Dual excitatory and inhibitory effects of opioids on intracellular calcium in neuroblastoma x glioma hybrid NG108-15 cells.

The intracellular free calcium concentration ([Ca2+]i) was measured in single NG108-15 cells using indo-1-based microfluorimetry. In cells differentiated for 6-14 days in serum-free, forskolin (5 microM)-supplemented medium, application of micromolar concentrations of [D-Ala2,D-Leu5]-enkephalin (DADLE) inhibited Ca2+ influx mediated by voltage-gated Ca2+ channels. DADLE, at concentrations ranging from 1 nM to 1 microM, also produced rapid transient increases in [Ca2+]i (EC50 = 10 nM). The [Ca2+]i increases elicited by DADLE did not correlate with the inhibitory effects of the peptide. DADLE-induced [Ca2+]i increases were blocked by naloxone. In single cells, sequential application of selective opioid agonists (30 nM) evoked responses of the rank order DADLE = [D-Pen2,D-Pen5]-enkephalin > (trans)-(+-)-3,4-dichloro-N-methyl-N-(2-[1-pyrrolidinyl]cyclohexyl) benzeneacetamide > [D-Ala2,N-Me-Phe4,Gly5-ol]-enkephalin, consistent with activation of a delta-opioid receptor. The response was completely blocked by removal of extracellular Ca2+ or application of 1 microM nitrendipine, indicating that the increase in [Ca2+]i results from Ca2+ influx via dihydropyridine-sensitive, voltage-gated Ca2+ channels. Substitution of N-methyl-D-glucamine for extracellular Na+ or application of 1 microM tetrodotoxin greatly reduced, and in some cases blocked, the DADLE-induced [Ca2+]i increase, consistent with amplification of the response by voltage-gated Na+ channels. The [Ca2+]i increase was mimicked by both dibutyryl-cAMP and phorbol 12,13-dibutyrate. These findings are consistent with a delta-opioid-induced depolarization, possibly mediated by a second messenger, that subsequently recruits voltage-sensitive Ca2+ channels. In contrast to differentiated cells, undifferentiated cells responded to DADLE with a modest [Ca2+]i increase that was not sensitive to nitrendipine. In these cells, activation of the same second messenger system may elevate [Ca2+]i by mobilization from intracellular stores rather than influx. In addition to previously described inhibitory coupling to adenylyl cyclase and Ca2+ channels in NG108-15 cells, these results suggest that a novel, excitatory, effector system may also couple to opioid receptors.

Cloning and expression of midecamycin 4"-acylase gene in spiramycin producing strains.

A recombinant plasmid p66B containing the midecamycin 4"-acylase gene was obtained by cloning this gene into plasmid vetor pIJ680 from the primary clone pCN6C5, presumably harboring the midecamycin biosynthetic gene. The expression of the midecamycin 4"-acylase gene (p66B) in spiramycin producing strains resulted mainly in the production of 4"-isovalerylspiramycin. Another positive clone pCN10F5 was discovered from the genomic library of S. mycarofacians 1748 by probing with p66B DNA BamHI-BamHI 2.3kb fragment. A BamHI-BamHI 8.0kb homologous region on pCN10F5 was determined by Southern hybridization and was subcloned into plasmids pWHM3 and pIJ680. Recombinant plasmids pWF5 and p6F5 with molecular size about 15.2kb and 13.3kb, respectively, were obtained. Transformation of spiramycin producing strains with these plasmids resulted in the production of two major components. Based on their physicochemical properties and spectral evidences, component I was identified as 4"-propionylspiramycin III, and component II as 4"-propionylspiramycin II. Southern hybridization confirmed that the BamHI-BamHI 8.0kb fragment was cloned in the spiramycin producing strain. Only pCN10F5 clone was identified from the genomic library of S. mycarofaciens 1748 when the 4"-isovaleryltransferase gene of carbomycin producing strain S. thermotolerans was used as a probe in colony hybridization. It suggests that there is a difference between the 4"-acyltransferase genes in the pCN6C5 and pCN10F5 clones.

[Isolation and characterization of mutactimycin-producing mutant].

