An evaluation of the 24-hour RAIU test performed with an Anger camera and a pinhole collimator.

RAIU measurements obtained with a conventional thyroid uptake probe have been compared with those obtained with an Anger camera and a pinhole collimator. Although the field distribution for the pinhole collimator deviates markedly from the IAEA recommendations, the group data from the comparative measurements in this series were identical, with a confidence level of 99%. However, in several cases the data from differing measurement regimes for individuals varied markedly.

Establishment of photoaffinity label derivatives of fluorene as probes in studies of chemical carcinogenesis in mammalian cell culture.

Several azido fluorenes, photosensitive analogs of the established carcinogen 2-acetylaminofluorene, have been synthesized to serve as probes in chemical carcinogenesis and mutagenesis studies. Unlike 2-acetylaminofluorene, these compounds do not require metabolic activation. However, alkylation to critical targets is achieved through the generation of nitrenes by photolysis at 360 nm in situ. We have found that the bifunctional azidofluorenes 2,5-diazidofluorene and 2,7-diazidofluorene were more toxic to and more transforming of the mouse embryo C3H 10T1/2 clone 8 cells when photolyzed in situ than were the monofunctional azides 2-azidofluorene and 7-bromo-2-azidofluorene. When the drugs were photolyzed in phosphate-buffered saline (pH 7.4) and then added to the cells these preirradiated derivatives were not transforming but were slightly toxic. However, there is not relationship between the number of photosensitive azido groups and the cytotoxicities of the preirradiated derivatives as measured by the plating efficiency method. The transformation experiments were performed under conditions in which photolysis was carried out for as little as 15 sec. This is significant because the major events that lead to oncogenesis apparently can occur within this narrow time channel, thereby making the drugs excellent probes in studies in chemical carcinogenesis. Although near ultraviolet light alone may transform the cells (8 to 16 min of irradiation), under the conditions studied ultraviolet light was neither toxic nor transforming.

Histophilus ovis infection in sheep in Western Victoria.

Organisms resembling Histophilus ovis were isolated from sheep in Western Victoria in association with synovitis, septicaemia and/or abscessation in 2 to 8 week old lambs. Histopathological lesions in these lambs were indicative of bacterial embolism with thrombosis. Similar organisms were also isolated from sheep in 2 flocks with a high neonatal mortality rate. These isolations were from vaginal swabs from the ewes, placentas and from one lamb which had meningitis, ependymitis, choroid plexitis, hepatic necrosis and focal myocarditis. The morphology, cultural characteristics and biochemical activity of the organisms are described and it is suggested that the organisms may be better classified in the genus Actinobacillus.

An outbreak of anal fibropapillomatosis in cows following rectal examinations.

An outbreak of fibropapillomatosis following rectal examination for pregnancy and involving the anus and to a lesser extent the rectum in a herd of beef heifers in western Victoria is described. Almost half the herd were involved at one time; most lesions regressed with the following 16 weeks. Lesions varied from 0.2 mm diameter polyps up to large multiple lesions up to 7 x 5 x 2 cm with a broad base. The presence of lesions did not affect the body condition or health of the animals.

Folates as inhibitors of glutamate dehydrogenase.

Folates and tetrahydrofolates inhibit beef liver glutamate dehydrogenase (EC 1.4.1.2). Double reciprocal plats indicate a competitive inhibition for alpha-ketoglutarate-glutamate by folic acid and methotrexate and a complex or mixed type for NAD-NADH site. Pteroic acid is not inhibitory at the concentrations studied. The addition of up to four gamma-linked glutamyl residues to folic and tetrahydrofolic acids increases the inhibition. Further chain elongation of the gamma-peptide had no effect on the inhibitory activity. The p-aminobenzoate poly-gamma-glutamates were less inhibitory than the corresponding folyl polyglutamates.

Production of frameshift mutations in Salmonella by a light sensitive azide analog of ethidium.

Frameshift mutations have been produced in specific repair-negative Salmonella tester strains by photoaffinity labeling technique using ethidium azide. Reversions requiring a +1 addition or a -2 deletion were specially sensitive. Mutagenesis was reduced by the simultaneous addition of non-mutagenic ethidium bromide, and was prevented by photolysis of the azide prior to culture addition. Identical tester strains active in DNA excision repaire were not mutagenized by the azide. These results are consistent with the interpretation that photolysis of the bound ethidium analog converts the drug from its noncovalent mode of binding (presumably intercalation) to a covalent complex with consequent production of frameshift mutations. Such photoaffinity labeling by drugs which bind to DNA not only confirms the importance of covalent drug attachment for frameshift mutagenesis, but also provides powerful techniques for studying the molecular deatils of a variety of genetic mechanisms.

Bovine nasal granuloma in Victoria -- a search for the causative allergens.

Cows with nasal granuloma showed skin sensitivity to a range of environmental allergens while clinically normal control animals showed little sensitivity. The allergens producing the greatest number of positive reactions included the pollens of dock, capeweed, clover, paspalum, sorrel, rye grass, pepper tree and wattle, the spores of penicillium, cladosporium, botrytis and rye grass rust and extracts of sheep's wool and mite. It is likely that different allergens are important in different animals and in different regions and their importance in the aetiology of nasal granuloma probably depends on their presence in the inhaled air in high concentration for long periods.

"Woolly everlasting daisy" (Helichrysum blandoskianum) toxicity in cattle and sheep.

Investigations into cattle mortalities suspected of being caused by the Woolly Everlasting Daisy (Helichrysum blandowskianum) revealed lesions of marked periacinar liver necrosis, vascular degeneration, widespread haemorrhages and oedema. Brains showed status spongiosus. These lesions were reproduced in cattle and sheep fed Helichrysum and in cattle given an intravenous injection of an extract of the plant.
