Twenty fresh Giardia intestinalis strains were isolated by inoculation of trophozoites obtained by in vitro excystation procedure or by duodenal aspiration. The strains were continuously cultivated in TP-S-1 or bile-supplemented BI-S-33 media without antibiotics. No correlation was found between the ease of isolation and axenization of Giardia strains and the source of material for inoculum preparation (faeces v. duodenal fluid and symptomatic patients v. asymptomatic cyst passers). Panmede and Trypticase used in TP-S-1 and TYI-S-33 media vary in their ability to support the growth of particular strains. Of the five sera used in TP-S-1 medium no variations were associated with bovine and calf serum. The bile-supplemented BI-S-33 medium supported the growth more consistently than did the TP-S-1 and TYI-S-33 media; depending on the strain, the mean number of trophozoites at 120 hours was 1.1 X 10(6) to 1.8 X 10(6), and the generation times were 8.1 to 16.6 hours.

Anti-Giardia antibodies were studied in 88 sera of patients with symptomatic and asymptomatic giardiasis by using the indirect immunofluorescent test and Giardia intestinalis trophozoites from an axenic culture as antigen. The sera of 40 healthy subjects were used as a control group and of 28 patients with other intestinal parasites (14 with Sarcocystis sp. and 14 with symptomatic Entamoeba histolytica infection) have been investigated for determination of test specificity. 82% of sera from patients with giardiasis presented G. intestinalis antibodies at titers of 1:16 to 1:256. Titers of at least 1:32, accepted as diagnostic and indicative of the infection, were found in 66% of patients with giardiasis. All of 18 patients with acute course of giardiasis had titers of at least 1:32. Only 5 sera (7%) from healthy control subjects or from patients infected with other intestinal parasites reacted at dilutions of 1: greater than or equal to 32. The indirect immunofluorescence test proves to be useful in detecting antibodies to G. intestinalis in the serum. However, the negative results occurring rather frequently in persons with chronic infection indicate a need for a more critical evaluation of serological tests in giardiasis.

The aim of the investigation was to determine the relationship between eosin exclusion, in vitro excystation and experimental infection, as determinants of Giardia sp. cyst viability. The results of experiments clearly show that the excystation induced in vitro, being a more sensitive indicator of cyst viability than is eosin exclusion, is, at the same time, technically more complicated and gives in some cases different and subjective results which are affected by the technique itself. The results of objective experimental infection of rats showed that there is no relation between percentage excystation in vitro, eosin exclusion and cyst infectivity. One may assume that in vitro excystation methods are not always objective determinants of Giardia cysts viability. However, the high excystation rate obtained exceptionally by Schaefer and co-workers showed the possibility for elaboration of an objective excystation procedure in vitro as a simple method for Giardia cyst viability determination.
