A genetic linkage map of five marker loci in and around the Duchenne muscular dystrophy locus.

Linkage analysis of five marker loci in and around the Duchenne muscular dystrophy (DMD) locus, DXS84, DXS206, DXS164, DXS270, and DXS28, was conducted with 499 families. Overall, the best multipoint distances were found to be DXS84-3.7 +/- 0.6 cM-DXS206-1.0 +/- 0.4 cM-DXS164-1.9 +/- 0.6 cM-DXS270-12.0 +/- 1.1 cM-DXS28. A comparison of this linkage map with the established physical map suggests the presence of hot spots for recombination in the DMD locus.

X-chromosomal DNA polymorphisms in two ethnic groups from India.

Four polymorphic sites of the short arm of the X chromosome were studied in two racial groups from India, the Assamese and the Malayalee. Since the allelic frequencies of the two groups did not differ markedly from each other, the data from the two populations were pooled. The frequency of the A2 allele was 0.57 for the L1.28 probe, 0.20 for the RC8 probe, 0.28 for the pD2 probe and 0.11 for the L754 probe. The A3 allelic fragment of the RC8 probe was not found among 67 Indians, and in one Assamese woman an additional 7.0-kilobase fragment was found. The differences between the Indian population and other ethnic groups were analyzed.

Linkage studies in a large fragile X family.

We have analyzed the segregation of five loci in the region Xq27/28 in a large family affected by the fragile X syndrome. The marker DXS115 (767) is shown to be polymorphic with the enzyme PstI, as well as with BstXI. This marker will be useful in the analysis of both fragile X and haemophilia A families. The data presented here are consistent with the following order of loci: Xcen-F9-DXS105(cX55.7,55E)-DXS98(4D-8)- FRAXA-DXS52(St14)-DXS115(767)-qter.
