A comparison of the behaviour to ions of the P3 component of the pigeon cone and rat rod electroretinogram.

1. Isolated pigeon and rat retinas were incubated in various media and the effect of changing ions on the P III component of the electroretinogram was studied, with particular regard to the influence of calcium on other ions.2. In pigeon, without calcium, reduction of sodium and chloride both reduce P III. This behaviour continues in the presence of calcium but only if the retina has been treated with ouabain. Under these conditions, the effect of calcium is to prevent the appearance of inverted responses.3. Under other conditions, calcium renders the pigeon P III insensitive to change in chloride. Sensitivity is rapidly restored by the use of ouabain, 2,4-DNP, or by cooling.4. Calcium does not alter the linear relationship between sodium and response amplitude.5. In pigeon, increasing potassium increases P III amplitude, though the exact relationship is altered by calcium.6. In the rat retina treated with ouabain, increasing sodium increases P III, and increasing potassium decreases P III. This result is similar to that of Sillman, Ito & Tomita (1969b). If calcium is absent, inverted P IIIs can be obtained.7. If calcium is present in the medium, reducing sodium also reduces rat P III.8. If calcium is reduced below 10(-7)M, then reducing sodium increases P III, and large responses are obtainable from solutions which contain almost no ions. These responses vanish very rapidly if the retina is cooled or treated with ouabain.

The effect of ions on the photoresponses of pigeon cones.

1. Pigeon cone receptor potentials have been identified in a preparation of isolated, incubated retina.2. There is an approximately linear relationship between response amplitude and sodium concentration of the bathing medium.3. A decrease of chloride ions produces results equivalent to a decrease of sodium ions. The response amplitude is a power function of the product of the sodium and chloride concentrations. For most experiments, the exponent is ca. 1.4. After treatment of the retina with ouabain, responses vanish, but can be temporarily restored, in normal, or inverted polarity, by appropriate changes in sodium or chloride concentration.5. Ammonium and potassium ions can substitute quantitatively for sodium ions. Lithium ions initially carry response currents, but later block all responses. Bromide, nitrate and thiocyanate can substitute for chloride ions.6. The response wave form, and the amplitude/light intensity relationship are independent of ions.7. Models of the photoreceptor are discussed. It is concluded that the membrane potential of the cone in light is chiefly determined by the distribution of anions across the membrane, and in darkness there is an increase in outer limb cation permeability.

Improvement of serological diagnosis of human cytomegalovirus infection in renal transplant recipients by testing for specific immunoglobulin E by ELISA.

The kinetics of human cytomegalovirus (HCMV)-specific immunoglobulin E (IgE), M (IgM), A (IgA) and G (IgG) were studied in 421 sera obtained from 19 renal allograft recipients by enzyme-linked immunosorbent assay (ELISA). Cytomegalic inclusion disease (CID) occurred in 11 (57.9%) patients. HCMV infection was diagnosed in all (100%) of these patients by testing for specific IgE. In contrast, increased levels of IgM and IgA class antibody against HCMV were detected in only 45.5% and 18.2% patients suffering from primary or recurrent HCMV infection, respectively. Concerning the time interval between the onset of clinical symptoms and the first positive test, no significant differences in the kinetics of HCMV-specific immunoglobulins E, M, A and G were observed. Elevated specific IgE levels persisted for longer time intervals than the other immunoglobulin classes. As shown by the present study, specific IgE proved to be a more reliable serologic marker than IgM and IgA for the serologic detection of HCMV infection in renal allograft recipients.
