In animal experiments the new imidazoline derivative N-(2-imidazolin-2-yl)-N-(4-indanyl)amine (indanazoline, E-VA-16, as monohydrochloride active substance of Farial) is characterized by a pronounced vasoconstrictive action after local or intravenous application. This is due to a direct action of the compound on alpha-adrenergic receptors. When applied systemically E-VA-16 being a peripherally acting alpha-sympathomimetic induces a rise in blood pressure and a reduction of heart rate and exerts antiphlogistic, spasmolytic, hyperglycemic and diuretic actions. When given by the intranasal route the substance influences blood pressure and heart rate only at concentrations considerably higher than those intended for use in therapy. After enteral administration the effective doses also markedly exceed the single therapeutic doses. There was no evidence of side-effects restricting the use of the drug as compared to other imidazoline derivatives. Studies on the isolated perfused rabbit ear, however, indicated a broader therapeutic range in local application.

[Chemistry and biological properties of substituted 3-amino-1H-isoindoles (author's transl)].

Several 1-, 3-, and 6-substituted 3-amino-1H-isoindoles have been synthesized and screened for biological properties. In pharmacological testing significant and long-lasting local-anesthetic activity was found even in low concentrations. Subsequently, investigations for antiarrhythmic properties were carried out. The same or even higher activity compared to quinidine sulfate was found on isolated guinea pig atria (refractory period prolongation), anaesthetized rats (aconitine induced arrhythmias), guinea pigs (electrically induced cardiac fibrillation) and in conscious dogs with arrhythmias due to coronary artery occlusion. Unfortunately, antiarrhythmic doses caused toxic symptoms in dogs.
