Extensive hepatic cell necrosis produced by the Shwartzman mechanism.

Acute, severe, and extensive necrosis of the liver was produced in pregnant and non-pregnant female adult rabbits by the Shwartzman mechanism. Shwartzman reagent (E. coli endotoxin) was administered in various combinations by three routes of injection, the portal vein (mesenteric vein), the bile duct, and the ear vein. Morphologic changes of the extrahepatic organs were minimal. The similarity to massive necrosis in human liver and the effect of pregnancy on hepatic necrosis in rabbit and man were discussed. The lesion is presented as a new animal model for acute massive hepatic necrosis and is proposed as a third category of Shwartzman reaction, designated the univisceral type.

Hashimoto's thyroiditis: a possible risk factor for lung cancer among Japanese women.

To study the epidemiologic association between Hashimoto's thyroiditis and malignancies, autopsy protocols (1959--1974) were examined at the Tokyo University Hospital. The study cases were classified according to major cause of death, including breast and lung cancer. The relationship between thyroid changes and breast cancer was not significant; however, a significant association between thyroid alteration and lung cancer was identified. This finding suggested that Hashimoto's thyroiditis and related changes could be one of the risk factors for lung cancer among Japanese women.

A study on the melanin-laden dendritic cell (melanocyte) in ovarian cystic teratoma.

To confirm the existence and to analyse the distribution of the melanocyte in cystic teratoma of the ovary, 51 cases were collected, among which 47 were found suitable for gross and histological examinations. The tissue components of cystic teratoma where the melanocyte was seen more or less associating were; the squamous, cuboidal and columnar cell epithelia especially the epidermal epithelium, the brain tissue principally the leptomeninx, and the interstitium. The frequency of melanocytes' association in the respective tissue was shown and compared between each other. One thing which was quite interesting for us was the relationship between the existence of brain tissue and the development of melanocyte in cystic teratoma. The melanocyte appeared in the above tissues, especially the interstitium, of cystic teratoma more frequently when the brain tissue was coexisting in the same tumor. These findings seem to be of meaningful suggestions concerning the melanocytogenesis not only of cystic teratoma of the ovary, but also in a physiological condition in man.

A morphological study of the circadian cycle of the pineal gland of the rat.

To seek a morphological expression of circadian rhythm, we investigated cytologically pineal glands taken from rats every 2 to 4 h under a lighting regime of 12 h of illumination (6:00 to 18:00) and 12h of darkness. The changes in the number of synaptic ribbons and ribbon fields was observed by electron microscopy. The number of these intracellular elements was greatest at 2:00 and lowest at 14:00, the difference being statistically significant. There were no significant local differences in numbers with respect to the part of the pineal gland examined. The data are similar to those of Vollrath from the pineal gland of a guinea pig, and seem to confirm a circadian function in the pineal gland in mammals.

Wound healing and complications of the liver after percutaneous transhepatic cholangiography.

A total of 10 autopsy cases who had received percutaneous transhepatic cholangiography (PTC) were examed and analysed from the view point of chronological changes of hepatic damage caused by the puncture. The conclusion obtained on the process of simple healing can be summarized as follows: (1) Hemocoagulation or hematoma formation on the hepatic surface and intra-peritoneal hemorrhage were seen until the 7th day after PTC but not in cases of the 13th day or later, while the scar of puncture was recognizable on the hepatic capsule in cases of up to the 16th day but was not remaining in ones of the 41st day and later. (2) Damage of the hepatic parenchyma was existing until the 7th day, which, however, was repaired by the 13th day. (3) Intra-hepatic hematoma was seen until the 13th day but faded away thereafter. Namely, intrahepatic trauma caused by PTC seems to be completely cured without leaving any scar in about a half month. However, the puncture trace on the diaphragm was seen in one case of the 42nd day after PTC examination, therefore the nature of diaphragmatic trauma seems to be somewhat different from that of the liver, including its capsule. Besides, discussions are also made on some hepatic complications caused by PTC, as well as on clinical significance of this kind of pathomorphological study.

Phospholipid metabolism in Ehrlich ascites tumor cells. I. Turnover rate of phosphatidylinositol.

1. Radioactive precursors, 32 PI, [1-14C]glycerol, and [1-14C]acetate, were individually injected into the peritoneal cavity of mice bearing Ehrlich ascites tumor, and the rates of incorporation into phospholipid fraction of Ehrlich ascites tumor cells were estimated. Although no distinct difference in specific activities was observed between phosphatidylinositol and other phospholipid classes as regards the incorporation of [1-14C]acetate of [1-14C]glycerol, a higher rate of incorporation of 32Pi into phosphatidylinositol was observed. The specific activity of phosphatidylinositol reached more than ten times that of phosphatidylcholine in the first hour. 2. The radioactivities incorporated into the phospholipids of Ehrlich ascites tumor cells and liver were estimated after simultaneous injection 32Pi and [2-3H]inositol. The incorporation of 32Pi into phosphatidylinositol of liver was similar in specific activity to those of other phospholipids. The ratio (3H/32Pi) of phosphatidylinositol only slightly in the ascites tumor cells, while an appreciable decrease of the ratio was observed in the liver during the first 3 hr. 3. These results suggest that phosphatidylinositol synthesis through pathways other than de novo synthesis is rapid in ascites tumor cells.

