Binding of [3H] 1,25(OH)2D3 and effects of 1,25(OH)2D3 on cell ultrastructure were evaluated in vascular smooth muscle cells (VSMC) primary cultures (aortic media). Specific reversible binding of [3H] 1,25(OH)2D3 by a 3.5 S macromolecule with DNA binding, KD 6.2 X 10(-10) M and Nmax 16 fmol/mg protein was demonstrated. Incubation of VSMC with 10(-8) M 1,25(OH)2D3, but not 25(OH)D3, in the presence of 10% FCS for up to three weeks caused rapid reversible appearance in the cytoplasm of membrane-bounded electron-dense lysosomal particles which on electronspectroscopic imaging contained Ca and Pi. VSMC are targets for vitamin D.
Eight subjects with the likely diagnosis of presenile or senile dementia of the Alzheimer type were tested on two frequently used primate learning tasks: a concurrent object discrimination task and a delayed non-match-to-sample task. In addition, various tests for cognitive, mnemonic, perceptual, and language functions were applied. The results suggest a severe decline of the Alzheimer subjects in all measures when compared with 10 control subjects matched for age, gender, and education. The two animal learning tasks revealed strong impairments, thus demonstrating a high sensitivity for the detection and assessment of human amnesic disorders. Implications of these findings for human neuropsychological, and especially comparative neuropsychological, research are discussed.
The integral dose and the dose to the organ at risk were calculated using the following angiographic techniques: 1. Intravenous DSA in the neck, 2. Intravenous DSA in the skull, 3. Intra-arterial DSA in the skull in searching for an aneurysm. This was compared with 4. 105 mm. camera technique and 5. An imaging plate technique. These methods have been compared with reference to dose and diagnostic value. Desirable developments for angiography in the future are discussed.
During the last year we diagnosed within the scope of preventive medical care of pregnant women in about the 30th pregnancy week, seven foetuses with abdominal cystic tumours by ultrasound examination. Postpartal ultrasound controls of the mature newborn confirmed the findings. After laparotomy we found ovarian pseudocysts with a volume up to 90 ccm. Histologically we could not identify ovarian tissue. In 66% of the cases we found small cysts on the contralateral ovary. Differential diagnosis and the possible complications must be considered. A laparotomy should be performed and only by high steroid serum level a conservative therapy with MPA should be considered. This is the case in premature newborn with an immature endocrine regulation mechanism.
The half-lives of 35S-labelled L-methionine and of 3H-labelled L-fucose of serum transferrin of rats were measured in pulse-chase experiments in vivo. Both L-[35S]methionine and L-[6-3H]fucose disappeared from transferrin with nearly the same half-lives of 33.8 h and 36.5 h, respectively. The data show that in this major serum glycoprotein peripheral carbohydrates and the protein moiety are degraded as a unit.

The particular uncertainties of the carcinoma of the urinary bladder results from diagnostic and therapeutic deficiencies the causes of which are to be found in the dignity and inclination to dissemination of tumours of the bladder. At present with the help of diagnostic methods which are at our disposal it is difficult to establish an exact clinical staging. Nevertheless it was tried to set certain therapeutic central points. For this purpose malignant tumours of the urinary bladder were subdivided into three groups, taking into consideration the tumour infiltration as well as the tumour grading and adequate therapeutic measures were discussed under a curative, adjuvant and palliative aim.

The authors describe the limited role of non-invasive imaging procedures in the differential diagnosis of splenic abscess versus infarction. Sonography and computed tomography often do not offer any characteristic findings in these splenic disorders. A case is reported as an example. By means of 111 In-labelled leukocyte and gallium-67-scintigraphy exclusion of acute inflammatory processes can be performed with great certainty, and unnecessary surgical interventions can thus be avoided.
On the basis of a clinical study an analysis about 86 patients with obstructing bilateral nephrolithiasis and chronic obstructive infection of the urinary tract is elaborated. In order to illustrate the change of resistance of the causative agents during a period of 10 years (1973-1982) two separated 5-year analyses are compared. It is made evident that the relations of sensitivity to seven chemotherapeutic drugs in Coli, Proteus and Pseudomonas infections have changed in favour of increasing resistances of causative agents. The results obtained are an expression of the unstableness of antibiotic therapies under palliative purpose when urological basic diseases are not removed. From the analysis the demand for changing the general problems of obstructive pyelonephritis without possible operative sanation becomes evident. early diagnosis and treatment of predisposing factors are the two keystones of our efforts, when the chronic obstructive infection of the urinary tract by resistant causative agents shall not remain a therapeutic problem in the same measure as hitherto.

A number of commercially available columns for ion-exchange and hydrophobic-interaction high-performance liquid chromatography (HPLC) have been tested, ranging from very fast columns with low capacity to preparative columns. The experiments represent a selection of column applications likely to occur in a biochemical laboratory. The use of several ion-exchange columns for separation of serum and membrane proteins has been demonstrated. Columns with a silica gel matrix and with a polymer matrix have proved to be of equal value in experiments with water-soluble proteins. In experiments with hydrophobic membrane proteins the choice of the column depends on the protein to be isolated. A combination of a cation-exchange and an anion-exchange column as a tandem as well as a mixed-bed column, were applied for the separation of serum proteins and for the isolation of a 175,000-dalton membrane glycoprotein. One problem of hydrophobic-interaction HPLC is the poor solubility of some proteins, for example serum proteins at high salt concentrations. This difficulty can be overcome by the use of columns with higher hydrophobicity which require lower initial salt concentrations. Less hydrophobic columns have been shown to separate hydrophobic membrane proteins by a combination of salt and detergent gradients.
