[Early use of Chinese drug rhodiola compound for patients with post-trauma and inflammation in prevention of ALI/ARDS].

OBJECTIVE: To evaluate the effects of the Chinese drug rhodiola on the preventive treatment of acute lung injury (ALI) caused by post-traumatic/inflammatory and thoracic-cardiovascular operations. METHODS: 104 patients who coincide with their diagnosis and standard scale were divided randomly in to single number groups A(1), B(1), C(1) and D(1) (early preventive treatment group EG) and double number groups A(2), B(2), C(2) and D(2) (control group CG). The treatment of primary disease both in EG and CG was similar, but in EG four grams of rhodiola compound (RCO) were used every day for a period of 3-15 days since admission and 5-7 days after operation. The indicators related to ALI in each groups were measured. RESULTS: PaO(2), AaDO(2) and hemorheology were significant in EG than in CG (P < 0.05) or (P < 0.01). The complication rate of ARDS was decreased about of 50.0% in EG than in CG. CONCLUSIONS: Early use of RCO may protect against major risk factors of ALI/ARDS caused by massive trauma/infection and thoracic-cardio-operations.

[Sex hormone receptor of hemangioma and vascular malformation in children].

OBJECTIVE: To investigate the level and difference of estrogen receptor (ER), progestogen receptor (PR) and androgen receptor (AR) in hemangioma tissues of Children. METHODS: 31 specimens of hemangioma and 36 specimens of vascular malformation were compound stained with hematoxylin-eosin and enzyme-linking affinitive immunohistochemistry. The prepared tissue slices were examined microscopically to find the ER, PR and AR in all the cells. RESULTS: The ER, PR and AR were detected in the two kinds of the tissue. However, their levels in the hemangioma were higher than those in the vascular malformation (P < 0.01) and the levels in the hemangioma of proliferating phase were higher than those in the hemangioma of regressive phase (P < 0.01). CONCLUSIONS: Hemangioma is one of the target tissues of estrogen. ER, PR and AR play an important role in the growth and development of hemangiomas. The binding site of the receptor may be occupied by extrogenous glucocorticoid so that hemangiomas gradually degenerates.

[Lactulose mannitol ratio in patients after operation].

OBJECTIVE: To study lactulose (lactulose, L) and mannitol (mannitol, M) ratio in urine following operation and investigate the correlation between severity of stress and L/M ratio. METHODS: 20 patients who had received operation were enrolled and each patient was evaluated by APACH II scores. L/M permeability study was performed before the operation and 5 and 10 days after the operation. The L and M concentration was analyzed by HPLC with pulsed electrochemical detection (HPLC-PED). The correlation between L/M ratio and the severity of surgical stress were related. RESULTS: The L/M ratio before the operation was (0.023 +/- 0.002), increased to (0.042 +/- 0.005) in the 5th post-operative day (P < 0.01), and then returned to (0.024 +/- 0.002) (P > 0.05) compared with pre-operation in the 10th Post-operation. A significant correlation between the severity of stress and the L/M ratio was observed (r = 0.762). CONCLUSIONS: The L/M ratio was increased 5 days after the operation and the L/M ratio was correlated with severity of operative stress.

[Vitrectomy for vitreous hemorrhage caused by retinal vein occlusion].

OBJECTIVE: To evaluate the efficacy and complications of vitrectomy for vitreous hemorrhage after retinal vein occlusion. METHODS: We retrospectively studied 47 patients (47 eyes) having undergone vitrectomy during the period from April, 1992 to December, 1996. Follow-up was made on 30 patients. RESULTS: (1) Visual acuity was improved postoperatively in 80% of the eyes. The patients with branch vein occlusion achieved more favorable final vision than those with central retinal vein occlusion (P < 0.05). (2) The most common complications were lens opacity, recurrent vitreous hemorrhage, iatrogenic rhegmatogenous retinal detachment and glaucoma. (3) Patients treated with endophotocoagulation had a lower rate of recurrent vitreous hemorrhage (P < 0.05). CONCLUSIONS: (1) More than half of the patients can achieve ambulant vision after vitrectomy for vitrous hemorrhage induced by retinal vein occlusion. (2) Recurrent vitreous hemorrhage can be effectively prevented by endophotocoagulation during the operation.

[LASIK for high and moderate hyperopia].

