A novel peptide hormone, atrial natriuretic factor/cardiodilatin (ANP/CDD), was recently isolated and characterized from mammalian heart. Its presence has been demonstrated in several organs that contribute to water and sodium homeostasis, such as salivary glands. This study demonstrates the presence of ANP/CDD immunoreactivity in the salt gland of Pekin ducks by high performance liquid chromatography, radioimmunoassay and immunocytochemistry, using a specific antibody against atriopeptide I. A small number of distinct, ovoid or cuboid shaped ANP/CDD-immunoreactive cells were localized in the connective tissue surrounding and separating the central secretory tubules, whereas no immunostaining was observed in the peripheral tubules. Salt glands of ducks that were adapted to salt water revealed a significant hypertrophy of their secretory lobules. However, no differences were found between the number or localization of immunoreactive cells in the salt gland of salt water-acclimatized ducks and nonstimulated glands of ducks that were housed with ad libitum access to fresh water. Our results indicate that ANP/CDD may play a role in the regulation of sodium secretion in the salt gland of aquatic birds.
Trophoblastic cells with their strictly controlled propensity for invasive growth hereby bear limited resemblance to malignant cells. Therefore, detailed description of molecular characteristics of this cell type in comparison to histologically related tumor cells may aid in defining physiologically relevant differences. In view of the potential importance of selective protein-carbohydrate interactions in recognitive processes, labelled neoglycoproteins were employed to evaluate systematically the presence and distribution of sugar receptors in tumor cells of chorionepithelioma and in trophoblastic cells of an early stage of gestation. Control reactions in glycohistochemistry, involving prolonged incubation, pH variation and use of nucleotides excluded the possibility that glycosidases or glycosyltransferases govern the sugar-specific binding to the tissue sections. Pronounced differences were detected in the expression of receptors (lectins) for alpha- and beta-glucosides as well as for N-acetylgalactosamine and N-acetylglucosamine. Further differences were revealed by probing both types of tissue with lactosylated, fucosylated, xylosylated and sialylated carrier protein. Upon developmental maturation of the normal cells in placenta the extent of binding of the neoglycoproteins decreased. The glycohistochemical differences between malignant and normally developing trophoblastic cells were found to be reflected in the alteration of expression of certain endogenous lectins, as ascertained by affinity chromatography and gel electrophoretic analysis. Consequently, we assume that the transient presence of certain endogenous lectins during early stages of gestation and their differential expression in relation to tumor cells of chorionepithelioma may be relevant for the special invasive and adhesive behavior of human trophoblastic cells.
The hepatoprotection induced by synthetic muramyl peptides was investigated using a model of lethal murine mouse hepatitis MHV-3 virus infection. MDP and a nonpyrogenic analog, Murametide, inhibited the steep elevation of serum transaminases induced by MHV-3 irrespective of whether the immunomodulators were administered before or after the infection. A significant proportion of MDP or Murametide-treated animals, in contrast to controls, survived the MHV-3 infection. The histopathological examination of the liver revealed marked necrosis of the hepatic parenchymal cells and infiltration of the inflammatory cells in controls but not in MDP-treated animals.
The influence of postpartal blood taking on health and growing of calves and the use of neonatal calf serum (NCS) for cultivation of mice embryos was investigated in 3 experiments with 134 calves. The results indicate, that the neonatal blood taking (max. 2 h p.p.) is without negative effects on health and growing of calves. It is possible to use the NCS like fetal calf serum (FCS) as additive in cultivation mediums for mice embryos. The utilization of NCS for cultivation of bovine embryos and for other laboratory purposes should be tested.
A technique of high-performance thin-layer chromatography (HPTLC plates) by which to identify the anabolic substance of trenbolone acetate (TBA) and its metabolite trenbolone is described in this paper. Plasma, bile, urine, faeces, liver, and meat can be used for testing. The sensitivity of the method is 1 ng. Additional reliability criteria are enumerated. Semiquantitative TBA determination has proved to be suitable in random sampling for residual analysis.
The presence and location of specific endogenous lectins in the anterior epithelia of the porcine eye were determined by histochemical staining using biotinylated glycosylated carrier molecules. Endogenous lectins with specificities for galactosides, alpha-mannosides and beta-xylosides were present in the conjunctival epithelium, but the corneal anterior epithelium appeared essentially free of histochemically demonstrable endogenous lectins. The corneal posterior epithelium contained a rich and complex set of endogenous lectins. The transitional area between conjunctival and corneal anterior epithelia at the limbus cornea contained, in striking contrast to either of the epithelia bordering it, cells expressing endogenous lectins in considerable wealth and variety.
For about 100 years retrobulbar injection of a local anesthetic has been regarded as being a safe, efficient anesthesia in cataract surgery. Today one of the most difficult steps in cataract surgery is the implantation of the IOL. Vis à tergo can lead to severe problems during this procedure. Therefore, the position of the iris (concave or convex) is an important result that depends on the quality of the block. Motor blockade and analgesia were also examined. The first investigation was done to answer the question of whether or not better results could be obtained by using a mixture of bupivacaine (0.75%) and mepicacaine (2%) or with bupivacaine alone. Injections were done observer-blind in all cases by the same operator. Pure bupivacaine provided the best anesthesia results. A second examination was done in the same way to measure the effects of the addition of hyaluronidase; motor blockade, analgesia und hypotonia of the globe were found to be significantly better with the addition of 75 IU of hyaluronidase.

