Several substance P-rich areas in rat CNS had increased levels of substance P-like immunoreactivity following the intraventricular injection of the protease inhibitors SQ 20881, SQ 14225, and leupeptin. There were significant differences in response patterns from region to region, possibly on account of an interaction of anatomical, biochemical, or physiological variables. Although the compound SQ 14225 appeared to be the most potent of the inhibitors examined, it had no apparent effect on CNS substance P-like immunoreactivity when administered peripherally.

The cerebrospinal fluid concentration of hydroxylase cofactor has been measured in patients with Parkinson's disease, the Shy-Drager and Steele-Richardson syndromes, adult onset focal dystonia, essential tremor, Huntington's disease and presenile dementia. The results were compared with age matched controls and low values were demonstrated for all disease groups studied except for focal dystonia.
The level of cerebrospinal fluid (CSF) hydroxylase cofactor activity was measured in 34 neurologically normal patients and analyzed according to age. Levels declined by approximately one-third over a 40-year span, probably reflecting loss of aminergic neurons. Fourteen patients with Parkinson disease all tended to have lower values than age-matched controls. Among Parkinson patients with the same order of disability, younger cases had higher CSF concentratons of hydroxylase cofactor than older patients.
Electrical stimulation (1--50 Hz, 100--500 muA, 0.3 msec pulse duration) of the dorsal or median raphe nucleus in anesthetized rats produces a transient increase in arterial blood pressure. Heart rate and respiratory rate do not appear to change systematically after stimulation. Transections rostral or caudal to the raphe nuclei or electrolytic lesions in the area of the nucleus linearis, pars caudalis abolishes the pressor response to stimulation. Depletion of brain serotonin with p-chlorophenylalanine significantly attenuates the pressor effect of both dorsal and median raphe stimulation. The p-chlorophenylalanine effect can be partially reversed by Ro4-4602, a peripherally acting decarboxylase inhibitor, plus 5-hydroxytryptophan. Fluoxetine, a specific serotonin uptake inhibitor, prolongs the duration of the raphe pressor response and slightly increases its magnitude. Injections of 2-bromolysergic acid diethylamide (BOL), a putative serotonin antagonist, into the anterior hypothalamus/preoptic area significantly attenuates the dorsal raphe pressor effect, whereas treatment of rats with the sympathetic postganglionic blocking agent bretylium prevents the stimulation-induced pressor effect. Taken together, these data suggest that stimulation of the ascending serotonergic neuronal system produces a phasic pressor effect which is mediated, at least in part, by synaptic serotonin.

Plasma dopamine-beta-hydroxylase (DBH) activity was studied in two different populations of chronic schizophrenic patients and assayed by two independent laboratories. No significant difference between schizophrenic patients and normal controls was found although in both groups chronic undifferentiated schizophrenics with paranoid features had a trend towards lower DBH activity than the other patients and controls. In addition, DBH and monoamine oxidase (MAO) activities were studied in 13 schizophrenic patients and available first degree-relatives. There was no association of low MAO and low DBH activities within the schizophrenic families.

A method for measuring hydroxylase cofactor activity in human cerebrospinal fluid is described. The hydroxylase cofactor content of cerebrsopinal fluid from Parkinsonian patients is approximately 50 percent that of normal subjects. A significant correlation between hydroxylase cofactor and the concentration of homovanillic acid in the cerebrospinal fluid was observed.