OBJECTIVE: To evaluate safety and efficacy of excimer laser in situ keratomileusis (LASIK) for high and moderate hyperopia. METHODS: LASIK was performed on 17 eyes of 9 patients using the SCHWIND KERATOM-F excimer laser. The range of preoperative spherical equivalent was + 5.50 to + 9.00 D, and its mean value, (+ 7.18 +/- 1.00) D. All the cases were followed up for 12 months or longer. RESULTS: There was no serious complication during the operation. No eye lost more than 1 line of the best corrected visual acuity after surgery. At 12 months, the mean postoperative spherical equivalent was (+ 1.77 +/- 1.56) D. 52.9% of eyes reached 0 to + 1.00 diopter. 88.2% of eyes achieved an uncorrected far vision of >or= 0.5. 100.0% and 76.5% eyes got uncorrected near visual acuities of >or= 0.5 and >or= 1.0 respectively. CONCLUSION: It is demonstrated the LASIK is a safe and effective technique for the treatment of high and moderate hyperopia. Further algorithm calibration must be made to avoid undercorrection.

[A study on immunogenicity and safety of bivalent inactivated vaccine against hemorrhagic fever with renal syndrome].

OBJECTIVE: To study side effects and effectiveness of bivalent inactivated vaccine against hemorrhagic fever with renal syndrome in population of a randomized controlled field trial. METHODS: Serum indirect immunofluorescent antibody in 167 persons and neutralization antibody (types I and II) in 69 persons, who received three-dose vaccine, were determined two weeks after immunization, and side effects in 657 vaccinees were observed within 72 houses after immunization. RESULTS: Serum positive conversion rate of indirect immunofluorescent antibody was 99.04% (166/167) with GMT of 24.51 +/- 2.06. Serum positive conversion rate of neutralization antibody was 100% (69/69), 91.30% (63/69) for types I and 88.41% (61/69) for type II, respectively. GMTs for type I and II neutralization antibody were 18.27 +/- 2.21 and 12.47 +/- 2.16, respectively. Side effects at local site vaccinated of the body and temperature rising could be seen in 1.48% and 0.36% of the vaccinees, respectively. No severe side effect and abnormal reaction was found in vaccinees on the field. CONCLUSION: Immune response to bivalent inactivated vaccine against hemorrhagic fever with renal syndrome was satisfactory with slight side effect.

[A study on the loss of heterozygosity of multiple tumor suppressor genes in gastrointestinal metaplasia].

OBJECTIVE: To investigate the role of multiple tumor suppressor gene abnormality in gastrointestinal metaplasia (IM). METHODS: The loss of heterozygosity (LOH) of YNZ22, APC, MCC and DCC genes was examined by PCR-RFLP technique in 60 IM cases. RESULTS: LOH of YNZ22, APC, MCC and DCC genes were found in 19.4% (6/31), 12.0% (3/25), 13.0% (3/23) and 4.3% (2/47) respectively. IM was classified into types I, II and III. It was found that the LOH rates of each tumor suppressor gene of type III IM were higher than those of types I & II IM, but the difference was not significant (P > 0.05). Two cases of type III IM were found to also have 2 gene alterations (APC, MCC or YNZ22, MCC) respectively. All LOH were found in IM of adjacent carcinoma. None were found in IM of cancer-free patients. CONCLUSION: The alteration of tumor suppressor gene may play a role in the development and carcinogenesis of IM.

[Epstein-Barr virus infection and expression of T-cell intracellular antigen-1 (TIA-1) in intestinal T-cell lymphoma].

OBJECTIVE: To study the status of Epstein-Barr virus (EBV) infection and the expression of TIA-1 antigen in 24 cases of intestinal T-cell lymphomas. METHODS: In situ hybridization for EBER1/2; immunohistochemical staining by three step ABC technique for TIA-1, LMP-1, CD30, CD45RO, CD3 AND CD20. 19 cases of intestinal B-cell lymphomas were used for comparison. RESULTS: (1) Of the 24 intestinal T-cell lymphoma cases, 23 were pleomorphic, medium to large cell type, and one cases was small cell type; in 20 of the 24 cases EBER1/2 signals were detected in the tumor cells; in 4 of the 20 EBER1/2 positive cases, tumor cells expressed LMP-1; EBER1/2 was not detected in 19 cases of intestinal B-cell lymphomas. (2) in 23 of the 24 cases, TIA-1 was expressed in most tumor cells, but not in the only case of small cell type; TIA-1 was negative in tumor cells of all 19 cases of intestinal B-cell lymphomas. CONCLUSIONS: A high level of EBV infection was present in this group of intestinal T-cell lymphomas, the expression of TIA-1 may be related to its cell origin. Intestinal T-cell lymphoma may be an EBV-associated extranodal NK/T cell lymphoma.

[Expression of WT1 gene mRNA in leukemia].