Trehalose dimycolate extracted from mycobacteria is a potent immunomodulator. Incorporation of trehalose dimycolate in a squalane-in-water emulsion leads predominantly to the formation of vesicular structures, which are observable by electron microscopy. The interaction between vesicles of trehalose dimycolate and the immunocompetent cells results in an enhancement of the host defence mechanisms and induction of non-specific resistance against viral, parasitic, and bacterial pathogens and certain tumors. A brief review of the pertinent observations is presented.

Histochemical detection of binding sites for sulfated polysaccharides believed to be important mediators within recognitive interactions was carried out by application of biotinylated probes such as heparin, native and desulfated fucoidan, dermatan sulfate, and two types of carrageenans. The probes were derivatized by mild cyanogen bromide activation and subsequent aminoalkylation to allow incorporation of biotin, inserted with an epsilon-aminocaproic acid spacer to reduce charge-related and steric impediments. Specific labeling could be detected in different cell types of human placenta, dependent on the developmental stage. Sulfated polysaccharides bound predominantly to leucocytes in full-term placenta, whereas demonstration of specific binding sites in decidua, syncytiotrophoblasts, and cytotrophoblasts was restricted primarily to heparin and, less intensely, fucoidan, although not desulfated fucoidan. Heparin binding in the placenta after 8 weeks of gestation was reduced for epithelia that, at this stage of development, revealed carrageenan binding sites. Fucoidan binding was at this developmental stage measurable only for leucocytes. These results provide definite histochemical evidence for the presence and developmental regulation of expression of receptors for sulfated polysaccharides in different cell types of human placenta.
Mice were administered nontoxic monophosphoryl lipid A (MPL) or refined standard bacterial endotoxin (RSE) prior to, simultaneously with, or after infection with Toxoplasma gondii. MPL and RSE given before or with the toxoplasma infection induced significant resistance. Administration of toxic RSE, but not nontoxic MPL, after the establishment of toxoplasma infection significantly shortened the survival time of mice.
Twelve patients suffering from haemorrhages from a superficial exulceration (Dieulafoy's disease) were treated on an emergency basis during the period from 1981 to 1987. Preoperative diagnosis by means of gastroscopy was possible in 3 cases only. The sites of predilection within the cardia and near to it, except the curvatures, determined the surgical access by means of high gastrotomy-independent of a verification of the diagnosis. In 11 cases the therapy consisted of the purse-string ligature, in one case endoscopic sclerosing was successful. The mortality rate was 16.6%.

The effect on natural killer (NK) cytotoxicity of splenic cells from BALB/c mice pretreated i.v. with squalane-in-water preparations of muramyl dipeptide (MDP), trehalose dimycolate (TDM), or the combination of MDP-plus-TDM was investigated. MDP or TDM augmented the NK cytotoxicity which peaked 48 h after the pretreatment whereas the combination of MDP and TDM induced an inhibition of the NK activity. Infection with influenza virus, a potent stimulator of NK cells, after the pretreatment with biological response modifiers resulted in a markedly enhanced NK activity on day 2 in MDP and control groups. Mice pretreated with TDM or the combination of MDP and TDM showed only moderate NK activity which peaked on day 3 after influenza infection. The NK activity was susceptible to asialo GM1 and complement treatment. The cytotoxicity of MDP-plus-TDM cells could be significantly enhanced after treatment with anti-macrophage monoclonal antibody and complement. NK activity induced by MDP or TDM was reduced by mixing MDP-plus-TDM cells. Addition of adherent cell-depleted MDP-plus-TDM suspension to MDP or TDM cells had a NK restorative effect. Splenic cells from mice pretreated 2 days earlier with MDP or TDM, but not MDP-plus-TDM, generated enhanced levels of luminol-dependent chemiluminescence.
