Further characterization of haptoglobin binding to streptococci of serological group A.

Certain group A streptococci with surface antigen T 4 possess surface receptors for human haptoglobin (Hp). Binding of 125I Hp 2-1 to two representative group A streptococcal cultures could be inhibited by unlabelled Hp 2-1, Hp 2-2 and Hp 1-1 but not by the alpha 1, alpha 2 or beta chains of Hp. Hp complexes formed with equine hemoglobin and asialo-Hp also reduced 125I-Hp 2-1 binding to group A streptococci. Hp binding proteins could be solubilized from streptococcal surface by hot acid treatment of the bacteria and purified by subsequent affinity chromatography on human Hp 2-1 sepharose. The isolated Hp binding proteins specifically inhibited 125I-Hp 2-1 binding to group A streptococci and retained their 125I-Hp 2-1 binding activity in a dot binding assay on nitrocellulose membranes. SDS-PAGE and protein blots of Hp binding proteins developed with 125I-labeled Hp 2-1 revealed numerous high molecular weight proteins with 125I-Hp 2-1 binding activity.

Microheterogeneity of alpha 1-acid glycoprotein in the sera of patients with cancer or inflammatory states of the ovaries.

alpha 1-Acid glycoprotein (AGP) and its concanavalin A-dependent four microheterogeneous fractions were measured in sera from women with ovarian carcinoma and those with an inflammatory ovarian disease. Total concentrations of AGP in inflammatory and cancerous sera were significantly higher than in control group, but did not differ from each other. In the fourth stage cancer the distribution of Con A-dependent fractions 1, 3 and 4 was significantly different as compared with control, second stage cancer and inflammatory groups.

Azimexone and cysteine hydrazide in the treatment of allergic encephalomyelitis.

In the course of allergic encephalomyelitis (EAE) in chickens, azimexone suppressed the production of the specific IgM immunoglobulins. Moreover, it decreased the level of haptoglobin and sialic acid but stimulated the activity of antitrypsin. It was also observed to reduce the production of the plasmatic cells in the spleen. L-cysteine hydrazide hydrochloride decreased the level of sialic acid in EAE; intensified the activity of trypsin inhibitor and exerted no effect upon the level of antimyelin antibodies.

Polyethylene glycol enzyme immunoassay for screening anti-haptoglobin monoclonal antibodies.

An enzyme immunoassay for the screening of anti-haptoglobin activities in supernatants of hybridoma cell cultures is described. The sandwich technique was greatly improved by the addition of 2% polyethylene glycol 6000 to all the reagents. It allowed a six-fold shortening in the incubation time compared with the standard method without sacrificing either sensitivity or accuracy.

Enzymes and acute phase reactants in serum of selenium treated rats bearing adenocarcinoma.

Sodium selenite was administered to the rats bearing subcutaneously transplanted adenocarcinoma. The following determinations were carried out in serum: gamma-glutamyl transferase, aminotransferases, alkaline phosphatase and its placental isoform, haptoglobin, protein- and lipid-bound sialic acid. Changes observed in catalytic activities and concentrations of the examined parameters could be ascribed rather to the presence of the tumor than to the effect of the selenium treatment.

Thymus factor X (TFX) in modulation of immune and inflammatory response in chickens immunized with myelin basic protein (MBP).

The immune response measured by the level of specific anti-myelin antibodies and inflammatory process indicator-haptoglobin, were studied in chickens immunized with myelin basic protein (MBP). The histopathological changes in lymphoid organs were also investigated. The occurrence of specific anti-myelin antibodies was shown in sera of immunized animals. High level of antibodies was found in IgG class. The concentration of anti-myelin antibodies in the remaining immunoglobulin classes showed different levels. High concentration of haptoglobin (up to 2 g/l) was regularly noted in the course of allergic encephalomyelitis. All the above parameters were also evaluated in the course of experimental therapy with thymus factor X (TFX). This preparation distinctly reduced haptoglobin level in serum and had only slight effect on humoral immunity. In the course of inflammatory process plasmatic cells stimulation in the spleen was observed microscopically: this was even enhanced by the thymus hormone. The lymphoid system did not undergo any significant changes. The Harder's gland did not seem to have any influence upon the induction or therapy of MBP-induced inflammation.

Affino-immunoelectrophoresis of haptoglobin with wheat germ agglutinin. Diagnostic significance in ovarian carcinoma.

