Expression of the TIGR gene in the iris, ciliary body, and trabecular meshwork of the human eye.

Mutations in the the glaucoma gene GCL1A, also known as trabecular meshwork glucocorticoid response (TIGR) or myocilin (Myoc), have been shown to be associated with juvenile-onset primary open-angle glaucoma. Very little is known about the pattern of expression of the TIGR gene in human ocular tissues. In-situ hybridization experiments demonstrated the localization of TIGR mRNA in cells throughout the iris, ciliary muscle, and the filtering portion of the trabecular meshwork of normal eye donors. The expression of TIGR protein was investigated by Western blot using an epitope-directed antibody to the carboxy terminus region of TIGR. This antibody was able to distinguish a recombinant TIGR fusion protein from a truncated TIGR form containing the naturally occurring Gln(368)-->stop mutation. In tissue extracts from the iris, ciliary body, and trabecular meshwork, the antibody recognized a major protein band of 57-kDa molecular mass. Deglycosylation treatment with PNGase F, NANase II, and O-glycosidase indicated that the 57-kDa protein in these tissues was unglycosylated. In agreement with this observation, in coupled in-vitro transcription/translation systems, the 57-kDa TIGR protein was unaffected by the presence of the processing and glycosylation activities of canine pancreatic microsomal membranes. These findings support the view that the expression of TIGR mRNA in cells of the iris, ciliary body, and trabecular meshwork correlates with that of TIGR protein, and that the 57-kDa TIGR protein was unglycosylated. These results, which are in contrast with earlier reports, raise the possibility that the TIGR protein might be processed into distinct forms in a tissue-specific manner.

Rat hepatocellular apoptosis induced by glycodeoxycholate.

OBJECTIVE: To explore the relationship between glycodeoxycholate (GDC) and rat hepatocellular apoptosis. METHODS: GDC was used to treat rat hepatocytes cultured in vitro and the apoptotic cells were observed and analyzed with light microscope, transmission electron microscope, TUNEL in situ hybridization, DNA agarose gel electrophoresis and flow cytometry. RESULTS: When rat hepatocytes were incubated with GDC (final concentration of 100 micromol/L) for 2h, apoptotic hepatocytes were observed with light and transmission electron microscope, and TUNEL in situ hybridization. When the hepatocytes were incubated with GDC (final concentration of 50, 100, 150 micromol/L, respectively) for 2h, or with GDC (final concentration of 100 micromol/L) for 2, 4, 6, 8h, respectively, agarose gel electrophoresis of the hepatocytes DNA demonstrated the typical ladder patterns. CONCLUSION: GDC with final concentration of 50, 100, 150 micromol/L could induce rat hepatocellular apoptosis.

[A long-term study of a small-caliber arterial autograft used for repair of a large-caliber arterial defect].

OBJECTIVE: The long-term fate of a small arterial graft used to repair a large diameter artery was studied in this study. METHODS: An end segment of the tail artery (0.3-0.4 mm in diameter) was transplanted to the defect of the carotid artery (1.0-1.4 mm in diameter) in 50 rats. The vessel grafts were examined histologically at 3 weeks and 5, 10 and 12 months after implantation. RESULTS: At 3 weeks, intimal thickening of the grafts was mild and present only in regions adjacent to the anastomosis. At 5 months, severe intimal thickening was seen along the entire graft wall. At 10 and 12 months, the intimal thickening reduced. Regression of the thickening was uneven within the graft wall. The adventitia thickened at the early postoperative period and reduced at the end of this study. The patency rate of the grafts at 12 months postoperation was 95% (19/20). Angiography indicated that the grafts stopped expanding after 5 months and were only slightly smaller than the recipient arteries at 12 months postoperation. CONCLUSION: These results suggest that a small, autogenous arterial graft may effectively repair a large-calibre arterial defect.

[Experimental study on revascularization of subdermal vascular network thin flap].

OBJECTIVE: To investigate the survival mechanism of subdermal vascular network thin flap (SVNF). METHODS: 24 rabbits were divided into three groups, in which the SVNF, subdermal vascular network skin graft and the traditional random flap were created respectively. Tissue hyalinization technique was used to find out the differences of vascular architecture between the three groups at the 1st-7th day, and the 14th day postoperatively. RESULTS: The results showed that the blood supply reestablished earlier in SVNF group than the other two groups. CONCLUSION: There is a zone of rapid and intensive angiogenesis in the mid-part of SVNF known as "intermediary pedicle". This is the anatomic basis of the early pedicle division and superior survival ratio of SVNF.

Effect of Q-switched alexandrite laser irradiation on dermal melanocytes of nevus of Ota.

OBJECTIVE: To investigate the effect of Q-switched alexandrite laser irradiation on dermal melanocytes of nevus of Ota. METHODS: Multiple biopsies were carried out on 4 patients with nevus of Ota before and after laser irradiation. Altogether 11 samples were examined under light microscope and 14 under transmission electron microscope. RESULTS: Immediately after laser irradiation, the dermal melanocytes were destroyed, the melanosomes were degenerated with central vesicle formation within most of them, and intradermal round vacuoles appeared. The epidermis remained intact. Three months to 1 year after irradiation, the degenerated melanosomes and cell debris were scavenged mainly by macrophages. Dermal melanocytes gradually decreased. No fibrosis was found. CONCLUSION: Q-switched alexandrite laser can selectively destroy dermal melanocytes of nevus of Ota and treat the disease safely.

Arg485Lys polymorphism of factor V increases the risk of coronary artery disease in a Chinese population.