Phospholipid metabolism in ehrlich ascites tumor cells. II. Turnover rate of ether phospholipids.

1. Radioactive precursors of phospholipids, i.e., 32Pi, [1-14C]glycerol, [2-3H]glycerol, and [1-14C]acetate, were individually injected into the peritoneal cavity of mice bearing Ehrlich ascites tumor cells and the rates of incorporation were estimated. 2. Although [2-3H]glycerol was not practically incorporated into ether phospholipids, the other three radioactive precursors were incorporated into diacyl, 1-O-alkenyl-2-acryl-, and 1-O-alkyl-2-acryl-GPE (GPC). 3. In the experiments on 32Pi or [1-14C]acetate incorporation, 1-O-alkyl compounds in the ethanolamine phosphoglyceride fraction showed high specific activities in comparison with 1-acyl compounds. In the case of [1-14C]glycerol incorporation, a high rate of incorporation into 1-O-alkyl compounds was not found. In the choline phosphoglyceride fraction, a high rate of incorporation of the above precursors into 1-O-alkyl compounds was not observed. 4. The specific activities of 1-O-alkenyl compounds were fairly low compared with those of 1-acyl- and 1-O-acyl- and 1-O-alkyl compounds throughout the incorporation experiments with [1-14C]glycerol and [1-14C]acetate, but in 32Pi incorporation, 1-O-alkenyl compounds showed higher specific activities than 1-acyl compounds in ethanolamine phosphoglyceride, suggesting an exchange reaction of the phosphorylethanolamine moiety. 5. From the above findings, it appears that alkyl ether phospholipids of ethanolamine from may have a significant role in ascites tumor cells, based on their rapid turnover.

Intimal thickening of intra-organ arteries and smoking habits.

Intimal thickening of the intra-organ arteries of the major organs have been examined in 253 autopsy cases and, the microscopical findings have been analyzed according to smoking habits. It was found that especially that small arteries of the lung present a significantly higher degree of fibrous intimal thickening in cigarette smokers.

Synaptic ribbon in the human pinealocyte.

An electron microscopic study of a pineal gland which had been obtained from a 3-year-old girl at autopsy revealed the presence of synaptic ribbon in it. Namely, in some parenchymal cells observed, synaptic ribbons, each of which was ca. 700 nm in length and with synaptic vesicles of up to 60 nm in diameter gathering on the surface, were found in the cytoplasmic area adjacent to the cell membrane. The substructure of the synaptic ribbon, i.e., parallel striae running inside along the longitudinal direction, was definitely observed. There is a hypothesis which has been almost accepted that the mammalian pinealocyte is homologous to the photoreceptor cell of the pineal organ in lower animals, and one of the morphological evidences which supports this speculation is the presence of sensory cell elements in both. Since, this kind of work has been hardly done with human materials, the above finding may offer a meaningful information which discloses a part of the real function of the pineal gland in man.

Immunological heterogeneity in human ferritinemia.

Hyperferritinemia in various diseases, mainly hematological, was confirmed by immunological methods. For ferritin detection, anti-human placental ferritin antiserum, anti-human hepatic ferritin antiserum, and anti-human leukemia cell ferritin antiserum were used and the result was compared with each other. Leukemia, malignant lymphoma, multiple myeloma, and aplastic anemia are hematological diseases which showed a positive reaction in this test, among which leukemia showed the highest positivity. Cases of hepatic diseases and non-hematological malignant neoplasms also showed a positive reaction. The positivity was quite low and almost negligible in other diseases and healthy individuals. Anti-human placental ferritin antiserum seemed to be suitable for cancer diagnosis and, antihuman hepatic ferritin antiserum for hepatic diseases. The results of analysis of purified human hepatic and placental ferritins highly suggested the presence of immunological heterogeneities between them. Also, a possibility was pointed out that one of the components of the so-called leukemia-specific antigens might sometimes be the isoferritin of leukemia cells.

Virus-like particles in human pinealoma.

Virus-like particles were seen in the tumour cells of a pinealoma in a male of 18 years. The particles were dense, spherical or hexagonal in shape, and 100-130 nm in size. They were found in the cytoplasm, but not in the nucleus. They occurred in aggregates or in isolation. They tended to form rows or circles. These particles may be viral in nature, and of one of the herpes-, leuro- or arena-groups, especially when judged from their size. However, greater certainty as to their nature is impossible from the morphological evidence.
