[Toxicology of carteolol].

Studies of the acute toxicity of 5-(3-tert-butylamino-2-hydroxy-propoxy)-3, 4-dihydro-2(1H)-quinolinone hydrochloride (carteolol hydrochloride, Endak, Endak mite) in various species by different modes of administration revealed comparatively slight necropsy and histological changes despite characteristic clinical features of poisoning, There were only minor discrepancies in LD50 between the different species, and no sex differences were observed. The LD50 was 2000-4000 times the therapeutic dose. Death is thought to be due to excessive enhancement of the pharmacological action of the sympathomimetic component with extreme abnormalities of blood distribution and interference with cardiac and lung function. In subchronic and chronic toxicity tests in rats no effects due to the drug were demonstrable for doses of up to 150 mg/kg. In dogs the highest dose free from side-effects was between 3 and 30 mg/kg. Above 10 times the therapeutic dose there was some increase in brown adipose tissue, but this is not thought to be of any pathological significance. No teratogenic effects were detected in experiments on mice, rats and rabbits, and no embryotoxic or fetotoxic activity was seen except for doses high enough to produce toxic effects in the mother animals. No carcinogenic properties were identified. Tests for mutagenic effects (S. typhimurium, E. coli, cytogenetic studies in rats, dominant lethal test) yielded negative results.

Mouse spleen cells are sensitized sheep red blood cells: an Fc-rosette-forming system allowing the detection of "activated" Fc structures.

From a variety of Fc receptor-bearing cell/sensitized red blood cell combinations, mouse spleen cells, and sensitized SRBC were selected as an Fc-specific EA rosette assay system because only this mixture combined a high percentage (about 50%) of rosette-forming cells with complete absence of spontaneous rosettes and showed no influence of complement on the rosette formation. From studies on the minimal structural requirement of IgG both for mediation and inhibition of EA rosettes using IgG and several well-defined fragments, it appeared that both the CH2 and the CH3 domain of Fc are needed for optimal interaction with the lymphocyte Fc receptor. Finally, it was demonstrated that the assay system is able to detect "activated" Fc structures (here: heat-aggregated IgG) and to differentiate between varying amounts of such structures.

Improvement of hyperlipidaemia by bezafibrate treatment in RDT patients.

The pharmacokinetics and therapeutic effects of bezafibrate were studied in 15 RDT patients in a placebo controlled trial. Serum half life of bezafibrate was prolonged to 17--21.5 hours compared to 1.6--2.1 hours in normals. Adequate dosage in RDT patients was found to be 200mg every 3rd day. Bezafibrate treatment resulted in significant decrease in the serum concentrations of triglycerides, total cholesterol and LDL-cholesterol, whereas HDL-cholesterol serum levels increased. Under this dosage regimen no adverse side effects were observed. Bezafibrate offers the possibility of correcting disturbances of lipid metabolism of RDT patients, possibly involved in the pathogenesis of atherosclerosis of these patients.

Praziquantel in clonorchiasis and opisthorchiasis.

A single stool examination revealed pathogenic intestinal parasites in 462 (58%) of 796 vietnamese and cambodian refugees. 56 (7.0%) were infected with Clonorchis sinensis and/or Opisthorchis viverrini. These patients received Praziquantel in a dosage of 20 mg/kg bwt. p.day on 3 consecutive days. Parasitological controls were completed after 12 months. No further excretion of eggs could be detected in 88% of the patients. Concurrent infections with other trematodes and cestodes were also cured. Nematode infections remained uninfluenced. No change of haematological and biochemical parameters could be observed during therapy. Diarrhea and epigastric pain were common side effects, which are probably not effects of the drug itself. They rather seem to be due to the release of parasitic antigens. This is also indicated by a further increase of circulating Ig E after therapy.

Proliferation kinetics of malignant non-Hodgkin's lymphomas related to histopathology of lymph node biopsies.

Since lymphomas of high and low malignancy have been differentiated by the Kiel-classification, the cell kinetics of malignant lymphomas have become an interesting subject. In 20 non-Hodgkin's lymphomas we have determined the initial labelling index, the mitosis index, the proliferation rate and the durations of DNA-synthesis and mitosis. The kinetic variables have been estimated by using an in vitro-incubation of fresh biopsies and double-labelling with radioactive DNA-precursors. in 8 lymphomas of low malignancy we found an initial labelling index of 7.8%, a mitosis index of 0.3% and a potential tumour doubling time of 100.2 h for a cell production rate without regard to cell loss. On the other hand, lymphomas of high malignancy showed a labelling index of 16.7%, a mitosis index of 0.7% and a potential tumour doubling time of 40.5 h. All these values could be differentiated with high statistical confidence. DNA-synthesis time and mitosis time of both lymphoma groups did not show any significant differences. From these findings we obtained confirmation of the cyto-morphological principles used in classifying lymphomas.