OBJECTIVE: To explore the relationship between genetic variation in coagulation factor V and the occurrence of coronary arterial disease (CAD). METHODS: Unrelated 86 patients with CAD and 102 healthy controls were analyzed by polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) to detect variations in the entire twenty-five exons of the factor V gene. RESULTS: Polymorphisms in exon 4 [642 G-->T (Ser156)], exon 10 [1628 G-->A (Arg485Lys)], exon 13 [4070 A-->G (His1299Arg)] and exon 16 [5380 G-->A (Val1736Met)] were documented. The study also identified a novel polymorphism in exon 2 (327 A-->G) which did not result in amino acid residue substitution. The Leiden mutation (Arg506Gln) was not detected in any of our 188 subjects. Among the 5 polymorphisms, the allele frequency of 1628 G-->A was significantly different between CAD patients and controls (0.69 vs 0.81, chi 2 = 6.908, P < 0.01). This is the first report of this finding in a Chinese population. CONCLUSION: 1628 G-->A polymorphism is associated with CAD and it may be a risk factor for CAD morbidity in the Chinese population.

Design and clinical application of cervical hollow threaded fixator.

OBJECTIVE: Cervical hollow threaded fixator (CHTF) was designed and applied to stabilize the cervical spine following anterior decompression due to cervical spondylosis or acute cervical disc herniation. METHODS: Medical use titanium alloy with no magnetism was made into a hollow threaded structure with holes on the wall of the fixator. Forty-two patients with cervical spondylosis and acute cervical disc herniation were stabilized using the CHTF technique after anterior decompression. X-ray films and CT scans were taken after the operation to observe the stability and fusion rate of the segments. RESULTS: Patients were allowed to walk one day after the operation and were followed up for 6 to 36 months. The operated segments were stable and solid fusion was observed 3-6 months after surgery. CONCLUSIONS: Immediate stability of the operated segments can be obtained using the CHTF technique. Patients were easy to manage after the operation and returned to their jobs in a short time. The CHTF technique is safe and simple to perform and eliminates the complications of an autograft. It may be an alternative to traditional cervical interbody fusion with bone graft.

[Significance of fucose expression in lung carcinoma and their brain metastases].

OBJECTIVE: To investigate the expression of fucose in brain metastases and their primary tumors. METHODS: Affinity histochemistry method was used to detect the expression of fucose in lung carcinoma and their brain metastases of 39 cases with available survival data. RESULTS: Of the 39 cases, 21 cases were positive for expression of fucose and 12 cases were negative both in the primary tumor and their metastases. 2 cases showed positive only in primary tumors and 4 cases only in metastatic tumors. The positive intensity (Dm) and percentage of the positive area (Sm) of fucose in brain metastases were stronger and higher than in their primary cancers (Dp, Sp), both were statistically significant (P = 0.0015 and P = 0.001 respectively). Multivariate analysis showed that the percentage of the positive area of the primary tumor and the positive intensity of the brain metastases was the significant predictor of survival after manifestation of the brain metastasis (Tm). CONCLUSION: Adenocarcinomas of the lung were prone to metastasize to the brain compared with other types of lung cancer. The Dm and Sm of fucose in brain metastasis was significantly stronger and higher than that of their primary tumor. The outcome of patients with brain metastasis could be predicated by Sp and Dm. Surgical resection of the primary lung cancer and their brain metastasis(es) resulted in better prognosis.

[The dynamic changes of postburn complement activation and its effects on the secretory function of macrophages in severely burned mice].

OBJECTIVE: To study the dynamic changes of postburn complement activation and its effects on the secretory function of macrophages in severely burned mice, so as to help explore the mechanism of postburn immunological disturbance. METHODS: The mice inflicted with 18% TBSA III degree scalding were employed as the model and randomly divided into normal control, decomplementizing control, scalding and decomplementizing scalding groups. The chemotactic activity of plasma, the production of nitride oxide (NO) and tumor necrosis factor alpha (TNFalpha) by peritoneal macrophages (MPhi) cultured by plasma from every groups were examined. RESULTS: The chemotactic activity of plasma complements increased quickly within 30 min. postburn, reached the summit level at 6 postburn hour (PBH) and declined since 12 PBH. Plasma from injured mice could induce peritoneal MPhis to produce more NO and TNFalpha. CONCLUSION: Complements were immediately activated postburn. The MPhis were activated by active complements. As a result, immunological active products increased significantly.

[Protective effects of brain-derived neurotrophic factor on injured retinal ganglion cells of rats].

PURPOSE: To investigate in vivo the survival of retinal ganglion cells(RGC) after optic nerve crush with intraocular injection of Brain-derived neurotrophic factor (BDNF). METHODS: Sprague-Dawly(SD) rats were divided into normal control, crush control, medium treatment and BDNF treatment group, and every group had 20 eyes. RGC of adult rats were labeled with fluorogold. Seven days later, the optic nerve was intraorbitally crushed and BDNF was injected into the vitreous. Five, 7, 14, 21 and 28 days after crush, the numbers of RGC were counted. RESULTS: The densities of RGC began to decrease 7 days after injury, the number of RGC was 70.2% and 40.5% of normal controls on days 14 and 28, respectively. In the group with BDNF injection, RGC densities decreased on days 7, but RGC densities were much higher than that of controls on 7, 14, 21, and 28 days after injury (P < 0.01). CONCLUSION: BDNF administered intraocularly at the time of crush can protect RGC from injury and cell death.
