Crk is required for apoptosis in Xenopus egg extracts.

Apoptosis is essential for the development and homeostasis of multicellular organisms. Recently, a cell-free extract prepared from Xenopus eggs was shown to recapitulate intracellular apoptotic pathways in vitro. While many stimuli have been shown to trigger apoptosis in a variety of cell types, the intracellular signaling pathways involved in apoptosis remain largely unknown. Here we show that addition of a recombinant protein containing the phosphotyrosine binding (SH2) domain from the adaptor protein crk, but not those derived from a panel of other signaling proteins, can prevent apoptosis in the Xenopus egg extract system. Furthermore, immunodepletion of endogenous crk protein from the egg extracts, or addition of anti-crk antisera to these extracts, prevents apoptosis. The ability to undergo apoptosis can be restored to these extracts by addition of recombinant crk protein. These results directly demonstrate that crk participates in apoptotic signaling.

Repeated amphetamine administration alters the expression of mRNA for AMPA receptor subunits in rat nucleus accumbens and prefrontal cortex.

Recent evidence suggests that behavioral sensitization to amphetamine is associated with alterations in excitatory amino acid (EAA) transmission in perikarya (ventral tegmental area) and terminal regions (nucleus accumbens [NAc]) of the mesoaccumbens dopamine system. The present study determined whether repeated amphetamine administration alters expression of mRNAs for AMPA receptor subunits. We studied the NAc, because it is the site of expression of amphetamine sensitization, and the prefrontal cortex (PFC), because it is the origin of EAA projections that regulate the mesoaccumbens dopamine system. Rats were treated for 5 days with 5 mg/kg/day amphetamine sulfate or vehicle (controls) and perfused 3 or 14 days after the last injection. We used a novel in situ hybridization method that allows quantification of mRNA levels [Lu et al. (1996) J. Neurosci. Methods, 65:69-76]. Repeated amphetamine administration decreased levels of GluR1 and GluR2 but not GluR3 mRNAs in both core and shell subregions of the NAc at the 14 day withdrawal time; no changes were observed after 3 days of withdrawal. In contrast, levels of GluR1 mRNA in the PFC were increased at 3 but not 14 days of withdrawal, while GluR2 and 3 mRNAs were unchanged. Levels of GluR4 mRNA were very low in both NAc and PFC. Functional properties of heteromeric AMPA receptors are determined by subunit composition. Thus, the observed changes in mRNAs for AMPA receptor subunits may result in altered AMPA transmission in NAc and PFC. This, in turn, may influence the responsiveness of the mesoaccumbens DA system to psychomotor stimulants and potentially contribute to behavioral sensitization.

Cyclooxygenase products contribute to endothelin-induced pulmonary hypertension and altered lung mechanics in sheep.

Endothelins have potent biological effect in vivo which may, in part, be mediated by stimulation of cyclooxygenase metabolism of arachidonic acid. We administered endothelins (ETs) intravenously to chronically instrumented awake sheep with and without pretreatment with meclofenamate (n = 8). 30 micrograms doses of ET-1, ET-2, and ET-3 caused similar degrees of acute elevation of pulmonary artery pressure (PPA), reduction of the dynamic compliance of the lungs (Cdyn), and increases in lung lymph flow. Pretreatment with meclofenamate inhibited the rise in PPA and reduction in Cdyn, but had no effect on lung lymph flow. We conclude that the biological effects of the endothelins on PPA and Cdyn, but not lung fluid balance, are mediated in part by cyclooxygenase products of arachidonic acid metabolism.

Cattle theileriosis in China.

Amongst the piroplasmoses of livestock in China, the commonest theilerioses are caused by Theileria annulata, T. sergenti and T. mutans. Research carried out over many years has shown these to be distributed mainly in Northern China but they also occur in Southern China. T. annulata and T. sergenti are the most troublesome, affecting large numbers of cattle, especially those of exotic breeds. The incident rate and numbers of fatalities vary widely between areas and breeds of cattle. T. annulata is the most virulent species having an incident rate of 11-46% for indigeneous cattle and causing death rates of between 11-40.8%. For exotic cattle in areas free of T. annulata the sickness rate is up to 61% and mortality rates up to 31%. T. sergenti is avirulent in cattle and clinical symptoms are rarely observed, however, in some areas the infection rate could possibly be as high as 97.5%. Dairy and hybrid cattle imported from other places proved highly susceptible, with mortality rates possibly reaching 14%. The pathogenic forms, clinical signs, tick vectors and the effects of chemical and immune intervention are also discussed in this paper.

Babesiosis in China.