UNLABELLED: WT1 gene expression was determined in acute leukemic patients by RT-PCR. RESULTS: 19 patients were found to have positive results among 28 patients, the positive rate is 67.9%, and in 16 ANLL patients, 11 cases got positive results, in 12 ALL patients, 8 cases got positive results. It suggested that WT1 was highly expressed in acute leukemia. It is a new and significant detective marker for monitoring the effect of chemotherapy, predicting the prognosis and in the research of minimal residual disease (MRD).

[Analysis of penicillins in whole blood by reversed high-performance liquid chromatography].

A reversed-HPLC method was established to determinate penicillins in whole blood. Thiamphenicol as internal standard was added to 0.5 ml blood. Proteins in blood were precipitated with acetonitrile, then the separate supernatant was directly injected onto the chromatography column. The range of five penicillins' linearities was 0.9918 to 0.9996, their range of relative recovery was 91.75% to 107.33%, their CV was 3.56% to 9.85%. Effects of different storage temperatures on the stability of analytes and interference of other antibiotics were also studied.

[Application of solid-phase microextraction technique to the detection of amphetamines in urine by GC].

A simple and rapid detection of nine amphetamines co-existing in urine was described. In the test, the method of solid-phase micro-extraction (SPME) by GC technique was used. Urine (1.0 ml), NaCl (0.3 g) and 4-phenylbutylamine (internal standard) were added into a vial (1.5 ml), then the sample was adjusted to pH 12 with 10% NaOH and sealed with a teflon-coated septum. After immersion of the SPME fiber (100 PDME) in the sample for 15 min, the SPME needle was inserted into the injection port of the GC and extruded for 3 min. The result showed that each peak from nine amphetamines compounds and internal standard was clearly separated. The calibration curves were linear from 0.2 to 15 micrograms/ml for most of five amphetamines with r between 0.9928-0.9995. The CV were less 10%. It is concluded that the method is simple, quick, accurate and useful for the practical detection of urine concentration of amphetamines.

[Segmental analysis of hair from meperidine abusers and evaluation of the results].

Using meperidine abusers as subjects for this study, we detected in hair meperidine and its metabolites normeperidine, N-OCH3-meperidine and N-AC-meperidine. The content of meperidine and normeperidine of 60 abusers were 103 +/- 130 ng/mg and 117 +/- 143 ng/mg respectively. Meperidine was stably present in hair for at least 20 months. Normeperidine level decreased from hair root to tip. The segmental analysis of hair revealed that there were correlations between the drug abuse history and the distribution of drug along the hair shaft, and between the dose of meperidine and its content presented in hair.

[Study on the combination of isotachophoresis and high performance liquid chromatography for the separation of complex samples].

A method on the combination of isotachophoresis (ITP) and high performance liquid chromatography (HPLC) for the separation and analysis of the complex samples containing proteins and metals is proposed. The instrumental system consisted of a self-made Model OCEP-1 isotachophoresis analyser, main parts of Waters high performance liquid chromatograph and a self-made Model IHI-1 interface. The interface is simple and convenient to use. The injection volume of the interface was determined accurately with a neutralization titration method. In ITP experiment the leading electrolyte was 2.5 mmol/L HCl containing 30 mmol/L of 18-crown-6; the terminating electrolyte was 5 mmol/L Tris+Cl-(pH 6.0); ITP current, 40 microA; detection of metal ions, potential gradient detector. In HPLC experiment the column was a mu-Bondapak C18, reversed-phase column, 4 mm x 200 mm, 10 microns, 12.5 nm; the mobile phase A, V(isopropanol with 5% volume fraction):V(butanol) = 2:1 containing 2.2 mmol/L HCl (pH 2.63); the mobile phase B, V(isopropanol with 50% volume fraction):V(butanol) = 2:1; low pressure gradient elution, mobile phase B changed from 0 to 70% in 17.5 minutes; flow rate, 1.0 mL/min; detection wavelength 225 nm; temperature, 25 degrees C. The experimental results showed that proteins were interfered in the determination of the metals with ITP method. It also showed that HPLC with the use of C18 column can separate the proteins but not the metal ions. By using this new combination system, a complex sample of the proteins and metal ions was injected to the HPLC, the proteins Lys, BSA and OVA were well separated in HPLC, while NH4+, K+, Na+, and Ca2+ were less retained and eluted first. Then the latter was introduced into the system of the ITP with the aid of the interface for separation. Satisfactory results were obtained.

[Simultaneous analysis of histamine and noradrenaline in human plasma by high performance liquid chromatography/electrochemical detection].