Affino-immunoelectrophoresis with wheat germ agglutinin (WGA) was applied to the analysis of microheterogenous fractions of human haptoglobin (Hp), present in cancerous ascitic fluids and in sera from patients either with ovarian carcinoma or with inflammatory ovarian disease. Normal and inflammatory sera contained two WGA-dependent Hp fractions: strongly- and weakly-retarded while cancer sera and ascitic fluids: non-retarded and weakly-retarded fractions. The content of WGA-dependent Hp fractions following curative surgery and chemotherapy of the patients was found to resemble the pattern of the control group.

Prediction of secondary structure of human haptoglobin.

Secondary structure of alpha (light) and beta (heavy) chains of human haptoglobin was proposed. The Chou-Fasman predictive method was used in conjunction with the hydrophobic character and hydropathy scale value assigned to each amino acid residue in the primary sequence of the protein. Moreover, by means of hydrophilicity values of amino acids, the location of antigenic determinants was predicted.

Synthetic factor for evaluation of the effectiveness of the therapy in ovarian carcinoma.

The synthetic factor based on determinations of 10 parameters in sera of patients with ovarian carcinoma, was constructed. The factor was found to be useful in evaluation of the effectiveness of the cytotoxic treatment and in indication of the recurrence of the malignancy. Reduction of the number of biochemical markers to five (haptoglobin, seromucoid, lactate dehydrogenase, alkaline phosphatase, aspartate aminotransferase), according to significance of particular markers in the laboratory diagnostics of ovarian cancer did not affect the diagnostic sensitivity of the synthetic factor.

Products of trypsin digestion of haptoglobin beta (heavy) chain.

Trypsin digestion of haptoglobin beta (heavy) chain resulted in five glycopeptides. The glycopeptides were characterized by carbohydrate and sulphydryl groups content; their molecular mass was determined by sodium dodecyl sulphate-polyacrylamide gel electrophoresis in the presence or absence of 2-mercaptoethanol. None glycopeptide possessed hemoglobin-binding capacity. glycopeptide I did not form any precipitate with antihaptoglobin serum but was shown to inhibit strongly the reaction of haptoglobin or beta chain with the antiserum. Glycopeptide II showed dominant antigenic determinants in relation to native haptoglobin and to beta chain. Reaction of this glycopeptide with concanavalin A was almost twice higher than the corresponding reaction of haptoglobin. Glycopeptides IV and V were inactive in the reaction with the lectin. Glycopeptide III exhibited relatively the strongest cross-reactivity with the specific antihaptoglobin serum while its inhibitory activity in the immunoreaction was the lowest.

Behavior of haptoglobin in experimental allergic encephalomyelitis in chickens.

Concentration of haptoglobin (Hpc) in serum of chickens with inflammation and allergic encephalomyelitis (EAE) was determined. Chickens with aseptic inflammatory state revealed 2.5 times higher concentration of Hpc while in the course of EAE only small increase was observed. Administration of immunomodulator--isoprinosine, resulted in elevation of Hpc level as compared to nontreated chickens.

Structural similarities among concanavalin A, haptoglobin, and trypsin.

Comparison of the primary structure of concanavalin A with those of trypsin and beta (heavy) chain of haptoglobin revealed a significant degree of chemical similarity. The amino acid sequence of concanavalin A was found to be 53.6% related to trypsin and haptoglobin. Certain analogy in the tertiary structure was shown by the use of thionine binding to the specific active site, characteristic for serine proteases.

Haptoglobin and lipoperoxides in rabbit plasma under turpentine- and endotoxin-induced inflammation.

Inflammation was induced in rabbits by injection of turpentine or/and endotoxin. Haptoglobin (Hp) and lipoperoxide (as malondialdehyde - MDA) concentrations in rabbit plasma, were measured. In some experiments indomethacin was administered, as a specific control of anti-inflammatory action. In the course of the reagents-induced inflammation there existed positive correlation between changes in Hp and MDA levels. Indomethacin was found to eliminate this effect. Under the model system of experiments, Hp as presumed endogenous inhibitor of prostaglandin synthesis, even in concentrations in rabbits' blood 15-20 times higher than normal, did not exert any protective action against inflammation.

Immunological comparison of glycopeptides obtained from haptoglobin types 1-1, 2-1 and 2-2.

Immunochemical properties of glycopeptides III and IV obtained from haptoglobin (Hp) types 1-1, 2-1, and 2-2, by trypsin digestion were studied by double immunodiffusion test, quantitative precipitation and crossed immunoelectrophoresis with antisera directed against Hp 1-1, Hp 2-1, and Hp 2-2, respectively. Trypsin digestion resulted in an exposition in the glycopeptide IV 1-1 of the antigenic determinant characteristic of Hp 1-1, which had been hidden inside the molecule of the native Hp 1-1. On the contrary, it was not possible to find an antigenic determinant characteristic of the product of alpha 2 gene.