The importance of babesiosis in livestock in China is discussed and mainly focused on bovine and equine babesiosis. Babesiosis is still one of the most important diseases affecting livestock and has caused great economic loss. Nine species of Babesia have been recognized in livestock: B. bigemina, B. bovis, B. major, B. motasi, B. ovis, B. perroncitoi, B. trautmanni, B. equi (Theileria equi), B. caballi. The distribution of Babesia follows the distribution of the tick vectors. The main vectors of bovine babesiosis are the one-host tick Boophilus microplus and the three-host ticks Rhipicephalus haemaphysaloides haemaphysaloides, Haemaphysalis punctata and Haemaphysalis longicornis. Bovine babesiosis has caused significant losses in milk and meat from cattle in most parts of China. The disease is also a barrier to improving productivity of local cattle by cross-breeding due to the high mortality of genetically superior but highly susceptible cattle, especially dairy cattle, imported from Babesia-free areas. Dermacentor nuttalli is the major vector of equine babesiosis and the tick is distributed in almost all parts of North China. Outbreaks of equine babesiosis have not been very common, but in some districts the disease has seriously affected horses, donkeys and mules.

Attempted transmission of Babesia major by Boophilus microplus.

Two experiments was carried out to determine if Babesia major could be transmitted by Boophilus microplus. In experiment 1, a Babesia-free batch of laboratory reared Bo. microplus larvae were applied to an intact calf infected by inoculation with a B. major stabilate. The calf showed a B. major parasitaemia while the larvae, nymphs and adult ticks were engorging. The engorged females were cultured and batches were incubated at one of the three following temperatures: 24, 28 or 32 degrees C. Approximately 10,000 larvae derived from each of the females were used to infest each of three splenectomized calves. In experiment 2, Babesia-free Bo. microplus larvae were applied to a splenectomized calf; the calf was injected with B. major stabilate and showed a B. major parasitaemia during the adult stage of tick development. The engorged females were incubated at room temperature and the resulting larvae (approximately 10,000) were used to infest a splenectomized calf. Examination of blood films for the presence of B. major from the four calves infested by the second generation larvae in the two experiments were negative.

Effects of ligustrazine on bone marrow microvessel system in the early period of acute radiation injury in mice.

Sublethally irradiated mice were immediately treated with 250 mg/kg Ligustrazine Phosphiatis intraperitoneally twice a day for seven days, and the bone marrow microvessels of the Ligustrazine group was much greater than that of the control group. On the 7th day, the amount of the control group decreased to normal, while the ligustrazine group was still increasing, and the microvessel area was enlarged obviously.. The percentage of the hematopoietic tissue volume in bone marrow between the two groups had no significant difference in the first 7 days. On the 7th day after irradiation, the peripheral neutrophilic granulocytes increased in the Ligustrazine group. The results suggested that early use of Ligustrazine after acute radiation injury might improve the blood supply of bone marrow, and be helpful for recovery of hematopoiesis.

The study on the relationship between the expression of calponin and gallstone formation.

In order to understand the molecular mechanisms of gallstone formation, the expression of calponin in animal model of gallstone disease was studied. High-cholesterol diet was given to the guinea pigs to induce gallstone formation. RT-PCR and Western-blotting were used to evaluate expression level of calponin gene. Down-regulation of calponin gene expression was observed in animals with gallstone, while myosin expression was relatively stable. Our results indicated that the decrease of calponin could increase the pressure of Oddi's sphincter, aggravate the stasis of bile and promote the gallstone formation.

Modified Evans blue fluorimetry for determination of pulmonary vascular permeability in rats sustaining burns, and delayed fluid resuscitation of burn shock.

The present experiment used modified Evans blue fluorimetry to determine changes in pulmonary vascular permeability using a delayed resuscitation model of burn shock in rats. The results showed that pulmonary vascular permeability in the immediate resuscitation (IR) group regressed to normal 12-24 h following the burn whereas it regressed slowly in the delayed resuscitation (DR) group. The study showed that Evans blue fluorimetry is a reliable and sensitive method for determining pulmonary vascular permeability, and dimethyl formamide is a preferable extracting solution which provides a quick and convenient means for scientific research.

Perinatal lethality with kidney and pancreas defects in mice with a targetted Pkd1 mutation.

PKD1 is the most common site for mutations in human autosomal dominant polycystic kidney disease (ADPKD). ADPKD is characterized by progressive replacement of kidney tissue by epithelial cysts and eventual renal failure. Hepatic and pancreatic cysts are also common. The PKD1 protein, polycystin, is a cell-surface protein of unknown function that is widely expressed in epithelia and in vascular smooth muscle and myocardium. None of the genetic forms of murine polycystic disease map to the murine Pkd1 locus. We introduced into mice by homologous recombination a Pkd1 truncation mutation, Pkd1-, that mimics a mutation found in ADPKD. Pkd1- heterozygotes have no discernible phenotype, whereas homozygotes die during the perinatal period with massively enlarged cystic kidneys, pancreatic ductal cysts and pulmonary hypoplasia. Renal cyst formation begins at embryonic day 15.5 (E15.5) in proximal tubules and progresses rapidly to replace the entire renal parenchyma. The timing of cyst formation indicates that full-length polycystin is required for normal morphogenesis during elongation and maturation of tubular structures in the kidney and pancreas.

