Effect of gentamicin on growth of viral, chlamydial, and rickettsial agents in mice and embryonated eggs.

Gentamicin, at concentrations up to 500 mug/ml, showed no effect on the replication, yield, or infectivity of seven viruses or on the agents of psittacosis and meningopneumonitis when grown in mice or embryonated eggs. At the 500 mug/ml level, lymphogranuloma venereum cultures had a slight reduction in infectivity. Rickettsia akari demonstrated susceptibility at the 5 mug/ml level, whereas R. rickettsii, R. mooseri, and R. canada grown in embryonated eggs were susceptible in varying degrees to gentamicin at or above the 50 mug/ml concentration.

KINETICS OF THE ACTION OF AMPICILLIN ON ESCHERICHIA COLI.

Bacteriol. 85:1160-1164. 1963.-The curve of the number of viable Escherichia coli after exposure to ampicillin can be divided into three phases: a lag phase, a rapid bactericidal phase, and a slow bactericidal phase. Some of the variables affecting the magnitude of the first two of these phases were investigated. Progressive lowering of drug concentration resulted in prolongation of the lag phase and decrease in slope and extent of the rapid bactericidal phase. The production of elongated gram-negative forms and the emergence of a mutant with increased penicillinase activity complicated interpretation of the lower dose curves. With sufficient drug concentration, the length of the lag phase and the slope of the rapid bactericidal curve were independent of the size of inoculum up to 10(8) organisms. Varying pH revealed that maximal activity, as measured by the shortest lag phase and steepest slope of the rapid bactericidal phase, was present at slightly acid pH levels. Increasing pH resulted principally in prolongation of lag phase. With greater acidity, decrease in slope of the rapid bactericidal phase was more prominent. Cultures studied under conditions of lessened metabolic activity exhibited prolonged lag phase and decreased slope and extent of rapid bactericidal phase.

Antibiotic therapy of abscess of the lung and bronchiectasis.

Since the fusospirochetal group of bacteria are the commonest etiologic agents in abscess of the lung, aqueous crystalline penicillin is the agent of first choice in the majority of cases. Streptomycin is indicated for a small group of cases in which Klebsiella is the etiologic agent. Aureomycin, chloramphenicol or terramycin may produce an excellent therapeutic response either initially or after therapeutic failure with penicillin. Administration of antibiotics by inhalation should be carried out in conjunction with systemic forms of treatment. In the treatment of bronchiectasis, the antibiotics are most useful in the control of acute exacerbations of pulmonary infection which punctuate the course of this disease.
