Screening system for the evaluation of preparations with potential cytostatic and immunotropic activity. I. Derivatives of 4,5-dihydro-2h-pyridazinone-3.

New screening system for the evaluation of preparations with potential cytostatic and immunotropic activity has been suggested. It consists in their interaction in relation to the mitogenic activity of phytohemagglutinin. A group of preparations, derivatives of 4,5-dihydro-2H-pyridazinone-3 was evaluated. It appeared that among the examined compounds there were preparations suppressing or stimulating the incorporation of 14C-thymidine to lymphocyte nuclei. Dependence between chemical structure and pharmacological activity in this group of compounds is discussed.

Bacteriophages and transplantation tolerance.

Our recent findings suggest that bacteriophages (phages) may not only eliminate bacteria, but also modulate immune functions. In this communication, we demonstrate that phages may strongly inhibit human T-cell activation and proliferation as well as activation of the nuclear transcription factor NF-kappaB in response to a viral pathogen. Phage administration in vivo can diminish cellular infiltration of allogeneic skin allografts. Thus, phage treatment should be considered in antibiotic-resistant posttransplantation infections. Furthermore, phages could find a broader application in clinical transplantation.

Laser scanning cytometry in pathology of solid tumors. A review.

OBJECTIVE: To review application of laser scanning cytometry (LSC) to analyze several different parameters of human solid tumors in relation to the cell cycle. STUDY DESIGN: Tissue sections of cytology specimens were stained for specific parameters and analyzed by LSC. RESULTS: Examples of LSC analysis of the expression of ER, Ki-67, cyclin B1, BrdUrd and ploidy are given. CONCLUSION: LSC provides rapid, high-precision measurement of the chosen parameters or constituents of each cell in a population of cells and correlates those measurements with the visual image of the corresponding cell.

Fine needle aspiration cytology of rare malignant tumors of the breast.

This report describes the fine needle aspiration (FNA) cytologic findings in 17 rare malignant breast tumors. The series consisted of invasive cribriform carcinoma, papillary carcinoma, apocrine carcinoma, carcinoma with pseudosarcomatous metaplasia, carcinosarcoma, fibrosarcoma, malignant phyllodes tumors, primary malignant lymphomas, plasmocytoma, metastatic melanoma and metastatic renal clear cell carcinoma. Besides cytomorphology, the results of immunostaining in eight cases are presented, as is a review of the literature. It is important for rare primary malignancies, as well as for metastatic tumors, to be diagnosed, or at least have the diagnosis suggested, preoperatively by FNA and immunocytochemistry, permitting better therapy planning.

Endoscopic brushing cytology of primary gastric choriocarcinoma. A case report.

A rare primary choriocarcinoma of the stomach developed in a 71-year-old woman admitted with epigastric pain and weight loss. Brushing smears revealed highly atypical malignant cells with bizarre nuclei and prominent nucleoli. Multiple histologic sections revealed pure choriocarcinoma. Human chorionic gonadotropin and keratin were demonstrated in malignant cells by immunohistochemistry.

Laminin production and fibronectin immunoreactivity in breast carcinomas.

Eighty-nine cases of primary breast carcinoma were evaluated immunohistochemically for basement membrane laminin and fibronectin (FN). 58 of the cases were also examined by transmission electron microscopy (TEM). Intraductal carcinomas were all positive for laminin and FN and by electron microscopy a continuous BM was found in all of them. Among invasive tumors, immunoreactivity for laminin was expressed in 31% of cases and FN in 66%, whereas TEM revealed BM in only 4 out of 52 (8%) studied cases. Tumors measuring 10 mm or less were more often laminin-positive and FN-negative than larger tumors. There was a tendency for lymph node negative tumors to express laminin more often, but there was no relation between lymph node status and FN. Laminin immunostaining proved to be very useful in identifying vascular invasion. The laminin and FN immunoreactivity showed no significant correlation with overall survival and disease-free survival (DFS) of the breast cancer patients. DFS for cases with laminin positive tumors was 50% and for laminin negative tumors 46.8%. For cases with tumors immunoreactive for FN, DFS was 50%, and for tumors lacking FN 43.5%.
