Activation of lymphocytes in the pulmonary inflammatory response to silica.

Lymphocytes were separated from the parenchymal lung tissue of mice administered either silica or titanium dioxide particles by intratracheal injection. There was an approximately 10-fold increase in the number of T-lymphocytes recovered from silica-treated animals at 2 weeks after injection. This was accompanied by an increase in the percentage of separated cells expressing the interleukin-2 receptor and by enhanced spontaneous DNA synthesis in serum-free culture. No such changes were demonstrable in cells obtained from animals administered titanium dioxide. These results confirm that an influx of T-lymphocytes occurs as part of the early pulmonary inflammatory response to silica particles. Furthermore, lymphocytes in the lung are functionally activated by at least two criteria. Activated T-lymphocytes may play a role in the induction of fibroblast proliferation and collagen synthesis that occurs in silicotic lung disease.

[The physiologic disposition and pharmacokinetics of guaiacol acetylsalicylate in rats].

Guaiacol acetylsalicylate (GASL) has been shown to possess significant anti-inflammatory, antipyretic and expectorant activities and has been used clinically in chronic bronchitis with fairly good results. In the present work, the absorption, distribution and excretion of metabolites of the drug were studied in rats using quantitative TLC scanning technique. GASL was shown to be unstable in rat gastrointestinal tract and guaiacol salicylate (GSLT) and salicylic acid (SLA) were found in plasma. The plasma SLA concentration-time curve obtained after oral administration of GASL to rats was shown to fit a one compartment open model with the following pharmacokinetic parameters: T1/2ka = 1.25 h, T1/2ke = 3.28 h, ka = 0.5554/h, ke = 0.2111/h, Tmax = 3.02 h, Cmax = 331.46 micrograms/ml, AUC = 2832.93 micrograms/ml.h The SLA concentration was found to be high in muscle, moderate in spleen, testicle, kidney, lung, plasma, heart and liver and low in brain. The GSLT concentration was found to be low in plasma and organs. Within 24 h following ig administration of GASL the total SLA excreted in urine and feces was 10.9 and 0.41 of the GASL dose, respectively.

[The evaluation of thyroid suppression test in predicting the outcome of Graves' disease treated by short-term antithyroid drugs].

One hundred and fifteen new patients with Graves' hyperthyroidism were received thyroid suppression test after cessation of 6 month course of drug therapy. The results of follow up for another 12 months were that one of them had lost, 48 cases were in remission, and 66 cases had relapsed. The present study shows that thyroid suppression test, including the 4th, 6th and 24th hour suppression rates, is useful in predicting the outcome of drug therapy. The effect of 6th hour suppression rate is better than that of 24th and 4th hours, with 75.8% sensitivity, 75.0% specificity and 75.4% accuracy when 30% of thyroid suppression rate is as out-off point which is decided by ROC curve. It is suggested that 6 hour 30% suppression rate is better than the traditional 24 hour 50% suppression rate in the prediction of outcome of drug therapy.

[Studies on man-cattle-man infection cycle of Sarcocystis hominis in Yunnan].

The present paper reports on the results of an experimental study on man-cattle-man infection of Sarcocystis hominis, found in Yunnan Province. About ten thousand sporocysts collected from the feces of persons naturally infected with Sarcocystis hominis were fed to a calf, which was dissected 150 days later. Numerous cysts of Sarcocystis hominis were found in the cardiac and skeletal muscles. By light microscopy, the cyst wall of fresh preparation showed numerous thick, finger-like projections, with maximum length of 7.9 microns. By electron microscopy, the cyst had a regularly folded, with primary wall forming palisade-like protrusions. Numerous sharp invaginations found in the protrusions were sawtooth-shaped, covering the whole surface of the protrusions. No fine fibrils were observed within the zone of ground substance beneath the primary cyst wall. Two rhesus monkeys were fed with beef infected with Sarcocystis hominis and sporocysts and oocysts were found in their feces 29 and 31 days later, the patent period of sporocyst excretion being 5 and 7 days, respectively. The senior author had taken voluntarily 60 g beef of the experimentally infected calf, and presented clinical symptoms such as anaemia, abdominal pain, diarrhoea, fatigue and dizziness on d3 post infection with sporocysts and oocysts found in the feces on d8. The patent period of sporocyst excretion was more than 42 days. The mean size of 50 sporocysts was 11.90 +/- 0.04 x 15.88 +/- 0.03 micron and that of 50 oocysts, 15.56 +/- 0.05 x 19.76 +/- 0.04 micron. On d50 he took acetylspiramycin tablets, the initial dose being 0.4 g, followed by 0.2g qid. for 15 days.(ABSTRACT TRUNCATED AT 250 WORDS)

[The effect of laser irradiation on arthritis in rats].