Natural non-antibiotic producing Streptomyces sp. 1254 was mutagenized by UV irradiation and two active mutants were isolated. Mutant 113 produced novel anthracycline compounds designated mutactimycins. Mutactimycin A was active against the bacteriophage of Bac. subtilis and some viruses in tissue culture. The mutant 2-6 synthesized a basic water-soluble antimicrobial antibiotic. Chemical analysis of the whole cell hydrolysate and the morphological characterization showed that the strain 1254 and its mutant 2-6 were of chemotype I, belonging to the genus of Streptomyces, and the mutant 113 was of chemotype IV without mycolic acid. Co-synthesis test of strain 1254 and a blocked mutant of strain 113 gave the active compounds identical with mutactimycins. Using the actI gene as a probe, the Southern hybridization revealed homology between the actinorhodin polyketide biosynthase gene and the total DNA of the strain 1254. Based on these data it was deduced that Streptomyces sp. 1254 should have a biosynthesis pathway for mutactimycin, but some of its genes might fail in expression and mutagenesis would make the silent gene(s) active.

A study of ventricular contraction sequence in complete right bundle branch block by phase analysis.

Twenty-four patients with complete right bundle branch block (CRBBB) combined with and without left axis deviation (LAD) on ECG, were compared with 17 normal subjects to evaluate the right ventricular contraction sequence and pattern in detail. Blood pool scintigrams were obtained in the left anterior oblique projection, and these images were analyzed by first component Fourier harmonics. In the normal subjects, the phase value distribution representing the pattern of ventricular contraction was almost homogeneous in both the right and left ventricles (RV & LV). In the CRBBB patients without LAD, the phase images showed apparent phase delay in the right ventricle. In the CRBBB patients with LAD, the phase images showed many different contraction patterns varying from normal to RV phase delay, owing to the effects of the hemi-block. Quantitative analysis of the absolute values, showed that the mean (RV-LV) value was 6.6 +/- 8.4 msec in the normal subjects. In the CRBBB patients without LAD, the duration of the QRS complex correlated with the mean (RV-LV) value, whereas no difference was observed between the duration of the QRS complex and the standard deviation of the right ventricle. Using phase analysis the degree of the RBBB can be determined from the phase images, and can be quantitatively analyzed as in electrical studies.

[An observation of protective effect of acupuncture on the gastric mucosa of Wistar rats and the relative histochemical changes of the neurotransmitters].

In this paper, changes of neurotransmitters and the protective effect of acupuncture on gastric mucosa have been studied by the histochemical methods of cholinesterase and catecholamine on 30 pairs of Wistar rats. 1. Sequential medical trials on rats of water-immersion induced gastric ulceration show that there was a protective effect of electro-acupuncture on gastric mucosa and it was statistically significant. 2. Under the protective effect of acupuncture on gastric mucosa, finally, both the cholinergic and adrenergic nerves undergo significant inhibition.

The fluid mechanics of a sac-type ventricular assist device.

Flow visualisation and velocity measurement studies have been carried out on the liquid side of a sac-type VAD. The objectives have been to identify both good (e.g. short residence times) and bad (e.g. high turbulence levels) flow characteristics. The flow was visualised using polystyrene micro-spheres illuminated with white light on selected transverse planes across the pumping chamber and its inlet and outlet connections. These results then served as a guide for velocity measurements using laser-Doppler anemometry. Flow tests were run using water or a water/glycerol mixture. Operating conditions were determined from the appropriate fluid mechanics modelling laws to ensure dynamical similarity. Initially, steady flow behaviour was investigated including the effects of removal and insertion of the tilting-disc inlet and outlet valves. In subsequent full pumping tests, using a pneumatic driver, regions of high shear, separation, and high turbulence intensity have been identified.

[Studies on antifertility constituents of Siegesbeckia glabrescens Mak].

Three diterpenes including a new compound were isolated from the whole plant of Siegesbeckia glabrescens Mak. The new one was named as neodarutoside and its structure was elucidated as ent-3 alpha, 15,16-trihydroxy pimarane 3,15-bis-(beta-glucopyranoside) based on the spectral evidence and chemical transformation. The other two were darutigenol and darutoside. Darutoside possesses the activity of termination of early pregnancy in experimental rats at a dosage of 20-40 mg/kg.