Fictive oesophageal peristalsis evoked by activation of muscarinic acetylcholine receptors in rat nucleus tractus solitarii.

The aim of the present study was to determine if muscarinic acetylcholine receptor-mediated peristaltic rhythmogenesis in the rat oesophagus is a central motor program that can be generated without peripheral sensory support. In anaesthetized male Sprague-Dawley rats, pressure-ejection of glutamate (10-20 pmol) and muscarine (5-10 pmol) in the sub-nucleus centralis of the nucleus tractus solitarii (NTSC) evoked monophasic pressure waves and rhythmic oesophageal peristalsis, respectively, but did not change mean arterial blood pressure or respiration. Application of muscarine (50-100 pmol) to the NTS extraventricular surface evoked rhythmic multi-unit burst discharges in the compact formation of the nucleus ambiguus (AMBC) that led to oesophageal peristalsis in a phase-locked manner. Evoked rhythmic AMBC activity persisted during neuromuscular blockade with curare, although the peak frequency of individual bursts was decreased. In a brainstem slice preparation, intracellular and whole cell patch recordings from AMBC neurones during focal stimulation of the NTSC region with muscarine revealed rhythmic depolarizing waves that showed a pattern similar to that of rhythmic oesophageal peristalsis. The present findings support the concept that medullary circuits comprising premotor neurones of the NTSC are intrinsically capable of generating rhythmic oesophagomotor output, but are subject to a powerful modulation by peripheral sensory feedback.

Distribution and developmentally regulated expression of murine polycystin.

PKD1, the gene that is mutated in approximately 85% of autosomal dominant polycystic kidney disease (ADPKD) cases in humans, has recently been identified (Eur. PKD Consortium. Cell 77: 881-894, 1994; also, erratum in Cell 78: 1994). The longest open-reading frame of PKD1 encodes polycystin, a novel approximately 460-kDa protein that contains a series of NH2-terminal adhesive domains (J. Hughes, C. J. Ward, B. Peral, R. Aspinwall, K. Clark, J. San Millan, V. Gamble, and P. C. Harris. Nat. Genet. 10: 151-160, 1995; and Int. PKD Consortium. Cell 81: 289-298, 1995) and several putative transmembrane segments. To extend studies of polycystin to an experimentally accessible animal, we have isolated a cDNA clone encoding the 3' end of Pkd1, the mouse homologue of PKD1, and raised a specific antibody to recombinant murine polycystin. This antibody was used to determine the subcellular localization and tissue distribution of the protein by Western analysis and immunocytochemistry. In the mouse, polycystin is an approximately 400-kDa molecule that is predominantly found in membrane fractions of tissue and cell extracts. It is expressed in many tissues including kidney, liver, pancreas, heart, intestine, lung, and brain. Renal expression, which is confined to tubular epithelia, is highest in late fetal and early neonatal life and drops 20-fold by the third postnatal week, maintaining this level into adulthood. Thus the temporal profile of polycystin expression coincides with kidney tubule differentiation and maturation.

The effects of hypoxia on the bioenergetics of liver in situ in chronic ethanol-treated rats: a noninvasive in vivo 31P magnetic resonance spectroscopy study.

OBJECTIVE: The effects of hypoxia on bioenergetic parameters of the livers of rats with and without chronic ethanol (CE) treatment has been studied in a totally noninvasive manner, using in vivo 31P magnetic resonance spectroscopy (MRS). METHOD: The effects of hypoxia (10% oxygen for 55 minutes) on the levels of ATP, inorganic phosphate (Pi), phosphomonoesters (PME) and phosphodiesters (PDE) were followed with time. The CE and control rats (four pairs) were starved for 16-20 hours to eliminate the effects of any ethanol still present within the liver. To complement the in vivo studies, perchloric acid extracts from rats (four pairs for each stage) at the three stages, initial (normoxic), hypoxic and recovery, were analyzed using in vitro high resolution 31P MRS. RESULTS: After hypoxic challenge, the hepatic Pi and PME levels increased much more significantly and more quickly for the CE rats compared to those of controls, and ATP levels decreased more dramatically for CE rats. The recoveries of Pi and PME levels after return to normoxic conditions were incomplete for the CE rats in contrast to the controls. The hepatic extract analyses showed that several different PMEs increased during hypoxia, including alpha-glycerophosphate, phosphocholine, AMP, IMP and glycolytic metabolites, and that total hepatic Pi increased during hypoxia in addition to cytoplasmic Pi. CONCLUSIONS: CE-treated rat livers in situ are bioenergetically more sensitive to a hypoxic challenge than are control rat livers, even in the absence of endogenous ethanol. This is consistent with a model of CE-induced liver damage in which CE-treated livers already have centrolobular hypoxia under normoxic conditions and cannot adequately compensate for additional hypoxic stress.