This paper reports a reversed-phase high performance liquid chromatographic method for the simultaneous separation and determination of histamine(Hist) and noradrenaline(NA) in human plasma by means of pre-column derivatization of Hist and NA with OPA in the presence of 2-mercaptoethanol. The analysis was carried out with Partisil ODS column, V[sodium acetate buffer solution(pH 4.90)]:V(acetonitrile) = 7:3 as mobile phase and 3-methylhistamine as internal standard. The detector potential was +0.7 V for both compounds to keep minimum background noise. A linear relationship between the electrochemical signal and the compound mass concentrations was obtained. The minimum detectable limit was 15 ng/L (S/N = 3). An example of application of the method to the determination of Hist and NA in human plasma is presented.

[Isotachophoresis analysis of hydrolytic products of DFP catalyzed by DFPase in porcine liver].

DFPase can hydrolyze the P-F bond of diisopropyl phosphate fluoride (DFP) to produce diisopropyl phosphate and HF. In this article, the activity of two kinds of DFPase from porcine liver was assayed by the methods of titration and F- ion sensitive electrode. The results showed that the activity of large molecular DFPase can be determined by both methods, but the activity of small molecular DFPase can only be determined by the titration and cannot be determined by F- ion sensitive electrode. The isotachophoresis analysis of the products of DFP hydrolyzed by the large and small molecular DFPase was studied. The results showed that the small molecular enzyme could catalyze the hydrolysis of DFP, but without the production of F- ion, and that this enzyme is one kind of phosphoesterase that hydrolyzed P-OR bond of DFP.

[Expression and significance of neuropeptide and neurotensin in the extract of platelet of chronic renal failure patients during hemodialysis].

The aim of this study was to investigate the change of neuropeptide Y(NPY) content and neurotensin (NT) content of the platelet from chronic renal failure (CRF) patients during hemodialysis (HD) and explore its mechanism of action. Platelet was separated from the patients' plasma and the NPY and and NT contents of the platelet and plasma were dynamically observed pre-HD and post-HD. 30 healthy people were chosen as the controls. The results showed the NPY and NT contents of the platelet extract were (58.18 +/- 21.29) ng/10(9) and (25.38 +/- 13.43) ng/10(9) respectively. The NPY content of the platelet extract of the CRF patients was obviously decreased and the NT content of the plasma was obviously increased however, the NT contents of the platelet extract and the plasma were both higher than those in the control group. The NPY content of the platelet extract and NT content of the plasma at the post-HD obviously increased as compared with pre-HD, but the NT content of the platelet extract and the NPY content of the plasma obviously decreased. There was an obvious correlation between NPY and NT. The authors conclude that during the period of CRF, the immunoreactions which mainly involve bio-active substances such as NPY, NT and 5-hydroxytryptamine released by the platelet to interfer the vasoconstrictive effect are the main pathologic factors to induce renal hypertensin and renal vasospasm.

[Dynamic changes in IL-4, IL-5 and IL-10 in liver and bone marrow of mice infected with Schistosoma japonicum].

AIM: To observe the changes in Th2 cytokines in the liver and bone marrow of mice infected with schistosomiasis japonica. METHODS: ABC immunohistochemical staining technique and multimedia pathological picture analysis were used. Meanwhile, IL-4, IL-5 and IL-10 derived from livers and bone marrows of infected mice were observed at wk 8, 10 and 12 after infection. RESULTS AND CONCLUSION: In the livers of infected mice, the levels of IL-4, IL-5 and IL-10 increased obviously with a prolongation of the infection duration, IL-4 being the highest. However, in the bone marrow of infected mice, IL-4 level increased slowly with the duration of the infection, being lower than those in the liver at wk 10 and 12 after infection. The level of IL-5 was higher than that in the liver within 12 wk. At wk 10 after infection IL-10 tended to increase, but decreased thereafter and was obviously lower than that in the liver. In mice infected with Schistosoma japonicum, the immune response occurred mainly in the liver. IL-5 in the bone marrow showed auto-secretion.

[Effect of liposomal albendazole on the ultrastructure of Echinococcus granulosus cysts in mice].

AIM: To observe the histopathological changes of Echinococcus granulosus cysts in mice treated with liposomal albendazole and co-administration with cimetidine by light microscopy and electron microscopy. METHODS: An oral dose of liposomal ABZ with different formulations was given at 200 mg/kg.d. Cimetidine was administered daily at an oral dose of 100 mg/kg.d. Sixty-seven mice were orally given different drugs six days per week for a total of twelve weeks. RESULTS: The histopathological changes indicated that there were significant differences (P < 0.01) between treated groups and control group. The degeneration and necrosis of E. granulosus cysts were marked in liposomal albendazole combined with cimetidin group. CONCLUSION: Liposomal albendazole was more effective against E. granulosus cyst than albendazole. Cimetidine had an apparent synergistic effect when given in combination with liposomal albendazle.