Based on the analgesic effect of laser therapy in clinic, the effect of laser irradiation at acupoints on the experimental arthritis in rats was further observed. The experiments were carried out on Wistar rats, which were divided into three groups: The normal group (n = 6): without any treatment. The laser group (n = 8): 24 hours after an injection of Freund's adjuvant, the typical symptoms of acute arthritis (red, swelling, pain and hyperalgesia) occurred. Then transcutaneous irradiation with a low power Helium-neon laser (3 mw, 6328 A) at ipsilateral Kun-Lun point was applied ten minutes every day for five days. The control group (n = 8): with the same treatment as the laser group except laser irradiation. All the indexes, such as foot volume, pain score and pain threshold (Heat-leg-withdrawal latency, HWL and electric-shock vocalization, VOC) were measured every day. Comparing with the symptoms in the control group, the intensity of pain as measured on pain score and the swelling of the ankles (foot volume) were significantly reduced by laser irradiation (P less than 0.01), but the pain threshold (HWL and VOC) did not show any improvement in laser group. However, immediately after laser irradiation, the average pain threshold (VOC) was significantly raised (P less than 0.01). It is concluded that low power laser irradiation at local points can produce the relief from arthralgia and reduction in the swelling of ankles, and it can also produce instant analgesic effect in test of pain threshold (VOC).

P210 BCR-ABL is complexed to P160 BCR and ph-P53 proteins in K562 cells.

The BCR gene (Groffen et al., 1984) plays a critical role in the pathogenesis of human leukemias that involve the Philadelphia chromosome (Ph1) (Rowley, 1973; Nowell & Hungerford, 1960). Cells containing the Ph1 contain a chimeric gene formed from the fusion of BCR (Collins et al., 1987; Lifshitz et al. 1988) and ABL genes that results from the reciprocal translocation of segments of chromosomes 9 and 22 (Shtivelman et al., 1985). The product of this chimera is a 210 kDa protein, termed P210 BCR-ABL, that possesses an activated tyrosine kinase activity (Konopka et al., 1984; Kloetzer et al., 1985). Studies using long-term marrow culture systems and retrovirus-mediated gene transfer have documented that P210 BCR-ABL can stimulate the growth of immature hematopoietic precursor cell types (McLaughlin et al., 1987; Young & Witte, 1984). We have previously reported that P210 BCR-ABL exists in cytoplasmic complexes in association with a 53 kDa protein termed ph-P53 (Maxwell et al., 1987; Li et al. 1988). Similarly, BCR proteins have been found in cytoplasmic complexes containing ph-P53 in cells lacking the Ph1 (Li et al., 1989). These BCR protein complexes possess an associated ser/thr protein kinase activity. In this same study, we found that P210-containing complexes phosphorylate BCR proteins on tyrosine residues in vitro (Li et al., 1989). We now present results which demonstrate that P210 BCR-ABL is tightly associated with P160 BCR and ph-P53 proteins in cytoplasmic complexes from cells containing the Ph1.

[Effect of qigong on sympathetico-adrenomedullary function in patients with liver yang exuberance hypertension].

By using the differential diagnosis of traditional Chinese medicine to determine the types of hypertension, using the diagnosis of western medicine (WM) to determine the phases of hypertension, 61 inpatients of Liver Yang exuberance type hypertension were randomly divided into Qigong group and WM group. The patients in the Qigong group were treated with both Qigong and antihypertensive drugs at low dosage, but those in the WM group were treated with the drugs alone. Several laboratory tests concerning sympathetico-adrenomedullary functions were conducted twice respectively at 1st and 9th week after hospitalization of the patients. The results indicated that the Qigong group after treatment of 9 weeks had more cases with normal sympathetico-adrenomedullary functions than it had before the treatment, and that their urinary CA, E, NE decreased, MHPG-SO4 increased, plasma cAMP and cGMP got down, but cAMP/cGMP ratio got up. It suggested that Qigong could modulate the sympathetico-adrenomedullary functions of patients with Liver Yang exuberance type hypertension.

[Iatrogenic central artery embolism].

This paper presented 3 cases of central retinal artery embolism (CRAE) caused by the drug injection at the head and neck. In two of the three cases, the emboli were formed by the suspended drug which entered CRA resulting in the occlusion. Another case was caused by the large amount of meglucamini diatrizoas which was injected too quickly during angiography. The CRA was filled up with fluorescein in a very short time, which lead to ischemia and hypoxia of the retina. This paper stressed the importance of early diagnosis and prompt treatment. The authors also Discussed the method of drug injection at the head and neck.

[Study on the fluorimetric determination of aluminum with H2QS and CTMAB in water].

A study on the fluorimetric reaction of Al-H2QS-CTMAB and conditions of the determination of aluminium in water by model 930 fluorimeter is reported. A good linear relationship between the strength of fluorescence and aluminum concentrations, ranging from 0 to 1.5 micrograms/10ml, is observed. Interferences from other constituents in water can be eliminated by the masking effect of 1, 10-phenanthroline and hydroxylamine hydrochloride and by using standard addition method. The coefficient of variation and recovery vary from 2.9% to 8.9% and from 87% to 105%, respectively. The detection limit is 3 x 10(-2) micrograms/ml. This method is rapid, sensitive, accurate and suitable for the determination of contents of aluminum in drinking and natural waters.