[Analysis of 37 cases of pneumonia of Legionnaires' disease].

OBJECTIVE: To heighten our awareness of pneumonia of Legionnaires' disease (LP). METHODS: 37 cases of LP were reviewed. RESULTS: Besides the characteristics of pneumonia, extrapulmonary manifestations, including gastrointestinal, hepatic, renal and neurologic abnormalities, occurred in 35% of the patients, pleural effusion was more common (40%), 38% and 17% of the patients showed hyponatremia and pulmonary abscess respectively. Roentgenographic changes appeared to last longer and resoluted later than the clinical improvement occurred. CONCLUSION: Erythromycin shows good effects in treating LP.

[Via the submandible: cervical glossopharyngeal neurectomy and histomorphology findings].

Three cases of serious idiopathic glossopharyngeal neuralgia are reported. They were treated with cervical glossopharyngeal neurectomy via the submandible. The resected nerve tissue was detected under light and microscope. The pathological change was found in the form of myelinoclasis. The present article is intended to introduce a method via the submandible, comparing the intracranial and extracranial approach. We think the procedure via the submandible is a method more safe, simple and therefore, worthy to be recommended.

[Study on dietary pattern and nutrients intakes of residents in areas of high and low incidence of esophageal cancer].

The dietary pattern and nutrients intakes of residents in areas with high and low prevalence of esophageal cancer were studied by 3 days' dietary survey. The results showed that the dietary pattern and nutrients intakes of residents in areas with low prevalence (ALP) of esophageal cancer was superior to that in area with high prevalence (AHP). The amount of soybean, meat and vegetable consumed by residents in the ALP was more than that in the AHP. Deep color vegetable and gaelic vegetable were dominant in ALP, but so were light color vegetables in AHP. The protein quality and quantity as well as the amount of Zn, Ca, RE and riboflavin consumed in the AHP were lower than that in the ALP. The energy from carbohydrate accounts for 78.14% of the total in the AHP. It is suggested that poor dietary pattern and the quality and amount of nutrients intake may be related to the development of esophageal cancer, and nutrition intervention in area of high prevalence seems rational.

[Study on liver targeting and sustained release hydroxycamptothecin polybutylcyanoacrylate nanoparticles].

The liver targeting and sustained release hydroxycamptothecin polybutylcyanoacrylate nanoparticles (HCPT-PBCA-NP) which were wrapped up with ployvinylpyrrolidone(PVP) were prepared by adsorption-enwrapping method. The morphology, size and size distribution, drug loading, release characteristics in vitro, distribution and pharmacokinetic parameters in animals of the PVP-HCPT-PBCA-NP were studied. The results showed that the average diameter was 81.2 nm, drug loading was 1.22%. The release characteristics in vitro was in accord with Higuchi equation: Q = 0.0615 + 0.0940 square root of t.64.5% of the HCPT were concentrated in liver 15 min after iv PVP-HCPT-PBCA-NP. The plasma drug concentration-time curve of the HCPT in 5 rabbits was fitted to a two-compartment open model. The Vc was 3.548 L; T1/2 was 146.99 h; CL was 0.178 L.h-1. The method of preparation presented in this paper seems to have important reference value for the preparation of PBCA-NP of the insoluble drugs both in water and in oil.

Dual reciprocity boundary element method for solving thermal wave model of bioheat transfer.

The newly developed dual reciprocity boundary element method (DRBEM) was extended to solve the thermal wave model of bioheat transfer (TWMBT) and a practical algorithm was established. A preliminary simulation of the temperature evolution in a two dimensional zone under certain boundary conditions and micro wave heating was conducted as a numerical illustration for the method and some meaningful conclusions were drawn. The validity of DRBEM was testified through a comparison with finite difference method (FDM) under one-dimensional calculation. Owing to its unique advantages like not confined by the complex shape of biological bodies, no need of discretization of the inner domain, save vast CPU time and easy to deal with different bioheat models, DRBEM may become an important approach for predicting and controlling the transient temperature field of biological bodies under hyperthermia or hypothermia.
